Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
Processes for the preparation of Stickstofflostverbindungen are described in large numbers and transferred to industrial practice. 4- {5- (bis (2-chloroethyl) amino] -1-methylm H-benzimidazolyl (2)} - butanoic acid hydrochloride hydrate (bendamustine) is the closest related cancer drug to the target products described below: in animal and human Among other things, bendamustin is biotransformed into the corresponding 3-hydroxybutanoic acid derivative (R. Preiss, R. Sohr, M. Matthias, B. Brockmann, H. Muller, Pharmazie 40 (1985) 782, R. Preiss, R. Sohr , H. Matthias, H. Muller, W. Werner, W. Ihn, H. Wolff, in the press.) The enzymatic degradation to the expected derivative of ethanoic acid have not yet been demonstrated.
The preparation of {5- [bis (2-chloroethyl) amino] -1-methylbenzimidazolyl (2)} ethanoic acid has been described by W. Ozegowski et al. Mitarb, starting from a condensation of N'-methyl-4-nitro-o-phenylenediamine with lactic acid and subsequent oxidation with CrO 3 in acetic acid solution to 2-acetyl-1-methyl-5-nitro-benzimidazole tried. However, the redox amidation of the above compound by Willgerodt-Kindler to the acetic acid derivative did not succeed.
Chem. [4] 29 [1965] 18-25).Another experiment described in the literature led to the condensation of o-phenylenediamine with cyanoacetic acid ethyl ester to 2-cyano-methyl-benzimidazole. 1-Methylation and acid ethanolysis of this compound gave ethyl 1-methyl-benzimidazolyl-2-ethanoate, which was nitrated at the 5 and 6 positions. Isomeran separation was followed by reduction of the 5-nitro group to the 5-amino derivative, to which ethylene oxide was added. Although the chlorination of the bis (2-hydroxyethyldimino derivative with SOCl 2 was successful, the acidic hydrolysis of the ester did not give the substituted ethanoic acid, but after decarboxylation only 5- [bis (2-chloroethyl) amino] -1,2-dimethyl benzimidazole (W.Ozegowski, D.Krebs; J. prakt. Chem. [4] 29 [1965] 18-25).
In these methods, it is disadvantageous that the synthesis of derivatives of the abovementioned substance group A starts from the unsubstituted o-phenylenediamine and the benzimidazole derivative later still N-methylated and nitrated, with 5- and 6-nitro-benzimidazole isomers which are subsequently to be separated (only the isomer with the nitro group in position 5 of the benzimidazole ring is of interest). In addition, the target compound eventually decayed by spontaneous decarboxylation.
Ziel der ErfindungObject of the invention
Die Erfindung verfolgt das Ziel, {5[Bis(2-chlorethyl)-amino]-1-methylbenzimidazol-(2)}ethansäurealkylester in effektiverer Weise herzustellen. The invention aims to produce {5 [bis (2-chloroethyl) amino] -1-methylbenzimidazole (2)} ethanoic acid alkyl ester in a more effective manner.
Darlequng des Wesens der ErfindungLoaning the essence of the invention
The invention has for its object to provide a chemical-synthetic process with which {5- (bis (2-chloroethyl) amino] -1-methyl-benzimidazolyl (2)} ethanoic acid in the form of the stable alkyl esters can be produced effectively ,
According to the invention, this object is achieved in that N'-methyl-4-nitro-o-phenylenediamine as the starting material first condensed with cyanoacetic acid ethyl ester to 2-cyanomethyl-1-methyl-5-nitro-benzimidazole (see 1 in Fig.2) and this intermediate in or with alcoholic hydrochloric acid to the respective associated alkyl ester (see, for example, 2 in Fig. 2) is solvolyzed. The nitrogen release group is then synthesized on these esters in a manner known per se by converting the nitro group to the amino group by hydrogenating reduction. 3 in Fig. 2), then ethylene oxide added to this intermediate (compare approximately 4 in Fig. 2) and in the last step by chlorination with thionyl chloride of the respective {5- [bis (2-chloroethyl) amino] -1-methyl benzimidazolyl (2)} ethanoic acid alkyl ester, d. H. the respective target compound A) (compare about 5 in Fig. 2) is obtained. In this way, stable compounds of a per se labile carboxylic acid are obtained. Advantageously, water-soluble hydrochlorides or analogous water-soluble salts can be prepared from the target compounds A) by dissolving si 3 initially in ether or in ethyl acetate or in a lower alcohol and introducing either hydrogen chloride gas in the second substep or an organic or inorganic acid, preferably glacial acetic acid or citric acid or sulfuric acid, added. The thus obtained derivatives of A) could also be administered intravenously.
