Low molecular saccharides, for example mono saccharides such as glucose, and oligosaccharides such as Sucrose are used as substrates for bio technological processes or in modifi adorned form as auxiliary substances for different Industries. For example, larger ones Amounts of glucose for chemical synthesis, at for example of sorbitol and glutamate, and for tech African purposes, including for alcoholic drinks tion, used. Sucrose, the economy The most important type of sugar, mainly becomes Can be used for food and preservation but also for plastics, varnishes, for synthesis of protein, amino acids, antibiotics etc. as well as Additive for hard PUR foams used the.
Polysaccharides such as Cel Lulose and starch, in native or modes form used in industry. Strength is not just the most important food of the People. Factory-made starch is also in the paper industry, including manufacturing of cardboard boxes or as paper aids, at for example for gluing paper, in the textile industry, for example as a size or for ap pretend and weight new tissues or as Stiffening agents for laundry and in pharmacy, for example as an explosive and filler for tab letten, as a lubricant and filler for powder, as Basis for ointments etc. used. cellulose is one of the most important raw materials for numerous Industries. The largest amounts of cellulose will be thereby from the paper and textile industry ver needs, for example the most common Textile fabric made of more or less pure, natural best or artificially converted cellulose hen. Polysaccharides are becoming increasingly important or polysaccharide derivatives also as Zu impact material in the construction industry.
Despite the numerous already established applications gen have both cellulose and starch gra four disadvantages. For example, the Manufacture of chemical cellulose derivatives, al so of products that, for example, via Ver esterification and / or etherification reactions, Substi tion, oxidation or crosslinking reactions generated, the use of numerous chemi reagent, especially the use of Solvents, their subsequent disposal for Part is associated with considerable costs. The Her The position of cellulose derivatives is therefore in space mean much more expensive than the production of Starch derivatives. The production of Starch derivatives associated with the problem that Strength is a heterogeneous connection that arises from the linear amylose and strongly branched ten amylopectin. Because of this heterogeneity it is not possible or only possible to a limited extent chemical starch derivatization reproducible and to carry out specifically. For years, therefore, al with moderate success so far, tried Grow plants that are exclusively amylose-haled deliver strength. Inexpensive, for the big technical use with suitable polysaccharides a predominantly linear structure is currently available not available on the market.
For applications in which a li near structure of carbohydrate polymers for the the desired property profiles Prerequisite is, therefore, make longer-chain buttocks lyfructane is an important alternative Polyfructans are polysaccharides, which mainly include fructose units are linked. Differentiate polyfructans the way in which fructose is linked Units. For example, the fructose Units of the main polyfructan inulin in furanoside form with a β-1,2 bond. At the bottom lyfructan levan are the fructose units above β- 2,6 bonds linked. Polyfructans are reserve Carbohydrates found in a number of monocotylers and dicotyledonous plants, for example composites, grains seeds and cereals, but also with algae and some against gram-positive and gram-negative bacteria are to be found.
Inulin is a predominantly linear polyfructan Structure, with fructose units above. β-1,2 Bonds are linked and the chain is true apparently from a non-reducing α-D Glucose unit is completed. Inulin finds alone or together with starch as a reserve Carbohydrate in dahlia tubers, arti shock, Jerusalem artichoke tubers, chicory roots and in the cells of Inula and other daisies (Compositae), less often in related plants families (Campanulaceae, Lobeliaceae). The inuline differ from different plant species essentially by their medium polymer onsgrad. For example, inulin has topiname bur an average degree of polymerization of 5-7, Inulin from chicory a medium polymer Degree of 10-12 and inulin from artichokes average degree of polymerization of about 25 (Beck, R. H. F. and Praznik, W., Inulin-Containing Plants Source of raw materials. Starch / Starke, 38 (1986), 391-394). Because of their linear structure, these can Inulins are relatively easy to make derivatives are, these derivatives largely biolo are biodegradable. Because of the relatively short ket ten are such inulins or from them manufactured derivatives, however, for use as polymeric surfactants, emulsifiers and plasticizers are not suitable. The use of I is also known nulin as "bulking agent" or as fat replacement material.
It is also known to inulin to fructooligosacchari the hydrolyze and this as prebiotic Le use food ingredients (Wang, X. and Gibson, G. R., J. Appl. Biochem., 75 (1993), 373-380). On serious disadvantage of these fructooligosaccharides however, is their relatively high glucose part. So fructooligosaccharides contain that from the inulin of Jerusalem artichoke were made, for example 40 to 20% glucose, while from the inulin of Zi fructooligosaccharides produced by chories about 8 contain up to 10% glucose. Such fructooligosac charids are only for Er suitable for diabetics.
From EP 657 106 A1 is the manufacture and Ver use of hydrogenated fructooligosaccharides known. Here, too, the relatively high glu is evident cos content of the fructooligosaccharides used as a disadvantage.
Difructosedianhydri are of particular interest de, which are also made from inulin can and which ver as food ingredients have it turned. They stand out particularly there by pronouncing a in the colon Develop prebiotic effects and thereby sustained contributes to a healthy intestinal flora and intestinal wall wear. There is therefore a big trend in industry interest in an inexpensive production of Difructosedianhydriden. Manufacturing process of difructosedian hydrides are known (Seki, K. et al., Starch / Starke, 40: 440-442 (1988); DE-PS 195 47 059; Uchiyama, T., in: Science and Technolo gy of Fructans (editors Suzuki, M. and Chatter ton, N.J. (1993), CRC Boca Raton, Florida). al However, the Difruc tosian hydrides the high glucose disadvantageous.
Plants containing polyfructan, like a number of microorganisms, are known to express fructosyl transferase (Ftf) proteins that catalyze the polymerization of linear or branched fructose polymers. Microbial fructosyltransferases have been detected, for example, in Strepto coccus, Bacillus, Pseudomonas, Xanthomonas, Acetobacter, Erwinia and Actinomyces strains. In microbial polyfructans, the fructose residues are linked via a β-1,2 and / or a β-2,6 bond. This means that microbial fructosyltransferases are either inulin sucrases or levan sucrases. While vegetable polyfructans have a relatively low molecular weight, with about 10 to 30 fructose units being linked per molecule, microbial polyfructans have a molecular weight of up to 10 6 to 10 8 , with more than 100,000 fructose units being linked. Studies on polyfructan biosynthesis have also shown that bio-synthesis is generally much easier in bacteria than in plants. For the reasons mentioned, there is a strong interest in using bacterial fructosyltransferases in particular for the production of polyfructanes.
Currently, however, only one bacterial fructosyltransferase is known, namely that from the gram-positive bacterium Streptococcus mutans, which can catalyze the formation of inulin based on sucrose as the starting molecule (Shiroza and Ku ramitsu, J. Bacteriol., 170 (1988), 810-816); Ro sell, K.-G. and Birkhead, D., Acta Chem. Scand., 28 (1974), 589). The inulin produced from sucrose with the S. mutans fructosyl transferase has a molecular weight of 20-60 × 10 6 g / mol. The glucose content of the inulin is therefore negligible, so that the inulin would in principle be suitable for the production of fructooligosaccharides. On the other hand, inulin produced in this way has about 7% branching and is therefore less suitable for use as a raw material for further chemical derivatization. In addition, the microorganism S. mutans is a human pathogenic germ, so that its use and multiplication on an industrial scale for enzyme production is associated with major problems.
Process for making carbohydrate Polymers, especially polyfructans, in transge plants using microbial fructo ftf genes encoding syltransferases, for example se of the fructosyltransferase gene from S. mutans, are also known.
Für das Fructo syltransferase-Gen aus Streptococcus mutans werden keine Sequenzmodifikationen beschrieben, so dass das Expressionsniveau der Fructosyltransferase ver gleichsweise sehr gering ist.PCT / NL 93/00279 discloses the transformation of Plants with chimeric genes that make up the sacB gene Bacillus subtiles or the ftf-Ge from Streptococ cus mutans included. In the case of a Levansucra se encoding sacB gene is used to increase the Ex levels of pressure in transformed plants Modification of the 5'-untranslated region of the bacterial gene recommended. For the fructo Streptococcus mutans syltransferase gene no sequence modifications described, so that the expression level of the fructosyltransferase ver is equally very low.
In addition, other fructosyltrans ferase genes of plant origin are used. just if described is the use of the trans plants produced oligosaccharides as Zu sugar substitute, as a dietary supplement and as bifidogenic agent in food as well as bi fidogenic agent in animal feed.
When using the fructosyltransferase gene of Streptococcus mutans in heterologous expressions onsystems, that means both heterologous bacteria expression systems as well as plant Systems, it has been found that the native protein produced in extremely small amounts and therefore the inulin production only in very limited. This low en zym production in heterologous host cells goes down forth with severe growth disorders Host cells.
The present invention is therefore techni underlying problem, procedures and means, esp special procedures and means on the basis of Streptococcus mutans fructosyltransferase gene, for the production of high molecular weight polyfructanes, especially inulin, with β-1,2 bonds and one essentially linear structure and a very to provide low glucose content, which is a simple and inexpensive production of the Polyfruc enable tane in large quantities, the above problems described in the prior art nik, especially the low expression bacterial Fructosyltransferase genes in heterologous hosts systems to be overcome.

The present invention solves this technical problem in particular by providing a modified fructosyltransferase gene from Strep tococcus mutans with a nucleic acid sequence as shown in SEQ ID No. 1 or a nucleic acid sequence that encodes an amino acid sequence as shown in SEQ ID No. 2, which is a at the N-terminus and / or at the C-terminus modified polypeptide encoded with the activity of a fructosyltransferase, in particular wherein the polypeptide has at least one deletion at the N-terminal and / or at the C-terminal end. In particular, the present invention provides a preferably isolated and completely purified nucleic acid molecule according to the main claim, which encodes a polypeptide with the activity of a fructosyltransferase (ftf) and which is in a nucleic acid sequence shown in SEQ ID No. 1 or in one of them Nucleic acid sequence coding for amino acid sequence given in SEQ ID No. 2 has at least one deletion selected from the group consisting of:
- a) Deletion der Nucleotide 4 bis 222;a) Deletion of nucleotides 4 to 222;
- b) Deletion der Nucleotide 1 bis 104; und b) deletion of nucleotides 1 to 104; and
- c) Deletion der Nucleotide 2254 bis 2385.c) Deletion of nucleotides 2254 to 2385.
By deleting nucleotides 4 to 222 approximately of nucleotides 1 to 104, the Sig nal sequence of the native fructosyltransferase gene completely or partially removed from S. mutans, so that the encoded signal peptide of the native S. Complete or partial mutans fructosyl transferase is wisely removed. Through the thus obtained in tracellular enzyme production will be the growth disorders of heterologous host cells, such as wise Escherichia coli, which is based on the expression of the S. mutans fructosyltransferase are eliminated and the enzyme can be in high volu yields can be obtained from these cells.
Surprisingly, it was also found that a dele comprising nucleotides 2254 to 2385 tion at the C-terminus of the fructosyltransferase a significant improvement in growth hetero leads host cells. The function of the deletier th hydrophobic region in the native protein not known. According to the invention, this leads De letion within the C-terminal region to egg ner significant improvement in growth hetero loger host cells and high volume yields of expressed protein.
According to the invention it was also found that the in the 5 'region of the nucleic acid sequence of the ftf gene deleted sequences by at least one Se range of another gene can be replaced nen, the other gene being preferred is the lacZα gene. Such a mutation too causes improved growth more heterologous Host cells and high volume yields of the expri protein.
Using the above invented nucleic acid molecules according to the invention can therefore Produce vectors in pro- and / or eukaryon cells express a high expression of a modifi graced polypeptide with the activity of a fructo Ensure syltransferase. According to the invention found that a particularly high volume prey of the expressed protein in bacterial Host systems is achieved when the fiction moderate, deleted at the 5 'end and / or at the 3' end Nucleic acid molecules or nucleic acid molecules in which the most 5'-deleted sequences through a sequence region of another gene in the Escheri expression cassette chia coli vector pJOE2702 (Volff et al., Mol. Further advantageous embodiments of the invention result from the remaining subclaims.
According to the invention, it is therefore preferred tion form, preferably insulated and free nended nucleic acid molecule provided that a bacterial fructosyltransferase (ftf) gene is and a polypeptide with the activity of a fructo syltransferase encoded and at the 5 'end and / or 3'-end contains at least one deletion.
In connection with the present invention under a bacterial fructosyltransferase gene or a nucleic acid molecule that is a bacterial The polypeptide with the activity of a fructo syltransferase encodes the coding DNA sequence understood a gene, the gene product of which Akti Quantity of sucrose: 2,1-β-D-fructosyltrans ferase (EC 2.4.1.9) and that originally from a bacterium, preferably from Streptococcus mutans. In connection with the present the invention means the term "fructosyl transferase "or" β-D-fructosyltransferase " Polypeptide or a protein that synthesizes a high molecular weight, from repeating fructose Catalyze units of existing carbohydrate polymer can, with sucrose as the starting substrate serves for further polymerization. In that way synthesized polymeric product are the themselves repeating fructose units above β-1,2- Bonds linked so that the Fruc tosyltransferase, which is the synthesis of this product catalyzed, also referred to as inulin sucrase that can. The synthesized, repeating itself the high-molecular fructose units Polymer preferably has a linear structure on, can be a terminal, from a sucrose Contain molecule derived glucose residue and around holds at least two fructose residues. Together Hang with the invention, this carbohydrate is considered Designated polyfructan. It is preferably the synthesized polyfructan is inulin.
The nucleic acid can be a DNA sequence play part of a genomic DNA sequence, or a RNA sequence, for example an mRNA sequence or be part of it. The nucleic acid can both of natural origin, that is, it can for example from Streptococcus mutans cells iso be lined, as well as synthetic origin his.
The nucleic acid molecule according to the invention has in the nucleic acid sequence shown in SEQ ID No. 1, which is the amino acid sequence shown in SEQ ID No. 2 encoded, at least one deletion selected from the group consisting of a) nucleotides 4 to 222, b) nucleotides 1 to 104 and c) the nucleus otides 2254 to 2385. In connection with the the present invention is called "deletion" understood a mutation in which part the nucleic acid sequence present in the wild-type gene is missing. For example, deletions can be performed in vitro generated using restriction enzymes if the area to be deleted is suitable Restriction interfaces are flanked. Deletion mutations can also be solved with the help tuned endonucleases, for example under Ver using the enzyme BAL-31. Such en zymes break down the ends by restriction zym cleavage were generated. A deletion mutage nese can also use a mutier ten primers can be achieved in a PCR reaction. The sequence of such a primer spans the area to be deleted, with the Primer binds to two areas of a target region, that flank the area to be deleted. In the The amplification is the one spanned by the primer Area not covered and therefore not amplified.
The fructo deletions according to the invention syltransferase gene preferably affect the 5'- Area and the 3 'area of the native fructo Streptococcus mutans syltransferase gene.
A particularly preferred embodiment with a Deletion in the 5 'region, which the nucleotides 4 to 222 comprises, the nucleic acid molecule with the in SEQ ID No. 3 shown nucleic acid sequence, the one Protein with the amino acid shown in SEQ ID No. 4 sequence encoded. In the original organism S. mutans a signal sequence mediates the secretion of the ene zymes from the cytoplasm, the signal sequence during the secretion process by a signal peptidase is split off. The signal peptide dependent protein export, which is used in gram positive and gram-negative bacteria works in a similar way, is a complex energy dependent process on which a variety of soluble and membrane-bound pro teine are involved (Schatz and Beckwith, Annu. Rev. Genet., 24 (1990), 215). At one aimed overproduction of fructosyltransferase Proteins in a heterologous host organism The signal sequence was therefore a nuisance growth of the host and thus for ver lead to a reduction in product yield if the Sig nal sequence is also functional in the heterologous host and the amount of protein produced is the capacity of the Secretion mechanism exceeds and / or when on their physiological intra- or extracellular pro processes in the area of the cell membrane through the recombi nante protein itself or by secretion of the recombinant protein.
Complete removal of the signal peptide coding nucleic acid sequences in the nuclein acid molecule with SEQ ID No. 3 leads to a complete removal of the signal peptide so that the expression of the modified fructosyltransfe rase protein in a heterologous host for intra leads to cellular production of the gene product. By intracellular production of fructosyltransfer The growth disturbances that occur in hosts systems that use native fructosyltransferase Express protein, almost completely eliminated and the desired protein product can be in high volume lumen yields are obtained.
A particularly preferred embodiment for a Deletion in the 3'-region of the native fructosyl tranbs ferasegens is the nucleic acid molecule with the in SEQ ID No. 7 shown nucleic acid sequence, the one Polypeptide with the amino shown in SEQ ID No. 8 acid sequence encoded. At this nucleic acid mole The nucleotides 2254 to 2385 have been deleted. This area has a high hydropathy index on how using the procedure of Kyte and Doo little (J. Mol. Biol., 157 (1982), 105-142) has been. Although no di right function can be assigned, show hete rologous host systems in the expression of a sol Chen modified fructosyltransferase protein versus host cells that contain the native fructo syltransferase protein express a significantly improved growth behavior. The modified Protein can also be used in high volume yields be isolated.
In a further preferred embodiment those in the 5 'region of the fructosyltransferase gene de lethal sequences by the sequence area of a other gene replaced. In a particularly preferred The third embodiment is the other Gene around the lacZα gene. In connection with the before lying invention means the term "replacement zen "that the ftf coding for the signal peptide Sequences completely or partially equivalent sequences of another gene replaced can be. When using the lacZα gene equivalent sequences of this gene with the am 5 'end of deleted fructosyltransferase gene fusio ned. A particularly preferred example of this is the nucleic acid molecule with the ge in SEQ ID No. 5 showed nucleic acid sequence that a in SEQ ID NO. 6 amino acid sequence shown encoded. At the This ftf gene variant became nucleotides 1 to 104 the native fructosyltransferase nucleic acid sequence against nucleotides 1 to 83 of the lacZα gene replaced. The nucleic acid molecule therefore has compared to the native ftf sequence Nucleotides 1 to 104. The exchange at the 5 'end leads to an expression of the fusion protein in heterologous host systems for localization of the gene product in the periplasmic space. With egg transformed such a plasmid host cells gene compared to the wild-type ftf gene a distinct improvement in the heterologous host cells growth and the fusion protein can also can be obtained in high volume yields.
Further preferred embodiments are the nuc Linseed acid molecule with the one shown in SEQ ID No. 9 Nucleic acid molecule, which is one in SEQ ID No. 10 amino acid sequence, and the Nuc Linseed acid molecule with the one shown in SEQ ID No. 11 Nucleic acid sequence, which is one in SEQ ID No. 12 provided amino acid sequence encoded. The nuclein acid molecule with SEQ ID No. 9 has at the 5 'end a deletion of nucleotides 4 to 222 on and on 3'-end a deletion of nucleotides 2254 to 2385. For the nucleic acid molecule with SEQ ID No. 11 are nucleotides 1 through 104 through the nucleotides 1 to 83 of the lacZα gene have been replaced and the 3'- End also shows a deletion of the nucleotides 2254 to 2385. Both ftf gene variants lead when expressing the corresponding gene products in heterologous host systems to a clear ver improvement in growth, taking the appropriate Gene products obtained in high volume yields that can.

The present invention further relates to vectors ent ent the nucleic acid molecules of the invention hold. In a particularly preferred embodiment, the present invention includes vectors in which at least one nucleic acid molecule according to the invention is coolly under the functional control of at least one regulatory element. According to the invention, the term “regulatory element” is understood to mean those elements which ensure the transcription and / or translation of nucleic acid molecules in prokaryotic and / or eukaryotic host cells, so that a polypeptide or protein is expressed. Regulatory elements can be promoters, enhancers, silencers and / or transcription termination signals. Regulatory elements which are functionally linked to a nucleotide sequence according to the invention, in particular the protein-coding sections of this nucleotide sequence, can be nucleotide sequences which originate from other organisms or genes than the protein-coding nucleotide sequence itself. Examples thereof are: T7 , T3, SP6 and other common regulatory elements for in vitro transcription; P LAC , P Ltet and other common regulatory elements for expression in E. coli; GAL1-10, MET25, CUP1, ADH1, AFH1, GDH1, TEF1, PMA1 and other regulatory elements for expression in the baker's yeast S. cerevisiae; Polyhedrin for expression in Baculovirus systems; P CMV , P SV40 and other common regulatory elements for expression in mammalian cells; Tissue- or organ-specific - in particular memory-organ-specific promoters, such as the Vicillin promoter from Pisum sativum, the Arabidopsis promoter AtAAP1 or the patatin B33 promoter, for expression in plant systems.
In a preferred embodiment it is provided that the regulatory elements derive from the L-rhamnose operon from Escherichia coli. In a particularly preferred embodiment, a nucleic acid molecule according to the invention is integrated into the expression cassette of the vector pJOE2702 (Volff et al., 1996; Stumpp et al., 2000). The expression cassette comprises the promoter rha P , which is derived from the two-stage regulated L-rhamnose operon rhaBAD from Escherichia coli (Egan and Schleif, J. Mol. Biol., 243 (1994), 821-829). The vector pJOE2702 is therefore an L-rhamnose-inducible expression vector which can be positively regulated over two stages and which is present in the host bacterium rium Escherichia coli in high copy numbers. The transcription termination and the translation initiation of the transcripts also take place via sequences of the expression cassette contained in the vector. Compared to most commercially available E. coli expression vectors, pJOE2702 has two decisive advantages. First, the vector in the non-induced state leads to a very low base expression of the polypeptide to be expressed. Second, transcription of the expression cassette is induced with a delay. The vector is therefore particularly suitable for cloning and producing proteins which negatively affect the vitality and the growth properties of the host cell, such as bacterial fructosyltransferases. Although Escherichia coli cells, which contain the vector pJOE2702 with the complete S. mutans fructosyltransferase gene, show complete growth inhibition after induction, the vector is particularly suitable for expression of the nucleic acid molecules according to the invention which 'End and at the 3' end contain at least one of the deletions according to the invention.

A polymerase chain reaction (PCR) was carried out for cloning the fructosyltransferase gene from S. mutans DSM20523, the primer ftf1 (upper) with the sequence: 5'-TATATATCATATGGAAACTAAAGTTAG-3 'and the primer ftf2 (lower) with the sequence: 5'- TAGGATCCTTATTTAAAACCAATGCT-3 'were used. The primer ftf1 (upper) contained an NdeI restriction site and the primer ftf2 (lower) contained a BamHI restriction site. Chromosomal DNA from S. mutans DSM20523, which had been isolated using a commercially available cleaning kit, served as a template in the PCR reaction. The PCR reaction was carried out in 40 μl reaction volume, each with 8 μmol primer, 50 ng DNA template and 2.5 units of polymer polymerase in 10 mM Tris-HCl, pH 8.85, 25 mM KCl, 5 mM ( NH 4 ) SO 4 , 2 mM MgSO 4 , 5% dimethyl sulfoxide, 0.2 mM dATP, 0.2 mM dTTP, 0.2 mM dGTP and 0.2 mM dCTP. The PCR reaction involved the following steps: 5-minute denaturation at 96 ° C, 5 cycles, each comprising a 1-minute denaturation at 63 ° C, a 30-second primer addition at 40 ° C and a 2 minute, 30 Polymerization lasting for seconds at 68 ° C, 25 further cycles, each comprising a 1 minute, 30 second denaturation at 92 ° C, a 1 minute, 30 second primer addition at 49 ° C and a 2 minute, 30 second polymerisation 68 ° C, and a 5-minute final polymerization reaction at 68 ° C. An approximately 2.4 kb fragment was amplified using the PCR reaction. The isolated fragment was digested with the NdeI and BamHI restrictionases and ligated with the vector pJOE2702 (Volff et al., Mol. Microbiol., 21 (1996) 1037-1047; Stumpp et al., Biospectrum, 1 (2000) 33-36) which had been cleaved with the same restrictionases. The coding sequence of the ftf gene was thus cloned in the correct reading frame into the L-rhamnose-inducible expression cassette contained in the vector, so that the transcription of the inserted nucleic acid is under the control of the promoter rha P contained in the vector pJOE2702. The transcription termination of the ftf gene and the translation initiation of the transcripts also take place via sequences of the vector. The E. coli strain JM109 was then transformed with the plasmid pDHE113 resulting from the ligation. Transformants were selected for ampicillin resistance.

The suspension was then adjusted to an OD 600 value of 10. The cells were disrupted by ultrasound treatment or by French press treatment. All extracts were then treated by centrifugation at 10,000 × g for 20 minutes.
After that, the total protein content was according to Bradford (Bradford, Anal. Biochem., 72 (1976), 248-254) true, with a calf serum albumin calibrated Reagent from Biorad (Biorad-Laboratories GmbH, Mün chen) was used. To compare the protein Sample of the crude extracts obtained with the protein patterns of non-induced control were the Extracts electrophoretically in 7.5% SDS Polyacrylamide gels separated (Laemmli, Nature, 227 (1979) 680-685).
The quantitative determination of the Ftf activities was done by measuring the specific Ftf activities in raw extracts. Cell cultivation, cell disruption and the preparation of the crude extracts were carried out as described above. Since each molecule of sucrose releases one molecule of glucose at each conversion, the activity was determined by determining the amount of glucose released per unit of time. The Ftf activities were determined over a period of 60 minutes at 30 ° C. using a crude extract amount containing about 1 to 3 mU of fructosyltransferase in a volume of 1 ml with 100 mM Scr in 50 mM K 2 HPO 4 / KH 2 PO 4 buffer, pH 6.5. One unit corresponds to the release of 1 µmol of glucose per minute in the presence of 100 mM sucrose at 30 ° C and a pH of 6.5. After an hour's incubation, the reaction was terminated by heating at 100 ° C for 10 minutes. This was followed by centrifugation at 10,000 × g for 10 minutes. The amount of glucose released was then determined in a 100 μl aliquot using the coupled glucose oxidase (GOD) / peroxidase (POD) enzyme test (Werner et al., Z. Analyt. Chem., 252 (1970) 224-228) , The GOD-Perid® test (Roche Diagnostics) was used, in which the oxidation of the dye 2,2'-azino-di- (3-ethylbenzthiazoline sulfonate) (ABTS®) is detected photometrically. The test was carried out in a total volume of 1 ml with 80 µg / ml GOD, 10 µg / ml POD and 1 µg / ml ABTS® in 50 mM K 2 HPO 4 / KH 2 PO 4 buffer, pH 6.5 , carried out, the extinction being measured at 578 nm after 30 minutes. The test was calibrated using standard glucose solutions. The results obtained for the crude extracts of the different expression strains are summarized in Table 1.

Das unter Verwendung des HPLC- GPC-Verfahrens ermittelte Molekulargewicht von Inu lin beträgt 40 × 106 g/mol, wobei der Verzweigungs grad 3 Mol% beträgt.60 l of a 10% sucrose solution, pH 6.5, were added after the addition of 640 U fructosyl transferase per liter of batch solution for 28 hours at 30 ° C. with stirring, using a crude extract of the E. coli strain JM109 (pDHE143) , The solution was then subjected to direct ultrafiltration (Sartocon Mini; 3 modules with a molecular retention capacity of 100,000 Daltons). The 4.5 l retentate obtained was diluted twice with 6 l of deionized water and finally concentrated to 4.5 l. Inulin was then precipitated with isopropanol (final concentration 62% by weight). The separated precipitate was washed with 5 l of the same isopropanol solution and then gently dried at 45 ° C. This gave 0.36 kg of a white product with a dry matter content (TS) of 93%. A yield of 8.5% was achieved in relation to sucrose consumption. The molecular weight of inulin, determined using the HPLC-GPC method, is 40 × 10 6 g / mol, the degree of branching being 3 mol%.
0,1 molarer Essigsäure mit Endo-Inulinase (0,13 ml/Ansatz;
SP 168, Fa.
bildeten Fructooligosaccharide mittels Ultrafiltra

Rückhaltevermögen von 10 000 Dalton) abgetrennt.
Das Permeat enthielt 7,5 g Fructooligosaccharide,
während das auf 1 l verdünnte Retentat entsprechend
11,6 g TS enthielt. Anschließend wurde das Retentat
nach erneutem Einstellen des pH-Wertes mit Endo-
Inulinase bei einem konstanten Enzym/Substrat-
Verhältnis inkubiert, wie zuvor beschrieben. Dieser
Vorgang wurde insgesamt 5-mal wiederholt. Mittels
einer Gelpermeations-Chromatographie wurde in den
vereinigten Permeaten die folgende Kettenlängenver
teilung bestimmt:
20 g of the Ftf-inulin obtained in Example 6 with 95% TS were after preparing a 1% solution by heating for 15 minutes at 95 ° C. and adjusting the pH to 5.0 using 0.1 molar Acetic acid with endo-inulinase (0.13 ml / batch; SP 168, Novo) incubated for 8 hours at 50 ° C. with stirring. After inactivating the enzyme by heating at 95 ° C. for 15 minutes, the fructooligosaccharides formed were separated by means of ultrafiltration (Sartocon Mini, module with a molecular retention capacity of 10,000 daltons). The permeate contained 7.5 g fructooligosaccharides, while the retentate diluted to 1 l contained 11.6 g TS. The retentate was then incubated with the endo-inulinase at a constant enzyme / substrate ratio, as previously described, after the pH had been adjusted again. This process was repeated 5 times in total. The following chain length distribution was determined in the combined permeates by means of gel permeation chromatography:
An analysis using the HPAEC method showed that fructose was the only carbohydrate building block, i.e. the isolated oligosaccharides are homoo ligomeric fructooligosaccharides of type F n with n = 1-25.
Endo-Inulinase (beispielsweise SP 168; NOVO) wurde
wie in Beispiel 5 für Fructosyltransferase be
schrieben, an EUPERGIT® C (Röhm) immobilisiert. Zu
1 l einer 10%igen Saccharoselösung, pH-Wert 6,5,
wurden bei 30°C 50 g immobilisierte Fructosyltrans
ferase (vergleiche Beispiel 2) und 20 g immobili
sierte Endo-Inulinase zugegeben und unter langsamen
Rühren inkubiert. Stündlich wurden Proben entnommen
und auf den Saccharosegehalt überprüft. Sobald kei
mittels Filtration entfernt. Mittels Gelpermeati
ons-Chromatographie wurde die Zusammensetzung der
dabei erhaltenen Produktlösung wie folgt bestimmt:
Endo-inulinase (for example SP 168; NOVO) was, as described in Example 5 for fructosyl transferase, immobilized on EUPERGIT® C (Röhm). To 1 liter of a 10% sucrose solution, pH 6.5, 50 g of immobilized fructosyl transferase (see Example 2) and 20 g of immobilized endo-inulinase were added at 30 ° C. and incubated with slow stirring. Samples were taken every hour and checked for sucrose content. As soon as sucrose was no longer detectable, the reaction was complete and the enzymes were removed from the mixture by filtration. The composition of the product solution obtained was determined as follows by means of gel permeation chromatography:
Compared to Example 7, products with egg get a much higher fructose content.
The homooligomeric fructooligosaccharide mixtures obtained in Examples 7 and 8 with a chain length of DP1-DP25 were adjusted by evaporation to a dry matter content of 10% in each case. 450 ml of each solution were hydrogenated in a laboratory autoclave in the presence of Raney nickel for 10 hours with hydrogen at 150 bar and 80 ° C. The solution obtained was pumped out of the autoclave, filtered and purified using an ion exchanger. An analysis showed that the solution contained (fructosyl) n- mannitol, (fructosyl) n -sorbitol (n = 1-24) as well as mannitol and sorbitol, which had been formed from the fructose present in the starting solution. Mannitol and sorbitol were separated using known chromatography methods, so that a product consisting of (fructosyl) n- mannitol and (fructosyl) n -sorbitol was obtained.
Cells of the strain Arthrobacter ureafaciens ATCC 21124 were placed in four shake flasks, each with 100 ml of a nutrient solution, containing 8 g chicory inulin (Raftiline®), 2 g Na 2 HPO 4 , 1 g KH 2 PO 4 , 1 g NH 4 NO 2 , 0.5 g MgSO 4 , 0.01 g Fe 2 SO 4 , 0.03 g CaSO 4 , 0.5 g yeast extract in deionized water, inoculated. A 24-hour incubation was then carried out at 27 ° C. with shaking at 150 rpm. The cells were then harvested by centrifugation and immobilized in polyvinyl alcohol gel particles.
To 1 l of a 10% sucrose solution, pH 6.5, 50 g immobilized Fructo were at 30 ° C. syltransferase (see Example 5) and 20 g in mobilized endo-inulinase and immobilized A. ureafaciens cells. Then was at 30 ° C. incubate with slow stirring leads. Samples were taken and opened every hour checked the sucrose content. As soon as no sac charose was more detectable, the reaction was be ends and the enzymes were removed from the batch using Filtration separated.
An HPLC analysis of the product solution obtained showed the formation of 18 g DFA III.
