Description
The invention relates to a process for the microwave production of glucose-isomerase-containing enzyme preparations, a product which is used to prepare fructose-containing solutions of D-glucose. These fructose syrups find application in the food and beverage industry due to nutritional physiological benefits. The field of application of the invention is thus in the technical-microbiological research and industry.
US 3645848; US 834988; DE OS 2400323; US 4315103; US 3813318; US 4532208; Brit. Pat. 1411763,1411764,1411765; US 4511430; US 4551430; US 4355103).The microbial production of glucose-isomerase or glucose-isomerase-containing enzyme preparations has been known since the end of the 1950s. The enzymatically active biomass is obtained by fermentation with the aid of high-performance processes whose economic efficiency is based on two process-specific features: On the one hand, the use of inexpensive raw materials, such as xylan-containing materials (British Patent 1410569) or D. Sorbitol (Biotechnol. Lett., 7, 1985, 597) instead of D-xylose, which are necessary as an inducer of enzyme formation, on the other bacterial strains of the genera Bacillus, Actinoplanes, Arthrobacter or Streptomyces are used, the D-xylose for the induction of glucose Do not require isomerase formation (US 3645848, US 834988, DE 2400323, US 4315103, US 3813318, US 4532208, British Patent 1411763, 1411764, 1411765, US 4511430, US 4551430, US 4355103).
Processes for the preparation of glucose-isomerase-containing enzyme preparations in the GDR have also been proposed (DD 244145, DD 241427), in which microorganisms of the genera Chainia or Streptomyces are used. Among them are strains of the species Streptomyces chrysomallus (ZIMET 43774, ZIMET 43775), which were selected from an antibiotic-producing wild strain as antibiotic inactive mutants (ZIMET 43774.43775). Co ++ -free nutrient media are used in this process which, in addition to glucose or starch as common C sources, contain D-xylose as well as Mg salts as essential components for the induction of glucose isomerase synthesis and enzyme stability.
The disadvantage of the specified method is that either constitutively constituting glucose isomerase-forming microorganisms are used, which are demanding in terms of organic N-substrates (meat protein) and Mg salts as exclusive enzyme stabilizing additives in the fermentation process or in the case of non-mycelial growth an elaborate separation technology for recovering the biomass require that when using cost-effective raw materials of the use of xylan hydrolyzing strains or enzymes or in mineral hydrolysis, the upstream special cleaning procedures with respect to the D-xylose are necessary, or that performance degradation occurs by increased ion contamination of the media.
The invention aims to produce glucose isomerase-containing enzyme preparations by an improved process.
Die Fermentationsbedingungen werden durch folgende Parameter charakterisiert:The invention has for its object to describe a microbiological process by means of which - using inexpensive starting materials and based on the use of new parent material - glucose-isomerase-containing enzyme preparations can be prepared with readily available nutrient media in good yields. According to the invention, the object is achieved in that microorganisms of the species Streptomyces chrysomallus, which were obtained by mutagenesis from the strain Streptomyces chrysomallus PL45, preferably the strain Streptomyces chrysomallus XK10, deposited in the depository for microorganisms of the Central Institute of Microbiology and Experimental Therapy AdW of the GDR , Jena, Beutenbergstrasse 11, under the registration number ZIMET 43808, or the strain Streptomyces chrysomallus XK12, deposited in the above-mentioned depository under the registration number ZIMET 43809, and a sterile and aerobic fermentation for a period of 24 hours to 48 hours, preferably 24 Hours to 36 hours using a medium containing glucose or glucose polymers as exclusive or predominantly C-substrate relative to D-xylose, with D-xylose added as a pure substance or as a neutralized mineral corn syrup hydrolyzate n becomes. In addition, organic and inorganic N-type substrates and mineral salts are added to the medium using magnesium salts and / or Na 2 SO 4 . The fermentation conditions are characterized by the following parameters:
Stirring: 120 to 800rpm; Aeration rate (v / v): 0.5: 1 to 1: 1; Temperature: 25 to 35 ° C .; Acidity of the medium: pH 5.5 to 8.5. The separation of the mycelium and the recovery of the glucose isomerase enzyme preparation ^ takes place in a conventional manner.
The advantages of this approach are that the production of glucose isomerase-containing biomass succeeds by fermentation by the use of the above Streptomyces strains on complex nutrient media, without or only to a small extent the cost-intensive inductor D-xylose and various mineral salts to use as enzyme stabilizers. The Streptomyces strains used have the beneficial property of either synthesizing glucose isomerase in glucose-free culture media in an economically acceptable amount or, in the presence of low levels of D-xylose in the medium, significantly higher volume-related performances at specific enzyme activities of at least 1.2 μg / mg mycelial protein to reach.

The corn husk hydrolyzate prepared according to Example 5 is separately sterilized after neutralization with NaOH to pH 6.5 in 25 ml portions in 100 ml bottles by autoclaving 1) at 120 ° C. for 30 minutes, or 2) Addition of 0, 0025% peracetic acid (Wofasteril-36% peracetic acid) takes place. These fractions are added separately to each 25 ml of main culture medium, which is presented in 50% of the total volume separately in the 500 ml bottles. The resulting 50 ml main culture media are mixed with 10 ml 48 hrs. Seed culture inoculated and 43h. Shaken at 27 0 C on a rotary table.
