Abstract
The invention relates to a process for the preparation of an orotropically active drug which can be administered orally and has improved bioavailability based on orotic acid. According to the invention, the object is achieved by producing orotic acid esters in a manner known per se by the esterification of orotic acid with aliphatic alcohols having a carbon number of 1 to 10, in particular 2 to 6, the chains being unbranched, branched or closed to form a ring and processed with the usual formulation auxiliaries and with the addition of polyvinyl alcohols to accelerate the absorption to pharmaceutical preparations that are suitable for oral administration.

The invention relates to a method for producing an orotropically active nootropic drug having improved bioavailability on the basis of orotic acid for use in human medicine.
Pharmazie 21* 309 (1979))·Orotic acid is only absorbed to a very limited extent when administered orally alloin or in the presence of conventional flavoring auxiliaries, such as lactose, sugar, starch, gelatin, ultra-amylopectin, talc, calcium stearate, etc. Due to their hydrophilicity, the absorption rate of orotic acid is 5 to 6 %. (Hinkel, G, K. and Ie Petit, G. Pediatrics 12, 63 (1973) and Walther, H., Köhler, E. and Meyer, PP Pharmacy 21 * 309 (1979))
Methylglucaminorotat (DD 116 036), 2-Aminoethanthiolorotat (KLosa, J. Arzneimittelfo"-8chung 26, 1532 (1976), Chinin- und Diisopropylorotat (Japan 16 E 461), Piperazin-, Piperidin-, Morpholin- undFor this reason a number of salts of orotic acid have been prepared, e.g. Methylglucaminorotat (DD 116 036), 2-Aminoethanthiolorotat (KLosa, J. Soweit pharmakokinetische Daten vorliegen, läßt sich ;jedooh einschätzen, daß durch die Salzbildung mit den genannten basischen Komponenten allein oder auch bei Einsatz von Formulierungshilfsstoffen keine nennenswerte Verbesserung der Pharmakokinetik der Orotsäure, insbesondere der Resorption bewirkt wird (ζ·Β· Walther, H·, Köhler, E· und Meyer, F.P·, Pharmazie 2i> 309 (1979) und Zbl· Pharmaz· Pharmakotherap· Laboratoriumsdiagnostik 1J6,, 399 (1976))·Insofar as pharmacokinetic data are available, it can be estimated that salt formation with the basic components mentioned alone or even with the use of formulation auxiliaries does not bring about a significant improvement in the pharmacokinetics of orotic acid, in particular absorption (ζ · Β · Walther, H ·, Köhler, E. and Meyer, FP., Pharmazie 2i> 309 (1979) and Zbl.
