Abstract
Die Erfindung betrifft ein Verfahren zur Gewinnung von alkalischer Phosphatase aus Kaelberdarm-Mukosa oder -Inhalt. Dabei wird die wasserloesliche Enzymform durch eine waeszrige 2-Phasenverteilung vom Hauptanteil der Fremdproteine und noch vorhandener Zelltruemmer getrennt und das Enzym nach einer Ionenaustausch-Chromatographie an DEAE-Cellulose durch Farbstoffligand-Affinitaetschromatographie gereinigt. Das mit einer hohen Ausbeute gewonnene Enzym besitzt die maximal erreichbare spezifische Aktivitaet und enthaelt keine stoerenden DNA-Endonukleasen und DNA-Nicking Aktivitaeten. Überraschenderweise ermöglicht das erfindungsgemrße 2-Phasensystem, bestellend aus 8-10% (w/w) PEG 4000 bzw. PEG 6000,0,5-2% (w/w) Dextran (M, = 30000-70000), Puffer (vorzugsweise 1OmM Tris/HCI, pH 7,5,2 mM MgCI2) und bis zu 75% (w/w) Butanolextrakt (50001E bzw. 1 g Protein pro 100g System), die Entfernung partikulärer Zellbestandteile und eines großen Teils der Fremdproteine in einem Schritt ohne Zentrifugation. Allerdings kann durch den Einsatz geringer Zentrifugalkräfte (3000xg) die Phasentrennung in einer Minute erfolgen. Günstig ist auch der Einsatz von Flüssig/Flüssig-Separatoren, er erlaubt eine kontinuierliche Prozeßgestaltung.
