Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Fremdproteinen in tierischen Zellen. Anwendungsgebiet der Erfindung ist die biotechnologische Produktion von Wirkstoffen und Vakzinen. Es werden Helferzellen fuer die Erreichung einer maximalen Genkopiezahl sowie zur effektiven Expression heterologer Promotoren mit einer prokaryotischen Polymerase hergestellt. Dieses doppelte Helferzellsystem mit einem UEberschusz an RNA-Polymerase und regulierbarer Kopiezahl des Fremdgens unter Kontrolle eines prokaryotischen Promoters gestattet die maximale Synthese eines Fremdgenprodukts in permanenten Zellen. Die Erfindung ist dadurch gekennzeichnet, dasz die prokaryotische Polymerase im Zellkern der Wirtszelle wahlweise durch Induktion der Vektorreplikation mit Genkopien gesaettigt wird und durch Ruecknahme der Induktion eine Erholung der Zellen gewaehrleistet ist. Die Verwendung tierischer Zellinien ist in dor biotechnologischen Wirkstoff- und Vakzineproduktion seit vielen Jahren üblich. Durch Einführung der entsprechenden Gene läßt sich der Ertrag gegenüber natürlicherweise produzierten Zellinien häufig, aber nicht immer, wesentlich steigern. Die Effektivität der Produktbildung durch fremde Gene wird durch drei Parameter bestimmt: (1) die Transkription (mRNA-Synthese), (2) die Translation (Proteinsynthese) und (3) den Proteintransport. Während der Transport zum größten Teil von der Struktur des Proteins selbst abhängig ist und damit kaum beeinflußbar zu sein scheint, sind die beiden anderen Parameter teilweise von den das Strukturgen flankierenden DNA-Sequeruan, aber auch von der Verfügbarkeit zellulärer Transkriptionsfaktoren abhängig. Der Promotor des Fremdgens konkurriert mit den zellulären Genen um diese Faktoren und um die RNA-Polymerase. Damit wird das Ausmaß der Transkription auf ein Niveau begrenzt, daß nicht wesentlich über dem von effektiv abgelesenen zellulären Genen liegt und z. B. weit hinter der Expression viraler Gene bei einer Virusinfektion zurückbleibt. Dabei wird bevorzugt das Groß-T-Antigen-Gen des Polyomavirus sowie der Promotor des Maus-Mamma-Tumorvirus (MMTV) verwendet. Die resultierende Rekombinante (z.B. pLT2-MMTV2, WP C12N/315016.8) wird zusammen mit einem Selektionsgen, bevorzugt dem Metallothionein-Gen in Form von pMT-l (Glanville et al. [1981] Nature 292,267), in eine beliebige tierische Zellinie transfiziert. Bei Verwendung des Polyoma-Groß-T-Antigens werdon Nagerzellinien (Maus, Ratte, Hamster) eingesetzt. Nach der Transfektion wird auf Anwesenheit des Metallothionein-Gens mit Schwermetallen, bevorzugt mit Cadmiumsulfat, selektiert. In den gewonnenen Zellkolonien wird T-Antigen mittels Immunfluoreszenz nachgewiesen und zwar sowohl in Anwesenheit als auch in Abwesenheit des Induktors für den Promotor, z. B. Dexamethasone (108M) im Falle des MMTV-Promotors. Ein Zellklon mit möglichst großer Differenz zwischen nichtinduziertem und induziertem T-Antigen wird für die weiteren Arbeiten ausgewählt.1. Dabei wird vorzugsweise das Kernlokalisierungssignal des T-Antigens von Virus SV40 in Form einer synthetisch hergestellten DNA-Sequenz und der Promotor des Metallothionein-Gens verwendet. Oie bevorzugt synthetisch hergestellte DNA-Sequenz kodiert die Aminosäuresequenz: Met-Asp-Lys-Val-Phe-Arg-Asn-Se'-Ser-Arg-Thr-Pro-Pro-Lys-Lys-Lys-Arg-Lys-Val-Glu-Arg-Leu-Asp.The RNA polymerase 3 gene, preferably that of phage T7, is previously modified at the N-terminus by adding a nuclear eukaryotic signal and an animal promoter. Nach Selektion mit Hygrcmyzin werden Kolonien isoliert und auf Vorhandensein der modifizierten RNA-Polymerase im Zellkern geprüft, bevorzugt m ttels Immunfluoreszenz und in vitro Transkription mit Kernextrakten. So wird die erfindungsgemäße Expressions; ellinie PMLTT7 N erhalten, die in Kombination mit einem geeigneten Vektor zur Expression von Fremdgenen eingesetzt wird.
