Description
Die vorliegende Erfindung betrifft eine Elektrospray-Ionisations-/Atmosphärendruck-Chemische-Ionisations-Ionenquelle (”ESI/APCI”).The The present invention relates to an electrospray ionization / atmospheric pressure chemical ionization ion source ("ESI / APCI").
Die geladene Spezies, die am häufigsten im positiven Ionenmodus gebildet wird, ist das Addukt zwischen dem Analytmolekül und positiven Wasserstoffionen (H+).Chemical ionization involves the transfer of charged species or species of reagent ions to analyte molecules for production of analyte ions, which can then be mass analyzed. The charged species, most commonly formed in positive ion mode, is the adduct between the analyte molecule and positive hydrogen ions (H + ).
The under atmospheric pressure performed chemical Ionization is called chemical ionization under atmospheric pressure ("APCI"). An analyte-containing sample is typically applied to a Ion source for implementation a chemical ionization under atmospheric pressure as a solution. The solution containing the analyte becomes then in a heated Sprayed pipe, by which a nebulizing or mist-forming gas also is directed.
The mist-forming gas causes the sprayed solution to be atomized into fine droplets is, which then impinge on the inner wall of the heated tube and be converted into the gas phase. With the transformation of the solution into the gas phase, the analyte molecules are desolvated. Hot gas with solvent in the mobile phase, microdroplets and desolvated analyte molecules, then enters or exits the heated tube and expands or expand in the direction of a corona needle. The analyte molecules become then ionized by chemical ionization with reagent ions which by a corona discharge in the presence of a reagent gas getting produced. In particular, analyte molecules are permeated Ion-molecule reactions ionized between reagent ions and analyte molecules in the gas phase.
Lediglich ein relativ kleiner Anteil der unter Atmosphärendruck gebildeten Analytionen wird tatsächlich durch eine kleine Öffnung in das Vakuumsystem des Massenspektrometers für eine nachfolgende Massenanalyse gezogen.at this conventional Arrangement pass analytes, which are in the form of neutral gaseous molecules, ions or charged microdroplets from the heated Pipe exit, immediately the corona needle, before entering the vacuum section a mass spectrometer over an ion sampling port enter. Only a relatively small proportion of the under atmospheric pressure actually formed analyte ions through a small opening into the vacuum system of the mass spectrometer for a subsequent mass analysis drawn.
Analytionen werden daher in der Region der Coronaentladung generiert, da dies auch der Ort ist, an dem die Reagenzionen gebildet werden.reagent ions, transfer the species charged to the formation of analyte ions to the analyte molecules, are produced as a result of corona discharge in the solvent vapor. The corona discharge is by application or application of a high voltage (eg 5 kV) on the tip of a sharp or pointed corona needle or a Corona pin generated. Analyte molecules are then processed by ion-molecule reactions with reagent ions in the gas phase in the region between the corona point and the ion sampling ionized. Analyte ions therefore become in the region of the corona discharge This is also the place where the reagent ions are formed become.
Of the large part of the gas exits the ion source via an outlet port, while a small proportion of the gas and analyte ions through the Ionenabtastöffnung in the vacuum system of the mass spectrometer for a subsequent mass analysis is or will be drawn.
In contrast, highly polar or ionic analytes typically a decrease in ion signal intensity when the Tension or the current to which the corona needle is applied elevated becomes. To guarantee a sufficiently high ion signal intensity for highly polar or ionic Analytes are conventionally these analytes using a other ion source than an ion source for performing a produced chemical ionization under atmospheric pressure, such as. an electrospray ionization ion source ("IT I").
It is believed that ionization sources for performing atmospheric chemical ionization exude highly polar or ionic analytes from the outlet of the heated tube in the form of ions or charged microdroplets before the analytes have had an opportunity to interact with the reagent ions. Since the corona needle is maintained at a relatively high positive potential (for positive ion analysis), an electric field is generated in the region of the corona needle. The electric field generated by the corona needle tends to retard and disperse and disperse the already positively charged analyte ions or microdroplets exiting the heated tube, causing the analyte ions or charged analyte microdroplets in the region the Ionenabtastöffnung be defocused. Accordingly, as the voltage or current applied to the corona needle is further increased, the positive analyte ions or microdroplets are simply retarded and dispersed to an even greater extent, thereby reducing even fewer analyte ions into the main body of the ion sampling port masses spectrometers for subsequent mass analysis and detection. Accordingly, the ion signal intensity for highly polar or ionic analytes is significantly reduced with an increase in the corona current.
It follows that the ion signal intensity for highly polar or ionic Analyte is optimized or will, if a relatively small current or a relatively small voltage is applied to the corona needle. In contrast, the ion signal intensity becomes low to moderately polar Analyte optimized when a relatively high current or a relative high tension is applied to the corona needle. This is in it justified that if a higher current or a higher one Tension is applied to the corona needle, a higher number is generated on reagent ions in the region of the corona needle. The increased Number of reagent ions interacts with the analyte molecules and generates a higher number on analyte ions. Because low to moderately polar analytes in general do not charge before they exit the heated tube and the Approach Coronanadel, become the low to moderately polar analyte molecules through the electric field generated by the corona needle is not retarded or dispersed. Thus, with an increase of the current or the tension applied to the corona needle, a higher number generated on analyte ions (due to the increased number of reagent ions generated), and these analyte ions pass through the ion sampling port for a subsequent one Mass analysis, whereby a larger or higher Ion signal intensity is detected or detected.
It will be understood, therefore, to analyze samples containing a Mixture of low to moderate polar analytes as well highly polar or ionic analytes using a usual Ion source for one chemical. Ionization under atmospheric pressure, it is necessary to perform a plurality of consecutive experimental runs, in which have different voltages or currents on the corona needle Be applied (for example, is a relatively lower Coronastrom set in a first experimental run, so that the ionization for highly polar analyte is optimized, and a relatively high corona current is set or set in a second experimental run, so that the ionization for low to moderate polar analytes is optimized). The implementation of a variety of experimental runs using different Voltages or currents on the corona needle gives a variety of records, which together, a relatively high ion signal intensity for each analyte in the sample deploy this independently from the polarities or the ionic nature of the analytes in the sample. The need the repetition of the data acquisition process under application however, increases from different voltages or currents on the corona needle both sample analysis time and sample usage volume. This can be a special problem, especially if only small amounts of a sample are available for analysis, and even if the sample given to the ion source will be in a short period of time changed dynamically, eg. in chromatography applications.

As will be understood by those skilled in the art, in a MRM experiment, a first mass filter (eg, a quadrupole rod set mass filter) is adjusted to pass or transmit parent ion (s) having a certain (specific) mass-to-charge ratio. The selected parent ions with a specific Mas The se charge ratio is then introduced into a collision or fragmentation cell in which the parent ions are fragmented into daughter or fragment ions. A second mass filter (eg, a quadrupole rod set mass filter) located downstream of the collision or fragmentation cell is then set up to transmit the daughter or fragment ions having a certain (specific) mass-to-charge ratio.
This and the MRM experiment described below, were verapamil parent ions with a mass to charge ratio of 455.1 transmitted through the first mass filter and in a collision or fragmentation cell fragmented. Corticosterone parent ions with a mass-to-charge ratio of 347.1 were transmitted through the first mass filter and fragmented in the collision or fragmentation cell. characteristic Daughter or fragment ions having a mass-to-charge ratio of 329.1 were set up to be transmitted through the second mass filter to become. Hydroxyprogesterone parent ions with a mass-to-charge ratio of 331.1 were transmitted through the first mass filter and fragmented in the collision or fragmentation cell. Characteristic daughter or fragment ions having a mass-to-charge ratio of 109.1 were established or arranged to be transmitted through the second mass filter to become. Finally were reserpine-starting ions with a mass-to-charge ratio of 609.1 transmitted through the first mass filter and in the collision or fragmentation cell fragmented. One first experimental run or a first acquisition was over a Period of 20 minutes (including column equilibrium) carried out, wherein while this time the four analytes within a time of seven minutes eluted and wherein a current of 0.2 uA applied to the corona needle has been. A second experimental run or a second acquisition was then over a another period of 20 minutes (including column equilibrium) carried out, wherein again the four analytes within a time of seven minutes eluted, but with a current of 5 μA was applied to the corona needle. The analytes in the order of their elution were verapamil, Corticosterone, followed by hydroxyprogesterone and finally reserpine. Verapamil and Reserpine are highly polar analytes / molecules, whereas corticosterone and hydroxyprogesterone are moderate polar analytes / molecules.

Ferner erhöht die Wiederholung des experimentellen Laufs unter Aufbringung verschiedener Ströme auf die Coronanadel das Probenverbrauchsvolumen.at this conventional Technique using two separate experimental runs which different streams applied to the corona needle is different for each of the two Types (i.e., polarities) of analytes in a sample during one or the other experimental run a sufficiently high Ion signal intensity achievable. However, because the analysis is effectively repeated while different streams Applied to the corona needle is the one for analysis needed a sample Time using a conventional Technology relatively long. For example. can the total analysis time for each Chromatogram for 20 minutes, including the column equilibrium. Further increased the repetition of the experimental run with application of various Currents the corona needle the sample usage volume.

The for reserpine shows ion signal intensity obtained using the preferred ion source, that the adverse effects associated with conventional APCI ion sources in the Ionization of highly polar or ionic analytes were observed, caused by the electric field generated by the corona needle can be substantially avoided if an ion source according to the preferred embodiment of the present invention is used.
