Abstract
Description
In areas of biology, physics or medicine there is often the task of suspensions, one or the other contain several types of suspended particles, to be classified precisely. Adequate Classification is achieved by specifying the number of particle types and the number of particles in the particle type achieved. Particles of this type are used by a magnetizable particle is functionalized with end groups in such a way that molecules of a different type are targeted at the add functionalized particles. The count of the target molecules is thus reduced to the count of magnetic or magnetizable particles functionalized with end groups.
Es besteht also neben der Identifizierung verschiedener Partikelarten die Auszählung der identifizierten Partikelarten als Aufgabe. Die Erfassung der Konzentration ist ein erster Schritt zum Erlangen einer die Suspension bestimmenden Größe, die Ermittlung der absoluten Zahl der suspendierten Partikel, ggf. deren Verteilung, ist eine wesentlich genauere und damit auch informationsreichere Aussage. In addition to the identification of different types of particles, there is a counting of the identified ones Particle types as a task. The acquisition of concentration is a first step in obtaining a die Suspension determining size, the determination of the absolute number of suspended particles, if necessary their distribution is a much more accurate and therefore more information-rich statement.
Die quantitative Analyse der Suspension läßt sich durch mehrere Verfahren realisieren:The quantitative analysis of the suspension can be carried out by several methods:
Lichtstreuung erlaubt die Bestimmung der Konzentration innnerhalb eines Streuvolumens, dabei fällt als Resultat gleichzeitig eine Information über die Gestalt und die Geschwindigkeitsverteilung der Partikel an. Nachteilig ist ein vergleichsweise komplexer Aufbau und eine Reihe an Voraussetzungen, die für eine korrekte Analyse getroffen werden müssen.Scattering of light allows the concentration within a scattering volume to be determined, thereby falling as The result also provides information about the shape and the speed distribution of the particles. The disadvantage is a comparatively complex structure and a number of requirements for a correct analysis must be made.
Durch Leitfähigkeitsmessungen erhält man Aufschluß auf Beweglichkeit und Konzentration der Partikel. Die Methode ist vergleichsweise einfach, erfordert jedoch spezielle Meßzellen mit direkten Elektroden, so daß ein geschlossenes System in jedem Fall an einer Stelle für die Messung geöffnet werden muß.Conductivity measurements provide information about the mobility and concentration of the particles. The method is comparatively simple, but requires special measuring cells with direct electrodes that a closed system must always be opened at one point for the measurement.
Methods that involve any sensor work very precisely and also in a particle-specific manner molecular end groups is functionalized and a reaction center for a specific particle type represents. Reaction with the (target) particle type usually changes the sensor signal very strongly and very low concentrations can be detected. This method is disadvantageous is the sensor, which is usually unusable after the measurement, since the rection centers by the Target molecule are occupied.
Eine sehr elegante Möglichkeit stellen Suszeptibilitätsmessungen dar, bei denen entweder die elektrische Suszeptibilität (Polarisierbarkeit) oder die magnetische Suszeptibilität ermittelt wird. Dies geschieht durch Anlegen eines externen Feldes (elektrisches oder magnetisches) und Messen der Reaktion der Suspension.Susceptibility measurements, in which either the electrical Susceptibility (polarizability) or the magnetic susceptibility is determined. This happens through Apply an external field (electrical or magnetic) and measure the reaction of the Suspension.
Die vorliegende Erfindung beschreibt ein Verfahren zur quantitativen Erfassung magnetischer Partikel. Eine geeignete Vorrichtung findet sich beispielsweise in WO 00/49407 beschrieben. Hierbei wird, wie in Fig. 2 gezeigt, ein von einer Suspension durchflossenes Rohr in das Feld einer Spule gebracht. The present invention describes a method for the quantitative detection of magnetic particles. A suitable device is described, for example, in WO 00/49407. Here, as shown in FIG. 2, a tube through which a suspension flows is brought into the field of a coil.
Falls die Suspension magnetisierbar ist im Sinne von Paramagnetismus oder Ferromagnetismus, so ändert sich dadurch die Induktivität der gesamten Spulenanordnung. If the suspension is magnetizable in the sense of paramagnetism or ferromagnetism, so this changes the inductance of the entire coil arrangement.
Zeigt die Suspension ein wie in Fig. 1 beschriebenes Verhalten, so läßt sich die Suspension durch geeignete Messung identifizieren. Die Magnetisierungskurve in Fig. 1 ist durch einen linearen Bereich bei kleinen Feldstärken gekennzeichnet, der bei größeren Feldstärken in eine Sättigungsmagnetisierung übergeht. If the suspension shows a behavior as described in FIG. 1, the suspension can be identified by suitable measurement. The magnetization curve in FIG. 1 is characterized by a linear range at small field strengths, which changes to a saturation magnetization at higher field strengths.
The method proposed in this invention takes advantage of the non-linearity of the magnetization curve for the detection of magnetizable (para- or ferromagnetic) particles in a suspension. In FIG. 4 and FIG. 5, the method is demonstrated by measurement of the magnetic particles with two different magnetic field strengths. In FIG. 4, the response of a suspension is illustrated in a sinusoidally modulated magnetic field of small amplitude. The amplitude of the modulated magnetic field is chosen so small that the linear region marked with in FIG. 1 is not left. The magnetization as a function of time in FIG. 4 is thus sinusoidal and the associated spectrum shows only one line at the frequency of the sinusoidal excitation.
If the amplitude of the sinusoidal excitation is increased in the further process by a magnetic field such that the non-linear part of the magnetization curve in FIG. 1 is reached, this means that the magnetizable particles achieve a saturation magnetization. Plotted over time, the curve shown in FIG. 5 (right) is obtained, which almost corresponds to a rectangular modulation because saturation has been reached. In addition to the line of the fundamental frequency, a line of higher frequency also appears in the spectrum of the magnetization signal. Lines of an even higher frequency naturally also appear according to the non-linearity of the curve in FIG. 1, but with a lower amplitude. The occurrence of the signal is used to detect a (magnetic) particle type. In addition to other methods, this has the advantage that the signal only differs from zero as soon as the substance to be examined is located in the probe head in the area of the coil.
The fact that the shifting of the Frequency of magnetization due to the change in inductance as the particles flow through is analyzed, but the occurrence of a spectral component (with symmetric Magnetization characteristic of at least third order), the amplitude of which is directly proportional to the Amount of particles detected. This improves the signal / background ratio extraordinary.
Eine weitere Ausprägung des Verfahrens besteht in der Möglichkeit, auch die höheren Frequenzen neben auszuwerten. Aus dem Verhältnis der Amplituden dieser Linien und einer Eichmessung der Magnetisierungskurven der jeweils reinen Substanzen läßt sich eine Mischung und deren Mischungsverhältnis an magnetischen Partikeln in einer Suspension nachweisen. A further form of the method is the possibility of also the higher frequencies next to evaluate. From the ratio of the amplitudes of these lines and a calibration measurement of the Magnetization curves of the respective pure substances can be a mixture and their Detect the mixing ratio of magnetic particles in a suspension.
By functionalizing magnetic or magnetizable particles with respect to a specific one another molecule / protein, the magnetic or magnetizable particle becomes specific Reaction center ("point of attraction") for the aforementioned molecule / protein. By suitable Fractionation of the suspension complexes are filtered out with the magnetic particles consist of one or more target molecules, depending on the functionalization. With help a device which magnetizes the complexes in a time-varying manner and a method which the Analyzing time-varying magnetization, it becomes possible to determine the amount of magnetizable particles to detect and thus, with knowledge of the functionalization of the particle, also the amount of To determine the target molecule. Target molecules can e.g. B. blood cells, bacteria, dyes, messenger substances, Viruses etc.
Als Vorteil des Verfahrens ist die Kombination mit bereits bekannten Vorrichtungen zur Magnetisierung und die daraus resultierende kostengünstige Anordnung eines Testsystems zu sehen. An advantage of the method is the combination with known magnetization devices and to see the resulting inexpensive arrangement of a test system.
Weiter ist von großem Vorteil die durch die Ausnutzung der Nichtlinearität der Magnetisierungskennlinie erzielbare hohe Auflösung bei der Ermittlung der Anzahl der detektierten magnetisierbaren Partikel zu werten.Another advantage is that by utilizing the non-linearity of the magnetization characteristic achievable high resolution when determining the number of detected magnetizable particles values.
