Abstract
本发明提供含有促生长激素释放因子受体(GHS-R)拮抗剂作为有效成分的糖尿病预防剂或者治疗剂、以及其特征在于使用GHS-R拮抗剂的血糖值降低方法。本发明还提供含有GHS-R拮抗剂作为有效成分的肥胖预防剂或者治疗剂、以及含有GHS-R拮抗剂作为有效成分的食欲抑制剂。

技术领域
本发明涉及新型的糖尿病预防剂或者治疗剂。进一步详细地,涉及含有促生长激素释放因子受体(GHS-R)拮抗剂作为有效成分的糖尿病预防剂或者治疗剂。另外,还涉及以使用GHS-R拮抗剂为特征的降低血糖值方法。本发明进一步涉及含有GHS-R拮抗剂作为有效成分的肥胖预防剂或者治疗剂,以及含有GHS-R拮抗剂作为有效成分的食欲抑制剂。
背景技术
近年,在我国伴随着膳食生活的欧美化迎来了“饱食时代”,并且,另一方面通过以汽车为代表的机械化文明的普及,慢性的运动不足状态在国民中间广泛的蔓延开来。在这样的社会背景下,最近的糖尿病人口的增加令人惊异,据说目前的数字如果包括潜在患者在内则达到500万或600万人。
糖尿病有2种类型。1型糖尿病(胰岛素依赖型糖尿病(IDDM))多见于儿童、年青人,该疾病是由于病毒感染等引起产生、分泌胰岛素的胰腺的郎格罕岛(胰岛)被破坏,变得完全不能分泌胰岛素,从而致糖尿病。一方面,多见于中老年人的2型糖尿病(非胰岛素依赖型糖尿病(NIDDM))占日本人糖尿病的绝大部分(约95%),对于有着胰岛素分泌量容易降低、易患糖尿病的体质的人来说,加上类似饮食过度、运动不足、肥胖、应激、年龄增长等妨碍胰岛素作用的诱因而出现症状。
糖尿病与肥胖有着很深的关系,根据调查,非胰岛素依赖型糖尿病患者中约2/3现在为肥胖或者过去有肥胖经历。实际上,已知肥胖者的胰岛素的血糖降低作用变弱。
肥胖是世界上特别在发达社会中正越来越普遍化的重要的健康问题。在美国,成人有半数以上体重过剩。Allison等在报告中称,1991年美国死亡人数的约280000人可以归因于肥胖。(Allison DB et al.,JAMA,280:1530-1538,1991)。肥胖的病理生理被认为是持续摄取超过消耗的过剩营养。有显示,脂肪含量高的“西式饮食”与肥胖的危险性增加有关。
体重调节的关键是食物摄入量与能量消耗的平衡,二者的对比关系引起肥胖、瘦。自明确了1994年发现的瘦素作为肥胖(机体脂肪量蓄积)信号与体重调节的根本有关以来,发现了位于瘦素下游的、很多新的与食欲调节相关的肽。特别是了解到,到那时为止被认为只能各自独立发挥功能的来自下丘脑的神经肽群,在瘦素下游各自发挥着功能,进而也在这些神经肽群相互之间密切地进行着信息交换。
在这些神经肽中,作为促进食欲的物质,据了解有神经肽(NPY)、食欲素类(orexins)、胃动素(motilin)、黑色素浓缩素(melanin-concentratinghormone:MCH)、刺相关蛋白质(agouti-related protein:AGRP)。另外,作为抑制食欲的物质,已知有α-黑色素细胞刺激素(α-melanocyte-stimulating hormone:α-MSH)、促肾上腺皮质激素释放因子(corticotropin-releasing factor:CRF)、***和安非他明调控转录物(***-and amphetamine-regulated transcript:CART)、胆囊收缩素(cholesystokinin:CCK)等。认为这些肽与控制食欲的生理学机制相关,且对能量恒常性有影响。
一方面,生长激素(GH)是从垂体前叶分泌的激素,其分泌被巧妙地控制着,受到丘脑下部的生长激素释放激素(GHRH)的刺激,并被生长激素抑制素所抑制。近年来,逐渐明确GHRH、生长激素抑制素是通过不同途径的GH分泌调节机构。该不同途径的GH分泌调节结构通过对作为具有促进GH分泌活性的合成化合物的促生长激素释放因子(growthhormone secretagogue:GHS)的研究被开展起来的。GHS以不同于GHRH的途径发挥作用。即,针对GHRH激活GHRH受体而使细胞内cAMP浓度上升,GHS激活不同于GHRH受体的受体、通过细胞内IP3系使细胞内Ca2+浓度上升。作为该GHS作用的受体的GHS-R,其构造通过1996年发现的克隆法而被弄清楚。GHS-R是7次贯穿细胞膜的典型的G蛋白共轭型受体,主要存在于下丘脑、垂体。
进而,因为存在与作为生物体内不存在的合成化合物的GSH相结合的受体,所以探索到与该GHS-R结合而激活的内因性配体。其结果,作为GHS-R的特异性配体,饥饿激素(Ghrelin)是从大鼠的胃精制、鉴别而来的(Kojima M.et al.,Nature,402:656-660,1999)。
饥饿激素是由28个氨基酸残基所组成的肽,第3位的丝氨酸残基被正辛酰化。另外,人的饥饿激素与大鼠的饥饿激素有2个氨基酸残基是不同的。下面显示大鼠以及人的饥饿激素的结构式。
化学合成的饥饿激素是纳摩尔级的,具有表达GHS-R的CHO细胞的细胞内Ca2+升高活性、在初代培养垂体细胞的生长激素释放活性。进而,即使在活体内(in vivo)也使大鼠的血液中生长激素升高。饥饿激素的mRNA在胃中显著表达,另外饥饿激素在血液中也存在。进而,GHS-R也存在于下丘脑、心脏、肺、胰腺、小肠、脂肪组织中(上述Kojima等)。另外,饥饿激素被报告具有摄食促进作用(Wren et al.,Endocrinology,141(11):4325-4328,2000)。本发明者等发现饥饿激素通过NPY和Y1受体显示了显著的食欲促进作用,并建议饥饿激素作为出现低营养症状的疾病的治疗剂是有作用的(PCT/JP02/00765)。

本发明的目的在于,开发新型的糖尿病预防剂或者治疗剂、以及肥胖预防剂或者治疗剂。本发明进一步的目的在于,提供新型的血糖值降低方法、肥胖预防剂或者治疗剂、以及食欲抑制剂。
本发明者等发现了促生长激素释放因子受体(GHS-R)拮抗剂明显降低血糖值,及显著抑制食欲,从而完成了本发明。
即,本发明提供含有促生长激素释放因子受体(GHS-R)拮抗剂作为有效成分的糖尿病预防剂或者治疗剂。
本发明进而提供以使用GHS-R拮抗剂为特征的血糖值降低方法。

具体实施方式
在本发明中作为有效成分使用的GHS-R拮抗剂,是指能够与GHS-R相结合来阻碍拮抗剂的效果的物质。作为GHS-R拮抗剂,能够使用饥饿激素或者饥饿激素类似物的拮抗剂。
饥饿激素或者饥饿激素类似物的拮抗剂,能够通过例如下面所述的筛选方法进行鉴别(参照PCT/JP02/00765)。
即,在饥饿激素或者饥饿激素类似物的存在情况下或者不存在情况下,把候补物质投放给动物,测定摄食量、NPY mRNA表达量、NPY与NPY的Y1受体的结合量、耗氧量、胃排空速度、或者迷走神经的活性,由此能够筛选饥饿激素或者饥饿激素类似物的拮抗剂,但是并不限于此。
能够将通过上述筛选方法得到的饥饿激素或者饥饿激素类似物的拮抗剂作为糖尿病预防剂或者治疗剂、肥胖预防剂或者治疗剂、及食欲抑制剂的有效成份而使用。
在本发明中,作为饥饿激素或者饥饿激素类似物的拮抗剂,也能够使用胃动素或者胃动素类似物的拮抗剂。
胃动素是由十二指肠、空肠上部的内分泌细胞分泌的22个氨基酸残基的肽(Itoh,Z.,Peptides,18:593-608,1997),与消化道的空腹期运动、胆囊收缩及胃、胰腺的酶分泌有关。有报告胃动素促进GH分泌,还有报告使用非肽性的胃动素拮抗剂来促进胃的运动(见上,Itoh)。如图1A所示,人饥饿激素和人胃动素的氨基酸彼此显示了36%的氨基酸同一性(存取编号A59316及P12872)。进而,如图1B所示,人饥饿激素受体和人胃动素受体在整体上显示了50%的氨基酸同一性(存取编号Q92847、Q92848及Q43193)。另外,最近,Tomasetto等从小鼠的胃中分离出新型肽,该肽与饥饿激素是一样的,被命名为胃动素相关肽(Tomasetto C.et al.,Gastroenterology,119:395-405,2000)。于鉴饥饿激素与胃动素的序列相同性以及饥饿激素受体和胃动素受体的序列相同性,作为饥饿激素或者饥饿激素类似物的拮抗剂,能够使用胃动素或者胃动素类似物的拮抗剂。
再者,饥饿激素是用式1所表示的大鼠饥饿激素或者人饥饿激素或者饥饿激素类似物。
在本发明所说的饥饿激素类似物中,只要具有食欲促进作用,也包括28个氨基酸中有1个以上的氨基酸缺失、取代或者附加的类似物,进而也包扩它们的各种衍生物[例如,取代肽组成氨基酸的衍生物(也包括在氨基酸之间***基团,例如亚烷基的衍生物)及酯衍生物]。
饥饿激素或者饥饿激素类似物可以用任何方法制造而,例如包括从人、大鼠细胞分离、精制而成的饥饿激素或者饥饿激素类似物、合成品、半合成品、由遗传工程得到的饥饿激素或者饥饿激素类似物等,且没有特别限制。
作为28个氨基酸中有1个以上的氨基酸缺失、取代或者附加的饥饿激素或者饥饿激素类似物的例子,去除了饥饿激素第14位的Gln残基的des-Gln 14-饥饿激素等具有代表性。大鼠des-Gln 14-饥饿激素是根据饥饿激素基因的剪接的不同而生成的,在大鼠胃中的量占饥饿激素的1/4,与生长激素释放活性的强度相同。
进而,在饥饿激素类似物中,也包括在J.MED.Chem.2000,43,4370-4376中记载的如下所述的物质。例如,可以举出:在饥饿激素的28个氨基酸当中具有从N末端第3位及第4位的氨基酸(优选N末端4个氨基酸)、且从N末端第3位氨基酸(Ser)的侧链被取代的肽及其衍生物,并具有生长激素游离促进作用。
作为从N末端第3位氨基酸的侧链的例子,可以举出作为饥饿激素的侧链的辛酰基以外的酰基及烷基(它们的碳原子数优选为6~18),作为具体的侧链,可以举出下述侧链:-CH2(CH2)9CH3、-CO-(CH2)6CH3、-CO-CH=CH-CH=CH-CH=CH-CH3、-CO-CH(CH2 CH2 CH3)2、-CO-(CH2)9CH3、-CO-(CH2)14CH3、-CO-(CH2)6CH2Br、-CO-CH(CH2)2CONH(CH2)2CH3、-COPh、

作为具有从N末端第3位及第4位的氨基酸、并从N末端第3位氨基酸(Ser)的侧链被取代的饥饿激素类似物为具体的例子,可以举出在第37届肽讨论会(2000年10月18日~20日)上报告的化合物:NH2-(CH2)4-CO-Ser(辛基)Phe-Leu-NH-(CH2)2-NH2。
在本发明中,作为GHS-R拮抗剂,使用[D-Lys-3]-GHRP-6(VeeraragavanK.et al.,Life Sci.50:1149-55,1992)及[D-Arg-1,D-Phe-5,D-Trp-7,9,Leu-11]P物质(Cheng,K et al.,Journal of Endocrinology,152:155-158,1997)来确认本发明的效果。
本发明的预防剂或者治疗剂、以及抑制剂可以进行中枢给药(例如脑室内给药、脊髓腔内注射)、也可以末梢给药。优选通过末梢给药而使用。食欲调节肽中的多数在进行末梢给药时不显示中枢给药那样的作用,但是本发明的预防剂或者治疗剂、以及抑制剂即使是末梢给药也使血糖浓度明显降低。从而,伴随本发明的预防剂或者治疗剂、以及抑制剂的给药,能够减少患者的痛苦,并能够简便服用,与以往的食欲调节性肽相比,其优点相当大。
GHS-R拮抗剂能够通过公认的制剂技术,单独或者与药理学上可以接受的载体、添加剂等一同做成通常的口服用制剂及非口服用制剂。例如,能够被做成溶液制剂(动脉注射、静脉注射或者皮下注射等的注射剂、点鼻剂、糖浆等)、片剂、含剂、胶囊、粉剂、颗粒剂、软膏、栓剂等。并且,也可以在药物输送***(Drug Delivery System)(缓释剂等)中使用。
本发明的预防剂或者治疗剂、以及抑制剂的给药量,因患者的年龄、体重、症状、给药途径等而不同,并根据医生的判断而决定。通常,为了静脉给药,作为GHS-R拮抗剂,每1kg体重大约0.1μg~1000mg,优选约0.01mg~100mg、更优选0.1mg~10mg。但是,给药量并不限定于此。
本发明的糖尿病预防剂或者治疗剂能够用于糖尿病的预防或者治疗,特别是作为2型糖尿病(NIDDM:非胰岛素依赖型糖尿病)的预防剂或者治疗剂是有用的。本发明的肥胖治疗剂对于因肥胖而产生的缺血性心脏病、变形性关节炎、腰椎病、脂肪代谢异常、睡眠呼吸暂停综合征、***等病患有效。本发明的食欲抑制剂对于贪食症、应激性贪食、糖尿病性贪食等病患有效。
本发明者等如后述的实施例所记载的那样,在高脂肪饮食下反复使用饥饿激素后测定体重、脂肪质量、葡萄糖、胰岛素、及白色脂肪组织中的瘦素(leptin)、脂粘素(adiponectin)、抵抗素(resistin)基因的表达。另外,通过Northern blot分析测定胃的饥饿激素基因的表达。进而,在GHS-R拮抗剂给药之后对能量摄取及胃排空(gastric emptying)进行测定。对ob/ob肥胖小鼠进行GHS-R拮抗剂的反复给药,并持续6天,试验其效果。
其结果是,饥饿激素在高脂肪饮食下引起明显的脂肪蓄积(adiposity)并使血糖控制恶化。饥饿激素使瘦素mRNA表达上升,并使抵抗素mRNA表达减少。胃饥饿激素mRNA表达因高脂肪饮食而增加。
虽然GHS-R拮抗剂在瘦小鼠(lean mice)、在饮食诱导性(diet-induced)肥小鼠、然后在ob/ob肥小鼠身上使能量摄取减少,但是其作用机制与胃排空减少有关。GHS-R拮抗剂的反复给药使ob/ob小鼠的体重增加减少且使血糖控制提高。
因而,饥饿激素与特别是在高脂肪饮食下的体重过度增加、脂肪蓄积以及胰岛素抵抗性有密切的关联,这点已被确认。进而,可期待GHS-R拮抗剂会成为用于肥胖及2型糖尿病的有前途的预防或者治疗剂。
关于GHS-R拮抗剂对摄食的作用的考察
基于GHR-R拮抗剂可能诱导能量的负平衡(negative energy balance)状态的预测,研究GHS-R拮抗剂对于摄食的作用。通过GHS-R拮抗剂给药,瘦小鼠及高脂肪饮食而致肥胖的小鼠摄食减少。根据过去的报告,GHS-R存在于下丘脑、心脏、肺、胰腺、小肠及脂肪组织。在下丘脑、GHS-R位于弓状核(ARC),此处2个增进食欲肽、神经肽Y(NRY)及刺相关蛋白质(AGRP)在神经元被合成。进而,非肽GH分泌促进物质作用于下丘脑,其作用是使ARC神经元的电活性发生变化并激活c-fos的表达(例如,Lawrence et al.,Endocrinology,143:155-162,2002)。迄今为止,有报告称其作用机制是饥饿激素刺激与ARC内(此处血脑屏障作用弱)的下丘脑NPY及AGRP神经元的直接激活相关的摄食行为(例如,Inui,NatRev Neurosci,2:551-560,2001)。但是,来自胃的饥饿激素信号的其他途径是借助通过迷走神经与脑干神经核(brainstem nuclei)最后到达下丘脑的上行神经网。在本发明中,中枢给药的GHS-R拮抗剂停止针对由末梢给药的饥饿激素所诱导的摄食的刺激作用。由这些结果提示饥饿激素在脑内似乎通过GHS-R而发挥作用。进而明确了末梢给药的GHS-R拮抗剂使胃排空速度减少,但这有助于其食欲不振诱发作用(食欲抑制作用)。相当的证据积累起来,显示胃扩张作为饱腹信号发生作用,从而阻碍食物摄取,迅速的胃排空与贪食和肥胖相关联、迟缓的胃排空与食欲不振和恶液质相关联(Inui,Mol Psychiatry,6:620-624,2001)。根据过去的研究,显示饥饿激素通过迷走神经途径增加胃排空速度及运动性(Inui,Nat Rev Neurosci,2:551-560,2001)。
通过本发明,显示GHS-R在摄食行为的控制上具有一定的作用,GHS-R的拮抗作用可以成为用于肥胖治疗的有希望的方法。
进而,通过本发明,已经明确末梢给药的GHS-R拮抗剂引起ob/ob鼠(被认为是胰岛素抵抗性及迅速的胃排空而作为肥胖及糖尿病的遗传模型)的食欲不振作用,使体重减少和血糖浓度降低。通过这种伴随中等程度的血清胰岛素水平减少的葡萄糖水平的显著减少,在胰岛素抵抗性的改善方面GHS-R拮抗剂的作用被显示出来。另一方面,显示通过阻碍与磷脂酰肌醇3-激酶活性的降低有关的作用机制的葡萄糖输送活性、由血清FFA的上升诱发胰岛素抵抗性。最近,循环性FFA浓度的上升成为中年男性突然死亡的独立危险因子,这是在长期群组调查中被报告出来的(Jouvenet al.,Circulation,104:756-761,2001)。在以下所示的实施例中,GHS-R拮抗剂引起ob/ob肥小鼠的FFA水平的显著降低。
作为结论,发现通过末梢给药的GHS-R拮抗剂、[D-Lys-3]-GHRP-6及[D-Arg-1,D-Phe-5,D-Trp-7,9,Leu-11]P物质,瘦小鼠、饮食诱导性肥小鼠及ob/ob肥小鼠的饮食摄取减少。另外,显示通过[D-Lys-3]-GHRP-6的反复给药,减少小鼠的体重增加、改善血糖控制。
本申请是主张基于特愿2002-197582的优先权的申请,上述申请的全部内容作为参照而包含在本申请中。
通过下面的实施例对本发明进行详细的说明,但本发明的范围并不限于此。本领域的技术人员可以进行基于本发明的记载的各种变更、修饰,这些变更、修饰也包含在本发明中。
实施例
试验材料和方法
(1)动物实验
使用ddy系雄性小鼠(34-37g、JAPAN SLC,Shizuoka,Japan)及obese(ob/ob)C57BL/6J小鼠(68-74g、Shionogi Co.,Ltd.,Shiga,Japan)。每只小鼠被收容在每只都被控制的环境中(22±2℃,55±10%湿度,12∶12小时的明∶暗循环、上午7:00开灯)。投给小鼠将总能量的12%作为脂肪而含有的标准饮食或者将总能量的45%作为脂肪而含有的高脂肪饮食(CLEA Japan,INC.,Tokyo,Japan)。只要没有记载其他内容,能够自由摄取饮食和水。整个实验是被我们大学的动物关心委员会承认了的。
(2)试验药剂
[D-Lys-3]-GHRP-6、[D-Arg-1,D-Phe-5,D-Trp-7,9,Leu-11]P物质及大鼠饥饿激素是从BACHEM CALIFORNIA INC.(Torrance,California)、NEOSYSTEM(Strasbourg,France)及肽取代物(Peptide Institute)(Osaka,Japan)分别购入的。就在给药之前,为了第三脑室(ICV)给药而在4μl的人工脑脊液(ACSF)中稀释各药剂,或者为了腹膜内(IP)给药而在100μl生理盐水中稀释各药剂。结果以平均值±s.e.m表示。使用方差分析(ANOVA)后继续的Bonferroni氏t-检验来评价各组间的差异。P<0.05认为在统计学上有意义。
(3)通过ICV的药剂给药
为进行ICV给药,将小鼠用戊巴比妥钠(80-85mg/kg IP)麻醉,在实验前7天,放于定位固定装置(SR-6,Narishige,Tokyo,Japan)内,在各颅骨上用针在中央缝合的侧方0.9mm处向前顶0.9mm后方开洞。将24轨距的插管(Safelet-Cas,Nipro,Osaka,Japan)一端倾斜埋置于第三脑室中3mm,用作ICV给药。齿科用水泥将插管固定于颅骨上,用硅做成盖子。用PE-20管道将将27轨距的注入用衬垫安装到微注射器上。
(4)摄食试验
在摄食试验前,小鼠虽然可自由饮水、但进行了16小时的断食(fooddeprived)。但是,在[D-Lys-3]-GHRP-6及饥饿激素的共同给药对食物摄取的影响的实验中,小鼠自由摄取食物和水。食物摄取在给药后20分钟、1、2及4小时进行,通过从一开始事先测定好的食物中减去没被吃掉的食物来测定。
(5)RNA分离及Northern blot分析
RNA是利用RNeasy Mini Kit(Qiagen,Tokyo,Japan)从胃及副睾脂肪中分离的。将全部RNA用甲醛变性,用1%琼脂糖凝胶进行电泳,在HybondN+膜上形成墨迹。用荧光素标记cDNA探针对膜进行杂交。通过光密度分析法求出杂交信号的总积分密度(total integrated densities)(AmershamPharmacia Biotech AB,Uppsala,Sweden)。数据被标准化成3-磷酸甘油醛脱氢酶(G3PDH)mRNA存在丰度(abundance)并作为相对对照的百分比进行显示。
(6)饥饿激素基因的表达
瘦小鼠对将全部能量的12%作为脂肪而含有的标准饮食或者将全部能量的45%作为脂肪而含有的高脂肪饮食进行2周的摄取。小鼠在8小时断食后通过颈椎脱位被杀死。其后立即切除胃,在干冰上冷冻、在-80℃下保存直到Northern blot的调制。
(7)胃排空
在进行胃排空实验前,小鼠自由摄取水的同时断食16小时。已断食的小鼠自由摄取预先称量好的小药丸1小时,然后被用以[D-Lys-3]-GHRP-6。在注入小鼠体内后,再在1或2小时内不给食物。通过称量没被吃掉的小药丸来测定食物的摄取。其后立即通过开腹暴露胃,并非常迅速地结扎幽门和贲门两侧,然后切除,然后测定干燥内容物的重量。胃排空通过以下面的式子计算出来:

(8)反复给药
瘦小鼠在高脂肪饮食或者标准饮食下持续进行反复IP给药5天,及ob/ob肥小鼠在标准饮食下持续进行反复IP给药6天,。小鼠在上午7:00和下午19:00接受给药。每天测定食物的摄取和体重。实验结束时(去除食物及最后一次IP给药后8小时),在***麻醉下、从在眼眶窦取得的血液中分离血清。鼠通过颈椎脱位被杀死。其后立即去除作为白色脂肪组织而估定的副睾脂肪垫(fat pad)块(mass)、及腓肠肌并测定重量。通过葡萄糖氧化酶法测定血中葡萄糖。分别通过酶免疫法及酶法(EIKENCHEMICAL CO.,LTD.,Tokyo,Japan)测定血清胰岛素及游离脂肪酸(FFA)。通过酶法(Wako Pure Chemical Industries,Ltd.,Tokyo,Japan)测定血清甘油三酸酯及总胆固醇。
实施例1:饥饿激素反复给药对高脂肪饮食下的体重增加及血糖控制的作用
通过饥饿激素IP给药一日2次、给药5天(1次给药量:3nmol/小鼠),伴随一天的能量摄取的增加,体重明显增加(图2、图3、表1)。与用加入了标准饮食以及高脂肪饮食的生理盐水处理的小鼠相比,脂肪垫块分别明显增加了45%及125%。骨骼肌未显示重量增加。血清胆固醇及胰岛素水平也增加,并伴随血中葡萄糖浓度的中等程度的增加。接着测定WAT中的瘦素、脂粘素及抵抗素的mRNA水平。通过饥饿激素的反复给药,在WAT中瘦素mRNA的表达增加,同时抵抗素mRNA的表达减少(图4)。
其次,测定在将全部能量的45%作为脂肪而含有的高脂肪饮食下的饥饿激素mRNA的表达。通过2周的高脂肪饮食,在食物剥夺小鼠的胃中的饥饿激素基因的表达与标准饮食相比,显著增加。

实施例2:GHS-R拮抗剂对能量平衡的影响
向食物剥夺小鼠进行GHS-R拮抗剂、[D-Lys-3]-GHRP-6 IP给药。如图6所示,通过[D-Lys-3]-GHRP-6用量依赖性地显著减少食物摄取。
接着,研究中枢给药的[D-Lys-3]-GHRP-6是否有同样的效果。通过被ICV以及IP给药的[D-Lys-3]-GHRP-6,引起摄食行为的强烈减少(图7)。
为了评价饥饿激素通过脑内的GHS-R而发生作用的可能性,调查饥饿激素及[D-Lys-3]-GHRP-6的同时给药对食物摄取的效果。被ICV给药的[D-Lys-3]-GHRP-6阻碍对通过饥饿激素的IP给药而诱导产生的摄食的刺激效果(图8)。
接着,调查针对胃排空速度的[D-Lys-3]-GHRP-6的IP给药的效果。通过末梢给药的[D-Lys-3]-GHRP-6,胃排空速度的显著减少发生在给药1小时(图9)。
使用食物剥夺小鼠,对其他GHS-R拮抗剂、[D-Arg-1,D-Phe-5,D-Trp-7,9,Leu-11]P物质(L-756,867)对于摄食的效果进行调查。末梢给药的[D-Arg-1,D-Phe-5,D-Trp-7,9,Leu-11]P物质与[D-Lys-3]-GHRP-6同样,用量依赖性地显著减少食物摄取(图10)。
由高脂肪饮食引起的体重增加通过赋形剂的反复给药而被极大地损坏(图2),因此研究[D-Lys-3]-GHRP-6在因饮食而造成肥胖的小鼠中的急性作用。通过[D-Lys-3]-GHRP-6的IP给药而大大减少食物的摄取,并减少体重的增加(图11)。
为了得到关于治疗效果的进一步预测,研究IP给药的[D-Lys-3]-GHRP-6在ob/ob小鼠中是否产生食欲不振诱发性作用。被给药的[D-Lys-3]-GHRP-6在ob/ob小鼠以及瘦小鼠中使食物摄取显著减少(图12)。

5.一种肥胖预防剂或者治疗剂,其特征在于,含有GHS-R拮抗剂作为有效成分。
6.一种食欲抑制剂,其特征在于,含有GHS-R拮抗剂作为有效成分。
7.一种糖尿病预防方法或者治疗方法,其特征在于,包括使用有效剂量的GHS-R拮抗剂。
8.一种肥胖预防方法或者治疗方法,其特征在于,包括使用有效剂量的GHS-R拮抗剂。
9.一种食欲抑制方法,其特征在于,包括使用有效剂量的GHS-R拮抗剂。
