Abstract
The conditioning of biologically active substances is a process that is applicable in the microbiological industry. The aim of the invention is the development of a method which allows to make a shape for biologically active thermolabile products with low specific equipment and energy costs. It is the object of the invention to produce biologically active products with performance properties (dust-free, free-flowing, narrow grain spectrum, abrasion-resistant, veined, sealed in accordance with the requirements of health protection). Surprisingly, it was found that under the conditions of the process according to the invention thermally induced activity losses are avoided in the thermolabile biologically active substances considered here. According to the invention, the process for the conditioning of biologically active substances in a fluidization regime is carried out by simultaneous drying and shaping without the addition of binders with subsequent sealing for the purpose of surface inactivation in a process step while maintaining the wetting range.
Description

Anwendungsgebiet ist die mikrobiologische Industrie. The invention relates to the conditioning of biologically active substances, in particular of enzymes. Field of application is the microbiological industry.
Charakteristik des bekannten Standes der TechnikCharacteristic of the known state of the art
For the shaping and sealing of biologically active products, there are numerous technical solutions, which are summarized and discussed below:
Besides the advantage of agglomerate formation at temperatures which are favorable for thermolabile substances, this technical solution has a number of disadvantages:
The limited water binding capacity results in products with low specific activity. In general, the activity ratio between end product and outer material, based on the mass unit, is <0.5.
The agglomerates are not wrapped closed by inert material.
- Als Folge des begrenzten Wasserbindever: iiögens sind sehr große Stoffströme zu verarbeiten, die zur Gewährleistung einer geringen Restfeuchte des Endproduktes häufig einer Konvektionstrocknung zugeführt werden (DE-OS 2042650, SU-UR 872549).- Due to the limited Wasserbindever: Äögens very large streams are processed, which are often supplied to ensure a low residual moisture of the final product convection drying (DE-OS 2042650, SU-UR 872549).
The laying of the mixing process in the fluidized bed according to DD-WP 151598 does not eliminate the principal disadvantages. In the so-called. Prillingverfahren (DE-OS 2060085), the biologically active substance is enclosed in hollow spheres of molten wax. This is based on largely anhydrous biologically active product, which was prepared for example by spray drying. The starting material suspended in the wax melt is sprayed in a spray tower, through which cold air is passed, whereby the atomized melt is obtained in the form of finely dispersed spheres. Disadvantages of this solution are the high cost-intensive wax portion (up to 50%), the need for thermal stabilization of the biologically active product and, resulting from both issues, the low specific activity of the final product. The ι alisierte activity ratio is well below 1. Finally, the biologically active substance before prilling in a finely dispersed dry product to be transferred. The goal of high specific activities in the end product can only be achieved by expensive purification steps of the biologically active product.
B. GB-PS 1 362 365, DE-AS 2137 043). Das erreichbare Aktivitätsverhältnis liegt bei 0,1 bis 0,6.Another technical solution is based on the extrusion of a mixture of filler, binder, active component and water, which is subsequently spheronized in a special apparatus for the "oarumerizer" formulation. The particles obtained are largely dust-free and have a narrow grain spectrum. Disadvantages are the high proportion of binder and filler and in particular the technically complex apparatus combination mixer-kneader-extruder-Marumerizer R dryer. Additions of stabilizers are necessary (see eg GB-PS 1 362 365, DE-AS 2137 043). The achievable activity ratio is 0.1 to 0.6.
Siehe hierzu:For the microencapsulation of biologically active substances, numerous proposed solutions are known and accessible in the specialist literature. See also:

B. DE-OS 2043110, DE-OS 2042650, DE-OS 2111411).The emulsion is prepared to stabilize the phase boundary with a film-forming polymer dissolved in the continuous phase. Waxes or synthetic resins dissolved in organic solvents are also widely used as wall material (eg DE-OS 2043110, DE-OS 2042650, DE-OS 2111411).

The problem with biologically active substances proves to be compliance with the necessary conditions for the capsule reaction conditions, which is only possible with the addition of adjuvants for stabilizing the active component. If the capsules contain the biologically active product in liquid form, a complex drying stage is required.
Der Anteil des Wandmaterials ist insgesamt hoch (DE-OS 2042650).Other disadvantages are the cost-intensive capsule material and the complex measurement and control technology that is required to comply with the reaction conditions. The proportion of wall material is high overall (DE-OS 2042650).
A further development of conventional methods for grain enlargement is the method of NOVO-Industri A / S (CH-AS 632788) for the production of enzyme granules. The teaching of the invention provides for the addition of fibrous cellulose with special properties, a binder and a Granulierhilfsmittels that in the entirety of their effects lead to a controllable grain growth in known granulating devices such as pelletizing drums or granulating plates.
The disadvantage of this solution is due to the high use of fibrous cellulose and the use of binders and Granulierhilfsmitteln. The final product is dried by a fluidized bed dryer and wrapped by molten waxes. The technical expenses for the multi-stage process are high.

Ensuring a closed envelope according to the requirements of health protection. The necessary addition of high filler proportions requires a highly active starting material, preferably in the form of dry product.
