Description
Die Erfindung betrifft neue Komplexe zur Transfektion die durch Zusammenlagerung von zu transfizierendem Material mit ober flächenmodifizierten Partikeln aus bei Raumzustand festen Lipiden oder aus Lipidmischungen aus bei Raumtemperatur festen Lipiden mit bei Raumtemperatur halbfesten oder flüssigen Lipiden hergestellt werden. The invention relates to new complexes for transfection Storage of material to be transfected with upper surface-modified particles from lipids that are solid when in space or from lipid mixtures of lipids solid at room temperature with semi-solid or liquid lipids at room temperature getting produced.
In der rekombinanten Gentechnologie und in der Gentherapie führt man Erbinformation (z. B. Polynukleotide) effizienter in Zellen ein, wenn man sie in einen Träger einarbeitet oder an ihn bindet. Leads in recombinant genetic engineering and in gene therapy genetic information (e.g. polynucleotides) is made more efficient in cells if you incorporate it into a carrier or bind it to it.
Für solche Zwecks eingesetzte Trägersysteme sind beispielsweise:
Carrier systems used for such purposes are, for example:
- 1. Viren1. viruses
- 2. Adenoviren2. adenoviruses
- 3. Liposomen3. liposomes
- 4. Polymerpartikel4. The polymer particles
- 5. Makromoleküle5. macromolecules
Ein Vorteil von Viren ist die hohe Effektivität, da die Infektiö sität von Viren ja gerade darauf beruht, virale DNA in Zellen einzuschleusen. Die Erbinformation wird zwar in das eukaryon tische Genom integriert, jedoch besteht dabei die Gefahr einer Mutation durch Integration des Gens in das Genom. Zusätzlich bedeutet die Verwendung von Viren ein hohes Sicherheitsrisiko für den Patienten. An advantage of viruses is their high effectiveness, because the infectious Virusity is based on viral DNA in cells infiltrate. The genetic information is indeed in the eukaryon integrated genome, but there is a risk of Mutation through integration of the gene into the genome. additionally the use of viruses means a high security risk for the patient.
Another advantage of adenoviruses is its high effectiveness. A disadvantage, however, is that they are not in the eukaryotic Be integrated into the genome but as extrachromosomal DNA in the Cell nucleus are present so that only transient transfection takes place. This requires repeated application of the viral gene transfer system for the entire duration of the disease, d. H. usually lifelong, because not once is finally treated.
Non-viral infection systems such as liposomes have the advantage that they avoid the dangers of viral systems. However, you have the disadvantage that the transfection efficiency is significantly lower is. In addition, with liposomes - as already known from liposomal drugs - problems with physical Stability. In addition, they are relatively expensive. "Cheap" Liposomes for pharmaceutical purposes do not yet exist.
So zeigten PLA-Partikel nach Aufnahme in Zellen Zytotoxizität (siehe A. Smith and I. A. Hunneyball, Evaluation of poly(lactic acid) as a biodegradable drug delivery System for parenteral administration, Int. J. Pharm. 30, 215-220 (1986)).Polymer particles are being examined intensively and have not yet found their way to the market for various reasons. Many of the polymers used are toxicologically problematic and have therefore not been approved by the responsible authorities. For example, polyalkylcyanoacrylates release potentially carcinogenic formaldehyde when broken down in vivo, polymethacrylates have a too long breakdown time (years). Polymers that have been approved as implant material in vivo, such as polylactic acid (PLA) and the copolymer of lactic and glycolic acid (PLA / GA), also have toxicity problems at the cellular level. For example, PLA particles showed cytotoxicity after uptake in cells (see A. Smith and IA Hunneyball, Evaluation of poly (lactic acid) as a biodegradable drug delivery System for parenteral administration, Int. J. Pharm. 30, 215-220 ( 1986 ) ).
Polymere werden auch in molekularer Form als Transfektionsagens eingesetzt, z. B. Polyethylenimin (pEI) (siehe Rudolph, C., Lausier, J., Naundorf, S., Müller, R. H., Rosenecker, J., In vivo gene delivery to the lung using polyethylenimine and fractured polyamidoamine dendrimers, The Journal of Gene Medicine 2, 269- 278, 2000). Die Problematik ändert sich wenig, wenn Polymere nicht in partikulärer Form vorliegen sondern als einzelne Makromoleküle. Im Gegenteil, erhöht sich bei gleicher Dosis teilweise die Toxizität, da das gesamte Polymer bereits gelöst vorliegt und nicht in kleineren Mengen verzögert durch Abbau der Partikel in Lösung geht.Polymers are also used in molecular form as a transfection agent used, e.g. B. polyethyleneimine (pEI) (see Rudolph, C., Lausier, J., Naundorf, S., Müller, R.H., Rosenecker, J., In vivo gene delivery to the lung using polyethyleneimine and fractured polyamidoamine dendrimers, The Journal of Gene Medicine 2, 269- 278, 2000). The problem changes little when polymers not in particulate form but as individual Macromolecules. On the contrary, increases at the same dose partly the toxicity, since the entire polymer is already dissolved is present and not delayed in smaller quantities by breaking down the Particle goes into solution.
Zur Umgehung der oben geschilderten Probleme wie Toxizität, Sicherheit, mangelnder physikalischer Stabilität und relativ hoher Kosten war es Aufgabe der vorliegenden Erfindung ein Transfektionssystem zu schaffen, daß diese Probleme minimiert oder vermeidet. To work around the issues outlined above, such as toxicity, Security, lack of physical stability and relative high cost, it was an object of the present invention To create a transfection system that minimizes these problems or avoids.
Diese Aufgabe wurde durch den erfindungsgemäß Transfektionskom
plex "TransoPlex" auf der Basis einer Kombination von
This object was achieved by the transfection complex "TransoPlex" based on a combination of
- 1. Lipidpartikeln1. lipid particles
- 2. Oberflächenmodifizierer für diese Lipidpartikel2. Surface modifier for these lipid particles
- 3. zu transfizierendem Material und3. material to be transfected and
- 4. gegebenenfalls Verstärker (Enhancer) der transfizierenden Wirkung (z. B. Chloroquin) sowie4. if necessary, enhancer of the transfecting Effect (e.g. chloroquine) as well
- 5. gegebenenfalls Elektrolyt (insbesondere CaCl2)5. optionally electrolyte (in particular CaCl 2 )
gelöst.solved.
Die Lipidpartikel in einer Größe von etwa 10 nm bis zu etwa 30 µm werden mit konventionellen Methoden der Lehrbücher und Literatur hergestellt. Sie bestehen - im Gegensatz zu Liposomen - aus Lipiden, die bei Raumtemperatur fest sind, bzw. Mischungen von bei Raumtemperatur festen Lipiden mit bei Raumtemperatur halbfesten oder flüssigen Lipiden. The lipid particles in a size of about 10 nm up to about 30 microns are using conventional methods of textbooks and Literature produced. They exist - in contrast to liposomes - from lipids that are solid at room temperature, or mixtures of lipids solid at room temperature with at room temperature semi-solid or liquid lipids.
Die Oberfläche dieser Lipidpartikel wird zur Erhöhung der physi kalischen Stabilität sowie der Komplexbildungsfähigkeit mit dem zu transfizierenden Material, mit Modifizierungsmittel, insbe sondere Tensiden oder Polymeren oder Mischungen derselben modifiziert. The surface of these lipid particles is used to increase the physi calic stability and the ability to form complexes with the material to be transfected, with modifiers, esp special surfactants or polymers or mixtures thereof modified.
Diese Lipidpartikel werden dann in einem geeigneten Verhältnis mit dem zu transfizierendem Material (z. B. Polynukleotide) gemischt, eine Zeitlang zur Ausbildung des Komplexes inkubiert und dann den Zellen zur Transfektion zugemischt. These lipid particles are then in an appropriate ratio with the material to be transfected (e.g. polynucleotides) mixed, incubated for a while to form the complex and then mixed into the cells for transfection.
Die Transfektionseffizienz kann durch Zumischung von sogenannten Enhancern verstärkt werden. Hierfür sind Arzneistoffe wie Chloro quin (CQ) oder Substanzen geeignet, die stark mit Membranen interagieren, z. B. Poly-L-Lysin.The transfection efficiency can be increased by adding so-called Enhancers. Medicines such as chloro are used for this quin (CQ) or substances that are strong with membranes interact, e.g. B. Poly-L-Lysine.
The tendency to form complexes via an aggregation mechanism can be slight in some mixtures of surface-modified lipid particles with material to be transfected, in particular in lipid particles with a low positive charge (<+20 mV) or negatively charged particles. Here, the aggregation can be promoted by adding electrolytes such as 1: 1 electrolytes (e.g. NaCl), 1: 2 electrolytes (e.g. CaCl 2 ) or 1: 3 electrolytes, the complex-promoting effect of the electrolytes being mentioned in the above Order increases.
The complex formation leads to condensation of the polynucleo tide. The condensation is necessary so that the success is true Probability of a transfection increases. The extent of Condensation in the complex can be tracked via particle size measurement become. Is the condensation insufficient or is for higher condensation may be required for certain purposes this also in the manner described above by Elek trolyte addition can be achieved.
Optional wird Komplex daher durch lysosomotrope oder endosomoly tische Substanzen und Elektrolyt ergänzt.Complex is therefore optional through lysosomotropic or endosomoly table substances and electrolyte added.
Lipide sind natürliche Bausteine des Körpers, somit ist die Toxizität der Lipidpartikel um ein mehrfaches geringer als die bisher eingesetzten Transfektionspromotoren - sowohl partikuläre als auch molekulare. Insbesondere trifft dies zu wenn Lipide aus Glyceriden von im Organismus bzw. der Zielzelle vorkommenden Fettsäuren verwendet werden.Lipids are natural building blocks of the body, so that is Toxicity of the lipid particles several times less than that Transfection promoters used to date - both particulate as well as molecular. This is especially true when lipids are out Glycerides from those occurring in the organism or the target cell Fatty acids are used.
Im Gegensatz zu Liposomen zeigen die erfindungsgemäßen Lipid partikel eine hohe physikalische Stabilität. Sie können sogar bei Temperaturen von etwa 121°C ohne Stabilitätsbeeinträchtigung durch Autoklavieren sterilisiert werden. In contrast to liposomes, the lipid according to the invention shows particles high physical stability. You can even go to Temperatures of around 121 ° C without impairing stability can be sterilized by autoclaving.
Die Transfektionseffizienz ist bei geeigneter Auswahl des Komplexes den bisherigen Systemen in der Regel überlegen, oder zumindest gleichwertig wobei TransoPlex gleichzeitig jedoch die oben erwähnten Vorteile aufzuweisen hat.The transfection efficiency is with a suitable selection of the Complex complexes generally superior to previous systems, or at least equivalent, but at the same time TransoPlex has the advantages mentioned above.
Der resultierende Transfektionskomplex zeichnet sich im Vergleich zu bisherigen Gentransfersystemen durch gute Transfektions effizienz bei gleichzeitig reduzierter Toxizität und hoher physikalischer Stabilität sowie der Möglichkeit zur Inkorporation von Hilfsstoffen aus.The resulting transfection complex stands out in comparison to previous gene transfer systems through good transfection efficiency with reduced toxicity and high physical stability and the possibility of incorporation from auxiliaries.

The lipid particles are produced by common methods. In the course of cold production, lipid particles - especially in the size in the lower micrometer range - can be produced by liquid grinding (colloid mill) or by gas jet grinding. During hot production, the lipid is melted, dispersed in an aqueous solution of the surface modifier (e.g. Tween 80 ) and converted into a fine-particle emulsion by high-speed stirring, for example with a propeller stirrer, blade stirrer, a toothed disk or a rotor-stator disperser.
Oberflächenmodifizierer können auch alternativ im geschmolzenen Lipid gelöst oder dispergiert werden. Beim Abkühlen kristalli siert das Lipid teilweise oder vollständig und es entstehen die für den Komplex notwendigen Lipidpartikel. Zur Herstellung sehr feiner Partikel im Nanometerbereich kann auch Hochdruckhomogeni sation eingesetzt werden, sowohl zur Heißzerkleinerung als auch Kaltzerkleinerung.Alternatively, surface modifiers can be melted Lipid can be dissolved or dispersed. Crystalline on cooling the lipid partially or completely and the result lipid particles necessary for the complex. Very much to manufacture Fine particles in the nanometer range can also be used for high-pressure homogeneity tion are used, both for hot grinding and Cold crushing.
Als Matrixmaterial können Lipide im weitesten Sinne eingesetzt werden, das heißt sämtliche lipophilen Substanzen einschließlich Wachse. Bevorzugt werden Lipide aus körpereigenen/zelleigenen Substanzen eingesetzt, wie beispielsweise Glyceride mit körper eigenen Fettsäuren.Lipids can be used in the broadest sense as matrix material be, that means all lipophilic substances including Waxes. Lipids from the body's own / cell's own are preferred Substances used, such as body glycerides own fatty acids.
A variety of different lipids can be used to make the Lipid particles are used. These are both chemical uniform lipids as well as their mixtures. When used Lipid mixtures can also be liquid lipids (e.g. oils, lipophilic Hydrocarbons, lipophilic organic liquids such as Oleyl alcohol) the solid lipids (e.g. glycerides, lipophilic Hydrocarbons such as hard paraffin) are added (so-called "lipid blends").
B. Cetylpalmitat und weißes Wachs (DAB).For example, the following lipids are used as the dispersed phase. They can be used as individual components or as a mixture. Suitable are: natural or synthetic triglycerides or mixtures thereof, monoglycerides and diglycerides, alone or mixtures thereof or with, for example, triglycerides, self-emulsifying modified lipids, natural and synthetic waxes, fatty alcohols, including their esters and ethers, and in the form of lipid peptides, or any mixtures thereof. Synthetic monoglycerides, diglycerides and triglycerides are particularly suitable as individual substances or as a mixture (e.g. hard fat), Imwitor 900 , triglycerides (e.g. glycerol trilaurate, glycerol myristate, glycerol palmitate, glycerol stearate and glycerol behenate) and waxes such as. Particles produced and the complex produced with them were characterized in terms of particle size and charge (quantified as zeta potential). In Abhängigkeit vom eingesetzten Herstellungsverfahren entstehen Lipidpartikel in unterschiedlichen Größenbereichen. Eingesetzt für die Herstellung des erfindungsgemäßen Transfektionskomplexes TransoPlex werden sowohl Mikrometerpartikel (ca. 1-30 µm) als auch Submikrometerpartikel (< 1 µm).Depending on the manufacturing process used Lipid particles in different size ranges. used for the production of the transfection complex according to the invention TransoPlex are both micrometer particles (approx. 1-30 µm) as also submicron particles (<1 µm).

Zetapotential:
The charge of the lipid particles differs depending on the surface modifiers used. In general, three groups can be distinguished, ie lipid particles in which the zeta potential:
- a) positiv ist, vorzugsweise < 0 mV bis +100 mV, insbesondere +20 mV bis 60 mV,a) is positive, preferably <0 mV to +100 mV, in particular +20 mV to 60 mV,
- b) im Bereich von -20 mV bis +20 mV liegt (d. h. ungeladene bzw. sehr schwach geladene Lipidpartikel),b) is in the range of -20 mV to +20 mV (i.e. uncharged or very weakly charged lipid particles),
- c) negativ ist, vorzugsweise < 0 mV bis -100 mV, insbesonde re -20 mV bis -60 mV.c) is negative, preferably <0 mV to -100 mV, in particular right -20 mV to -60 mV.
Zur Herstellung der Komplexe werden die oberflächenmodifizierten Lipidpartikel mit dem zu transfizierendem Material gemischt, z. B. Polynukleotide wie DNA, rekombinante DNA, insbesondere zirkuläre oder linearisierte Plasmide zur Expression therapeutischer Proteine, RNA, insbesondere mRNA, Polynukleotidanaloga, Anti sensenukleotide, und in der Regel für 15 Minuten inkubiert. To produce the complexes, the surface-modified ones are used Lipid particles mixed with the material to be transfected, e.g. B. Polynucleotides such as DNA, recombinant DNA, especially circular or linearized plasmids for the expression of therapeutic Proteins, RNA, especially mRNA, polynucleotide analogs, anti sensenucleotides, and usually incubated for 15 minutes.
negativen Bereich stärker aggregationsfördernde Elektrolyte in
unterschiedlicher Konzentration eingesetzt werden müssen, d. h.
analog zu a)-c) oben vorzugsweise:
Positive particles often - but not always - show good complexation behavior, it decreases with uncharged particles and is less pronounced with negatively charged lipid particles. The complexation can be promoted by targeted addition of electrolyte or triggered in the case of non-complexing lipid particles. If the complexation is promoted, denser condensed complexes are obtained, generally advantageous for the trans fection. For this, weakly aggregating 1: 1 electrolytes are sufficient. Electrolytes can only initiate complex formation via aggregation, whereby with increasing displacement of the particle charge from the positive over the uncharged area to the negative area, more aggregation-promoting electrolytes must be used in different concentrations, ie preferably analogously to a) -c) above:
- a) 1 : 1 Elektrolyte wie NaCl und KCL, vorzugsweise in isoto nischer Konzentration oder geringer konzentriert,a) 1: 1 electrolytes such as NaCl and KCL, preferably in isoto concentrated or less concentrated,
- b) 1 : 2 Elektrolyte wie CaCl2 oder 2 : 1 Elektrolyte, vorzugsweise in isotonischer Konzentration,b) 1: 2 electrolytes such as CaCl 2 or 2: 1 electrolytes, preferably in isotonic concentration,
- c) 1 : 3 Elektrolyte wie AlCl3 oder 3 : 1 Elektrolyte, vorzugsweise in isotonischer Konzentration.c) 1: 3 electrolytes such as AlCl 3 or 3: 1 electrolytes, preferably in isotonic concentration.
Nach Herstellung des Komplexes mit Elektrolytzusatz kann der Elektrolyt in der Suspension verbleiben oder - falls erforder lich - auch der überschüssige Elektrolyt wieder entfernt werden (z. B. durch. Dialyse), der Aggregationsvorgang zum Komplex ist meistens irreversibel. After preparation of the complex with the addition of electrolyte, the Electrolyte remain in the suspension or - if required Lich - the excess electrolyte can also be removed (e.g. by dialysis), the aggregation process to form a complex mostly irreversible.
Certain drugs can also be used to enhance transfection substances - in particular substances that are suitable to avoid lysosomal degradation of the polynucleotides - like Chloroquine, monensin, weakly basic nitrogen compounds or other weak bases can be added. Alternatively you can Use substances that interact with membranes show and influence their structure and fusion properties, z. As dimethyl sulfoxide (DMSO), azones and membrane destabilizing Peptides such as amino acid sequences from the In fluenzavirus haemagglutinin or the bee venom mellitin.
Die Struktur der gebildeten Komplexe wird in Beispiel 2 und Abb. 2 beschrieben. Primär sind die Komplexe dadurch gekennzeichnet, daß DNA nicht alleinig auf der Oberfläche komplexiert eines Partikels ist sondern sich primär Aggregate bilden, bei denen Lipidpartikel über DNA Stränge verbunden sind. The structure of the complexes formed is described in Example 2 and Fig. 2. The complexes are primarily characterized in that DNA is not solely complexed on the surface of a particle, but rather aggregates are formed in which lipid particles are connected via DNA strands.
The complexes are aggregates, the Aggregate structure consists of strands of transfection material, the lipid particle is linked. The aggregates exist thereby from subunits assembled. Smallest sub units that also have their own complex (aggregate) represent, sometimes consist of only a few lipid particles plus linking transfection material (e.g. DNA strands). It also happens that only one or two lipid particles one Form complex with the material to be transferred, preferably the lipid particles have a size of <500 nm, in particular <1 µm.
The complexes formed can be used both in vitro (e.g. cell culture) and in vivo. When used in vivo, the application can be done in a variety of ways, e.g. B. orally via the gastrointestinal tract (GIT) or parenterally in various ways, from intravenous injection to lymphatic absorption after injection near lymphatic vessels. In order to achieve an improved uptake from certain organs (from the GIT, increasing bioavailability) or into certain organs (e.g. brain, heart, bone marrow, kidney after intravenous injection), the complexes produced can themselves be provided with surface modifiers , This ranges from lectins for improved uptake from the GIT to polymers (Poloxamer 407 for bone market targeting, Poloxamine for blood targeting) and nonionic surfactants (Tween 80 , brain targeting) including antibodies.
Die erfindungsgemäßen Komplexe können daher beispielsweise zusätzliche Stoffe enthalten, insbesondere Lektine, Antikörper oder Adhäsionsfaktoren, die die Bindung und die Aufnahme durch die zu transfizierenden Zellen verbessern. The complexes according to the invention can therefore, for example contain additional substances, especially lectins, antibodies or adhesion factors that affect binding and uptake improve the cells to be transfected.
Ferner können die erfindungsgemäßen Komplexe zusätzlich Stoffe, insbesondere Peptide, enthalten, die die Fähigkeit haben, eine Translokation der transfizierten DNA in den Zellkern zu induzie ren, wobei diese zusätzlichen Stoffe an den Lipidpartikel oder die DNA gekoppelt sind.Furthermore, the complexes according to the invention can additionally contain substances especially peptides, which have the ability to To induce translocation of the transfected DNA into the cell nucleus ren, these additional substances on the lipid particles or the DNA is coupled.

Zur Herstellung des Komplexes wurde die Lipidpartikeldispersion mit pCMVβ Plasmid in HEPES-Puffer (pH 7,4) gemischt, in der Endmischung betrug die Konzentration an Lipid 600 µg/ml, die an Plasmid 10 µg/ml.The lipid compritol was melted in water with 4% of a surfactant mixture of Tween 80 / Span 85 (ratio 7: 3) and 1% EQ1 (N, N-Di (β-stearoylethyl) -N, N-dimethylammonium chloride) by stirring dispersed at the same temperature and then the emulsion obtained (lipid content 4%) at 85 ° C high pressure homogenized (LAB 60 homogenizer, APV Homogenizer GmbH, Lübeck). Homogenization took place in the circulation mode over 30 minutes at 500 bar at the main pressure valve. The PCS diameter of the lipid particles was 101 nm, the associated polydispersity was 0.101, the zeta potential was +42 mV. To prepare the complex, the lipid particle dispersion was mixed with pCMVβ plasmid in HEPES buffer (pH 7.4), the concentration of lipid in the final mixture was 600 μg / ml, that of plasmid was 10 μg / ml.
The structure of the complex can be influenced by the type of production, z. B. lipid particle size, ratio lipid: plasmid, and order, type and concentration of added electrolytes. The complex products from Example 1 were examined using atomic force microscopy (AFM). The complexes were adsorbed on the surface of a silicon wafer and examined with AFM in contact mode with a purple nanoscope (Digital Instruments). The cantilever's spring constant was 34 N / m, the scan speed 0.5 Hz with a resolution of 512 × 512 pixels. The overview ( Fig. 1A) shows the complexes, in addition individual lipid particles with a size of approx. 100 nm can be seen. There are two types of complexes, the most frequently occurring complex type I with a more rounded shape with a size of approx. 300-800 nm and complex type II with a size of <1 µm. The higher-resolution images ( Figs. 1B and 1C) show that the complexes consist of subunits that have fused, Fig. 1C shows the comparison of the original, individual lipid particle to the complex produced. Fig. 1 D shows a snapshot of the complex's formation process. Free DNA strands, DNA strands already filled with lipid particles and free SLN are in the process of fusing into a large complex (aggregate in the lower left half).
Transfektionskomplexe wurden durch Mischen von Lipidpartikeln und pCMVβ Plasmid in 200 µl HEPES-Puffer (pH 7,4) bei einer Plasmid konzentration von 20 µg/ml analog Beispiel 1 hergestellt.Transfection complexes were made by mixing lipid particles and pCMVβ plasmid in 200 ul HEPES buffer (pH 7.4) with a plasmid concentration of 20 µg / ml prepared analogously to Example 1.
Oberhalb eines Lipidpartikel verhältnisses von 50, 100% der DNA war zu höheren Molekularge wichten verschoben bzw. komplett immobilisiert (Abb. 2) Different ratios of lipid particles: pCMVβ DNA (from 200 to 5) were mixed and one-dimensional gel electrophoresis on agarose gel was carried out with these mixtures (2 hours at 5 V / cm). The gel obtained was examined with a UV transilluminator and the Geldoc2000 gel documentation system (Biorad, Munich, Germany), the bands and background correction were integrated using the Molecular Analyst Version 1.1 software (Bioread). Binding of the DNA in a complex leads to a change (shift) in the migration in the gel towards higher molecular weights or DNA is immobilized in such a way that it remains in the gel at the application site (well). Above a lipid particle ratio of 50, 100% of the DNA was shifted to higher molecular weights or completely immobilized ( Fig. 2)
Increasing ratios of lipid particles to pCMVβ DNA (from 10:90 to 90:10) were mixed to give the invention To produce complexes. DNA was in 50 mM Hepes buffer (pH 7.4) diluted to a concentration of 10 µg / ml, then with mixed different amounts of particle dispersion and make up to 2 ml refilled. After 3 minutes of incubation, the complex was formed Add 1 ml EB solution (800 ng / ml) to half the sample, 1 ml Water to the other half (background measurement) and the fluo Resence at λ = 366 nm (desiccation) and λ = 590 nm (emission) in measured on an F-2000 fluorescence spectrophotometer.
Freie (zugängliche) DNA interkaliert mit EB, das Fluoreszenz signal von interkalierten EB wird zur Quantifizierung genutzt, d. h. je geringer das Signal um so mehr ist EB ausgeschlossen und DNA gebunden (Abb. 3).Free (accessible) DNA intercalates with EB, the fluorescence signal from intercalated EB is used for quantification, ie the lower the signal, the more EB is excluded and DNA bound ( Fig. 3).
The transfection efficiency was investigated on African Green Monkey Kidney fibroblast-like Cos-1 cells. Complexes were prepared by mixing as described in Example 1, the lipid particle concentrations varied, the final concentration of plasmid in the mixture was 20 μg / ml. The complexes were diluted 1: 1 with buffer or double-concentrated cell culture medium (Dulbeco's modified eagle's - DME), and the monolayers of the Cos-1 cells were incubated in the well of the microtiter plates with 100 μl of this mixture for 4 hours at 37 ° C. The cells were washed with buffer and cultured for a further 48 hours in DME with the addition of 10% fetal calf serum (FCS). The pCMVβ plasmid causes β-galactosidase expression. The transfection efficiency was therefore subsequently determined via the activity of the β-galactosidase using the Luminescence Galacto-Star assay system (PE Biosystems, CCC). The cells were washed with buffer, lysed and incubated with 100 μl reaction solution over 90 minutes at room temperature in order to achieve maximum luminescence signals. The luminescence was determined using a Wallac beta (PE Biosystems) over an integration time of one second. Purified recombinant β-galactosidase was used as the standard. The transfection efficiency was maximal at a ratio of approx. 40-60 ( Fig. 4).
Die Herstellung und Testung erfolgte wie in Beispiel 5 beschrie ben, zusätzlich enthielt das Komplexsystem 100 µM Chloroquin. Das Komplexsystem aus Lipidpartikeln, Plasmid und Chloroquin hat eine weit höhere Transfektionseffizienz, maximale Transfektion tritt bereits beim kleinsten Gewichtsverhältnis von 10 auf (Abb. 4).The preparation and testing was carried out as described in Example 5, additionally the complex system contained 100 μM chloroquine. The complex system of lipid particles, plasmid and chloroquine has a far higher transfection efficiency, maximum transfection occurs even at the lowest weight ratio of 10 ( Fig. 4).
To determine the membrane damage, the Cos-1 cells were incubated with increasing concentrations of the transfection agent for 4 hours and then, after washing with buffer, examined directly with the LDH assay (lactate dehydrogenase) (Boehringer Mannheim, Mannheim Germany). With increasing membrane damage, the LDH release increases. The transfection agents lipid particles from Example 1 were examined in comparison to the conventional agent polyethyleneimine (pEI) (molecular weight 60 kDa) and to poly-L-lysine. The ED50 in the LDH assay was 3000 µg / ml for the lipid particles, but 20 µg / ml for pEI (ie pEI has approximately 150 times higher membrane toxicity) and 10 µg / ml for poly-L-lysine.
The Cos-1 cells were with increasing concentrations of Transfection agent incubated for 4 hours, washed and - in Contrary to example 7 - not examined directly but after washing with buffer for a further 48 hours in DME medium Addition of 10% FCS cultivated. To determine cell viability the WST-1 assay was used (Boehringer Mannheim, Mannheim Germany), an improved form of the MTT test. That was determined Dose that causes 50% of the cells to die (LD50).
Untersucht wurden die Transfektionsagentien Lipidpartikel aus Beispiel 1 im Vergleich zu dem konventionellen Agens Poly ethylenimin (pEI) sowie zu Poly-L-Lysin als Standard. Die LD50 war 600 µg/ml bei den Lipidpartikeln, jedoch 20 µg/ml bei pEI und 10 µg/ml bei Poly-L-Lysin.The transfection agents lipid particles were examined Example 1 compared to the conventional agent poly ethyleneimine (pEI) and poly-L-lysine as standard. The LD50 was 600 µg / ml for the lipid particles, but 20 µg / ml for pEI and 10 µg / ml for poly-L-lysine.
Lipidpartikel wurden wie in Beispiel 1 hergestellt, jedoch aus 4% festem Paraffin als Lipid. Die Mebranschädigung wurde analog Beispiel 7 bestimmt: ED50 < 5000 µg/ml (pEI 20 µg/ml), die Viabilität analog Beispiel 8 bestimmt: LD50 700 µg/ml (pEI 20 µg/ml).Lipid particles were prepared as in Example 1, but from 4% solid paraffin as lipid. The membrane damage became analogous Example 7 determines: ED50 <5000 ug / ml (pEI 20 ug / ml), the Viability determined analogously to Example 8: LD50 700 µg / ml (pEI 20 µg / ml).
Lipid particles were produced analogously to Example 1, as lipid the behenic acid-glyceride mixture Compritol was used (company Gattefossé, France), as additional cationic surfactant 1% Cetylpyridinium chloride (CPC). The PCS diameter was 105 nm, the polydispersity index 0.112 and the zeta potential +40 mV (i.e. identical physical characteristics to the lipid particles from example 1). Simply mix these lipid particles with HEPES buffer without complex-promoting electrolyte addition led to no complex formation, in the EMSA no "mobility shift" detected.
Die in Beispiel 1 hergestellten Partikel wurden durch Autoklavie ren bei 121°C für 15 Minuten sterilisiert. Der PCS-Durchmesser vor der Sterilisation betrug 103 nm, der Polydispersitätsindex 0,110, die Werte nach der Sterilisation 101 nm und 0,101.The particles produced in Example 1 were autoclaved sterilized at 121 ° C for 15 minutes. The PCS diameter before sterilization was 103 nm, the polydispersity index 0.110, the values after sterilization 101 nm and 0.101.
