Abstract
Die Erfindung betrifft ein Verfahren zur enzymatischen Herstellung von Indolpyruvat, einer Biochemikalie, die fuer Forschungen auf dem Gebiet des Tryptophanstoffwechsels und des Metabolismus anderer Indolverbindungen verwendet werden kann. Zur enzymatischen Herstellung der erfindungsgemaeszen Verbindung wird ein aufbereiteter Enzymrohextrakt aus der Hefe Candida maltosa (ZIMET HI 28) mit Tryptophanaminotransferase als dem wirksamen Enzym eingesetzt. Das waehrend der enzymatischen Umsetzung von Tryptophan mit 2-Oxoglutarat als Aminogruppenacceptor und Pyridoxalphosphat als Coenzym gebildete Indolpyruvat wird durch Aufarbeitung mit verschiedenen Anreicherungs- und Reinigungsverfahren (Extraktion; Saeulenchromatographie; Umkristallisation) in reiner, kristalliner Form gewonnen.{Indolpyruvat; Biochemikalie; Candida maltosa (ZIMET HI 28); Tryptphanaminotransferase; Isolierung; Umkristallisation}The invention relates to a process for the enzymatic production of indolpyruvate, a biochemical which can be used for research in the field of tryptophan metabolism and the metabolism of other indole compounds. Proben mit einem zu hohen Gehalt an Indolpyruvat (mehr als 30OnMoI) mußten vor Zugabe des Reagenz entsprechend verdünnt werden. Nach 2stündiger Inkubation bei 370C wurde das Reaktionsmedium mit konz. HCI auf pH 2 angesäuert und danach 20 Minuten bei 16000 χ g zentrifugiert. Der Überstand wurde 2mal mit Ethylacetat extrahiert. Die vereinigten Ethylacetat-Extrakte wurden bei 300C eingeengt und der verbleibende Rückstand in 10ml Ethanol ,in diesem und in allen weiteren Schritten wurden Ethanol mit 50% Volumenanteil verwendet) aufgenommen. Diese Ethanol-Lösung wurde auf eine Säule (2x 10cm) von DEAE-Sephadex A-25 (Acetat-Form) gegeben. Nach Waschen der Säule mit 30ml Ethanol wurde mit 200 ml eines Gradienten (0-0,5 M) Essigsäure in Ethanol eluiert. Es wurden Fraktionen von 6,6ml gesammelt. Die r-idktionen, die Indolpyruvat enthielten, wurden vereinigt, und bei 3O0C eingeengt. Der Rückstand wurde in Ethanol aufgenommen, erneut eingeengt, und dieser Schritt wurde ein weiteres Mal wiederholt. Nach Resuspenslon des verbleibenden Rückstandes in Ethanol und langsamer Evaporation der Ethanol-Lösung bildeten sich bräunliche Kristalle.
