Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung monoklonaler Antikoerper gegen spermaspezifische Antigene im Molekulargewichtsbereich um 30 kDa. Anwendungsgebiete der Erfindung sind die Kriminalistik, Gerichtsmedizin und medizinische Diagnostik. Das erfindungsgemaesze Verfahren ist dadurch gekennzeichnet, dasz man zunaechst in an sich bekannter Weise Milzzellen von BALB/c-Maeusen, die mit Spermaplasmaantigenen immunisiert wurden, mit BALB/c-Myelomzellen fusioniert, die antikoerperproduzierenden Hybridome mit einem Enzymimmunoassay selektioniert und erfindungsgemaesz zur Produktion der monoklonalen Antikoerper durch Kultivierung oder Transplantation einsetzt. BALB/c-Mäuse wurden mit dem durch Ammoniumsulfatfällung aus geporitem Spermaplasma hergestellten Antigonpräparat immunisiert. Milzzellen dieser Mäuse wurden mit Maus-Myelomzellen der Zellinie X63-Ag'8.653 (Karney, J. F. et al.; J. Immunol. 123,1548,1979) mittels Polyethylenglykol fusioniert (Peters, H. H., et al.: .Monoklonale Antikörper, Herstellung und Charakterisierung", Springer, Berlin, 1985).BALB / c mice were immunized with the antisense preparation prepared by ammonium sulfate precipitation from geporitized seminal plasma. Danach wurde mit PBS-Tween-20 Lösung gewaschen und die Kulturüberstände unverdünnt sowie 1:2 verdünnt zugegeben (Inkubationszeit: 90 Minuten, 250C). Nach erneutem Waschen wurde 90 Minuten bei 25°C mit phosphatasekonjugiertem Anti-Maus-Ig inkubiert. An ein weiteres Waschen schloß sich die Zugabe von 50μΙ Substratlösung (1 mg P-Nitrophenylphosphat/1 ml Diethanolamin-Pufferlösung) zu jedem Napf an.2.
