Abstract
To deliver ergosine to a broader pharmaceutical use, claviceps strains capable of producing saprophytic ergosine, which is almost free of side alkaloids, and characterized by stability, high alkaloid products and good fermentation properties, are required. The task is solved by using the strain Claviceps purpurea (Fr.) Tul. To produce Ergosin. IBP 179, its mutants or variants used. The new strain forms a total alkaloid mixture (about 900-1300 * g / ml nutrient medium), of which at least 50% of the alkaloids can be extracted with organic solvents (about 80-90% ergosin / ergosinin and 10-20% easily separable substances) , The strain is grown in aerobic fermentation in a liquid nutrient medium containing an assimilable carbon source, an assimilable nitrogen source and mineral salts in an emersed and / or submerged culture at a temperature of 20-30 degrees C and a pH of 4.0-6 , 2 cultivated during a period of 7-35 days. 95% of the alkaloids formed are released into the nutrient medium from which ergosine / ergosinin is isolated.
Description
Die Erfindung betrifft ein Verfahren zur Herstellung von ErgosineThe invention relates to a process for the production of ergosins
·Of the more than 50 previously known natural ergot alkaloids, the representatives of the peptide group have great importance in medicine, such. Ergotamine, Ergocornin, Ergokryptin and Ergocristin. The Ergosin is very close to the ergotamine and differs from the latter by replacing the amino acid phenylalanine with the amino acid leucine in the cyclol side chain. Like other ergot alkaloids of the peptide type, ergosine has an uterotonic and vasoconstrictive effect and is therefore of importance to medicine. The ergosin or its 9> 10-dihydro derivatives can be used as agents for migraine or for the treatment of arterial hypertension, peripheral vascular diseases, disorders of brain circulation or cardiac arrhythmias (see, for example, DDR patent 128612, DDR patent 128613). · Ergosin is also a key compound for partial syntheses. ·
So far, hardly any methods have become known according to which saprophytic ergosine can be obtained.
In einem britischen Patent ist beschrieben, daß durch Claviceps purpurea OKI 88/72 neben Ergokryptin, ErgocorninIn a British patent is described by Claviceps purpurea OKI 88/72 in addition to ergocryptine, Ergocornin

Patent 1 401 406).and Ergometrin as the main alkaloids (JO%, 28 % and 22 %, respectively) and ergosine in amounts of 5% is formed (Brit. Patent 1 401 406).
Patent 1 184 039)οIn other patents, co-production of ergocornine and ergosine is protected side by side. The protected strain forms both alkaloids or their isomers ergocorninin and ergosinin in approximately equal amounts with a total alkaloid content of about 900 - 1100 ng / ml nutrient solution (Brit. Patent 1 184 039) ο
(DDR-Patentschrift 129 801),Another patent describes the microbiological production of ergosine free of other peptide alkaloids. Clavinal alkaloids occur as adjuvant alkaloids (about 10% of the total alkaloids compared to 90% Ergofin / Ergosinin), which are easily separable. (DDR patent 129 801),
The disadvantages of the said methods are that either closely related peptide alkaloids are present side by side, which must first be separated from each other in a complicated way, or that only small amounts of ergosin are produced after long fermentation times.
It is the object of the invention to prepare the alkaloid Ergosin according to a favorable process and thus to enable a pharmaceutical use.

TuI. IBP 179> seine Mutanten oder Varianten verwendet.According to the invention for the production of ergosine the strain Claviceps purpurea (Fr.) TuI. IBP 179> uses its mutants or variants.
Der neue Stamm Claviceps purpurea (Fr.) TuI. The new tribe Claviceps purpurea (Fr.) TuI. IEP 179 is deposited with the Central Institute for Microbiology and Experimental Therapy of the Academy of Sciences of the GDR in Jena under the name IMET PA 136. It forms a total alkaloid mixture (about 900-1300 μg / inl of nutrient medium), of which at least 5.0 % of the alkaloids can be shaken out with organic solvents and of which more than 80-90% are represented by the isomer pair ergosine / ergosinine. The remaining 10 - 20% are easily separable from the main alkaloids substances (Clavinalkaloide or ergometrine). Ergokryptin occurred partially in small traces. Lysergic acid remains in the nutrient media after shaking out. The alkaloids can be isolated from the nutrient medium by known methods. A conversion of the isomeric alkaloids into each other is possible.
The strain is characterized by the fact that it releases more than 95% of the alkaloids formed into the surrounding nutrient media and that it forms only Begleitalkaloide next to ergosine and its isomers ergosinin, which are easily separable.

To identify the alkaloids, 10 ml of the nutrient media are alkalized and extracted with chloroform. The extract is concentrated to 5 ml. Of these, 2 ml are separated by preparative layer chromatography on silica gel with fluorescence indicator (solvent system chloroform: ethanol = 8: 2). The alkaloid stains are scraped off, taken up with a mixture of water: methanol: glacial acetic acid = 45: 45: 10 and then centrifuged with van Urk's reagent. The supernatant is irradiated with a quartz lamp for 5 min and determines the extinction in the photometer. Γ*2ψ = + 394 ° (c = 1 % Chloroform). Hochaufgelöstes Massenspektrum m/e:The preparative recovery of the peptide alkaloids was carried out in the following manner: 2.5 liters of nutrient solution were made alkaline (pH 9), extracted exhaustively with chloroform, and the combined extracts were evaporated in vacuo. This results in 1300 mg of crude halide mixture. This material was dissolved in a little chloroform and chromatographed on a silica gel column. The eluent was chloroform, to which increasing amounts of ethanol (up to 15%) were added. It formed two separate, in the UV light blue fluorescent zones. The faster moving zone yielded ergosinine. After recrystallization from methanol, 290 mg ergosinin (C 30 H 57 O 5 N 5 ) were obtained, mp 225 0 O. Γ * 2ψ = + 394 ° (c = 1 % chloroform). High-resolution mass spectrum m / e:
