Abstract
Řeší se výroba thymosinu frakce V z bo-. vinních fetálních thymů, které je možno použít z exsanquinovaných fetů při výrobě bovinního fetálního séra, nebo novorozených telat rovněž exsanquinovaných^pro přípravu novorozeneckého séra. Tkán thymů se homogenizuje, centrifuguje a získaný supernatant se tepelně denaturuje při 80 °C, pak následuje precipitace síranem amonným do saturace 30 %. Získaná frak-- ce se ultrafiltruje s nominální odddělovací hranicí 15 až 20 tis., následně odsolí a lyofilizuje. Získaný thymosin frakce V obsahuje zisk až trojnásobného množství z 1 kg tkáně bovinních fetálních thymů oproti vzrostlým telatům. Ultrafiltrát se lyofilizuje, získaná látka (frakce IV) se rozpustí v deionizované vodě (2 g v 10 ml), pokud je třeba, upraví se pH na 7,6 a odstřeluje se 5 min. při 15 000:rpm.
