Agents that relax vascular smooth muscle may be used for the treatment of arterial hypertension, as such patients suffer from increased peripheral resistance to blood flow. Compounds that interfere with vascular smooth muscle activity have been used clinically for several years. However, their usefulness has often been limited due to inadequate efficacy and adverse effects. Side effects (outside the cardiovascular system) have often been associated with properties of the agent that are not relevant to the smooth muscle relaxing effect. Sometimes the vasodilating agents also had a negative effect on the contractility of the heart.
It appears that the development of special smooth muscle relaxants that have no adverse effects can give a therapeutic benefit in arterial hypertension and for the treatment of ischemic heart disease as well as the acute failing heart. In addition, such agents may also be useful in the treatment of other symptoms with excessive activation of smooth intestinal musculature.

In the preparation of pharmaceutical compositions containing a compound of the invention in the form of dosage units for oral administration, the selected compound may be combined with a solid, powdered carrier material such as lactose, sucrose, sorbitol, mannitol, starch such as potato starch, Corn starch or amylopectin, cellulose derivatives or gelatin and with an anti-friction agent, such as magnesium stearate, calcium stearate, Polyathylenglykolwachsen or the like are mixed and pressed into tablets. When coated tablets are desired, the core prepared as above may be coated with concentrated sugar solution, and this solution may contain, for example, gum arabic, gelatin, talc, titanium dioxide or the like. In addition, the tablets may be coated with a varnish dissolved in a volatile organic solvent or solvent mixture. To this coating, a dye may be added to easily distinguish between tablets having different active compounds or different amounts of active compound contained.

Vegetable oil mixed. Hard gelatin capsules may contain granules of the active compound in combination with a solid, powdered carrier material such as lactose, sucrose, sorbitol, mannitol, starch (such as potato starch, corn starch of amylopectin), cellulose derivatives or gelatin.
Dosage units for rectal administration may be prepared in the form of suppositories containing the active substance in admixture with a neutral fat base, or they may be prepared in the form of gelatin rectal capsules containing the active substance in admixture with a vegetable or paraffin oil ,
Liquid preparations for oral administration may be in the form of syrups or suspensions, such as solutions containing from about 0.2% to about 20% by weight of the described active substance, glycerol and propylene glycol. Optionally, such liquid preparations may contain colorants, flavors, saccharin and carboxymethyl cellulose as thickening agents.
Die Herstellung pharmazeutischer Tabletten für perorale Verwendung erfolgt gemäß der nachfolgenden Methode:The preparation of pharmaceutical tablets for peroral use is carried out according to the following method:
Das Befeuchten des pulverförmigen GemischesThe respective solid substances are ground or sieved to a certain particle size. The binder is homogenized and suspended in a certain amount of solvent. The therapeutic compound and required auxiliaries are mixed with the binder solution under continuous, constant mixing and so moistened that the solution is evenly distributed in the bulk without over-wetting any parts. The amount of solvent is usually chosen to give the mass a consistency reminiscent of wet snow. The moistening of the powdery mixture
243010 2243010 2
Dieses Trocknen erfolgt während 10 Stunden und ist sorgfältig zu standardisieren, da der Dämpfungsgrad des Granulateswith the binder solution causes the particles to adhere somewhat to aggregates, and the actual granulation process is carried out by forcing the mass through a sieve in the form of a stainless steel mesh having a mesh size of about 1 mm. The mass is then laid in thin layers on trays for drying in a drying chamber. This drying takes place for 10 hours and must be carefully standardized, since the degree of damping of the granules
In diesem Fall wird die Masse nicht auf Bleche gegeben, sondern in einen Behälter mit einem Netzboden geschüttet.IQ is of utmost importance for the following procedure and for the characteristics of the tablets. Drying in a fluidized bed is also possible. In this case, the mass is not placed on metal sheets, but poured into a container with a net bottom.
After the drying step, the granules are sieved to obtain the desired particle size. Under certain circumstances, powder must be removed.
Merging agents, anti-friction agents and detackifying agents are added to the so-called final mixture. After mixing, the mass should have its proper composition for the tabletting step.
Wenn die richtige Einstellung erreicht ist, beginnt die Herstellung der Tabletten und wird mit einer Geschwindigkeit von 20 000The cleaned tablet punching machine is equipped with a specific set of punches and mouthpieces, whereupon the proper setting for the weight of the tablets and the degree of compression are tested. The weight of the tablet is critical to the size of the dose in each tablet and is calculated based on the amount of therapeutic agent in the granules. The degree of compression affects the size of the tablet, its strength and its ability to melt in water. Especially with regard to the latter two properties, the selection of the compression pressure (0.5 to 5 t) means something like a compromise. When the right setting is achieved, the production of the tablets begins and is at a rate of 20,000
243010 2243010 2
bis 200 000 Tabletten je Stunde durchgeführt. Das Pressen der Tabletten erfordert unterschiedliche Zeiten und hängt von der Größe des Ansatzes ab.up to 200,000 tablets per hour. The pressing of the tablets requires different times and depends on the size of the batch.
The tablets are freed from adhering powder in a special apparatus and then stored in closed packages until they are delivered.
. .Many tablets, especially those that are harsh or bitter, are provided with a coating. This means that they are coated with a sugar layer or any other suitable coating. , ,
The tablets are usually packaged by machines with an electronic counter. The different types of packaging are made of glass or plastic pots, but you can also use boxes, tubes and special packages for dosing.
The daily dose of the active substance varies and depends on the type of administration, but as a rule of thumb it can be said that the daily dose is 100 to 1000 mg per day of the active substance when administered perorally.

Consciousness and tested without restraint. The animals had been prepared by previously implanting catheters in the abdominal aorta via the femoral artery. Mean arterial blood pressure (MABP) and heart rate were recorded continuously. After a two-hour control period, the compound to be tested was administered by oral intubation at two-hour intervals while being suspended in Methocel's solution (5 ml per kilogram of body weight). The cumulative doses were 1.5 and 25 μΜοΙ per kilogram of body weight, the antihypertensive response, i. the BP reduction, on each dose, was expressed as a percentage of the initial BP control level and plotted against the dose on a logarithmic scale. The dose that would give 20% BP reduction was then determined by interpolation. The results are shown in the table below.
Ein "Trennungswert" wurde für jede Verbindung bestimmt, indem die Unterschiede der -Logarithmus ED -Werte für GefäßerweiterungSpecificity against smooth muscle relaxation was tested as follows: The isolated venous venous preparation of Wistar rats was mounted in an organ bath together with a paced isolated papillary myocardial preparation of the same animal. The integrated contraction activity with respect to the smooth muscle of the portal vein and the peak force amplitude of the papillary cardiac muscle preparation were recorded. The activities involved during a 30-minute control period were assumed to be 100%, and the activities occurring under the influence of an agent to be tested were expressed as a percentage thereof. The agent was administered at 10 minute intervals, and the vasodilation ability (Pfortvene's logarithm ED ^ n ) and that of cardiac muscle suppression (logarithm EDr n of the papillary muscle) were determined by interpolation from the concentration-activity curve in each experiment. A "separation value" was determined for each compound by comparing the differences in the -Logarithm ED values for vascular dilation
