Es ist seit langem bekannt, dass biochemische Reaktionen, insbesondere enzymatische Reaktionen, wenigstens teilweise über die Temperatur gesteuert werden können, da viele biologische Makromoleküle, insbesondere Enzyme, deutliche ausgeprägte Maxima ihrer Funktionsfähigkeit und Effizienz bei bestimmten Temperaturen aufweisen. Allerdings ist diese Steuerung meist auf einen relativ engen Temperaturbereich beschränkt und größere Temperaturänderungen nach oben oder unten führen in der Regel zum Abbruch der Reaktion. Hebt man diese Blockierungen oder Reduzierungsmaßnahmen auf, so reagiert das System durch, um im thermodynamischen Gleichgewicht zu enden. Diese letzte geordnete Reaktionskette wird genutzt, in dem man ein oder mehre mRNA-Typen in großer Menge zugibt, deren codierten Proteine dann in dem Zelllysat produziert werden. Auf diesem Wege wurden bereits mehrere hundert Proteintypen synthetisiert, die durchaus auch toxische Eigenschaften aufweisen können, da keine lebende Zelle zur Synthese benutzt wird. Das Prinzip ist allerdings bisher von einer Vielzahl technologischer Schwierigkeiten begleitet. Das sind einmal der schnelle Verfall der Reaktionsfreudigkeit des Zelllysats, zum anderen die mangelnde Kontrolle der Reaktionen, wenn diese einmal in Gang gekommen sind, letztendlich auch die Einmaligkeit der Nutzung. Des Weiteren bereitet die Skalierung hin zu größeren Volumina und Mengen der Produkte Schwierigkeiten. Insbesondere wäre es sehr hilfreich, wenn man die Inaktivierung/Aktivierung der Zelllysate und entsprechend komplexer Reaktionsgemische auf einfache Weise steuern könnte. Ähnlich kurzlebigen Isotopen ist die Handhabung der Reaktionsgemische nur zeitlich sehr begrenzt möglich und daher jede Art von Steuerung und Synchronisation willkommen. Dies ist erfindungsgemäß über die Beimengung oder auch komplette Nutzung von Komponentenketten oder Zelllysaten aus hochdruckadaptierten Tiefseeorganismen gelöst.In the last 10 years, the field of cell-free biosynthesis has developed rapidly. Das bedeutet, bei einem Organismus aus 6000–10000 m Tiefe, liegt der optimale Arbeitsbereich der makromolekularen Komponenten bei einem Druck von 60 bis 100 MPa (600 bis 1000 atm). Für viele dieser Reaktionskomponenten ist deren Reaktionsfähigkeit bei Normaldruck auf wenige Prozent oder darunter reduziert bzw. komplett unmöglich. Ungeachtet dessen tolerieren sie eine derartige Druckänderung über zwei Dekaden nahezu reversibel für bestimmte Zeiten (Stunden bis Tage). Somit kann ein derartiges Zelllysat, ein reaktives Vielkomponentensystem oder auch ein Zellorganellensystem über einfachen Druckwechsel zwischen z. B. 80 MPa und 0,1 MPa an- und abgeschaltet werden (
Im Fall der Nutzung eines HP-Zelllysats kann dessen Reaktionsfähigkeit neben den bereits genutzten Prinzipen nahezu vollständig über die Druckänderung gesteuert werden. Bei NP ist das System reaktionsblockiert und beginnt zu arbeiten, wenn der Druck sprungweise oder langsam erhöht wird (HP-Modus). Auf diese Weise lassen sich Zelllysate an unerwünschter oder zu früher Reaktion hindern. Insbesondere energiereiche Moleküle werden daher abgebaut oder in der Konzentration reduziert bzw. reagieren unerwünscht mit anderen Komponenten. Da man Biobatterien in einem breiten physiologischen Temperaturbereich benutzen möchte, wäre es hilfreich, bei Nichtnutzung der Energie, die Reaktionen zu stoppen. Zudem sollten die üblichen Lade- und Pflegeprozesse abgekoppelt werden können. Dies ist über die Verwendung von zyklischen HP/MP/NP-Reaktionselementen möglich. Nur wenn das System in einem dieser Modi oder einem permanenten Wechsel zwischen HP und NP oder auch MP betrieben wird, kann Energie entnommen werden. In
- a) „Laden” = HP: Nur die HP-Reaktionen laufen ab. Angeregtes A und angeregtes B werden im Kompartiment I gebildet und nach Kompartiment II bzw. III transportiert. Dort kann jedoch keine Auskopplung der Energie erfolgen. Folglich werden A* und B* in II und III angereichert, was einem Ladevorgang entspricht.
- b) „Arbeitsbetrieb = MP oder zyklischer Wechsel zwischen HP und NP: Wechselt man beständig zwischen HP und NP oder wählt man MP, einen Mitteldruck an dem alle Reaktionen mit ausreichender Effektivität ablaufen, dann liefert das System Strom, regeneriert sich dazu noch gleichzeitig. Die Regeneration und der Transport der Komponenten speist sich aus dem eingebrachten Potential der Ausgangskomponenten. Eine Zelle muss bekanntlich von Zeit zu Zeit spaltbare Grundkomponenten aufnehmen und Abbauprodukte heraus transportieren. Dies ist in analoger Weise auch hier der Fall und durch die senkrechten breiten schraffierten Pfeile veranschaulicht. In diesem Modus laufen alle Prozesse ab, die Dauer der Energiebereitstellung ist optimal.
- c) „Entladen” = NP: Betreibt man das System im NP-Modus, kann einmal der in den Kompartimenten II und III angereicherte Energieanteil mit maximaler Effizienz ausgekoppelt und an die Elektroden gebracht werden. Die Recktanten A und B werden auch zurück in das Kompartiment I transportiert. Alle anderen Vorgänge stehen jedoch. Dies entspricht einem einmaligen Entladen, allerdings mit besonders hoher Lastfähigkeit. Da die HP-Reaktanten A und B bei NP entladen werden, ist diese Reaktion bei NP nicht optimal. Da ggf. im System Reaktionswärme frei wird, ist dies für das Entladen einer Batterie mit hoher Kapazität sogar günstig (innere Kurzschlussfestigkeit).
