Abstract
血管内皮生长因子-B(VEGF-B)和血管内皮生长因子-C(VEGF-C)是血管生成多肽,已显示特别是与bFGF组合时,VEGF-B和VEGF-C是体外血管生成性的。VEGF-C还在牛内皮细胞系中增加纤溶酶原激活物(PA)活性,这伴有PA抑制剂-1的相伴增加。向用bFGF和VEGF-C共处理的牛内皮细胞中加入α-2-抗纤溶酶能部分抑制胶原凝胶侵袭。

发明背景
血管生成是通过以预存在的血管的萌发过程而形成新的毛细血管的过程,并发生在发育过程中及大量生理及病理状态中。因此血管生成为组织生长,伤口愈合及女性生殖功能所必需,且也是一些病理进程如肿瘤生长,血管瘤形成及眼新生血管化的一部分(见Folkman,New Engl.J.Med.333:1757-1763(1995);Pepper,Arberioscler.Thromb.Vasc.Biol.17:605-619(1997))。一相似的但未深入研究的进程也发生于淋巴***,并有时指作***生成。
血管生成起于母血管基膜的局部破损处,随后内皮细胞迁移并派出入胞外基质周围,导致毛细萌芽形成。随后形成腔,构成功能血管萌芽形成中的一基本要素。在基膜重构后完成血管萌芽成熟。在此系列事件期间,至少发生三种内皮细胞功能改变:1)与胞外基质相互作用的调节,其要求改变细胞-基质接触及降解基质的蛋白酶解酶(纤溶酶原激活物(PAs)及基质金属蛋白酶)的产生;2)移动(迁移)开始增高随之降低,这使细胞转位于血管生成刺激物附近,且一旦达到目的地则停止转位;3)增殖增加,此为血管生长及延长提供新细胞,且一旦血管形成则随之恢复为静止状态。总之,这些细胞功能在毛细管形态发生,即三维未闭合或开放的类管结构的形成中起作用。许多新形成的毛细管随后经历血管壁成熟的过程(即平滑肌细胞富集基质及外膜形成),其它的则经历退化(即缺乏血流时)[见Pepper et al.,Enzyme Protein,49:138-162(1996);Risau,Nature 386:671-674(1997)]。
通常阐述的是除应答组织损伤或女性生殖器官发生的血管生成之外,在健康成年机体内内皮细胞转化非常低。推想内皮保持静止是由于内源性负调节物的存在,因为正调节物通常在成熟组织中被检出,其中明显没有血管生成,此认识的反面也是正确的,即正及负调节物经常共存于组织中,其中内皮细胞转化提高。由此引出“血管生成开关”的见解,其中内皮激活状态通过正及负调节物间的平衡而决定。在激活的(血管生成)内皮中,正调节物占优势,而通过负调节物的优势控制得以实现并保持内皮静止[Hanahan et al.,Cell.86:353-364(1996)]。“开关”最初用于肿瘤渐进的描述以描述从血管前期至血管期,“开关”这一见解还可用于发育,生理及病理性血管生成。尽管其仍需在体内最后证实,现行研究假设:“开关”包含正调节物的诱导和/或负调节物的缺失。
许多多肽生长因子或细胞因子已证实是体内血管生成的[见Klagsbrun et al.,Ann.Rev.Physiol.,53:217-39(1991);Leek et al.,J.Leukocyte Biol.,56:423-435(1994);Pepper et al.,Curr.Topics Microbiol.Immunol.,213/II:31-67(1996)]。这些包括血管内皮生长因子(VEGF),也称作血管通透性因子或vasculotropin,及碱性成纤维细胞生长因子(bFGF)。但尽管VEGF在胚胎血管发育及肿瘤血管生成中的作用已明确证实(见Carmeliet et al.,Nature 380:435-39(1996);Ferrara et al.,Nature,380:439-442(1996);Dvorak et al.,Amer.J.Pathol.,146:1029-1039(1995);Ferrara et al.,Endocrine Rev.,18:4-25(1997)),bFGF在血管生成的内源性调节方面的实际作用仍需证实。然而,有两项观察报告指出这两种细胞因子在血管生成调节中的相互作用的潜在重要性。首先,VEGF及bFGF已证实在体外血管生成诱导中协同作用[Pepper et al.,Biochem.Biophys.Res.Commun.189:824-831(1992);Goto et al.,Lab.Invest.69:508-517(1993)],且此项观察在兔后肢局部缺血模型中得以体内证实。[Asahara et al.,Circulation 92(Supp.II):II.365-71(1995)],及在大鼠海绵移植模型体内证实[Hu et al.,A.Br.J.Pharmacol.,114:262-268(1995)]。其次,VEGF的体外血管生成作用及其诱导PA活性的能力均依于由内皮细胞产生的内源性bFGF。

VEGFR-1的配体包括VEGF及胎盘生长因子(PlGF);VEGFR-2的配体包括VEGF及VEGF-C;而VEGFR-3只报道有一个配体VEGF-C[见Mustonen et al.,J.Cell Biol.129:895-898(1995);Thomas,J.Biol.Chem.,271:603-606(1996)]。VEGF-C是血管生成细胞因子的VEGF家族的最近阐述的一成员,其是与VEGF结构同源的蛋白质,是在探查VEGFR-3的配体期间,从人***腺瘤细胞系PC-3分离的[见Joukov et al.,EMBOJ.,15:290-298(1996)]。在另一项VEGFR-3配体的探查中,VEGF相关的蛋白(VRP)分离自人G61神经胶质瘤细胞DNA文库,此文库用基于编码具有与VEGF高度相似性的氨基酸序列的表达序列标记(EST)的探针筛选[Lee et al.,Proc.Natl.Acad.Sci.USA 93:1988-92(1996)]。VEGF-C和VRP是相同的蛋白,并从此称作VEGF-C。VEGF-C呈现与VEGF高度相似性,包括参与分子内及分子间二硫键的8个半胱氨酸残基的保守。半胱氨酸丰富的C-末端部分,其增加了与该家族其它配体相关的VEGF-C/VRP多肽的长度,显示出提示为Balbiani3环蛋白重复的半胱氨酸残基的间距图式。此C末端前肽还包括VEGF类似区的短基序,其可促进分泌的VEGF-C与胞外基质间的相互作用。VEGF-C与VEGFR-3的胞外区结合,并诱导VEGFR-3酪氨酸磷酸化。除VEGFR-3之外,VEGF-C还与VEGFR-2结合并诱导其磷酸化。VEGF-C促进人及牛内皮细胞生长,尽管在此分析中其比VEGF活性低。已报道VEGF-C在三维胶原凝胶中可引起内皮细胞迁移[Joukov,et al.,EMBO J.,16:3898-911(1997)]。VEGF-C转录物可在许多成人及胎儿组织中及在一些细胞系中检测。人VEGF-C已定位于染色体4934[Daavonen et al.,Circulation 93:1079-82(1996)]。
VEGF-C的显著特征之一是其mRNA首先翻译为前体,从中成熟配体通过细胞相关的蛋白酶解加工衍生。在生物合成之后,通过二硫键及非共价键连接将VEGF-C快速缔合成kDa反平行的同源二聚体。随后在分泌途径的末端部分及在细胞膜,N-末端及C-末端前肽经蛋白酶解加工,产生一些未完全加工的中间产物,然后,成熟的VEGF-C作为含二个经非共价相互作用连接的VEGF-同源区的21kDa同源二聚体从细胞中释出。此加工结果是VEGF-C获得与VEGFR-2结合并激活其的能力,并也提高与VEGFR-3的亲和性及激活性。细胞内蛋白酶裂解非VEGF-C分泌所必需。基于这些观测报告已推断VEGFR-3作为前体的合成使其信号优先地传至VEGFR-3[Joukov et al.,EMBO J.,16:3898-911(1997)],VEGFR-3在发育早期限于静脉内皮,并在发育后期及出生后变为限于淋巴内皮内[Kukk et al.,Development,122:3829-3827(1996)]。在加工机制被激活的某些情况下,可推断VEGF-C可获得额外的信号导向VEGFR-2的能力,从而提供额外的VEGF-C活性调节水平。信号导向VEGFRs要求受体二聚化。蛋白酶解加工通过间接促进VEGFR-2/VEGFR-3异源二聚体的形成可提供进一步调节机制。
Montesano等在Cell 42:469-477(1985)中阐述了应用血管生成体外模型分析胞外基质侵袭及管形成,其报道bFGF及VEGF诱导生长于三维胶原或血纤蛋白凝胶上的胎儿微血管、淋巴及动脉内皮细胞侵袭其下基质,在其中它们形成类毛细管结构[见Montesano et al.,Proc.Natl.Acad.Sci.USA.83:7297-7301(1986);Pepper et al.,Exp.CellRes.,210:298-305(1994);Pepper et al.,J.Cell Sci.,108:73-78(1985)]。这表明bFGF及VEGF的血管生成诱导性可经直接作用于内皮细胞而介导。但越来越多证据表明特异性细胞因子引起的应答的性质是前后相关的,即取决于应答细胞的周围环境中其它调节分子的存在与否。就血管生成而言,已先期证实VEGF及bFGF在体外模型中是协同的[Pepper et al.,Biochem.Biophys.Res.Commun.,189:824-831(1992)],并在相同模型中,TGF-β1具有双相作用[Pepper et al.,Exp.Cell Res.,204:356-363(1993)]。
尽管在现有技术中已有深入细致的研究,但仍需要影响血管生成的方式。尤其需要增强、调节和/或抑制血管生成的方法,及刺激或抑制蛋白酶如纤溶酶原激活物活性的方法。发明概述
本发明一个目的是鉴定在牛内皮细胞系中的VEGF受体的表达。
本发明另一目的是研制出一种协同诱导内皮细胞的血管生成应答的方法。
本发明又一目的是提供一种影响内皮细胞的蛋白酶解性质的方法。
本发明再一目的是提供一种抑制内皮细胞中血管生成的方法。
本发明再一目的是提供一种抑制蛋白酶活性的方法。
本发明再一目的是提供一种抑制纤溶酶原激活物活性的方法。
本发明再一目的是提供一种抑制肿瘤生长的方法。
现已发现,血管内皮生长因子B(VEGF-B)及血管内皮生长因子C(VEGF-C)刺激内皮胞外蛋白酶解活性,尤其刺激纤溶酶原激活物的活性,且也与碱性成纤维细胞生长因子(bFGF)协同诱导内皮细胞的血管生成,通过施用或共施用VEGF-B和/或VEGF-C以及其它生长因子,可调节内皮细胞血管生成活性。
尤其现已发现当与bFGF或VEGF共施用时,VEGF-C诱导体外内皮细胞的血管生成,且VEGF-B及VEGF-C协同地促进内皮细胞的血管生成。
VEGF-B或VEGF-C与bFGF的协同血管生成诱导作用可用于调节生理及病理性血管生成,以及用于抗肿瘤或其它病症的治疗方案。
用于以下实验的牛细胞示出对人VEGF,VEGF-B及VEGF-C的明显应答的事实表明:牛受体与人受体的同源性足以产生高度的物种间交叉反应性。由此,在表达人受体的人组织中可期望相似地或更多地应答。
VEGF-B及VEGF-C诱导各种蛋白酶如纤溶酶原激活物(PAs)的基因表达,其可破坏周围组织,并从而促进肿瘤细胞对那些组织的侵袭。同样,由于VEGF-C可作为内皮细胞的化学引诱物,这可导致***(其由内皮细胞组成)的侵袭及渗透,并最后支持肿瘤转移的开始,因而,VEGF-B和VEGF-C拮抗剂可治疗性地应用以抑制VEGF-B和/或VEGF-C的功能,从而阻止或预防渗透、内皮细胞侵袭和/或转移,尤其,如以下实施例14所示,α2-抗纤溶酶可抵抗VEGF-B和/或VEGF-C的血管生成功能,并从而可根据本发明的一方面用作抗转移剂。
本发明的另一方面包括提供一反义核苷酸序列,其至少互补于可促进内皮细胞增殖的VEGF-B和/或VEGF-C的DNA序列的一部分。本发明因而包括反义寡核苷酸,其选择性地与编码VEGF-B和/或VEGF-C蛋白的核酸分子结合,并分别用于降低VEGF-B或VEGF-C基因的转录和/或翻译。这在希望降低VEGF-B和/VEGF-C基因产物的表达的任何疾病中均是合乎需要的,包括降低由于VEGF-B和/或VEGF-C基因的表达所致的肿瘤细胞表型的任何方面。反义分子可以此方式用于阻止或抑制肿瘤细胞的此方面。
本文所用的术语“反义寡核苷酸”或“反义”意为此寡核苷酸是一寡核糖核苷酸,或修饰的寡脱氧核糖核苷酸,其在生理条件下与含有特定基因的DNA或与此基因的mRNA转录物杂交,并从而抑制此基因的转录和/或此mRNA的翻译。反义分子被标记以便在与靶基因杂交时干扰靶基因的转录或翻译。本领域技术人员将认识到反义寡核苷酸的实际长度,且其与其靶互补程度将取决于所选择的特异靶,包括靶的序列及含有此序列的特定碱基。优选反义寡核苷酸的构建及排列是使得在生理条件下选择性与靶结合,即在生理条件下与靶序列比与靶细胞中其它序列杂交的多。基于公布的VEGF-B及VEGF-C的DNA序列,本领域技术人员根据本发明能容易地选择及合成任何数目的合适的反义分子以应用。为有足够的选择性及有效抑制,该反义寡核苷酸应包括至少7个,优选至少15个互补于靶的连续碱基[见Wagner et al.,Nature Biotechnology,14:840-844(1996)]。更优选地,该反义寡核苷酸包括一20~30个碱基的互补序列。尽管可以选择反义于基因或mRNA转录物的任何区域的寡核苷酸,在一优选的实施方案中,反义寡核苷酸相应于N-末端或5’上游位点如翻译起始、转录起始或启动子位点,另外,3’未翻译区也可被靶击。本领域也使用靶击mRNA剪接位点的反义寡核苷酸,但如果发生可变mRNA剪接则非十分优选。另外,该反义序列优选靶击于不会出现mRNA二级结构的位点[见Sainio et al.,Cell MolNeurobiol.,14(5):439-457(1994)],以及预期不结合蛋白质的位点。
由于本领域熟练技术人员可容易地衍生相应于cDNA的基因组DNA或相应于基因组DNA的cDNA,因而本领域技术人员能提供互补于所需基因的cDNA的反义核苷酸,或提供与所需基因的基因组DNA杂交的反义核苷酸。以相似方式,不经过分试验可容易地获得等位或同源DNA的反义序列。
本发明所用反义寡核苷酸可包括“天然”脱氧核糖核苷酸,核糖核苷酸或其任何组合,即一天然核苷酸的5’末端与另一天然核苷酸的3’末端可以共价键合,如在天然***中,通过磷酸二酯核苷酸间连键连接。这些寡核苷酸可通过本领域公知的方法手工地或经自动合成仪进行制备。还可通过载体重组地产生。但在优选的实施方案中,用于本发明的反义寡核苷酸还可包括“修饰”的寡核苷酸,即此寡核苷酸可以各种方式修饰,这种修饰不阻碍其与其靶杂交,但可增强其稳定性或靶击能力或另外增强其治疗功效。
本文所用术语“修饰的寡核苷酸”阐述了一寡核苷酸,其中(1)其至少2个核苷酸通过合成的核苷酸间连键共价键合(即在一核苷酸的5’末端及另一核苷酸的3’末端之间除了磷酸二酯键之外的键合),和/或(2)与核苷酸非天然相连的一化学基团共价附着于寡核苷酸。优选的合成的核苷酸间连键是硫代磷酸酯、烷基膦酸酯,二硫代磷酸酯、磷酸酯、烷基硫代膦酸酯、亚磷酰胺、氨甲酯、碳酸酯、磷酸三酯、乙酰胺化物、肽及羧甲基酯。术语“修饰的寡核苷酸”也包括具有共价修饰的碱基和/或糖的寡核苷酸。例如修饰的寡核苷酸具有主链糖的寡核苷酸,所述的糖在3’位与非羟基的低分子量的有机基团及在5’位与非磷酸酯基团的低分子量的有机基团共价附着。因此修饰的寡核苷酸可包括2’-O-烷基化核糖基团。另外,修饰的寡核苷酸也可包括如以***糖代替核糖的糖。修饰的寡核苷酸也可包括碱基类似物,如C-5丙炔修饰的碱基[见Wagner et al.,Nature Biotechnology,14:844(1996)]。因而本发明涉及含有互补于编码VEGF-B和/或VEGF-C蛋白的核酸,并在生理条件下与之杂交的修饰的反义分子,以及药物学可接受载体的药物制剂。
反义寡核苷酸可作为药物组合物一部分而施用,如此药物组合物可包括与任何本领域已知的标准生理学和/或药物学可接受载体组合的反义寡核苷酸,该组合物应是无菌的并含有适于施用于患者的单位重量或体积的治疗有效量的反义寡核苷酸。术语“药物学可接受的”意为不干扰活性成分的生物活性的非毒性物质。术语“生理学可接受的”指与生物***如细胞、细胞培养物、组织或生物体相容的非毒性物质。载体的特征将依于施用途径。生理学及药物学可接受的载体包括稀释剂,充填剂,盐,缓冲剂、稳定剂、溶解剂及其它本领域熟知的物质。
含有如此反义序列的载体可用于抑制或至少降低相关细胞因子的表达。在细胞因子的表达与疾病相关的情况下,应用抑制一或多种血管生成细胞因子表达的此型载体是有利的,例如肿瘤产生VEGF-B和/或VEGF-C以提供血管生成的疾病。用含反义核苷酸序列的载体转化如此的肿瘤细胞将抑制或阻止内皮细胞增殖活性,并从而能抑制渗透、内皮侵袭和/或转移,且因此抑制或阻止肿瘤生长。附图的简要描述

实施例1:微血管内皮细胞的细胞培养
牛肾上腺皮质衍生的微血管内皮(BME)细胞(Furie et al.,1984)得自M.B.Furie博士和S.C.Silverstein博士,并生长于补加15%热灭活性供体小牛血清(DCS)(Flow Laboratories,Baar,Switzerland),青霉素(500u/ml)及链霉素(100wg/ml)的α改良极限必需培养基(Gibco AG,Basel,Switzerland)。所用细胞在16及19代之间。
实施例2:动脉内皮细胞的细胞培养
将分离自成熟牛胸主动脉刮屑并通过有限稀释克隆的牛动脉内皮(BAE)细胞(如Pepper et al.,Am.J.Physiol.262:C1246-57(1992)所述),在补加10%DCS及抗生素的低糖Dulbecco’s修改的极限必需培养基(DMEM,Gibco)中培养。所用细胞在10及15代之间。
实施例3:淋巴内皮细胞的细胞培养
牛淋巴内皮(BLE)细胞分离自肠***并由S.Wasi博士提供。第3代细胞如Pepper et al.,Exp.Cell Res.,210:298-305(1994)中所述通过有限稀释克隆。将BLE细胞在补加1mM丙酮酸钠,10%供体小牛血清及抗生素的DMEM中培养,所用细胞在21及26代之间。
实施例4:肺动脉内皮细胞的细胞培养
小牛肺动脉内皮(CPAE)细胞购自美国典型培养物保藏中心(Rockville,MD),并在补加20%DCS的199培养基中培养,所用细胞为第23代。
将所有内皮细胞系保存在1.5%明胶覆盖的组织培养烧瓶(FalconLabware,Becton-Dickinson Company,Lincoln Park,NJ)中,并以1/3或1/4分离比传代培养,所有4种细胞系的内皮性质已由DiI-Ac-LDL(Paesel and Lorei,Frankfurt Germany)吸收及用抗人von Willebrand因子的兔多克隆抗血清(Norclic Immunology,Tivburg,TheNetherlands)免疫染色预先证实。
实施例5:在BME细胞中由单独VEGF或VEGF-C或与bFGF组合刺激血管生成
为测定体外血管生成诱导,如Montesano et al.,Cell 42:469-477(1985)所述制备三维胶原凝胶,将8体积来自大鼠尾腱的I型胶原的溶液(大约1.5mg/ml)在冰上与1体积10倍极限必需培养基(Gibco)及1体积碳酸氢钠(11.76mg/ml)迅速混合,分散于18mm组织培养孔中(Nunclon,A/S Nunc,Roskilde,Denmark),并在37℃10分钟使之胶凝。然后将牛微血管内皮(BME)细胞于500μl培养基中以0.5-1.0×105细胞/孔种植在胶原凝胶表面上,细胞生长至铺满(3-5天),在此点将血清减至2%,并单用30ng/ml浓度的VEGF或0,1,3,10,30及100ng/ml浓度的VEGF-C或与10ng/ml牛成纤维细胞生长因子(bFGF)组合处理细胞单层。每2-3天更新一次培养基及处理一次,并在4天后固定培养物及照相。
在每孔中以在表层之下一致水平随机选择1.0mm×1.4mm测定区域,应用Nikon Diaphot TMD倒置光学显微镜,以105×放大通过相差显微镜照相,图1a是未处理对照组的相差视图,图1b是用30ng/mlVEGF处理的样品,胶原凝胶内形成的细胞索的结构十分清楚,焦点面在凝胶表面之下,图1c示出用30ng/ml VEGF-C处理的胶原凝胶,图1d示出只用10ng/ml bFGF处理的样品,同样可见清楚的侵袭应答,图1e示出用30ng/ml VEGF及10ng/ml bFGF组合处理的凝胶,图1f示出用30ng/ml VEGF-C及10ng/ml bFGF组合处理的凝胶,VEGF和bFGF或VEGF-C和bFGF共用而得的协同作用显而易见。
如Pepper et al.,Biochem Biophys Res.Commun.,189:824-831(1992)所述,通过检测已显而易见地以单细胞或细胞索形式穿过表面单层细胞的所有细胞结构的总附加长度定量细胞索状侵袭。图2a及2b示出的结果以平均总附加萌芽长度(μm)±s.e.m表示,且至少源自9个照相区域,即每种条件源自至少3个独立试验的每个试验的三个区域,1P<0.000,2P<0.000(Student’s不成对t-检验)。用Student’s不成对t-检验对比平均值,显著性p值<0.05。为对比之目的,单独使用30ng/ml VEGF以及30ng/ml VEGF与10ng/ml bFGF组合应用所得结果在每图右侧以条状(V-A)显示。
实施例6:在BME细胞中通过VEGF-B与bFGF组合刺激血管生成
重复实施例5的步骤,除了BME细胞单层用1,10及100ng/ml浓度的VEGF-B167单独处理,用1,10,100ng/ml浓度的VEGF-B186单独处理及与10ng/ml bFGF组合处理。VEGF-B较短的167个氨基酸形式缺失见于较长的186氨基酸形式的疏水区及O-糖基化位点。结果示于图3,尽管在此实验中单用VEGF-B不能产生可测的血管生成,但100ng/ml浓度的VEGF-B167及所有浓度的VEGF-B186当与10ng/ml bFGF组合时可产生不止加性的血管生成效应,为对比的目的,单用VEGF及VEGF-C及与10ng/ml bFGF组合应用的效果分别在每图右侧以V-A,V-C条示出。
实施例7:在BAE细胞中单用VEGF-C及与bFGF组合刺激血管生成
重复实施例5的步骤,除了在每孔凝胶表面种植牛主动脉内皮(BAE)细胞培养物,并单用VEGF-C及与1ng/ml和10ng/ml bFGF组合进行实验,结果分别图示于图4a,图4b及图4c,可见不仅单用VEGF-C可在BAE细胞中刺激血管生成,当细胞培养物用VEGF-C和bFGF组合处理可观测到不止加性的血管生成刺激效应。同样用VEGF处理的结果在每图右侧以条状(V-A)示出。
实施例8:在BAE细胞中通过VEGF-B与bFGF组合刺激血管生成
重复实施例6步骤,除了每孔凝胶表面种植牛主动脉内皮(BAE)细胞培养物,并应用3ng/ml浓度的bFGF,结果示于图5a及5b。同样,在此实验中单用VEGF-B不能产生明显血管生成效应,但当与bFGF组合时,VEGF-B167和VEGF-B186均产生不止加性(即协同)的血管生成效应,为了对比,单用VEGF(V-A)及VEGF-C(V-A)及与bFGF组合应用所得结果示于每图的右侧。
实施例9:在BLE细胞中通过VEGF-C刺激血管生成
将如实施例5所述用BLE细胞培养物种植三维凝胶形成的牛淋巴内皮(BLE)细胞单层用VEGF-C和/或VEGF处理,在处理4天后,如上述随机选择凝胶区域在单层表面之下一致水平照相。结合前述实施例所述定量内皮细胞侵袭,结果示于图6,可见VEGF-C产生明确的血管生成活性刺激。
前述实施例5-9表明:VEGF-B和VEGF-C的血管生成诱导活性可通过对内皮细胞的直接作用而介导,且在血管生成诱导方面,VEGF-B或VEGF-C和bFGF之间存在强协同效应。
如图1c及2a所示,当将VEGF-C加至三维胶原凝胶上的BME细胞的铺满单层细胞时,由VEGF-C诱导的侵袭应答勉强可测,相反,图4a和6示出,VEGF-C分别在BAE及BLE细胞中诱导明确的剂量依赖性侵袭应答,其伴随着分支类管结构的形成,此结构通过相差显微镜聚焦于单层细胞表面之下可见。假定VEGF-C的Mr为42,000,当在等摩尔(0.6-0.7nM)浓度对比时,VEGF-C(30ng/ml)比VEGF(30ng/ml)效力稍低,是bFGF(10ng/ml)效力的一半(参见图4a及4b,6及7)。
当与bFGF共用时,VEGF-C诱导一协同体外血管生成应答,因而,当加入至BME细胞时(其中单用VEGF-C效力很小或没有),VEGF-C增强bFGF的效力,如图1f及2所示30ng/ml VEGF-C可增加效力最大至2.5倍。相似地,在BAE及BLE细胞中单用VEGF-C诱导血管生成活性,共用VEGF-C和bFGF诱导不止加性的应答,如图4所示。
实施例10:共用VEGF及VEGF-C
共用VEGF及VEGF-C的效应也被评价,1p<0.000,2p<0.000(Student’s不成对t-检验)。在BME细胞中,其中只有VEGF诱导侵袭,发生应答(14天之后测定)只由于VEGF。在BLE细胞中,其中VEGF及VEGF-C均诱导侵袭,对共用30ng/ml VEGF及30ng/mlVEGF-C的应答基本上是加性的(见图6)。在BAE细胞中,VEGF及VEGF-C单独均诱导侵袭,应答(4天后测定)远比加性高,见于图7所示。
单用VEGF,VEGF-B及VEGF-C和组合应用的效应的三种方式对比示于图8a和8b,图8a示出三种细胞因子的单用效应,可见单用30ng/ml VEGF-B产生一可忽略的效应,单用VEGF(30ng/ml)产生一显著的效应,但最大单用效应由30ng/ml VEGF-C产生。从图8b可见,当VEGF和VEGF-B共用时,结果明显高于其单独应用的效应总和,同样地,共用VEGF-B和VEGF-C明显增强VEGF-C的胶原凝胶侵袭诱导活性,但最惊人的效应是30ng/ml VEGF与30ng/ml VEGF-C共用产生。
实施例11:在BME和BAE细胞中通过VEGF-C刺激PA和PAI-1活性
将制备自BME和BAE细胞的细胞提取物和培养物上清液暴露在0,1,3,10,30及100ng/ml浓度的VEGF-C中15小时,进行酶谱及反相酶谱分析。为对照的目的,分别暴露于30ng/ml VEGF及10ng/ml bFGF的BME及BAE细胞的测试培养基也被分析。用无血清培养基冲洗35mm明胶覆盖的组织培养盘中的铺满单层内皮细胞二次,并加入在含Trasylol(200 KIU/ml)的无血清培养基中的各细胞因子。15小时后,制备细胞提取物,并如Vassalli et.al.,J.Exp.Med.159:1653-68(1984)及Pepper et.al.,J.Cell Biol.,111:743-755(1990)所述通过酶谱及反相酶谱加以分析。
图9a示出来自BME细胞的细胞提取物的酶谱分析,图9b示出在37℃温育较长时期的相同酶谱,图9c示出BAE细胞的培养物上清液的反相酶谱分析。
实施例12:在BME及BAE细胞中通过VEGF-B制激PA和PAI-1活性
重复实施例11步骤,除了培养物上清液取自暴露于0,1,3,10,30及100ng/ml浓度的VEGF-B,结果示于图10a和10b。为对照的目的,暴露于30ng/ml VEGF及10ng/ml bFGF的结果也示出。
示于图9a-9c的酶谱及反相酶谱结果表明VEGF-C在BME及BAE细胞中诱导uPA,tPA及PAI-1活性。相似地,示出图10a和10b的酶谱及反相酶谱结果表明VEGF-B在内皮细胞中诱导uPA和PAI-1活性,uPA和PAI-1活性的诱导比约等摩尔浓度的VEGF所见的要小,酶谱结果也证实bFGF对tPA的表达没有或有轻微作用。也测试了VEGF-C对BME细胞中PAI-1活性,BAE细胞中uPA活性及BLE细胞中tPA和PAI-1活性的作用,并发现比图9a-9c所示效应低。
实施例13:RNA制备,体外转录,及Northern印迹杂交
将细胞因子加入铺满内皮单层细胞中,其中24小时前已加入新鲜完全培养基。根据厂商指导,应用Trizol试剂(Gibco BRL LifeTechnologies,Paisley,Scotland)在所示时间后制备总细胞RNA,将制备于暴露于VEGF-C或VEGF-8B(30ng/ml)0,1,2,9,24,48小时的BME细胞的铺满细胞单层的含总细胞RNA(5μ/泳道)的影印滤膜,用32P一标记的cRNA探针杂交。示出0,9,24及48小时的对照。加样的RNA完整性及均匀性通过在转移及交联后用亚甲兰染色滤膜确定,如图11及12底部的28S和18S核糖体RNAs所示。如Pepper et.al.,J.Cell Biol.,111:743-755(1990)所示进行Northern印迹,UV交联及滤膜的亚甲兰染色,体外转录,杂交及杂交后冲洗。从牛尿激酶型纤溶酶原激活物(u-PA)[Kraetzschmar et.al.,Gene,125:177-83(1993)],牛u-PA受体(u-PAR)[Kraetzschmar et.al.,Gene,125:177-83(1993)],人组织型PA(t-PA)[Fisher et.al.,J.Biol Chem.,260:11223-11230(1985)]及牛PA抑制剂-1(PAI-1)[Pepper et al.,J.CellBiol.,111:743-755(1990)]cDNA,如Pepper et al.,J.Cell Biol.,111:743-755(1990)和Pepper et al.,J.Cell Biol.,122:673-684(1993)所述制备[32P]-标记的cDNA探针,可见VEGF-C在BAE细胞中提高uPA,tPA及PAI-1 mRNA的稳态水平。
示于图11的Northern印迹分析表明,VEGF-C在BAE细胞中提高uPA,uPAR,tPA及PAI-1 mRNAs的稳态水平。PA,uPAR及PAI-1诱导的动力学是迅速(1小时内)且瞬间的(uPA和uPAR基线在3-9小时间达到,PAI-1在9-24小时间达到)。就uPA及uPAR而言,这与有关VEGF及bFGF的更延迟的增加的报道相反[见Pepperet.al.,J.Cell Biol.,111:743-755(1990);Mandriota et al.,J.Biol Chem.,270:9709-16(1995)]。就PAI-1而言,结果类似于VEGF及bFGF。使用VEGF-B的试验的Northem印迹实验结果示于图12。
细胞外蛋白酶活性的共诱导(其是以uPA合成的增加而表现),及蛋白酶抑制作用的共诱导(其是以PAI-1合成的增加而表现),是体内及体外血管生成的标志之一,并与“蛋白酶解平衡”理论相一致,该理论认为尽管蛋白酶为细胞迁移及形态发生所必需,但蛋白酶抑制剂也通过保护胞外基质免于非适合的破坏而发挥相当重要作用。
实施例16:在VEGF-C及bFGF存在下,α2-抗纤溶酶对内皮细胞血管生成的抑制
通过bFGF和VEGF-C以及0、1、3、10和30μg/ml的α2-抗纤溶酶共处理BME单层细胞4天,然后随机选择处理的凝胶区域在单层细胞表面之下一致水平照相,如实施例5中所述定量内皮细胞的侵袭,图13示出结果。可见随α2-抗纤溶酶浓度的提高,血管生成活性明显降低,因而表明α2-抗纤溶酶通过以剂量依赖形式与VEGF-C和bFGF共处理可抑制血管生成的诱导。

21、如权利要求19的方法,其中所述内皮细胞选自微血管内皮细胞,动脉内皮细胞及淋巴内皮细胞。
22、一种刺激内皮细胞蛋白酶解活性的方法,包括用VEGF-B,VEGF-C或者VEGF-B或VEGF-C与至少一种选自VEGF及FGF的其它细胞因子组合处理细胞的步骤。
23、如权利要求22的方法,其中所述内皮细胞选自微血管内皮细胞,动脉内皮细胞及淋巴内皮细胞。
24、一种抑制患者内皮细胞通透、缺陷和/或转移的方法,包括给所述患者施用有效抑制内皮细胞增殖量的VEGF-B或VEGF-C拮抗剂。
25、一种调节内皮细胞血管生成活性的方法,包括用含有VEGF-B或VEGF-C的反义核酸的载体转染或转化所述细胞。
Claims (25)
1、在内皮细胞中刺激血管生成的方法,包括向所述细胞共同施用至少二种选自VEGF,VEGF-B,VEGF-C及FGF的细胞因子。
2、如权利要求1的方法,其中所述的内皮细胞选自微血管内皮细胞,动脉内皮细胞及淋巴内皮细胞。
3、如权利要求2的方法,其中所述的内皮细胞是牛内皮细胞。
4、如权利要求2的方法,其中所述的内皮细胞是人内皮细胞。
5、如权利要求1的方法,其中所述细胞以体外培养物在培养基中处理,且通过将细胞因子掺入培养基中而施用细胞因子。
6、如权利要求1的方法,其中细胞因子以纯化蛋白质形式施用。
7、如权利要求1的方法,其中通过用至少一种载体转染或转化内皮细胞而施用细胞因子,所述载体包含与至少一种启动子序列可操纵连接的编码所述细胞因子的核苷酸序列。
8、如权利要求1的方法,其中所述的至少两种细胞因子为VEGF-C及FGF。
9、如权利要求1的方法,其中所述的至少两种细胞因子为VEGF-B和FGF。
10、如权利要求1的方法,其中所述的至少两种细胞因子为VEGF及VEGF-C。
11、如权利要求1的方法,其中所述的至少两种细胞因子为VEGF-B和VEGF-C。
12、如权利要求1的方法,其中所述的至少两种细胞因子为VEGF及VEGF-B。
13、一种通过释放协同组合的至少两种选自VEGF,VEGF-B,VEGF-C及FGF的血管生成细胞因子的细胞调节内皮细胞血管生成的方法,所述方法包括中和所述至少两种血管生成细胞因子中的一种。
14、如权利要求13的方法,其中所述中和是用血管生成细胞因子的抗体处理细胞而进行。
15、如权利要求14的方法,其中被中和的细胞因子是VEGF-C。
16、如权利要求14的方法,其中被中和的细胞因子是VEGF-B。
17、如权利要求14的方法,其中所述述抗体是单克隆抗体。
18、如权利要求13的方法,其中所述细胞是实体肿瘤细胞。
19、一种在VEGF-C及FGF存在下,通过内皮细胞抑制血管生成的方法,所述方法包括用抗纤溶酶处理所述细胞。
20、如权利要求19的方法,其中所述抗纤溶酶包括α2-抗纤溶酶。
21、如权利要求19的方法,其中所述内皮细胞选自微血管内皮细胞,动脉内皮细胞及淋巴内皮细胞。
22、一种刺激内皮细胞蛋白酶解活性的方法,包括用VEGF-B,VEGF-C或者VEGF-B或VEGF-C与至少一种选自VEGF及FGF的其它细胞因子组合处理细胞的步骤。
23、如权利要求22的方法,其中所述内皮细胞选自微血管内皮细胞,动脉内皮细胞及淋巴内皮细胞。
