Unter Angiogenese versteht man die Ausbildung neuer Blutgefäße durch das Aussprossen von Kapillaren aus einem bereits bestehenden Gefäßsystem (Hertig, 1935). Die im Rahmen der Angiogenese stattfindende Neovaskularisierung erfolgt in mehreren Teilschrit ten (Folkman und Shing, 1992). Dabei wird zunächst die die Blutgefäße umgebende Basalmembran mit Hilfe verschiedener proteolytischer Enzyme, zum Beispiel Kollagenase, abgebaut und zusätzlich wird die ext razelluläre Matrix im perivaskulären Raum fragmen tiert. Anschließend erfolgt die Differenzierung der Endothelzelle, wobei sich die Zellmorphologie än dert und damit verbunden Pseudopodien ausgebildet werden. Die Sequenzierung erfolgte mit Hilfe des Dye Terminator Cycle Sequencing Kits und des ABI PRISM 377A DNA-Sequenators. Die dabei erhaltenen vollständigen und partiellen Sequenzen von ARNA I(s), ARNA I(as) und Pseudo-ARNA I sind in SEQ ID Nr. 1 bis SEQ ID Nr. 5 und 8 bis 15 gezeigt. In Fig. 3 wird in schematischer Weise die Pseudo-ARNA I-Sequenz gezeigt. Der obere Bereich stellt die 5'- 3' Orientierung eines Sinntranskripts von der Pseu do-ARNA I in prä-mRNA Form dar, also das potentiel le L27a Exon I und das potentielle Intron umfassend die ARNA I-Nucleotide Nr. 64-205 und 1-63, beide in Antisinn-Orientierung, L27a Exon II und Poly A Schwanz (in 5' zu 3' Richtung). Dabei stellen die Nucleotide 50-63 der ARNA I(as) einen gemeinsamen Sequenzabschnitt zwischen ARNA I(as) und dem Be reich des Pseudo-ARNA I Transkripts dar, der homo log zum 5'-Ende des Exon II von L27a ist. Die 3' Spleißstelle der prä-mRNA von L27a beziehungsweise möglicherweise auch der Pseudo-ARNA I(s) Sequenz liegt im mit dem Doppelpfeil gekennzeichneten Be reich. Im darunter gelegenen Bereich wird in 3'-5'- Orientierung (Antisinn-Transkript) dargestellt, wie das entsprechende Antisinn-Transkript der Pseudo- ARNA I-Sequenz aufgebaut ist, nämlich (in 3' zu 5' Richtung): L27a Exon I in Antisinn, Intron aus ARNA I Nt 142-1 und ARNA I Nt 205-143 beide in Sinn- Orientierung, L27a Exon I (Antisinn). Claims (42)
- a) einem Nucleinsäuremolekül, das erhältlich ist mittels reverser Transkription von Plazenta- Gesamt-RNA mit den ARNA I-spezifischen Primern mit den in SEQ ID Nr. 6 oder/und 7 dargestellten Nucleotidsequenzen, und einem Fragment davon;
- b) einem Nucleinsäuremolekül, das mindestens eine der in SEQ ID Nr. 1 bis 5 oder 8 bis 15 darge stellte Nucleotidsequenz umfasst;
- c) einem Nucleinsäuremolekül, das erhältlich ist durch Substitution, Addition, Inversion und/oder Deletion einer oder mehrerer Basen eines Nuc leinsäuremoleküls nach a) bis b);
- d) einem Nucleinsäuremolekül, das mit einem Nuc leinsäuremolekül nach a) bis c) und einem Frag ment davon hybridisiert, und
- e) einem Nucleinsäuremolekül, das zu einem Nuclein säuremolekül nach a) bis b) komplementär ist, und einem Fragment davon.
- a) a nucleic acid molecule which is obtainable by reverse transcription of total placental RNA with the ARNA I-specific primers with the nucleotide sequences shown in SEQ ID No. 6 or / and 7, and a fragment thereof;
- b) a nucleic acid molecule which comprises at least one of the nucleotide sequences shown in SEQ ID Nos. 1 to 5 or 8 to 15;

- e) a nucleic acid molecule which is complementary to a nucleic acid molecule according to a) to b), and a fragment thereof.
- - Modifikation des zu transportierenden Nucleinsäu remoleküls, das natürlicherweise nicht mit dem Angiotropin-Komplex und/oder ähnlichen Komplexen in Zusammenhang steht, durch Anlagerung von Se quenzen eines Nucleinsäuremoleküls nach einem der Ansprüche 1 bis 7, die unter geeigneten Bedingun gen zur Ausbildung einer Haarnadelschleife mit der Konsensus-Sequenz 5'-CUG-3' geeignet sind;
- - Herstellung eines ternären Komplexes durch in vitro Zugabe von Metall-Ionen und mindestens ei nem Protein zu dem modifizierten Nucleinsäuremo lekül; und
- - Applikation des hergestellten ternären Komplexes auf Zellen oder Gewebe, in die das modifizierte Nucleinsäuremolekül eingebracht werden soll.
