Description
本发明涉及一种用于制备固定化脂酶的方法、涉及所述的固定化脂酶自身并涉及一种在所述固定化脂酶存在的情况下进行酶催化转化的方法。
可以将脂酶在溶液中用作转化底物的酶催化剂。固定化脂酶通过具有的增加的稳定性和在进行连续和间歇式反应时的有效期并通过在间歇式反应中催化活性物质容易回收而与游离的脂酶区别开来。
公知固定化脂酶通过吸附固定在固相支持体上。还公知通过使聚烯烃颗粒与一种纯化脂酶的水溶液接触来制备固定化脂酶。
EP 232 933中描述了通过将水溶液吸附在诸如例如脂族聚烯烃类这样的疏水性热塑性聚合物上来固定来自水溶液的纯化的脂酶的技术方案。将所述的固定化脂酶用于脂肪的水解。
WO 90/15868中公开了通过将水溶液吸附在已经用有机溶剂预处理过的脂族聚烯烃上来固定来自水溶液的纯化的Candida antarctica脂酶的技术方案。将所述的固定化脂酶用于酯的合成。
在WO 94/28118中,在将纯化脂酶固定在疏水性支持体上之前加入非离子表面活性物质。
所有现有技术方法具有的缺陷在于在固定在固相支持体上之前将脂酶纯化以从粗脂酶溶液中除去其它的蛋白质、酶类和其它的细胞组成成分。这种经沉淀和层析过程的纯化步骤费时而昂贵。
此外,按照现有技术制备的固定化脂酶比游离脂酶具有显著降低的活性且必须通过添加例如表面活性物质来使其再活化。
还有必要添加表面活性物质以便使在按照现有技术从粗脂酶溶液中纯化的有机溶液中的游离脂酶活化(WO 95/17504)。
此外,按照现有技术制备的固定化脂酶的有效期仍然不是最理想的。
本发明的一个目的是改进所述的缺陷并提供一种用于制备固定化脂酶的新型简化方法和具有最佳特性并通过简化方法制备的新型固定化脂酶。
我们已经发现这些目的通过一种用于制备固定化脂酶的新型方法来实现,其中将一种粗脂酶溶液与聚烯烃颗粒接触。
例如,粗脂酶溶液指的是含有大于2%重量、更优选大于5%重量、特别优选大于15%重量杂质的脂酶溶液,所述的杂质诸如例如其它的蛋白质、其它产生脂酶的生物体的细胞组成成分或营养介质的残余物。所述的脂酶可以存在于水溶液中,或者还可以存在于含水缓冲***或诸如例如任意的卤代脂族或芳香烃(例如甲苯)这样的有机溶剂中。优选粗脂酶的水溶液。
优选的粗脂酶的水溶液是例如培养用肉汤,它通过在含水营养介质中培养产生脂酶的生物体而获得、或通过在含有(如果合适)缓冲液或其它使脂酶稳定的组分的含水溶剂中分散和/或匀化产生脂酶的生物体或产生脂酶的(诸如、例如)动物器官或植物的细胞组织而获得。
在与聚烯烃颗粒接触前,优选将粗脂酶溶液纯化以通过本身公知的方法、诸如离心法或过滤法来除去细胞。
例如,通过将聚烯烃颗粒引入粗脂酶溶液来进行接触过程。
当将所述的粗脂酶溶液与聚烯烃颗粒接触时,所述的脂酶被吸附在聚烯烃颗粒上。令人意外的是在这种接触中聚烯烃颗粒对脂酶具有非常高的选择性使得粗脂酶溶液中只有脂酶和(如果合适)其片段吸附在聚烯烃上而不吸附其它蛋白质-或仅吸附很小程度的(通常<2%重量)其它蛋白质。
吸附步骤由此也是一种从所述粗脂酶溶液中的其它蛋白质和酶类中纯化脂酶的步骤,从而可以省略固定在固相支持体上之前用于粗脂酶溶液的另一个纯化步骤。除制备过程简化外,在本方法中制备的固定化脂酶还具有下列超过现有技术固定化脂酶的优点:
-所述的固定化脂酶具有较长的有效期。
-活化物质诸如例如油酸的添加不再是必要的且不会导致活性的提高。
原理上也能够使用另外的纯化粗脂酶溶液用于本发明方法。例如,可以将所述的粗脂酶溶液纯化以达到一个点,在这个点向所述固定化脂酶中添加油酸则再次产生一次活性跳跃。
合适的纯化步骤是用于蛋白质纯化的所有常规工艺,诸如例如离子交换层析法、分子筛层析法、疏水层析法和沉淀法。
然而,优选在本发明的方法中使用未纯化的不含细胞的粗脂酶溶液。
因此,用于制备固定化脂酶的一个优选方法是这样一种方法,其中所述的粗脂酶溶液是一种不含细胞的培养用肉汤,这种肉汤通过下列方法获得:
a)培养一种产生脂酶的生物体,
b)如果合适随后在溶液中分散和/或匀化所述的生物体,和
c)随后除去细胞。
产生脂酶的生物体指的是能够自然或通过基因修饰、例如通过将一种脂酶基因***所述生物体的基因组而生产脂酶的生物体。生物体指的是微生物、植物和动物、以及来源于动物或植物的细胞组织。
优选的细菌和真菌脂酶来自于下列属的生物体:曲霉属、节杆菌属、产碱杆菌属、芽孢杆菌属、短杆菌属、假单胞菌属、伯克霍尔德氏菌属、色杆菌属、假丝酵母属、镰孢属、地霉属、腐质霉属、毛霉属、毕赤氏酵母属、青霉属、Rhizomucor、根霉属或栖热菌属。
特别优选的细菌和真菌脂酶是来自下列属和种的脂酶:节杆菌属的各种、产碱杆菌属各种、黑色曲霉、米曲霉、蜡状芽孢杆菌、枯草芽孢杆菌、凝结芽孢杆菌、产氨短杆菌、植物伯克霍尔德氏菌、Candidaantarctica、Candida cylindracea、解脂假丝酵母、产朊假丝酵母、皱摺假丝酵母、粘稠色杆菌、茄病镰孢、白地霉、Humicola lanuginosa、毛霉属的种类、Mucor japonicus、Mucor javanicum、米赫毛霉、豆酱毕赤氏酵母、变黑色根霉、米根霉、无根根霉、根霉属的种类、Rhizomucormiehei、无根根霉、德列马根霉、雪白根霉、Penicillium acylase、娄格法尔特氏青霉、水生栖热菌、黄栖热菌、嗜热栖热菌、粘稠色杆菌、假单胞菌属的各种、恶臭假单胞菌、荧光假单胞菌、葱头假单胞菌、Pseudomonas burkholderia或铜绿假单胞菌。
优选的动物和植物脂酶是猪胰腺脂酶(PPL)和麦胚脂酶。
特别优选的情况是来自Pseudomonas burkholderia(曾用名:植物伯克霍尔德氏菌)或铜绿假单胞菌的脂酶并将Pseudomonasburkholderia或铜绿假单胞菌用作产生脂酶的生物体。
可以按照本身公知的方式来进行微生物或植物或动物细胞培养物的培养,例如通过在一种营养培养基中发酵,所述的培养基除含有营养物、微量元素和(如果合适)抗菌素外还含有例如用于使蛋白质和酶类保持稳定的缓冲***。在这种情况中通常能够省略步骤b)-分散和/或匀化。
可以以一种本身公知的方式,例如在营养介质中或动物体内培养植物、动物和来源于动物或植物的细胞组织诸如植物的器官或部位并以一种本身公知的方式来收集或分离它们。然后优选以一种本身公知的方式通过在溶剂、优选在水或含水缓冲溶液中分散和/或匀化所述植物或细胞组织来制备培养用肉汤且随后除去细胞。
聚烯烃颗粒指的是聚烯烃类的颗粒。优选的聚烯烃是任意取代的烯烃类的均聚物或共聚物,所述的烯烃类诸如例如乙烯、丙烯、丁二烯、丁烯或辛烯。特别优选的聚烯烃颗粒是聚丙烯颗粒,诸如例如以商品名ACCUREL(R)得到的聚丙烯颗粒(来自Akzo via Enka AG,Obernburg,德国)。
聚烯烃颗粒的大小和空隙比不是关键。因为它们较容易加工,所以优选颗粒具有的大小为100μm-2000μm,且特别优选的颗粒具有的大小为200μm-1000μm。聚烯烃颗粒的空隙比在40%-80%较为有利,特别优选60%-70%,更特别优选65%。
聚烯烃颗粒的孔径大小优选为0.01μm-1μm,特别优选0.05μm-0.5μm。
聚烯烃颗粒中脂酶的负载量不是关键。优选的负载量取决于聚合物的性质并且可以通过常规试验发现,在优选负载量时最大量的脂酶被吸附且没有过多的脂酶过度损耗。在聚丙烯颗粒的优选应用中,优选的情况是每g聚烯烃颗粒负载2mg-6mg脂酶,且特别优选的情况是每g聚烯烃颗粒负载4.2mg脂酶。
粗脂酶溶液的pH对负载程度的影响不是关键。在pH 4-7时实现高程度的负载量。优选的pH为4.5-5.5,且特别优选的pH为4.8。
粗脂酶溶液的离子强度对负载程度的影响同样不是关键的。使用小于500mM的离子强度实现高程度的负载量。特别优选的离子强度小于300mM。
负载过程的最佳期限取决于脂酶和聚烯烃颗粒的性质且可以通过常规试验来测定。在聚烯烃颗粒与粗脂酶溶液之间接触的时间为4-6小时后通常达到最终的负载程度。
在下文所述的酶反应中可以直接使用由这种方法制备的固定化脂酶。活化过程例如通过添加油酸的过程是不必要的。
在反应中使用前,对于所要纯化的固定化脂酶来说,例如通过用一种合适的溶剂诸如例如水洗涤可有利地除去未吸附的物质。然后,如果合适,通过本身公知的方法、例如通过在空气中干燥来干燥所述的固定化脂酶。
本发明进一步涉及一种通过上述制备方法获得的固定化脂酶。
本发明进一步涉及一种通过使底物在本发明固定化脂酶存在情况下反应而酶催化转化或对映选择性转化底物的方法。
因此,将本发明的固定化脂酶用作催化剂。
酶催化转化指的是所述脂酶在能够催化的非固定化的游离态溶液中底物的化学反应。可以提及下列反应作为实例:
醇类的酰化或对映选择性酰化;
胺类的酰化或对映选择性酰化;
氨基酯类诸如例如氨基酸的酯类的酰化或对映选择性酰化;
羧酸酯类的水解或对映选择性水解;
氰醇类的酰化或对映选择性酰化;
氰醇酯类的水解或对映选择性水解;
内消旋二醇类的不对称反应或内消旋二酯类经水解的不对称反应。
优选的方法是用于下列反应的方法:
醇类的酰化或对映选择性酰化;
胺类的酰化或对映选择性酰化;
氨基酯类诸如例如氨基酸的酯类的酰化或对映选择性酰化;
或用于羧酸酯类的水解或对映选择性水解的方法。
在这些方法中特别优选使用的固定化脂酶是来自Pseudomonasburkholderia(曾用名:植物伯克霍尔德氏菌)或铜绿假单胞菌的一种脂酶,它通过上述本发明的制备方法来制备。
用于底物的酶催化转化或对映选择性转化的方法包括使所述底物在有固定化脂酶存在的情况下反应。
如果反应类型需要,优选添加另外的试剂。因此,例如,酰化过程需要添加酰化剂,而例如水解过程不需要添加其它试剂。
底物指的是可以经脂酶用酶催化反应、即以化学方式改变的化合物。在对映选择性转化中,其中只有一种反应的立体异构体的混合物同样也是底物。
可提及的底物的实例是醇类、胺类、氨基酯类、酰胺类、羧酸酯类、硫酯类、硫醇类、氰醇类、氰醇酯类和内消旋二醇类及其立体异构体的混合物。优选的底物是醇类、胺类、氨基酯类和羧酸酯类、以及外消旋醇类、胺类、氨基酯类和羧酸酯类。
反应优选在溶液中进行,就液体底物来说,使用或不使用溶剂。可以使用的溶剂的实例是水、有机溶剂,否则就是水/有机两相混合物。
优选使用的有机溶剂是二噁烷、THF、***、甲基叔丁基醚(MTBE)、甲苯或庚烷。优选使用的水/有机两相混合物是任意合适比例的水/MTBE混合物。
当在溶液中进行反应时,底物的浓度并不关键,但是优选在溶液中占0.5%重量至50%重量之间,特别优选20-30%重量。用于进行反应的温度同样不是关键的,但是可以通过脂酶在聚合物中的热稳定性来测定上限值。
反应优选在0℃-60℃、特别优选在15℃-40℃进行。
反应可以以连续或间歇方式进行。为了连续进行反应,例如使液体流动相以本身公知的一种方式通过反应器中的固定化脂酶床。流动相可以是底物(和试剂)的溶液或可以是不含溶剂的液体底物(和试剂)。流速并不关键且取决于反应的技术方式诸如床的高度、直径和颗粒大小并取决于反应器的设计。
优选用于连续方法的反应器是通常用于连续非均匀催化方法(液体/固体反应)的反应器(J.Hagen,Chemische Reaktionstechnik,VCH,Weinheim 1992,pp.165-169)。可以提及的实例是流化床反应器和固定床反应器,诸如管状反应器、柱反应器、全空间反应器、骤冷管反应器、管簇反应器和平板接触反应器。
当进行间歇式方法时,在反应器中以一种本身公知的方式将固定化脂酶悬浮于底物(和试剂)的溶液或含或不含溶剂的液体底物(和试剂)中并混合悬浮液。优选用于间歇式方法的反应器是通常用于间歇式非均匀催化方法(液体/固体反应)的带有震摇、混合或搅拌装置的反应器。可提及的实例是搅拌容器和由此生成的设计容器以及带有震摇装置的反应容器。
在反应完全后(达到热动平衡),例如通过滗析、离心或过滤和洗涤来分离固定化脂酶并用于进一步的反应。
在该方法的一个优选的实施方案中,含有可酰化官能团诸如例如羟基或氨基的底物诸如例如醇类、胺类或氨基酯类在有固定化脂酶作为催化剂和酰化剂存在的情况下被酰化或对映选择性酰化。
这种酶催化转化优选在有机溶剂诸如例如二噁烷、THF、***、甲基叔丁基醚(MTBE)、甲苯或庚烷中进行。
特别优选的方法是用于在有酰化剂和来自Pseudomonasburkholderia或铜绿假单胞菌的固定化脂酶存在的情况下醇类、胺类或氨基酯类或外消旋醇类、胺类或氨基酯类的酰化或对映选择性酰化的一种方法。

可以将使用本发明固定化脂酶的底物对映选择性酶催化转化的方法用于除去立体异构体且特别是用于从底物的立体异构体混合物中除去对映体或非对映体。特别优选用于从外消旋底物中除去对映体或非对映体且由此用于由相应的外消旋混合物制备旋光化合物。
固定化脂酶的对映选择性底物的特异性指的是(例如)仅转化外消旋底物中的一种对映体、而另一种对映体不反应。以一种本身公知的方式通过化学、物理和机械分离法可以很容易地分离所得的产物。可提及的实例是结晶法、沉淀法、两相溶剂***中的萃取法、层析分离法诸如HPLC、GC或硅胶柱层析法或诸如蒸馏法这样的热分离法。
因此,本发明进一步涉及一种用于制备旋光化合物的方法,该方法包括下列步骤:使可以由脂酶进行酶催化反应的底物的立体异构体或外消旋物的混合物在有本发明固定化脂酶存在的情况下以对映选择性方式反应且然后对所述的混合物进行分级分离。
可以将本发明的方法优选用于制备旋光化合物,这些化合物可以与参与反应的立体异构体化合物的混合物一样作为脂酶的底物、或这些立体异构体混合物中的至少一种立体异构体可以作为脂酶底物参与反应。
优选将用于醇类、胺类或氨基酯类的对映选择性酰化的方法用于拆分外消旋醇类、胺类或氨基酯类其且由此用于制备旋光醇类、胺类或氨基酯类。
优选将用于羧酸酯类对映选择性水解或水解的方法用于拆分外消旋羧酸酯类其由此用于制备旋光羧酸酯类。
下列实施例用来解释本发明:
实施例1
固定化脂酶的制备
1.1采用Pseudomonas burkholderia发酵实例的产生脂酶的生物体的培养
在14升发酵罐内使用菌株Pseudomonas burkholderia。预培养物中含有:1 g/l MgSO4*7H2O3.5 g/l KH2PO43.5 g/l K2HPO45.0 g/l (NH4)H2PO40.02 g/l CaCl2*2H2O10 g/l酵母提取物(Difco)
在每种情况中,在121℃下在带2个基底挡板的1升摇瓶中将200ml培养基灭菌30分钟,且向每个瓶中添加1ml微量元素(浓缩2倍)。对于14升发酵罐来说,在30℃和200rpm下在2个摇瓶内用Pseudomonasburkholderia培养7-9小时。
发酵罐构造:
标准构造的Techfors发酵罐,它带有:
pH电极(酸性和碱性对照);
pO2电极和
消泡电极(Russell)。
外设:
-用约2kg的大豆油充满带有Masterflex管和连接针的2升玻璃瓶并在121℃下灭菌30分钟。
-在121℃下将带有连接针的2个空1升玻璃瓶(用于计量酸和碱)灭菌30分钟。然后将500ml氨的水溶液(25%)和500ml硫酸(20%)盛入无菌瓶。
-在121℃下将带有连接针的充满了Tegosipon 3062(约500ml)的1个玻璃瓶灭菌30分钟。
-带有秤机的沙多利斯计量控制管和Watson-Marlow泵用于添加的大豆油的油的计量。
用于发酵培养基的混合物由下列成分组成:110 g 酵母提取物(Difco)38.5 g KH2PO438.5 g K2HPO455 g (NH4)H2PO411 g MgSO4*7H2O0.22 g CaCl2*2H2O
用去离子水加至11升。
在121℃下将所述的混合物灭菌30分钟且然后添加55ml的微量元素(2倍浓缩物)。
给这种培养基接种400ml的Pseudomonas burkholderia预培养物并在下列条件下使发酵过程在14升发酵罐内进行60小时: 温度 30℃旋转速度 1000rpm气体流速 11升/分钟压力 0.1巴pH 6.5
在发酵过程中计量大豆油,所计量的量随时间成指数增加。发酵完全后,将发酵罐冷却至15℃并排出液体。
所排出的来自Pseudomonas burkholderia的粗脂酶溶液的脂酶活性为8900U/ml。
1.2固定
在固定前,在1升容器中用RC-3B外旋离心机以5000-6000rpm离心从所述发酵罐排出的来自实施例1.1Pseudomonas burkholderia的粗脂酶溶液。
在离心从所述发酵罐排出的来自Pseudomonas burkholderia的粗脂酶溶液后,浑浊的不含细胞的粗脂酶溶液的活性为10,500U/ml。
8.3mg的脂酶相当于250,000U。
用不同的浓度(稀释度)、根据不同的接触时间和在不同条件下(pH、离子强度)用Accurel(R)1004(聚丙烯颗粒<400μm)和Accurel(R)1001(聚丙烯颗粒400μm-1000μm)培养不含细胞的浑浊的上层液。

将由来自铜绿假单胞菌的粗脂酶溶液制备的固定化脂酶(泳道1)过滤(粗脂酶溶液的滤过物=未吸附的粗脂酶溶液的组成成分=泳道3)、洗涤(泳道4)并与解吸附SDS缓冲溶液在95℃下接触5分钟。所述的缓冲溶液(泳道2)中含有已经再次解吸附且已经从粗脂酶溶液中的其它组成成分中纯化的脂酶溶液(泳道3)。
附图2表示经凝胶电泳分级分离中的相应泳道1-4。
标准=分子量标准:207kDa;123kDa;86kDa;44.6kD;31.4kDa;18.7kDa;7.2kDa。
实施例2
实施例2.1
间歇式方法
首先,在2升MTBE中由500g(4.1mol)1-苯基乙醇和246g(2.46mol)丙酸乙烯酯制备储备溶液。将5ml等份的该溶液加入到相当于10mg游离脂酶的固定化脂酶等份中。室温下将混合物振摇12小时并过滤以除去固定化脂酶并通过GC测定酰化了的对映体的转化率和对映选择性并将它们列在表3中。将滤器上的残余物用MTBE洗涤并重新使用以便测定有效期。将实验过程重复至多10次。所用的固定化脂酶是来自Pseudomonas burkholderia的实施例1.1.2d)中制备的固定化脂酶。使用游离脂酶进行类似的实验以便比较。

连续法
用来自实施例1.2.2d)的固定化脂酶填充层析柱(1.8×15cm)并将母液的液流以30-35ml/小时泵压在固定物上。以固定的间隔取样品并通过GC测定转化率和对映选择性并将它们列在表3.4中。使用游离脂酶的冻干物作为比较。

作为底物的外消旋反式-2-甲氧基环己醇与作为酰化剂的琥珀酐在有实施例1.2.2d)中制备的固定化脂酶存在的情况下的对映选择性酰化
实施例4.1
间歇法
首先,在6.2升MTBE和2.2升碳酸丙烯酯的混合物中由1100g(8.46mol)反式-2-甲氧基环己醇和470g(4.65mol)琥珀酐制备储备溶液。将5ml等份的该溶液加入到相当于10mg游离脂酶的固定化脂酶等份中。在室温下将混合物振摇12小时并过滤以除去固定化脂酶并通过GC测定未酰化对映体的转化率和对映选择性并将它们列在表4中。将滤器上的残余物用MTBE洗涤并重新使用以便测定有效期。将实验过程重复至多10次。所用的固定化脂酶是在实施例1.1.2d)中制备的脂酶。使用游离脂酶进行类似的实验以便比较。

Claims (11)
1.一种用于制备固定化脂酶的方法,通过使粗脂酶溶液与聚烯烃颗粒接触来进行。
2.一种如权利要求1中所述的方法,其中所述的粗脂酶溶液是不含细胞的培养用肉汤,它可通过下列方法获得:
a)培养产生脂酶的生物体;
b)如果合适随后在溶液中分散和/或匀化所述的生物体和
c)随后除去细胞。
3.一种如权利要求1或2中所述的方法,其中所用的脂酶是来自Pseudomonas burkholderia或铜绿假单胞茵的脂酶或所用的产生脂酶的生物体是Pseudomonas burkholderia或铜绿假单胞菌。
4.一种通过权利要求1中所述方法可获得的固定化脂酶。
5.一种用于底物的酶催化转化或对映选择性转化的方法,该方法包括使所述底物在有如权利要求4中所述固定化脂酶存在的情况下进行反应。
6.一种如权利要求5中所述的方法,其中所用的底物是醇类、胺类或氨基酯类,且将它们在有酰化剂存在的情况下进行酰化或对映选择性酰化。
7.一种如权利要求5中所述的方法,其中所用的底物是羧酸酯类,且将它们进行水解或对映选择性水解。
8.一种用于制备光学活性化合物的方法,该方法包括下列步骤:在权利要求4的固定化脂酶存在的情况下,将可被酶催化反应的底物的立体异构体或外消旋物的混合物被脂酶对映选择性反应,然后将混合物分级分离。
9.一种如权利要求8中所述的方法,其中所用的底物是醇类、胺类或氨基酯类,且将它们在有酰化剂存在的情况下进行酰化或对映选择性酰化。
10.一种如权利要求8中所述的方法,其中所用的底物是羧酸酯类,且将它们进行水解或对映选择性水解。
11.如权利要求4中所述的固定化脂酶作为化学反应中的催化剂的用途。
