发明背景
发酵方法被用来制造大量具有商业价值的产品。工业上,发酵被用来生产简单的化合物如酒精(尤其是乙醇);酸,如柠檬酸、衣康酸、乳酸、葡糖酸、赖氨酸;酮;氨基酸,如谷氨酸,但也用于生产更为复杂的化合物例如抗生素,如青霉素、四环素;酶;维生素,例如核黄素、B12,β-胡萝卜素;难以用合成的方法生产的激素,例如胰岛素。发酵方法也用于酿造(啤酒和酒工业)、乳品、皮革、烟草工业中。
关于发酵产品例如乙醇的生产有大量的文献披露,其中包括WO2002038787A2。
需要进一步改进发酵方法,也需要经改进的包括发酵步骤的方法。相应地,本发明的目的在于提供经改进的用于生产例如乙醇的发酵方法。
发明概述
本发明涉及经改进的生产发酵产品尤其是乙醇的方法,此发酵产品还有例如“发明背景”部分提到的产品。根据本发明,可以预期饮品例如啤酒的生产。
第一个方面,本发明提供了生产发酵产品的方法,所述方法包含在β-葡聚糖酶存在下非淀粉多糖的预液化(preliquefaction),然后在热稳定性β-葡聚糖酶和木聚糖酶的存在下进行喷射蒸煮(jet cooking)和液化。
第二个方面提供生产发酵产品的方法,所述方法包含步骤:(a)提供糖化醪,该糖化醪包含含淀粉的物质和水;(b)在β-葡聚糖酶存在下将步骤(a)的糖化醪预液化;(c)使步骤(b)的糖化醪凝胶化;(d)在α-淀粉酶、β-葡聚糖酶和木聚糖酶存在下液化步骤(c)的糖化醪;和(e)糖化并发酵步骤(d)的糖化醪以生产发酵产品。
第二个方面提供β-葡聚糖酶和木聚糖酶在生产乙醇的方法中的用途。
本发明的方法中,应用稀释酶(thinning enzyme)如β-葡聚糖酶和木聚糖酶降解葡聚糖和木聚糖,以此减小糖化醪的粘性。减小的粘性增强了液化糖化醪的流加速率,由此,尤其是通过改善热转移以及促进液化糖化醪通过糖化醪冷却器,提高了工厂的生产能力。如此,本发明的方法有利于发酵中利用更高的干物质百分率,而且仍然确保有效冷却,确保输送到发酵槽中的糖化醪的温度正确且一致。
类似***糖基木聚糖和β-葡聚糖的非淀粉多糖的前期水解对蒸馏过程的作用是使生产量全面增加,以及获得更好的热转移和相转移。
非淀粉多糖的前期水解对副产品如蒸馏器干颗粒(distiller`s dry grain)的作用是使食品转化全面改善,营养物质如矿物质、蛋白质、脂类和淀粉的消化性更好。
发明详述
本发明的方法可用于大量发酵产品的生产中,所述发酵产品包括但不限于醇(尤其是乙醇);酸,如柠檬酸、衣康酸、乳酸、葡糖酸、赖氨酸;酮;氨基酸,如谷氨酸,也包括更为复杂的化合物如抗生素,如青霉素、四环素;酶;维生素,如核黄素、B12、β-胡萝卜素;激素,如胰岛素。优选可饮用的乙醇,还有工业乙醇及燃料用乙醇。
原料
任何含淀粉的材料均可用作本发明方法中的原料。在一个实施例中,含淀粉的材料是从谷物(cereal)中获得的完整谷粒,优选选自玉米、小麦、大麦、燕麦、水稻、木薯(cassava)、高粱、黑麦、买罗高梁(milo)和粟(millet)组成的组中。此外,含淀粉的材料可由马铃薯、甘薯(sweet potato)、木薯、木薯粉(tapioca)、西米(sago)、香蕉、糖用甜菜(sugar beet)和/或甘蔗(sugar cane)获得。甘蔗或糖用甜菜可以如GB 2115820A和US4886672A1所述加以利用。本发明优选谷类,例如小麦、大麦、燕麦、黑小麦(triticale),尤其是燕麦和大麦,以及源于谷物例如小麦、大麦、燕麦、黑小麦、尤其是燕麦和大麦的麦芽(malt)。由小麦、大麦、燕麦和黑小麦制成的高粘性浆经稀释是有利的。
方法步骤
在一个实施例中,本发明的主要方法步骤可以分为以下主要处理阶段描述:(a)糖化醪形成;(b)预液化;(c)凝胶化;(d)液化;和(e)糖化及发酵,其中步骤(a),(b),(c)和(d)以(a),(b),(c),(d)和(e)的顺序进行。步骤(e)可以是同时进行糖化及发酵(SSF)或作为两个分离的子步骤进行。
酒精生产的单个方法步骤可以一批批地进行或者作为连续的流程来进行。对于本发明,所有方法步骤均以批次方式进行,或者所有方法步骤均以连续的流程方式进行,或者一个或多个方法步骤以批次方式进行而一个或多个方法步骤以连续的流程方式进行,这些方法都是同等优选的。

磨碎(milling)

糖化醪形成

典型地,浆桶(包含磨碎的全部谷粒)中的干燥固体%(干燥固体百分比)在1-60%范围,特别是10-50%,如20-40%,如25-35%。
预液化
预液化步骤中,含淀粉的材料(前端糖化醪)保存在有稀释酶如β-葡聚糖酶或木聚糖酶存在的环境,稀释酶优选β-葡聚糖酶,温度在45-70℃,更优选53至66℃,最优选55至60℃,例如58℃。预液化步骤的持续时间优选5至60分钟,更优选10至30分钟,如15分钟左右。
凝胶化
在凝胶化步骤中淀粉被凝胶化。可以通过将含淀粉的浆加热至高于所使用的特定淀粉的凝胶化温度的温度来达至凝胶化。优选在适当条件下通过喷射蒸煮达至凝胶化,例如在95-140℃之间的温度,优选105-125℃,如在120℃条件下完成淀粉的凝胶化。通过非压力蒸煮达到凝胶化也是优选的。凝胶化期间,在预液化步骤中加入的酶将被置于升高的温度条件下,可能会全部或部分失活。因此,根据本发明,优选在凝胶化步骤之后加入新的稀释酶。
在一个实施例中,液化处理在pH4.5-6.5的条件下实施,尤其是在5和6之间的pH条件下实施。
喷射蒸煮期间,在预液化步骤中加入的酶将被置于升高的温度条件下,可能会全部或部分失活。因此,根据本发明,优选在喷射蒸煮步骤之后加入新的稀释酶。
液化
液化步骤中,凝胶化的淀粉(下游糖化醪)被打断(水解)为麦芽糊精(maltodextrins)(糊精)。为使淀粉水解,需加入适当的酶,优选α-淀粉酶。
根据本发明,向糖化醪中加入β-葡聚糖酶和木聚糖酶。在一个实施例中,进一步加入内切葡聚糖酶(endo-glucanase)。
液化步骤期间的温度为60-95℃,优选80-90℃,优选在70-80℃例如85℃,时间为1-120min,优选2-60min,例如12min。令人吃惊的是,液化步骤期间,在这样高的温度发挥酶功能。
在一个实施例中,步骤(d)的液化在约4-7的pH范围内进行,优选约4.5-6.5的pH。在一个优选的实施例中,液化期间的pH至多约为5。浆的pH可以调整,也可不调整,其依赖于所用的酶的特性。因此,在一个实施例中,加入如NH3将pH上调约1个单位。在加入α-淀粉酶时可方便地调整pH。在另外一个实施例中没有调整pH,α-淀粉酶具有相应适合的pH-活性谱,例如在pH约为4时具有活性。
在本发明的一个实施例中,稀释酶与α-淀粉酶一起加至凝胶化的糖化醪。
糖化与发酵
糖化步骤和发酵步骤可以作为分离的方法步骤分开进行,或者糖化与发酵步骤同时进行(SSF)。在存在糖化酶,例如葡萄糖淀粉酶、β-淀粉酶或麦芽糖淀粉酶的情况下进行糖化。任选加入植酸酶(phytase)和/或蛋白酶。
发酵生物可以是真菌,例如酵母,或者是细菌。合适的细菌可以是例如发酵单胞菌属(Zymomonas)物种,例如运动发酵单胞菌(Zymomonas mobilis)和大肠杆菌(E.coli.)。丝状真菌的例子包括青霉菌属(Penicillium)菌株。优选的乙醇生产生物为酵母,例如毕赤酵母属(Pichia)或者糖酵母属(Saccharomyces)酵母。根据本发明的优选酵母为糖酵母属物种,尤其是啤酒糖酵母(Saccharomyces cerevisiae)或者面包酵母。酵母细胞的加入量可以是每ml发酵肉汤105至1012,优选107-1010,尤其优选5×107活酵母数。在乙醇生产阶段酵母细胞数应优选在107-1010范围,尤其优选约2×108。有关使用酵母进行发酵的进一步指导可以参见例如,″The alcohol Textbook″(Editors K.Jacques,T.P.Lyons and D.R.Kelsall,Nottingham University Press,UnitedKingdom 1999),此处一并作为参考。
将用于发酵的微生物加至糖化醪,发酵过程一直进行到生产出所需的发酵产品的量;在一个实施例中,将乙醇作为发酵产品来回收,此时间可能为例如24-96小时,如35-60小时。发酵期间的温度和pH处于适合于所讨论的微生物的温度和pH,并且要考虑发酵产品将来的用途,例如,在一个实施例中,发酵微生物是酵母,所要回收的发酵产品是乙醇,优选的温度在约26-34℃,如约32℃,pH在约3-6范围,如约4-5的pH。
在另一个实施例中,发酵微生物是酵母,发酵的糖化醪将被用作啤酒,糖化醪的温度优选约12-16℃,如约14℃。
在一个优选实施例中使用同时糖化与发酵(SSF)的方法,其中没有针对糖化的保温阶段,其意思是酵母和糖化酶本质上是一起加入的。一个实施例中,当实施SSF时,在刚刚要发酵前,在高于50℃的温度条件下增加预糖化步骤。
蒸馏
本发明的方法可以进一步包含回收发酵产品如乙醇;因此,酒精可以从发酵的材料分离纯化。
这样,在一个实施例中,本发明的方法进一步包含步骤:(f)蒸馏以获得乙醇。
由蒸馏得到的副产品
通过例如离心可将蒸馏过程中产生的副产品水溶液(Whole Sillage)分为两个组分:1)Wet Grain(固相),和2)Thin Stillage(悬浮物)。
将Wet Grain干燥,通常是在转鼓式干燥器中干燥。干燥后的产品称为“蒸馏器干颗粒”,可用作动物食物。
可以将Thin Stillage组分蒸发,得到两个组分:-4-6%干燥固体(主要是淀粉、蛋白质和细胞壁成分)的冷凝组分,和-浆状组分,主要由有限糊精和不能发酵的糖组成,其可以与Wet Grain(来自Whole Stillage分离步骤)一起放入干燥器以获得称为“蒸馏器干颗粒”的产品,该产品可用作动物食物。
本领域中,术语“Whole Stillage”用于指来源于发酵糖化醪蒸馏产生的副产品。
本领域中,术语“Thin Stillage”用于指Whole Stillage离心所得的悬浮物(上层)。典型地,Thin Stillage包含4-6%的干燥固体(主要是淀粉和蛋白质),温度为约60-90℃。Thin Stillage是粘性的,难以处理。通常Thin Stillage在再循环至浆桶之前先盛放在存储桶中至数小时。再循环前,可以用适当的酶如β-葡聚糖酶和木聚糖酶稀释stillage。
有关如何实施液化、糖化、发酵、蒸馏和回收乙醇的详细信息是熟练技术人员所熟知的。
由本发明的方法所生产的产品的用途
发酵产品是乙醇的实施例中,可以从发酵的糖化醪回收由本发明的方法所获得的乙醇,并用作例如燃料乙醇;饮用乙醇,如适于饮用的酒精,或者工业用乙醇,包括燃料添加剂。
发酵产品是乙醇,且不从发酵的糖化醪回收由本发明的方法所获得的乙醇的实施例中,包含乙醇的糖化醪可以用作啤酒。所述啤酒可以是包括ales、strong ales、bitters、烈性啤酒(stouts)、porters、lagers、export beers、麦芽酒(malt liquor)、大麦酒(barley wine)、happoushu、high-alcohol beer、low-alcoholbeer、低热量啤酒(low-calorie beer)和轻淡啤酒(light beer)的任何啤酒。
在发酵产品并非乙醇的实施例中,产品可以用于任何适当的目的。
酶活性
β-葡聚糖酶(E.C.3.2.1.4)
β-葡聚糖酶可以是微生物来源的,例如源于细菌(例如芽孢杆菌(Bacillus))或丝状真菌(例如曲霉属(Aspergillus)、木霉属(Trichoderma)、腐质霉属(Humicola)、镰刀霉属(Fusarium))的菌株。

可以0.01-5000BGU/kg干燥固体的量加入β-葡聚糖酶,优选以0.1-500BGU/kg干燥固体的量,最优选以1-50BGU/kg干燥固体的量,在液化步骤(下游糖化醪)中以1.0-5000BGU/kg干燥固体的量,最优选以10-500BGU/kg干燥固体的量。
木聚糖酶(EC3.2.1.8和其他)
在有效量的合适的木聚糖酶存在下实施本发明的方法,该木聚糖酶可源于多种微生物,包括真菌和细菌,例如曲霉、Disporotrichum、青霉、链孢霉(Neurospora)、镰刀霉和木霉。
合适的木聚糖酶的例子包括源于H.insolens(WO 92/17573);塔宾曲霉(Aspergillus tubigensis)(WO 92/01793);黑曲霉(A.niger)(Shei等,1985,Biotech.and Bioeng.Vol.XXVII,pp.533-538,和Fournier等,1985,Bio-tech.Bioeng.Vol.XXVII,pp.539-546;WO 91/19782和EP 463706);棘孢曲霉(A.aculeatus)(WO 94/21785)的木聚糖酶。
木聚糖酶也可以是1,3-β-D-xylan xylanohydrolase(EC.3.2.1.32)。
特定实施例中的木聚糖酶是WO 94/21785公开的木聚糖酶II。
可考虑的商业上可获得的包含木聚糖酶的组合物包括SHEARZYMES200L、SHEARZYME500L、BIOFEED WHEAT、PULPZYMETM HC(来自Novozymes)和GC 880、SPEZYMECP(来自Genencor Int)。
可以以1.0-1000FXU/kg干燥固体的量加入木聚糖酶,优选以5-500FXU/kg干燥固体的量,再优选以5-100FXU/kg的量,最优选以10-100FXU/kg干燥固体的量。
α-淀粉酶(E.C.3.2.1.1)
优选的α-淀粉酶为真菌或细菌来源。
根据本发明,芽孢杆菌α-淀粉酶(通常指“Termamyl样α-淀粉酶”)、其变体和混合物是优选的。熟知的Termamyl样α-淀粉酶包括源于地衣芽孢杆菌(B.licheniformis)(商业上可获得,商品名TermamylTM)、解淀粉芽孢杆菌(B.amyloliquefaciens)、和嗜热脂肪芽孢杆菌(B.stearothermophilus)菌株的α-淀粉酶。合适的细菌α-淀粉酶可以是源于嗜热脂肪芽孢杆菌的α-淀粉酶,其具有WO99/19467中SEQ.NO:4的氨基酸序列。
合适的真菌α-淀粉酶可以源于曲霉属,例如源于黑曲霉的酸性真菌α-淀粉酶。
商业α-淀粉酶产品和含有α-淀粉酶的产品包括TERMAMYLTM SC、FUNGAMYLTM、LIQUOZYMETM SC和SANTM SUPER,(Novozymes A/S,Denmark)和DEX-LOTM、SPEZYMETM AA、和SPEZYMETM DELTA AA(来自Genencor Int.)。
液化步骤中,可以以0.001-1.0AFAU/g干燥固体的量加入真菌α-淀粉酶,优选以0.002-0.5AFAU/g干燥固体的量,优选以0.02-0.1AFAU/g干燥固体的量。
可以以本领域熟练技术人员熟知的有效量加入芽孢杆菌α-淀粉酶。
麦芽糖淀粉酶
α-淀粉酶可以是麦芽糖α-淀粉酶。麦芽糖淀粉酶(葡聚糖1,4-α-麦芽糖水解酶,E.C.3.2.1.133)能够将直链淀粉和支链淀粉水解为α构型的麦芽糖。此外,麦芽糖淀粉酶还能够水解麦芽三糖以及环状糊精。特别要考虑的麦芽糖淀粉酶包括EP专利no.120,693所公开的来自嗜热脂肪芽孢杆菌C599的一个麦芽糖淀粉酶。商业上可获得的麦芽糖淀粉酶是来自Novozymes A/S的MALTOGENASETM。
葡萄糖淀粉酶
可以在葡萄糖淀粉酶存在下实施糖化步骤或同时糖化与发酵步骤(SSF)。葡萄糖淀粉酶可以是任何来源,例如来源于微生物或植物。选自由黑曲霉葡萄糖淀粉酶,尤其是黑曲霉G1或G2葡萄糖淀粉酶(Boel etal.(1984),EMBO J.3(5),p.1097-1102)或其变体,例如WO92/00381和WO00/04136所公开的;泡盛曲霉(A.awamori)葡萄糖淀粉酶(WO 84/02921)、米曲霉(A.oryzae)葡萄糖淀粉酶(Agric.Biol.Chem.(1991),55(4),p.941-949)或其变体或片段所组成的组的真菌或细菌来源的葡萄糖淀粉酶是优选的。
商业产品包括SANTM SUPERTM和AMGTM E(来自Novozymes A/S)。在一个实施例中,可以在糖化及发酵步骤(e)以0.02-2AGU/g干燥固体的量加入葡萄糖淀粉酶,优选以0.1-1AGU/g干燥固体的量,假如0.2AGU/g干燥固体的量。
蛋白酶
在糖化步骤、SSF步骤和/或发酵步骤,蛋白酶的加入提高FAN(游离氨基酸)水平,增强酵母代谢率,并且可以提高发酵效率。
合适的蛋白酶包括微生物蛋白酶,例如真菌和细菌蛋白酶。优选的蛋白酶是酸性蛋白酶,例如,以在低于7的酸性pH条件下水解蛋白质的能力为特征的蛋白酶。
合适的酸性真菌蛋白酶包括源于曲霉属、毛霉属(Mucor)、根霉属(Rhizopus)、假丝酵母属(Candida)、革盖菌属(Coriolus)、内座壳属(Endothia)、Enthomophtra、耙齿菌属(Irpex)、青霉菌属、小菌核属(Sclerotium)和球拟酵母菌属(Torulopsis)的蛋白酶。特别要考虑的是源于黑曲霉(参见,如,Koaze等,(1964),Agr.Biol.Chem.Japan,28,216)、斋腾曲霉(Aspergillus saitoi)(参见,如,Yoshida,(1954)J.Agr.Chem.Soc.Japan,28,66)、泡盛曲霉(Hayashida等,(1977)Agric.Biol.Chem.,42(5),927-933)、棘孢曲霉(WO95/02044)、或者米曲霉、例如pepA蛋白酶;和来自Mucor pusillus或Mucormiehei的酸性蛋白酶。
ALCALASETM是地衣芽孢杆菌蛋白酶(subtilisin Carlsberg)。根据本发明,可以优选以10-7至10-3克活性蛋白酶蛋白/g干燥固体的量加入ALCALASETM。尤其以10-6至10-4克活性蛋白酶蛋白/g干燥固体的量加入,或者以0.1-0.0001AU/g干燥固体的量加入,优选以0.00025-0.001AU/g干燥固体的量加入。
FLAVOURZYMETM(可由Novozymes A/S获得)是源于米曲霉的蛋白酶制剂。根据本发明,可以优选以0.01-1.0LAPU/g干燥固体的量加入FLAVOURZYMETM,优选以0.05-0.5LAPU/g干燥固体的量加入。
蛋白酶的合适剂量在10-7至10-3克活性蛋白酶蛋白/g干燥固体的范围,尤其是10-6至10-4克活性蛋白酶蛋白/g干燥固体的范围。
植酸酶:
根据本发明,所用的植酸酶可以是能够从植酸(myo-inositolhexakisphosphate)或其(肌醇六磷酸)任何形式的盐中释放无机磷酸的任何酶。
植酸酶的合适剂量在0.005-25FYT/g干燥固体的范围内,优选0.01-10FYT/g干燥固体,例如0.1-1FYT/g干燥固体。
材料与方法
方法
木聚糖酶活性(FXU)的测定
通过测试测定木聚糖内切酶的活性,该测试中,将木聚糖酶样品与remazol木聚糖底物(用Remazol亮蓝R染色的4-O-methyl-D-glucurono-D-xylan,Fluka)在pH6.0的条件下一起保温。50℃保温30分钟。用乙醇沉淀未降解的染色底物背景。585nm处以分光光度法测定悬浮物中的剩余蓝色物质,其与木聚糖内切酶活性成比例。与标准酶对照以测定样品的木聚糖内切酶活性。
出版物AF 293.6/1-GB中进一步描述了该测试,其可以从Novo NordiskA/S,Denmark得到。
β-葡聚糖酶活性(BGU)的测定
可以用β-葡聚糖酶单位(BGU)的方式测量纤维分解酶活性。β-葡聚糖酶与β-葡聚糖反应生成葡萄糖或还原性碳水化合物,将其作为还原性糖,用Somogyi-Nelson方法测定。标准条件下,1个β-葡聚糖酶单位指每分钟释放相当于1μmol葡萄糖还原能力的葡萄糖或还原性碳水化合物的酶量。标准条件为0.5%β-葡聚糖作为底物,在pH7.5的条件下,30℃反应30分钟。该分析方法(EB-SM-0070.02/01)的详细描述可由NovozymesA/S获得。
内切葡聚糖酶活性(EGU)的测定
可以用内切葡聚糖酶单位(BGU)的方式测量纤维分解酶活性,其在pH6.0条件下以纤维素(CMC)为底物测定。
所制备的底物溶液为pH6.0的0.1M磷酸缓冲液中含有34.0g/lCMC(Hercules 7 LFD)。所要分析的酶样品溶解于同样的缓冲液中。将5ml底物溶液和0.15ml酶溶液混合,转至振动粘度计(例如来自Sofraser,France的MIVI 3000),自动调温至40℃处理30分钟。
一个EGU定义为在这些条件下将粘度减小至一半的酶量。需将反应混合物中酶样的量调至0.01-0.02EGU/ml。arch标准定义为880EGU/g。
葡萄糖淀粉酶活性(AGU)的测定
Novo葡萄糖淀粉酶单位(AGU)定义为,37℃及pH4.3条件下每分钟水解1微摩尔麦芽糖的酶量。
在使用来自Boehringer Mannheim,124036的Glucose GOD-Perid试剂盒后,用改进的方法(AEL-SM-0131,可由Novozymes获得)测定以AGU/ml计的活性。标准:AMG-standard,7-1195批,195AGU/ml。375microL底物((50mM醋酸钠中1%的麦芽糖,pH4.3)37℃保温5分钟。加入25microL于醋酸钠中稀释的酶。10分钟后加入100microL 0.25M NaOH终止反应。将20microL转至96孔微量滴定盘,加入200microL GOD-Perid溶液(124036,Boehringer Mannheim)。室温30分钟后,650nm处测吸光值,从AMG-standard计算以AGU/ml计的活性。该分析方法(AEL-SM-0131)的详细描述可由Novozymes获得。
α-淀粉酶活性(KNU)的测定
可以使用马铃薯淀粉作为底物测定淀粉分解活性。该方法以通过酶打断修饰的马铃薯淀粉为基础,该反应之后将淀粉/酶溶液样品与碘溶液混合。最初,形成蓝黑色物质,但在淀粉打断期间,蓝色变弱,并逐渐变成红褐色,将其与有色玻璃标准对比。
一个Kilo Novoα-淀粉酶单位(KNU)定义为:标准条件(即,37℃+/-0.05;0.003M Ca2+;pH5.6)下将5260mg淀粉干物质Merck Amylum soluble转化为糊精的酶量。

蛋白酶(LAPU)的测定
1个亮氨酸氨肽酶单位(LAPU)为在下述条件下每分钟分解1 microM底物的酶量:26mM的底物L-亮氨酸-p-硝基苯(nitroanilide),0.1M Tris缓冲液(pH8.0),40℃,反应时间10分钟。
实施例
实施例中所用的酶:
包含源于解淀粉芽孢杆菌的β-葡聚糖酶的组合物;1200BGU/g。
包含公开于WO94/21785的木聚糖酶II的组合物;该木聚糖酶II为1-4-β-木聚糖内切酶;521FXU/g。
包含内切葡聚糖酶活性和一些木聚糖酶和β-葡聚糖酶活性的源于里氏木霉(Trichoderma reesei)的组合物;700EGU/g,50FXU/g,和60BGU/g。
可由Genencor Int.获得的称为GC 880“工程化纤维素酶复合体”的组合物,至少包含β-葡聚糖酶和木聚糖酶活性;59BGU/g和222FXU/g。
实施例1
在装有700ml水的1公升瓶中,用300g粉碎的大麦粉制备前端浆。pH调至5.2,在温控型水浴中将糖化醪从室温(25℃)加热至54-60℃。
依照表1的剂量测试不同的酶组合。用Haake Viscotester VT-02测量粘性。

实施例2
在用细菌α-淀粉酶液化过的浆中,测试了用上述提到的非淀粉降解酶得到的粘性减小。28%干燥固体的浆为DE 16,pH为5.0。将浆分装到1公升的瓶中,保持在84℃的温控型水浴中。依照表3的剂量测试了不同的酶组合。用Haake Viscotester VT-02测量了粘性,粘性为时间的函数,见表4。
β-葡聚糖酶+木聚糖酶II+内切葡聚糖酶组合比单独的产品GC 880或β-葡聚糖酶或木聚糖酶II+内切葡聚糖酶更有效地发挥作用。

实施例3

实施例4
在实验室里,为模拟下游程序,将磨碎的黑麦移入相应量的水中,制得具有20%谷粒干物质的1kg浆。75℃下,用0.5kg Termamyl SC/tons谷物(按目前的样子)的剂量进行液化。然后,70℃下用减小粘性的酶处理浆,在70℃,pH=6-6.5的条件下测量。这个作为测试模型来实施,目的在于测试减小粘性的酶的效果。工业上,Termamyl+降解非淀粉多糖的粘性减小的酶需同时操作。
除了温度为70℃外,按上述方法在t=3、15、30、60分钟时用HAAKEViscotester VT-02测量粘性。测量粘性后,在样品上测量温度。

Claims (19)
1.生产发酵产品的方法,所述方法包含在β-葡聚糖酶的存在下,非淀粉多糖的预液化,然后凝胶化,接下来在热稳定性β-葡聚糖酶和木聚糖酶的存在下液化。
2.生产发酵产品的方法,所述方法包含在β-葡聚糖酶的存在下,非淀粉多糖的预液化,然后在热稳定性β-葡聚糖酶和木聚糖酶的存在下喷射蒸煮及液化。
3.生产发酵产品的方法,所述方法包含步骤:
a.提供包含含淀粉的材料和水的糖化醪;
b.在β-葡聚糖酶存在下预液化步骤(a)的糖化醪;
c.将步骤(b)的糖化醪凝胶化;
d.在α-淀粉酶、β-葡聚糖酶和木聚糖酶的存在下液化步骤(c)的糖化醪;和
e.糖化及发酵步骤(d)的糖化醪以生产发酵产品。
4.前述任一项权利要求的方法,在液化步骤(d)之后和步骤(e)之前进一步包含预糖化步骤。
5.前述任一项权利要求的方法,进一步包含发酵产品的回收。
6.前述任一项权利要求的方法,其中发酵产品是:醇(尤其是乙醇);酸,例如柠檬酸、衣康酸、乳酸、葡糖酸、赖氨酸;酮;氨基酸,例如谷氨酸;抗生素,例如青霉素、四环素;酶;维生素,例如核黄素、B12、β-胡萝卜素;激素,例如胰岛素。
7.前述任一项权利要求的方法,其中木聚糖酶源于曲霉菌菌株,优选源于棘孢曲霉菌株。
8.前述任一项权利要求的方法,其中β-葡聚糖酶源于芽孢杆菌菌株,优选源于解淀粉芽孢杆菌菌株。
9.前述任一项权利要求的方法,其中在液化步骤(d)中还存在内切葡聚糖酶,所述内切葡聚糖酶优选源于木霉菌菌株,优选源于里氏木霉菌株。
10.前述任一项权利要求的方法,其中含淀粉的材料从谷物和/或块茎获得。
11.前述任一项权利要求的方法,其中含淀粉的材料选自由玉米、小麦、大麦、黑麦、粟、高粱和买罗高粱组成的组。
12.前述任一项权利要求的方法,其中含淀粉的材料选自由马铃薯、甘薯、木薯、木薯粉、西米、香蕉、糖用甜菜和甘蔗组成的组。
13.前述任一项权利要求的方法,其中用微生物,例如细菌和真菌(包括酵母)如发酵单胞菌属物种和糖酵母属物种如啤酒糖酵母实施步骤(e)的发酵。
14.前述任一项权利要求的方法,其中在植酸酶和/或蛋白酶的存在下进行发酵。
15.前述任一项权利要求的方法,其中步骤(b)的预液化在45-70℃,更优选在53-66℃,最优选在55-60℃,例如58℃的温度进行5-60分钟,更优选10-30分钟,例如约15分钟。
16.前述任一项权利要求的方法,其中步骤(d)的液化在60-95℃,优选在80-90℃的温度进行10-120min,更优选在83-85℃的温度进行15-80min。
17.热稳定性β-葡聚糖酶和木聚糖酶在乙醇生产方法中之液化步骤中的应用。
18.根据前一项权利要求的应用,其中用于乙醇生产方法中的含淀粉的原材料如权利要求6-8任一项所定义。
19.根据前一项权利要求的应用,其中乙醇用作饮用酒精、燃料酒精和/或燃料添加剂。
