R. Liss Inc., New York, 1987, S. 257–288) erhalten; die Methode wurde leicht modifiziert (S. Perovic, C. Schleger, G. Pergande, S. Iskric, H. Ushijima, P. Rytik, W. E. G. Müller, Eur. J. Pharmacol. (Mol. Pharmacol. Sec.) 1994, 288, 27–33). Zusammenfassend wurden die Neuronen aus den Cerebral-Hemisphären isoliert und diese in "Hank's balanced salt solution" ohne Ca2+ und Mg2+ (HBSS) überführt. Die Zellen wurden in HBSS mit Hilfe von 0,025% (w/v) Trypsin (10 Min.; 37°C) dissoziiert. Die proteolytische Reaktion wurde mit 10% (v/v) fötalem Kälberserum (FKS) beendet. Die Einzelzell-Suspension wurde zentrifugiert und das resultierende Pellet mit DMEM/HG/10% FKS-Medium aufgenommen. Die Zellen wurden in eine mit Poly-L-Lysin (5 μg/ml, 300 μl/cm2) beschichtete Kammer mit einer Zelldichte von 2,0 × 105 Zellen/cm2 ausplattiert. Das DMEM/HG/10% FKS-Medium wurde zwei Tage nach der Isolierung entfernt und durch DMEM/HG/serumfreies Medium ersetzt. Mit einem "inverted-stage"-Mikroskop Olympus IX70 mit apochromatisch reflektierendem Licht und dem Fluoreszenzobjektiv UApo40X/340 wurden die Fluoreszenzmessungen durchgeführt. Die Zellen wurden alternierend mit Licht der Wellenlängen 340 und 380 nm mit Hilfe eines computergesteuerten Schmalband-Interferenzfilters vor einer 100-W-Xenon-Lampe beleuchtet. Zusätzlich wurde ein 0,25-ND-Filter für 380 nm benutzt. Die Fluoreszenzemissionen bei 510 nm wurden mit einer CCD-Kamera (Modell C2400-87; Hamamatsu, Herrsching, Deutschland) aufgezeichnet. Die Bilder wurden computergestützt, mit dem Imaging-System Argus 50, Hamamatsu, digitalisiert als 256 × 256 pixels mit 8-bit-Arrays.
