Abstract

技术领域
概括而言,本发明涉及生物相容的、可生物降解的聚合物领域。更加具体地,本发明描述了一种用于纯化低分子量聚合物的方法,所述方法利用聚合物溶液的降低温度的液-液相分离,其中溶剂由甲醇、乙醇和/或异丙醇组成。用于所述方法的合适聚合物包括聚乳酸(PLA)。本发明经纯化的聚合物的独特之处在于它们的高纯度,其部分地通过具有比粗聚合物更窄的分子量分布表现出来,从而它们特别适合用于持续释放制剂或生物相容聚合物。
背景技术
诊断剂和药物,不管是蛋白质或是小分子,在患者体内各自具有确定的半衰期。很多时候,通过延长其半衰期可以使所述试剂或药物的效果最大化。一种方法就是将所述试剂或药物包裹于与受用者相容的材料中,所述材料在受用者体内缓慢破坏或溶解,从而试剂或药物在比单独的试剂或药物的半衰期长的时间内持续释放。
已经证明,可以将生物活性的或者药学活性的试剂包裹在生物相容的、可生物降解的成壁材料例如聚合物中,以提供持续释放或缓释。在这些方法中,典型地使用搅拌器、混合器或其它动态混合技术将试剂或药物溶解、分散或乳化于一种或多种含成壁材料的溶剂中。然后除去溶剂,形成包括所述试剂或药物的微粒。然后可以将这些微粒给予患者。
可生物降解的聚合物早已被广泛用于控制药物递送。它们的优点是在完成它们的目标任务之后不需要外科手术除去,因为它们或者酶降解掉或者化学降解掉,例如水解。对于各种药学和生物医学应用,已经对聚酯例如聚乳酸(PLA)、聚乙醇酸(PGA)和乳酸-乙醇酸共聚物(PLGA)进行了广泛研究。PGA、PLA和特别是它们的共聚物PLGA是最常用的聚合物族。这些聚合物的每一种都展示出期望的生物相容性特性并且当注入患者中时是可生物降解的,因此它们已被广泛地被接受作为药物组分并特别用于持续释放制剂中。Chaubal,Drug DeliveryTechnology,2002,2:34-36和Anderson et a1.,Adv.Drug Deliv.Rev.,1997,28:5-24。
包裹于PLA微粒中的药物通过水性环境的扩散作用或者通过上面提及的聚合物的降解释放。影响由聚合物制成的微粒的体外和体内药物释放速率的一个变量是其分子量。特别地,聚合物的分子量影响生物降解速率。对于活性剂释放的扩散机理,聚合物应该保持完整直到所有的活性剂从微粒释放,然后才降解。随着聚合物基体材料的生物侵蚀,活性剂也可以从微粒释放。
已经发现,与高分子量聚合物相比,低分子量聚合物倾向于以更快的方式释放活性剂(Asano et al.,Biomaterials,1989,vol.10:569)。因此,通过选择较低分子量的聚合物,可以制造其中所得到的微粒显示出加速释放的制剂。当活性剂需要在更短的时间内或者以更高的浓度进行递送时,这是合乎需要的。但是,目前没有相应的技术来制造具有低多分散性的相对纯的低分子量聚合物(Asano et al.,Biomaterials,1989,vol.10:569;Hyon et al.,Biomaterials,1997,vol.18:1503)。
用于持续释放制剂中的聚合物制备中的另一个因素是聚合物混合物的多分散性。目前没有相应的方法可以制造或纯化具有相对低多分散性的小分子量聚合物。因此,本领域中存在对制造用于持续释放药物制品(例如微粒、棒、膜等等)的聚合物微粒的改进方法的需求,其中所述方法得到具有低多分散性的低分子量聚合物。
发明概述
本发明教导了一种用于纯化低分子量聚合物的方法,所述方法利用聚合物溶液的降低温度的液-液相分离,其中溶剂由甲醇、乙醇和/或异丙醇的混合物组成。在具体的实施方案中,溶剂是甲醇、乙醇或异丙醇。
本发明涉及一种制造低分子量和低多分散聚合物的方法。更加特别地,本发明提供一种在单相溶剂中利用聚合物的降低温度的相分离作用纯化低分子量聚合物的方法,其中溶剂选自甲醇、乙醇和异丙醇。更加具体地,所述方法包括以下步骤:将粗聚合物与甲醇、乙醇或异丙醇混合直到聚合物溶解,降低溶液的温度直到形成两层,将上层液体与下层分离并分离聚合物。所述溶剂也可以是甲醇、乙醇或异丙醇相互之间的或是与其它液体的混合物,只要所使用的溶液能作为本发明聚合物(例如PLA)的溶剂并且能够如本文所述发生相分离。
在一个实施方案中,溶剂主要是甲醇、乙醇或异丙醇。主溶剂还可以与其它溶剂混合。例如,根据本发明的方法甲醇和乙醇可以混合并使用,或者甲醇可以与另一种溶剂例如二氯甲烷混合,其中所述溶剂混合物仍然允许纯化低分子量、低多分散的聚合物。
在某些实施方案中,聚合物的选择是聚乳酸,也称为PLA。在特定实施方案中,使用本发明的方法进行纯化后,通过Mw除以Mn测量的低分子量聚合物的P值小于1.6。因此,本发明的一个实施方案是通过本发明的方法纯化的聚合物与常规制造的聚合物相比具有更窄的分子量分布。
聚合物可以经控制而形成适于注入需要它们的患者中的微粒、棒、膜等等。因此,本发明还涉及本发明纯化的聚合物的药学可接受制剂以及使用所述聚合物的方法。
发明详述
本发明涉及一种纯化低分子量聚合物的方法,所述方法利用在甲醇、乙醇或异丙醇中所述聚合物的降低温度的液-液相分离,涉及包含所述聚合物的组合物和涉及使用所述组合物的方法。在一个实施方案中,聚合物的选择是聚乳酸,也称为PLA。所得到的聚合物具有更窄的分子量范围,并且比使用当前的方法得到的那些聚合物显著更纯。因此,本发明还涉及经纯化的PLA聚合物的新组合物,以及它们在标准药物组合物例如微粒中的用途。所述经纯化的低分子量、低多分散聚合物可以用于制造包裹试剂和/或药物的微粒,所述微粒用于注入需要它们的患者中。本发明的所述经纯化的低分子量、低多分散聚合物当被制成微粒时将提供试剂和/或药物的持续释放。
如在本文中使用的那样,应该理解术语“低分子量”是指其中平均分子量低于5000道尔顿的分子量范围。在可选择的实施方案中,低分子量表示低于3000道尔顿的平均分子量。当该术语与聚合物联用时,应该理解的是优选的实施方案是聚乳酸,也称为PLA。在一个实施方案中,PLA聚合物基本由含乳酸酯的部分组成。
在一个实施方案中,本发明的纯化方法产生低分子量聚合物,所述聚合物与先前公开的聚合物纯化步骤相比具有更低的多分散性。在一个实施方案中,通过Mw除以Mn测量的聚合物的P值小于1.6。更优选地,P值低于1.55,更优选P值低于1.5,更优选P值低于1.45,更优选P值低于1.4,最优选P值低于1.3。此外,本发明的方法得到了自由流动细白色粉末的聚合物形式。在另一个实施方案中,本发明经分级的聚合物当干燥后是非常白的,具有雪白色的外观。
在另一个实施方案中,根据本发明制造的聚合物具有800-10000,800-5000,800-4000,800-3000,800-2000,800-1500,800-1200,或1000-2000,或1000-1500,或1000-1200的重均分子量。
虽然代表性的实施例使用甲醇、乙醇或异丙醇,但是本领域技术人员很容易理解,也可以使用这三种醇的混合物或者含有低于100%的任一种这三种醇的溶剂。例如,可以用另一种溶剂例如异丙醇稀释乙醇以得到90%乙醇、10%异丙醇溶剂,其根据本发明的教导也能够实现本发明。还可以加入除这三种主要溶剂以外的溶剂以得到溶剂混合物,例如二氯甲烷。本领域的技术人员能够确定用其它溶剂稀释主要溶剂即甲醇、乙醇和异丙醇的适当限度,从而所得到的混合溶剂可以根据本发明的教导进行使用。
在进一步的实施方案中,本领域的技术人员将会理解未经纯化的聚合物在本发明溶剂中的初始溶解将取决于聚合物的溶解度。因此,溶剂-聚合物混合物可能需要升温到室温以上以使聚合物溶解。后续的相分离步骤,其是通过温度的降低促进的,可以在更低的温度进行,例如在室温或者接近室温的温度。因此,其中聚合物溶解于溶剂中的起始温度可以是约60℃,相分离可以在室温或接近室温的温度(例如18℃-28℃)。在另一个实施例中,其中聚合物溶解于溶剂中的起始温度可以是室温,相分离可以在约10℃或更低的温度进行。
本领域的技术人员,使用本文的教导利用常规实验,即可以容易地确定合适的溶解温度和相分离温度。因此,在进一步的实施方案中,开始温度可以比相分离温度高约10℃。在一个实施例中,如果聚合物在室温溶解,那么相分离温度比溶解温度低至少约10℃,例如约10℃,或者可选择的约5℃,约0℃,约-5℃,约-10℃,约-15℃,约-20℃,约-30℃,约-40℃,约-50℃,约-60℃或更低,从而发生相分离。基于本发明的教导,本领域的技术人员利用常规实验即可以容易地确定最佳聚合物溶解温度和最佳引起相分离的温度。
如在本文中所使用的那样,术语“约”是指高达列举值20%的可变性,不论列举值是上面描述的温度还是其它值。
最佳溶解度需要通过实验确定,因为溶解度太高将导致不能实施液-液相分离,而溶解度太低将是不实用的。方便地,可以使用USP溶解度定义,其中微溶是1份聚合物对100-1000份溶剂,溶解度增加是30-100份溶剂,溶解度进一步增加是10-30份溶剂,溶解度继续增加是1-10份溶剂,非常可溶是1份聚合物对不足1份溶剂。
如在本文中所使用的那样,术语“微粒”是指体积平均颗粒尺寸为约1-1000微米的颗粒。此外,术语“非溶剂”是指基本不溶解材料的物质,“溶剂”是指溶解本发明聚合物的液体。如在本文中所使用的那样,试剂和/或药物的“持续释放”是从本发明组合物的释放,其中该释放发生在比直接给药而能得到试剂和/或药物的期间长的期间内。
据设想,包裹在由根据本发明纯化的聚合物制成的微粒中的试剂的持续释放,在大于1天的期间内发生。持续释放可以是连续或不连续的释放,具有相对恒定的或变化的释放速率。释放的连续性和释放的水平可受下列因素影响:使用的聚合物组合物的类型(例如单体比率、分子量、嵌段组成和聚合物的不同组合)、蛋白质负载和/或制造期望效果的赋形剂的选择。
可根据本发明的方法纯化的合适生物相容聚合物,可以是可生物降解的聚合物或非生物可降解的聚合物,或者是它们的共混物或共聚物。如果聚合物和任何降解产物对接受者都是非毒性的,那么聚合物是生物相容的。更加具体地,非毒性包括在本发明聚合物的正常使用期间对接受者的身体没有显著的有害或不利(untoward)作用,例如由于聚合物导致的对注入的显著免疫反应。
可根据本发明纯化和使用的一种合适生物相容、可生物降解聚合物包括例如聚乳酸(PLA)。适合根据与本发明的方法类似的方法进行纯化的本领域已知的其它聚合物包括聚乙交酯(PGA),乳酸-乙醇酸共聚物(PLGA),聚乳酸,聚乙醇酸,聚碳酸酯,聚酯酰胺,polyanydrides,聚氨基酸,聚原酸酯,poly(dioxanone)s,poly(alkylene alkylate)s,共聚物或聚乙二醇和聚原酸酯,可生物降解聚氨酯,它们的共混物,和它们的共聚物。
术语“可生物降解”是指组合物将体内降解或侵蚀而形成更小的化学物种。降解可以由例如酶、水解或其它化学机理,和/或物理方法产生。经包裹的生物活性剂从本发明的可生物降解的PLA制剂的释放是通过扩散和聚合物组合物的降解(即酶降解或水解降解)的结合实现的。
如本文中所使用的那样,术语“生物活性剂”是一种试剂或其药学可接受的盐,其当在体内释放时,具有期望的生物活性,例如体内治疗、诊断和/或预防性质。本发明的持续释放组合物可以含有约0.01%(w/w)-约90%(w/w)的活性剂(组合物的干重)。试剂的数量可取决于试剂的期望效果、计划的释放水平和试剂被释放的时间跨度而发生变化。合适的生物活性剂的例子包括蛋白质、肽、突变蛋白质(muteins)和它们的活性片段,以及小分子,下面将进行更充分地描述。
如在本发明中所使用的那样,术语“蛋白质”和“肽”应理解为包括酰胺键连接的氨基酸聚合物。典型地,肽由少于约50个氨基酸组成,更加典型地少于约30个氨基酸残基,甚至更加典型地少于约20个氨基酸残基。而蛋白质典型地由多于50个氨基酸组成,并具有结构和生物活性。蛋白质的生物活性可以是酶性质的或者其可以是赋予构想变化的结合活性。这些术语进一步包括模仿蛋白质或肽的组分的化学结构的类似物和衍生物。类似物的例子包括含一个或多个非天然氨基酸的肽或蛋白质。衍生物的例子包括含已经被衍生的氨基酸侧链、肽主链和/或端-氨基或-羧基的肽或蛋白质。
适合用于本发明制剂的肽包括但不限于enfuvirtide(Trimerisand Roche as Fuzeon销售)、血管紧张素、Amylin、ACTH、血管紧张素原、Ceropin A-蜂毒肽酰胺、Cecropin B、Magainin 1、肾素抑制剂肽、铃蟾肽、Osteocalcin、缓激肽、B1抑制剂肽、胰激肽、降钙素、胆囊收缩素、促肾上腺皮质激素释放因子、强啡肽A、Endomorphin、Sarafotoxin、脑啡肽、Exendin、血纤肽、促生长激素神经肽、促胃液素、促胃液素释放肽、类似高血糖素的肽、生长激素释放因子、OVA肽、黄体化激素-释放激素、心房肽、黑色素浓缩激素、脑钠素、Vasonatrin、神经激肽、神经介素、神经肽Y、神经降压肽、Orexin、催产素、后叶加压素、副甲状腺激素肽、催乳素释放肽、生长激素抑制素、生长激素抑制素瘤抑制类似物、促甲状腺激素释放激素,以及它们的变体和衍生物(可参见Latham,(1999)Nat.Biotech.,17:755)。
合适的蛋白质、突变蛋白质和它们的活性片段的例子包括但不限于免疫球蛋白、抗体、细胞因子(例如淋巴因子、单核因子、趋化因子)、白细胞介素、干扰素(β-IFN,α-IFN和γ-IFN)、***、核酸酶、肿瘤坏死因子、集落(colony)刺激因子、胰岛素、酶(例如超氧化物歧化酶、组织纤维蛋白溶酶原激活剂)、肿瘤抑制剂、血蛋白质、激素和激素类似物(例如生长激素、促肾上腺皮质激素和黄体化激素释放激素(LHRH))、疫苗(例如瘤的(tumoral)、细菌和病毒抗原)、抗原、血液凝结因子;生长因子;肽例如蛋白质抑制剂、蛋白质拮抗剂和蛋白质激动剂;核酸,例如反义分子;寡核苷酸;和核酶。
适合用于本发明的小分子量试剂包括抗肿瘤试剂例如盐酸博来霉素、卡铂、甲氨蝶呤和阿霉素;抗生素例如庆大霉素、盐酸四环素和氨苄西林;解热剂、止痛剂和抗炎剂;盐酸甲麻黄碱、盐酸那可丁和磷酸可待因;镇静剂例如盐酸氯丙嗪、盐酸prochloperazine和硫酸阿托品;肌肉松弛剂例如氯筒箭毒碱;抗癫痫剂例如苯妥英钠和乙琥胺;抗溃疡剂例如甲氧氯普胺;抗抑郁剂例如氯米帕明;抗过敏剂例如苯海拉明;强心剂例如theophillol;抗心律不齐剂例如盐酸***;血管舒张剂例如盐酸地尔硫卓和硫酸巴美生;hypotensive diuretics例如潘托铵和盐酸乙肼苯哒嗪;抗利尿剂例如二甲双胍;抗凝血剂例如柠檬酸钠和肝素钠;止血剂例如凝血酶、亚硫酸氢钠甲萘醌和维生素K4;抗结核剂例如异烟肼和ethanbutol;激素例如强的松龙磷酸钠和甲巯咪唑;抗紧张剂例如利培酮;和麻醉拮抗剂例如盐酸烯丙***。
“稳定剂”,如该术语在本文中使用的那样,是指以共价或非共价方式与生物活性剂结合或发生相互作用或者被包含的任何试剂。适合用于本发明中的稳定剂描述于美国专利5716644、5674534、5654010、5667808和5711968中。
此外,可以加入赋形剂以保持生物活性剂在释放期间内的效力(potency)和改变聚合物降解。所述赋形剂可以加入到经分散的体系中,该经分散的体系然后被粉化,或者可以加入到混合物中,该混合物在干燥物质碎裂之前或之后进行碎裂化以得到生物活性剂的颗粒。合适的赋形剂包括例如碳水化合物、氨基酸、脂肪酸、表面活性剂和填充剂,是本领域技术人员公知的。酸性或碱性的赋形剂也是合适的。使用的赋形剂的数量是基于按重量计其与生物活性剂的比率。对于氨基酸、脂肪酸和碳水化合物,例如蔗糖、海藻糖、乳糖、甘露糖醇、葡聚糖和肝素,碳水化合物对生物活性剂的比率典型地在约1∶10-约20∶1。对于表面活性剂,表面活性剂对生物活性剂的比率典型地是约1∶1000-约2∶1。典型地,填充剂包括惰性材料。合适的填充剂是本领域技术人员公知的。
赋形剂也可以是单独地分散在聚合物基体内的金属阳离子组分。该金属阳离子组分用于调节生物活性剂的释放,并不与生物活性剂络合。如果存在的话,金属阳离子组分可任选含有与包含于金属阳离子稳定的生物活性剂中的金属阳离子相同种类的金属阳离子,和/或可含有一种或多种不同种类的金属阳离子。金属阳离子组分用于调节生物活性剂从持续释放组合物的聚合物基体的释放并且可以增强生物活性剂在组合物中的稳定性。使用调节释放的金属阳离子组分的例子包括或包含,例如,Na、K、Mg、Zn和Ca。阳离子对聚合物的最佳比率取决于所使用的聚合物和金属阳离子组分,可以容易地由本领域的技术人员确定。含有分散的金属阳离子组分以调节生物活性剂从聚合物基体的释放的聚合物基体进一步描述于美国专利5656297中。
在另一个实施方案中,在持续释放组合物中可以包括至少一种成孔剂,例如水溶性盐,以改变微结构,例如如美国专利6531154中教导的那样。加入到悬浮液中的成孔剂的比例是约1%(w/w)-约30%(w/w),其中所述悬浮液包含分散于含至少一种生物相容聚合物和至少一种聚合物溶剂的溶液中的生物活性剂的亚微粒。
已知有多种方法可以用于形成聚合物/活性剂基体。在很多这些方法中,待包裹的物质被分散到含成壁材料的溶剂中。在所述方法的一个单独阶段,从微粒除去溶剂从而得到微粒产品。例如,用于形成持续释放生物活性剂的组合物的方法描述于美国专利5019400和5922253中。虽然对本发明的方法而言最合适的聚合物是PLA,但是本领域的技术人员将认识到在持续释放制剂的制造中,通过本发明的方法纯化的PLA聚合物可以与其它聚合物进行混合。这些聚合物共混物可以用于形成适于药物递送的基体。还将理解的是,这些共混物因而与本发明使用的PLA分子的纯形式相比通常将具有不同的性质。
适于冻结小滴的方法包括将小滴引导进入或靠近液化气体,例如液体氩气或液体氮气,以形成冷冻微滴,然后从液体气体分离所述冻结微滴。然后将所述冻结微滴暴露于液体或固体非溶剂,例如乙醇、己烷、与己烷混合的乙醇、庚烷、与庚烷混合的乙醇、戊烷或油。作为固体和/液体,冻结微滴中的溶剂被提取进入非溶剂中,形成包含生物相容聚合物和生物活性剂的聚合物/活性剂基体。将甲醇、乙醇或异丙醇与其它非溶剂例如己烷、庚烷或戊烷混合,可以增加从某些聚合物萃取溶剂的速率,高于单独使用甲醇、乙醇或异丙醇能实现的速率。
通过改变小滴尺寸,例如,通过改变超声喷嘴直径,可以制造各种尺寸范围的持续释放组合物。如果持续释放组合物是微粒形式,并且希望得到非常大的微粒,那么可将微粒例如通过注射器直接挤到冷液体中。增加聚合物溶液的粘度也可以增加微粒的尺寸。可以通过该方法制造的微粒尺寸的范围是例如直径从大于约1000微米到约1微米。
从含生物相容聚合物和生物活性剂的悬浮液形成持续释放组合物的另一种方法包括例如在模具中的薄膜注塑,以形成膜或形状。例如,在将悬浮液放入模具之后,通过本领域中公知的方式除去聚合物溶剂,或者降低聚合物悬浮液的温度直到得到具有一致的干重的膜或形状。
常规微囊化方法及其制备的微粒的进一步的例子公开于美国专利3737337中,其中制备了成壁或成壳聚合物材料在溶剂中的溶液。所述溶剂在水中只是部分可混溶的。固体或核芯物质被溶解或分散于含聚合物的混合物中,然后含核芯物质的混合物被分散于在所述有机溶剂中不混溶的水性液体中以从微粒除去溶剂。
其中溶剂被从含有物质的微粒除去的方法的另一个例子公开于美国专利3523906中。在该方法中,待包裹的物质在溶液中乳化,其中所述溶液是聚合物材料在与水不混溶的溶剂中的溶液,然后所述乳液在含有亲水胶体的水溶液中乳化。然后通过蒸发实现从微粒除去溶剂,得到产品。
在美国专利3691090所公开的另一个方法中,优选地在减压下,从在水性介质中的微粒分散体蒸发有机溶剂。类似地,美国专利3891570的公开给出了一种方法,其中通过施加热或通过使微粒置于降低的压力之下,使来自在多元醇介质中的微粒分散体的溶剂从微粒蒸发。溶剂去除方法的另一个例子示于美国专利3960757中。
Tice等人在美国专利4389330中描述通过包含如下步骤的方法制造含有活性剂的微粒:(a)将活性剂溶解或分散于溶剂中并将成壁材料溶解于该溶剂中;(b)将含活性剂和成壁材料的溶剂分散于连续相加工介质中;(c)从步骤(b)的分散体蒸发一部分溶剂,从而在分散体中形成含活性剂的微粒;和(d)从微粒提取剩余的溶剂。
不拟囿于特定的理论,据信生物活性剂的释放可以通过两种不同的机理进行。第一,生物活性剂可以经由在聚合物基体内产生的水溶液填充的通道通过扩散释放,例如通过生物活性剂的溶解或者通过由在持续释放组合物制造期间聚合物溶剂的除去产生的孔隙。第二机理是由于聚合物的降解导致的生物活性剂的释放。降解速率可以通过改变影响聚合物水合速率的聚合物性质而进行控制。这些性质包括,例如,交酯与乙交酯的比率,包括聚合物;单体L-异构体而不是外消旋混合物的使用;和聚合物的分子量。这些性质可影响亲水性和结晶性,而亲水性和结晶性控制聚合物的水合速率。
本发明的聚合物及其药学可接受的变体可以通过注入、植入(例如皮下、肌肉内、腹膜内、颅内和真皮内)、给药到粘膜(例如鼻内、肺内、含服或通过栓剂)或者原位递送(例如通过灌肠法或气溶胶喷雾剂)体内给药到例如人或动物,以基于已知的治疗参数提供期望剂量的生物活性剂用于利用特定的试剂治疗各种医疗病症。如在本文中所使用的那样,“治疗有效量”、“预防有效量”或“诊断有效量”是给药后引出希望的生物、预防或诊断响应所需的生物活性剂的量或生物活性剂的持续释放组合物的量。
下面的实施例应理解为是代表性的实施例,并不是限制本发明要求保护的整个范围。下述的PLA聚合物的数均和重均分子量(Mn,Mw)通过端基滴定和凝胶渗透色谱法(GPC;普适标定)测定。
实施例
低分子量聚乳酸(PLA)聚合物的合成
通过乳酸单体缩聚合成PLA是在没有任何催化剂存在的情况下通过在高温和降低的压力下从85wt%的乳酸水溶液蒸馏出水进行。例如,将412g乳酸水溶液加入500ml三口烧瓶中,所述烧瓶配有内搅拌棒、通过带有稳定剂的蒸馏头配有水冷冷凝器、针状入口(连接有气体鼓泡器并***橡胶隔片中以通过干燥氮气)。在大气压下,氮气速率大约是每分钟280泡。
冷凝器连接到接管,接管连接到气体鼓泡器和接收瓶。烧瓶上部缠绕有玻璃纤维。烧瓶浸入油浴中直到液体平面与油平面持平。可变变压器一直设定为70和140v。加热板的搅拌位置是8。在50分钟内烧瓶在油浴上从室温加热到140℃。当水开始冷凝时,温度在约2小时时间内逐渐升至160℃。然后接管连接到Buchi Rotavapor泵***而不是连接到气体鼓泡器,并且用干冰浴冷却接收瓶。在40分钟时间内,压力从大气压降低到400毫巴(mbar)并且油浴温度逐渐增加到170℃。氮气速率降低到每分钟2-10泡。在大于55分钟内,***压力进一步降低到100毫巴并且油浴温度逐渐增加到188℃。以防止蒸馏暴沸降低压力必须是渐进的,并且蒸馏头温度不超过120℃。
反应在这些条件下搅拌1、3、5和7小时,分别制备Mn约为700、1000、1500和2000的PLA。除去油浴,用氮气吹扫烧瓶并冷却至室温。烧瓶存储在冷冻箱(-40℃)中以备第二天纯化。

方法B:
使用一种现有方法制备聚合物。向烧瓶中加入220ml二氯甲烷,混合物在温和回流下在油浴上在55℃加热,直到聚合物完全溶解(约2-3小时)。然后将溶液倒入在1升烧杯中的400ml去离子(DI)水中,搅拌混合物0.5小时。加入额外的二氯甲烷(180ml)以帮助漏斗中分层。在分液漏斗中分离有机层(约450ml)。通过减压旋转蒸发除去二氯甲烷。凝胶状的聚合物在真空下进一步干燥三天。得到153g凝胶PLA(Mn;670)。
Mn约1000
在100毫巴真空下在188℃3小时制备的聚合物
方法E:
向烧瓶中加入220ml二氯甲烷,混合物在温和回流下在油浴上在55℃加热,直到聚合物完全溶解(约2-3小时)。然后将溶液倒入在1升烧杯中的400ml去离子(DI)水中,搅拌混合物0.5小时。加入额外的二氯甲烷(180ml)。在分液漏斗中分离有机层(约450ml)。通过减压旋转蒸发除去二氯甲烷。凝胶状的聚合物在35℃水浴温度通过低于2mmHg真空的旋转蒸发进一步干燥3小时。
将粗聚合物转移到500ml塑料容器中,在室温混合到320ml乙醇中并在-40℃存储4小时。形成两层混合物。快速除去上层液体。在室温将另外200ml乙醇与在剩余底层中的聚合物混合,然后在-78℃冷却。形成白色固体,通过滗析溶液分离白色固体。在-78℃用200ml戊烷洗涤聚合物并冷冻干燥5天。得到136.4g白色固体PLA(Mn:1042)。将上层溶液和洗涤液体合并。通过减压旋转蒸发除去上层的乙醇溶剂,残余物真空干燥5天。得到27.1g凝胶状PLA(Mn:679)。
Mn约1500
在100毫巴真空下在188℃5小时制备的聚合物
方法E:
向烧瓶中加入220ml二氯甲烷,混合物在温和回流下在油浴上在55℃加热,直到聚合物完全溶解(约2-3小时)。然后将溶液倒入在1升烧杯中的400ml的60℃DI水中,搅拌混合物0.5小时。加入额外的二氯甲烷(130ml)。在分液漏斗中分离有机层。在2.5小时内,PLA溶液的二氯甲烷(约440ml)在机械搅拌下通过注射泵滴加入在4升烧杯中的3400ml乙醇中,所述烧杯用干冰/丙酮浴冷却。一种白色固体沉淀出来。在完全加入之后,将混合物静置1-2小时并将绝大部分溶液倒掉。将聚合物分开放进两个500ml的容器中并再次在-78℃冷却。固体形成,除去溶液。
在室温向每一个容器混入200ml乙醇。混合物在-40℃存储4小时。形成两个浑浊的层。除去上层(在该温度底层轻微固化)。将底层在-78℃冷却以除去更多的剩余乙醇并在78℃用2×200ml戊烷洗涤。
聚合物在真空下干燥10天得到白色固体(119g,Mn:1589PLA)。
Mn约2000
在100毫巴真空下在188℃7小时制备的聚合物
方法E:
本过程与上述过程(Mn约1500)相似,得到白色固体(112g,Mn:约2000)。结果示于表2中。

通过聚合物溶液的相分离纯化的低分子量PLA
在文献中甲醇和乙醇被报道为是PLA(Mn>800)的非溶剂。下面的实施例证明:取决于聚合物的MW在室温和升高的温度(约40-50℃),低分子量(MW)PLA(Mn<2000)可以溶于MeOH(甲醇)和EtOH(乙醇)中。而且,取决于PLA起始材料的MW,异丙醇(IPA)显示出在室温至高达50℃的温度溶解PLA。普通PLA材料溶解度的比较显示在这些溶剂中PLA溶解性的一般顺序是MeOH>EtOH>IPA。换言之,测试的PLA聚合物在甲醇中溶解性更大,乙醇次之,在异丙醇中的溶解性最小。
通过降低温度在醇溶剂***中观察到聚合物溶液的液-液相分离,所述醇溶剂***例如MeOH、IPA和MeOH-甘油(表3)。在单一溶剂***中PLA溶液此类相分离的临界温度并不限于低于室温;也可以在室温进行,例如在PLA-IPA***中在室温相分离。
通过在这些醇溶剂***中的相分离方法的连续纯化,在相分离以后在上部相(较低MW)和底部相(较高MW)中的低MW PLA的分子量分布可以进一步变窄。通过结合有在线双检测器(RI和粘度测定)的GPC分析的结果列在表3中。
在具有强溶解力(solubilizing power)的溶剂例如二氯甲烷(DCM)、丙酮、乙腈(ACN)和乙酸乙酯中没有观察到温度降低导致的PLA溶液相分离。虽然通过向这些溶剂中加入非溶剂来降低它们的溶解力可以导致聚合物沉淀,但是因为聚合物在底部相中几乎完全沉淀,所以在这些***中聚合物分级非常差(表3)。

5)在双溶剂/非溶剂(1∶1)混合物例如DCM/己烷和乙酸乙酯/己烷中的PLA在RT分离,在上层相中几乎没有发现PLA,也就是说PLA几乎完全沉淀在底部相中。
虽然已经用优选实施方案对本发明进行了描述,但是应该理解对本领域的技术人员来说存在变化和改变。因此,后附的权利要求函盖所有在保护范围内的此类等价变化。
Claims (16)
1.一种分级低分子量聚乳酸(PLA)聚合物的方法,包括以下步骤:
A)将PLA聚合物溶解在溶剂中,其中所述溶剂是含有甲醇、乙醇或异丙醇的混合物;
B)冷却步骤A的溶液以引起液-液相分离;
和
C)分离步骤B的上层和下层。
2.权利要求1的方法,进一步包括以下步骤:
D)向分离的上部和/或底部层中加入溶剂;
E)使步骤D的混合物形成固体,和
F)从每一层分离固体。
3.权利要求1的方法,其中所述溶剂由选自甲醇、乙醇或异丙醇的溶剂组成。
4.权利要求1的方法,其中所述溶剂由甲醇组成。
5.权利要求1的方法,其中所述溶剂由乙醇组成。
6.权利要求1的方法,其中所述溶剂由异丙醇组成。
7.权利要求3、4、5或6的方法,进一步包括重复步骤A-C的步骤。
8.从权利要求1或2任一项的层分离的固体聚合物。
9.由权利要求7的方法制备的固体聚合物。
10.权利要求9的聚合物,其中多分散性小于1.6。
11.权利要求10的聚合物,其中多分散性小于1.3。
12.权利要求8的聚合物,其中聚合物的数均分子量是800-2500。
13.包含权利要求2所述经纯化的聚合物的药学可接受组合物。
14.固体PLA聚合物,其中多分散性小于1.6。
15.权利要求14的聚合物,其中多分散性小于1.3。
16.一种治疗病症的方法,包括给药有效量的包含微粒的药学组合物,其中所述微粒由权利要求14的PLA聚合物制成。
