Description
The present invention relates to a method for separating mixtures of substances and to an apparatus for carrying out this method.
For the separation of mixtures of substances, in particular of complex mixtures of organic compounds, the high pressure liquid chromatography (HPLC) has been proven for years. It is advantageous that the separation of mixtures of substances with high throughput volumes can be carried out by the use of classical HPLC methods.
A disadvantage, however, is that in all hitherto known HPLC techniques, the separation is completed only when the last substance of interest, the least polar in the case of reverse phase chromatography, has left the separator. In these HPLC techniques, one can speak of a time fractionation (time fractionation) of the given via the separation device (in the case of HPLC, an HPLC column) mixture. These mixtures are characterized by different polarity.
This results in that the separation of a substance mixture take a long time and thus the residence time of the substances on the column can be large. For sensitive substances just a time-extended contact with the column material is not desirable. In addition, for the purification of substances towards the analysis purity often multiple disconnections are necessary, which in turn take time fractionation in the time fractionation and damage sensitive substances.
Furthermore, in the prior art, a chromatographic method is known in which so-called "column tubes" are used to separate substance mixtures. The principle of this method is that the chromatography process is stopped after the separation of the substance mixture into its components and there is no elution of the substances. The moment the separation on the column material has taken place, the components are localized on the chromatographic column and are isolated by extraction from the column material. This chromatography method may be referred to as "fractionation by site". It is advantageous in this approach that the components of the substance mixture are isolated immediately after their separation in the chromatography column and the residence time of the substances on the chromatography material is therefore much lower compared to time fractionation methods.
A disadvantage of this known in the prior art method that the isolation of the substance components is very uncomfortable. Thus, after completing the separation, the column tube must be cut manually with extreme caution to prevent loss of substance and remixing of the substances fixed on the column material. Subsequently, the substances are isolated from the column material by methods known in the art.
Since the column tubes used must be cut after the separation, the materials used have only a limited stability. A transfer of this method to Hochdruckchromatographieverfahren is therefore not possible. Furthermore, due to the fact that when using these column tubes also necessarily manual operations such as the cutting of the hoses must be performed, an automated performance of separations is not possible. Associated with this, carrying out this large throughput process would be extremely time consuming and labor intensive.
A use of this chromatography method has not yet succeeded in separations with large throughputs, in combination with HPLC methods and in automated processes.
The automatable implementation of the method of local fractionation in the context of Hochdruckflüssigchromatographieverfahren requires a completely new design of the chromatography columns. It is imperative that the chromatographic columns consist of individual segments, which can also be moved in an automated process, separated from each other and reconnected.
As a preferred use of these segmented columns, use in methods utilizing carrier-bound reagents such as proteins and enzymes is disclosed. According to the invention, a plurality of chemical separation and reaction processes are to be made possible within a combination column. The disclosed combination column is composed of cylindrical column segments, which carry thread at both ends, as well as also on both sides threaded bearing separators, which are provided with frits. The connection of the column segments is then via this thread through which the items are firmly bolted together. An easily detachable, even in an automated process, connection of the column segments, as required by automated local fractionation, is not disclosed. The separation of the column fragments from each other must be done manually here.
Through the use of the sieve-equipped separators on both sides of a column segment, the combination columns can be used as sieve cages.
A use for the location fractionation is not disclosed and this would mean no improvement over the column tubes described above due to the tight screwing of the column segments on the separators in their handling.
Moreover, the disclosed combination columns are preferably made of plastic and their use in high pressure liquid chromatography methods is not disclosed.

Furthermore, it is preferred that an industrial robot is integrated in the device. Surprisingly, it has been found that it is possible to combine the advantages of HPLC methods with the advantages of the method of local fractionation.
The object is further achieved by a method for the separation of mixtures, wherein the separation is carried out as a local fractionation by means of HPLC technology.

It is also advantageous to carry out an assessment of the substance mixture before the fractional fractionation in order to obtain information about the polarity of the substances and thus the probable local fixation during the local fractionation.
According to the invention, the evaluation is carried out by means of analytical HPLC.
Furthermore, it is preferable to use a detection method in order to obtain information about the spatial separation of the substance mixture in the separation column. It is preferred that one uses for detection of a standard in HPLC detector.
It is also preferred that the detection is carried out after a successful separation of the substance mixture and before repeating the separation on a further separation column.
Particular preference is given to a method in which one or more method steps are carried out automatically. It is preferred that one carries out the automation by means of an industrial robot. Furthermore, it is preferred that the method is computer-aided.
By means of the method according to the invention and the device according to the invention, it is possible to separate large throughput quantities, even of sensitive substance mixtures, without requiring elution of the substances at each separation step. It is even possible to carry out multidimensional local fractionations, that is to say successive separations, which take much less time compared to known HPLC methods and are considerably gentler. Moreover, an automated implementation of the method according to the invention by means of the device according to the invention, even using commercial industrial robots, possible.
Verfahrenmethod
In the method according to the invention, in contrast to the methods of time fractionation, the fractional fractionation is already completed shortly before the first substance of interest leaves the separation device and the last substance of interest has reached the separation device. This absolutely necessary separation of the substance mixture is fulfilled in the process according to the invention by the choice of the material in the separation device, the choice of eluent and its composition, the determination of the time until completion of the fractionation of the site and the choice of gradients or isocrates. It is advantageous here that all known in the prior art column material, normal phase material as well as reverse phase material can be used. The selection of the eluent or the solvent mixture is subject in the process according to the invention no special requirements and depends only on the individual properties of the substance mixture. As a result, the process is not subject to any particular restrictions and can be used almost indefinitely.
After completion of the fractionation, the individual substances of the mixture are according to their polarity on demarcated Areas (locations) of the separator. The fractionation has led to a distribution of the substances of the mixture to different locations in the separating material used along the spatial axis in the direction of the exit from the separation device (separation column). This first separation of the substance mixture is referred to as separation in the 1st dimension in the context of the present invention. In the first step of the method, therefore, a separation takes place along the direction of flow and the substances are spatially fixed in the separation column without having left the separation material. The method according to the invention can then be continued by a further separation of the spatially fixed substances, or the fixed substances can be separated from the separation column after separation in the 1st dimension and isolated from the separating material. For isolation from the column material, various methods known in the art can be used. For example, the substances can be separated off from the column material by standard extraction methods, they can be flushed out of the material in fractions or isolated into fractions by conversion to customary chromatography columns.
If a further local fractionation of the substances fixed locally on the segmented separation column is to be carried out according to the method according to the invention, the substances fixed on the phase in the first dimension are selectively transferred from the mobile phase by the respective segment to downstream separation devices. The substance fractions are then subjected to a second local fractionation, which is referred to as spatial fractionation of the second dimension. Analogously to the location fractionation of the 1st dimension, components of the transferred substance fraction are distributed on the second separation device according to their polarity and after separation have been spatially fixed on the second separation device. Also in the 2nd dimension, the separation is completed before the first fraction of the mixture leaves the separation column.
Analogously to the above-described procedure of separation of the 1st dimension, the separated substances can now be isolated or the process steps described above can be repeated until the substances are present in the desired purity. The location fractionation can be repeated as often as desired, up to the nth dimension. After completion of the separation, the substances are isolated as described above.
The site fractionation according to the invention can be carried out using high pressure liquid chromatography. In addition, an automated implementation of the method according to the invention, with high throughputs is possible. Here, the steps of the substance task, the separation, the transfer of the locally fixed substances in another separation device for local fractionation in the next dimension, the detection and the purging of the substances can be automated. In addition, the method can also be carried out computer and / or robot-assisted.
To carry out the method according to the invention, it is necessary to use a device which, in particular in the case of automated process control, differs in essential features from chromatography devices known in the prior art.
Thus, for example, the automated transfer of the fixed-location substances into the next dimension requires a special embodiment of the chromatography columns in order to be able to dispense with manual isolation of the substance and introduction onto the separation columns of the next dimension.
In order to carry out the method according to the invention with high throughput and preferably automated, the device according to the invention can be used, which will be described in detail below.
A component of the device according to the invention is a combination of separable segment columns, which replace the chromatography columns known in the prior art. These chromatographic columns according to the invention consist of column segments which are coupled together. Preferably, the column segments are combined such that a chromatographic column is used which is similar in length to a conventional long HPLC column. By constructing coupled column segments, the chromatographic column filled with separation material can be separated into the segments after a separation operation and these segments containing substance-loaded separation material can then be used to selectively transfer the substance to the next dimension of the fraction or flush the substance be used.
In order to enable a simple coupling and again separation of the column segments, they are movably connected. Special requirements for the coupling of the column segments are made in particular when the method according to the invention is carried out automatically. On the one hand, the coupling of the column segments must be easily solvable, but on the other hand, it must be done so that the tightness meets the requirements of high pressure liquid chromatography. For this reason come in the Device according to the invention novel HPLC-segment columns and coupling techniques are used.

As a further embodiment, an extract for distribution may be analyzed for polarity and distribution of its ingredients by analytical grade HPLC analysis. The information on which segments of the 1st dimension substances are located and whether individual segments are not occupied. This information is available for deciding whether all or only part of the segments from the 1st dimension to the 2nd dimension must be redistributed. For the distribution of these parts extracts from the 1st dimension, an assessment of the starting extract can no longer be used safely. Here, an online detection must be carried out, which allows an assessment of the occupancy of the segments of the second dimension with substances. This makes it possible to identify any occurring, not occupied with substances segments and from fractionation into fractionation vessels.

Automatisierung durch den Einssatz kommerzieller IndustrieroboterAutomation through the use of commercial industrial robots
In a preferred embodiment of the device according to the invention, an industrial robot is used for automation. If a commercially available industrial robot is used instead of linear technology or portal technology, the structure of the automated device according to the invention for multi-dimensional spatial fractionation can be simplified and the degree of automation can be increased.

Die Datenverarbeitung besteht folglich aus drei Komponenten:As a further preferred embodiment, the apparatus of the present invention is combined with an on-line data processing system to automate fractionation and control of elution in a fraction collector. On-line detection in distribution with on-line data processing ensures effective fractionation of ingredients of up to 20 segments of a substance mixture. During the distribution in the second dimension, software-controlled peak detection is used to obtain data that is automatically used to fractionate the individual segments from the second dimension. This data is combined with the information obtained from profiling for the distribution in the 1st dimension, which can be entered into a control software. Thus, segments occupied with substances are distinguished from empty ones. As a result, only those segments are fractionated in the fraction collector, which can expect substances. All other segments can be rinsed off immediately and conditioned again. The data processing therefore consists of three components:
Enter the conditions for distribution in the 1st dimension with the data obtained from the profiling.
Online recording of the distribution in the 2nd dimension by a bypass between task segment and target segments with automatic peak detection by means of chromatography software.
-
Automatic evaluation of the data obtained in
1 and 2 for the targeted elution of exclusively loaded segments.points
