Při vývoji syntézy generického lapatinibu API byly prováděny experimenty s cílem najít optimální poměr jednotlivých komponent jak z hlediska reakční rychlosti, tak z hlediska čistoty intermediátu vzorce I, ale i z hlediska ekonomiky výsledného procesu. Překvapivě jsme zjistili, že při použití různých šarží boronové kyseliny od různých výrobců bylo nutné k dosažení optimálních výsledků použít různé množství boronové kyseliny vzorce VI. 5-Formylfuran-2-yl boronová kyselina (1040 mg, 7,43 mmol) byla suspendována v THF (15 ml), následně byl přikapán hexylenglykol (0,86 ml, 791 mg, 6,69 mmol , 0.9 ekv.) a reakční směs byla míchána při laboratorní teplotě 2 h. K esteru Vila (1,8 g, 8,35 mmol, 1.3 ekv.) byl přidán III (3,3 g, 6,42 mmol, ), 10 % Pd/C (78 mg, 0,07 mmol, 1,2 mol. %), methanol (45 ml), A/,A/-diisopropylethylamin (2,2 ml, 12,85 mmol, 2 ekv.) a vzniklá suspenze byla refluxována po dobu 8 hodin. Po ochlazení na laboratorní teplotu byla žlutá suspenze přefiltrována a promyta methanolem (3 X 20 ml). Produkt byl následně rozpuštěn vTHF (45 ml) a zfiltrován přes vrstvu křemeliny čímž se po odpaření rozpouštědla získalo 2,6 g (87 %) titulní sloučeniny (I) jako žluté práškovité látky. K esteru Vile (4,7 g, 21,3 mmol, 1,35 ekv.) byl přidán III (8,0 g, 15,8 mmol, 1 ekv.), 10 % Pd/C (0,2 g, 0,19 mmol, 1,2 mol. %), křemelina (740 mg), roztok N,Ndiisopropylethylaminu (5,5 ml, 31,6 mmol, 2 ekv.) v směsi methanol/ethanol 1 : 1.2 (140 ml) a vzniklá suspenze byla refluxována (teplota lázně 90 °C) po dobu 2,5 hodin. Po ochlazení na laboratorní teplotu byla žlutá suspenze přefiltrována přes křemelinu a promyta methanolem (4 X 90 ml). Produkt byl rozpuštěn přímo na filtru přídavkem horkého THF (2 X 70 ml) a zfiltrován, čímž se po odpaření rozpouštědla získalo 6,75 g (90 %) titulní sloučeniny (I) jako žluté práškovité látky. Čistota dle HPLC 99 %.To the Vile ester (4.7 g, 21.3 mmol, 1.35 equiv) was added III (8.0 g, 15.8 mmol, 1 equiv), 10% Pd/C (0.2 g, 0.19 mmol, 1.2 mol.%), diatomaceous earth (740 mg), solution of N,Diisopropylethylamine (5.5 ml, 31.6 mmol, 2 equiv.) in methanol/ethanol 1 : 1.2 (140 ml) and the resulting suspension was refluxed (bath temperature 90 °C) for 2.5 hours. After cooling to room temperature, the yellow suspension was filtered through diatomaceous earth and washed with methanol (4 X 90 mL). The product was dissolved directly on the filter by addition of hot THF (2 x 70 mL) and filtered to give 6.75 g (90%) of the title compound (I) as a yellow powder after evaporation of the solvent. Purity according to HPLC 99%.

K směsi boronové kyseliny VI (120 mg, 0,86 mmol, 1,35 ekv.), Ill (322 mg, 0,64 mmol, 1 ekv.), 10 % Pd/C (8,8 mg, 0.008 mmol, 1,3 mol. %) byl přidán N,N diisopropylethylamin (0,13 ml, 0,73 mmol, 1,15 ekv.) a ethanol (4,8 ml) a vzniklá suspenze byla refluxována po dobu 3 hodin. Po ochlazení na laboratorní teplotu byla žlutá suspenze přefiltrována přes křemelinu a promyta methanolem (4X3 ml). Produkt byl rozpuštěn přímo na filtru přídavkem horkého THF (2X3 ml) a zfiltrován, čímž se po odpaření rozpouštědla získalo 238 mg (79%) žluté práškovité látky obsahující dle HPLC 88 % titulní sloučeniny I a 11 % nezreagované výchozí látky III.To a mixture of boronic acid VI (120 mg, 0.86 mmol, 1.35 equiv), III (322 mg, 0.64 mmol, 1 equiv), 10% Pd/C (8.8 mg, 0.008 mmol, 1.3 mol %) N,N diisopropylethylamine (0.13 mL, 0.73 mmol, 1.15 equiv) and ethanol (4.8 mL) were added and the resulting suspension was refluxed for 3 h. After cooling to room temperature, the yellow suspension was filtered through diatomaceous earth and washed with methanol (4X3 mL). The product was dissolved directly on the filter by the addition of hot THF (2X3 ml) and filtered, whereby, after evaporation of the solvent, 238 mg (79%) of a yellow powder containing, according to HPLC, 88% of the title compound I and 11% of the unreacted starting material III was obtained.

Ke směsi boronové kyseliny VI (104 mg, 0,74 mmol, 1,5 ekv.), Ill (250 mg, 0,49 mmol) a 10 % Pd/C (13 mg, 0,012 mmol, 2,5 mol. %), DME (4 ml) a methanolu (2 ml) byl přidán triethylamin (0,2 ml) a vzniklá suspenze byla míchána při teplotě 50 °C po dobu 14 hodin. Po zpracování se získalo 184 mg (78%) žluté práškové látky obsahující dle HPLC 91 % titulní sloučeniny I a 8 % nezreagované výchozí látky III.To a mixture of boronic acid VI (104 mg, 0.74 mmol, 1.5 eq.), IIl (250 mg, 0.49 mmol), and 10% Pd/C (13 mg, 0.012 mmol, 2.5 mol % ), DME (4 mL) and methanol (2 mL) was added triethylamine (0.2 mL) and the resulting suspension was stirred at 50 °C for 14 h. After processing, 184 mg (78%) of a yellow powder substance containing, according to HPLC, 91% of the title compound I and 8% of the unreacted starting substance III was obtained.
Příklad 11 - Syntéza lapatinibuExample 11 - Synthesis of lapatinib
K suspenzi aldehydu I (1 g, 2,11 mmol) a hydrochloridu 2(methylsulfonyl)ethanaminu (0,54 g, 3,37 mmol, 1,6 ekv.) v THF (10 ml) byla za laboratorní teploty přikapána kyselina octová (0,5 ml, 8,44 mmol, 4 ekv.) a Nethyldiisopropylamin (1.47 ml, 8,44 mmol, 4 ekv.). Reakční směs byla míchána při 40 °C 1 h, ochlazena na 20 °C, byl přidán roztok NaBH(OAc)3 (0,9 g, 4,25 mmol, 2 ekv.) v THF (4,2 ml) a výsledná směs byla míchána při 20 °C 3 h. Následně byl přikapán 1,25 M roztok NaOH (16 ml), směs míchána při 20 °C 30 min, produkt extrahován do ethylacetátu (3x8 ml), organická vrstva sušena nad MgSO4, zfiltrována a odpařena za sníženého tlaku. K odparku byl přidán THF (5 ml), výsledná suspenze byla zahřáta na 65 °C, přikapán roztok PTSA (1622 mg, 8,44 mmol, 4 ekv.) ve vodě (0,6 ml), směs míchána při 65 °C 1 ha další 3 h při 20 °C. Suspenze byla odsáta, produkt promyt ethanolem (20 ml) a sušen v sušárně (40 °C) po dobu 3 h čímž se získalo 1,54 g (79 %) titulní sloučeniny II s čistotou dle HPLC 97,1 %.To a suspension of aldehyde I (1 g, 2.11 mmol) and 2(methylsulfonyl)ethanamine hydrochloride (0.54 g, 3.37 mmol, 1.6 equiv.) in THF (10 mL) was added dropwise acetic acid at room temperature (0.5 mL, 8.44 mmol, 4 equiv.) and Nethyldiisopropylamine (1.47 mL, 8.44 mmol, 4 equiv.). The reaction mixture was stirred at 40 °C for 1 h, cooled to 20 °C, a solution of NaBH(OAc)3 (0.9 g, 4.25 mmol, 2 equiv) in THF (4.2 mL) was added and the resulting the mixture was stirred at 20 °C for 3 h. Subsequently, a 1.25 M NaOH solution (16 ml) was added dropwise, the mixture was stirred at 20 °C for 30 min, the product was extracted into ethyl acetate (3x8 ml), the organic layer was dried over MgSO 4 , filtered and evaporated under reduced pressure. THF (5 mL) was added to the residue, the resulting suspension was heated to 65 °C, a solution of PTSA (1622 mg, 8.44 mmol, 4 equiv.) in water (0.6 mL) was added dropwise, the mixture was stirred at 65 °C 1 ha for another 3 h at 20 °C. The suspension was suction filtered, the product washed with ethanol (20 mL) and dried in an oven (40 °C) for 3 h to give 1.54 g (79%) of the title compound II with an HPLC purity of 97.1%.
