Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Arzneimitteln, insbesondere die Herstellung von Gastropharm-Produkten zur Verwendung in der pharmazeutischen Industrie. Ziel und Aufgabe der Erfindung ist es, ein Gastropharmprodukt mit besseren Gebrauchswerteigenschaften herzustellen. Dabei soll in einem kontinuierlichen Prozess bei entsprechender Reinheit des Produktes eine ueber einen laengeren Zeitraum dosierte Abgabe von Wirkstoffen erreicht werden. Gleichzeitig soll die Rieselfaehigkeit und Lagerstabilitaet verbessert sowie der Einsatz an Energie und Transportkosten gesenkt werden. Erfindungsgemaess wird dieses dadurch erreicht, dass Gastropharmtrockenmasse mit Wasser und Sojaoel angemaischt und Kristallzucker zugegeben wird, so dass sich eine verzuckerte Gastropharmloesung ergibt. Diese Loesung wird durch Verspruehen mit gleichzeitiger Zugabe von arteigenem agglomerisierbarem Produkt auf fluidisierte Granulate, bei gleichzeitiger Granulationstrocknung und Agglomerisation, in eine Wirbelschicht mit nachfolgender Klassierung aufgegeben. Die Herstellungsbedingungen sind im Ausfuehrungsbeispiel dargestellt.The invention relates to a process for the preparation of medicaments, in particular the production of gastro-pulp products for use in the pharmaceutical industry. Gastropharmproduktes aus verzuckerter Gastropharmlösung zu entwickeln, bei dem das Produkt in einem kontinuierlichen Prozeß mit wahlweise einstellbarem Korngrößenspektrum hergestellt wird. Dabei soll außerdem bei entsprechender Reinheit des Produktes eine kontinuierliche, über einen längeren Zeitraum dosierte Abgabe von Wirkstoffen erreicht werden. Erfindungsgemäß wurde die Aufgabe dadurch gelöst, daß Gastropharmtrockenmasse mit Wasser und Sojaöl angemaischt und Kristallzucker zugegeben wird, so daß sich eine verzuckerte Gastropharmlösung ergibt. Diese Lösung wird durch Versprühen mit gleichzeitiger Zugabe von arteigenem agglomerierbarem Produkt auf fluidisierte Granulate, bei gleichzeitiger Granulationstrocknung und Agglomerisation, in eine Wirbelschicht mit nachfolgender Klassierung aufgegeben. Die verzuckerte Gastropharmlösung wird mit einem Trockensubstanzanteil von 5-30% kontinuierlich und gleichmäßig auf die Wirbelschicht eingedüst. Die Agglomerate verbleiben bei einer Wirbelschichttemperatur 370 K maximal 50 min in der Wirbelschicht und verlassen über einen klassierenden Abzug mit einer Temperatur von 340 K bis 370 K, vorzugsweise 343 K, die Wirbelschicht. Der mittlere Korndurchmesser beträgt dabei 2 bis 6mm mit einem Feuchtigkeitsgehalt von 5 bis 10Ma.-%. Der Mengenstrom von 30kg/h wird in zwei Produkströme aufgeteilt, wobei 10kg/h als Endprodukt mit einem Wassergehalt von 7,5 Ma.-% und einem Durchmesser von 3 mm zur Lagerung gefördert werden. Der restliche Produktstrom von 20kg/h wird in einer Zerkleinerungsmaschine auf einen Korndurchmesser von 0,5 bis 1,5mm zerkleinert und zur Aufrechterhaltung des Granulierprozesses über eine Zeil en radschleuse der Wirbelschicht zugeführt. Das aus der Zyklonabscheidung anfallende staubförmige Fertigprodukt wird der Wirbelschicht wieder zugeführt. Durch die erreichte Kugelform des Präparates entsteht ein Produkt mit der geringsten möglichen Oberfläche. Die langsame Aufgranulierung läßt außerdem keine Lufteinschlüsse zu.
