Abstract
Die Erfindung betrifft ein Verfahren zur Milchsaeuregaerung mittels immobilisierter Mikroorganismen und dient der Durchfuehrung von Hochleistungsgaerungen, bei denen hohe Milchsaeurekonzentrationen in kurzen Gaerzeiten erreicht werden. Dies gelingt erfindungsgemaesz dadurch, dasz ein Gelimmobilisat eingesetzt wird, das nach seiner Herstellung in Abhaengigkeit vom Bakterienstamm mit mindestens (0,2 bis 1,0)1010 viablen Zellen/ml Gel und nach der sich anschlieszenden Aktivierung mit mindestens (1,0 bis 5,0)1010 viablen Zellen/ml Gel beladen ist. (Biotechnol. Bioeng. 30 (1987) 788-793) beträgt bei einem Gelanteil am Arbeitsvolumen von 0,5 die Gärzeit für die Bildung von 62-73g Milchsäure/l B-8d. Die spezifische Produktivität der bei den o.g. Laborverfahren eingesetzten Immobilisate liegt überwiegend unter 10g Milchsäure/l Gel · h, wobei der Höchstwert für die Beladung mit viablen Zellen unmittelbar nach der Immobilisierung 9 · 107/g Gel betrug und nach 20tägiger intermittierender Präfermentation 1,2 · 10*/g Gel (P. Linko et al.: Ann. New York Acad. Sei. 434 [1984] 406-417). B. in Rührreaktoren, eingesetzt werden kann. Das erfindungsgemäße Verfahren kann in Reaktoren bekannter Konstruktion und Betriebsweise durchgeführt werden. Beim wiederholten Einsatz eines Lactobacillus spec./Ca-PektaMmmobilisats zur Milchsäuregärung unter Verwendung eines nährund wirkstoffreichen Mediums in einem Ruhrreaktor kann bereits bei einem Immobilisatanteil am Arbeitsvolumen von 0,15 bei 95%igem Zuckerumsatz Milchsäure in einer Konzentration von 80g/l in Gärzeiten von 8 bis 20 h erzeugt werden. In Reaktoren mit höherem Immobilisatanteil und bei Verwendung von Streptococcus spec, sind noch kürzere Gärzsiten möglich.
