1.发明领域
本发明涉及营养滋补品和药品片剂的含水薄膜涂层领域,特别是有关以右旋糖为基础的用作营养滋补品涂层的白色光亮薄膜涂层,例如:维生素片剂和钙/牡蛎壳片剂,以及药品片剂,例如:乙酰氨基酚(APAP)、阿斯匹林(ASP)和布洛芬
2.现有技术的描述
羟丙基甲基纤维素(HPMC)曾经用于涂覆如药品片剂等基底。例如:由Colorcon生产的涂层组合物制备的涂层,包括白色涂层(含有二氧化钛为颜料的涂层),它的制备方法见美国专利No.4,543,370和No.4,683,256,上述专利在本发明中作为参考,该涂层已经证明用于药品片剂是非常有效的。

按照本发明,用于形成涂层分散体以在营养滋补品和药品片剂等基底上进行薄膜涂覆的固体薄膜涂层组合物含有右旋糖,辅助薄膜形成物和二氧化钛。
可选择但优选的是本发明的固体薄膜涂层组合物还可以包括一种或多种的下列组份:增塑剂,表面活性剂,助流剂和防腐剂。
根据本发明,在营养滋补品和药品片剂等基底上涂覆的方法包括在水中混合右旋糖,辅助薄膜形成物和二氧化钛生成本发明的含水涂层分散体,将本发明的涂层分散体涂在所说的基底上形成白色薄膜涂层,并干燥所说基底上的白色薄膜涂层。可选择但优选的是将下列组份:增塑剂、表面活性剂、助流剂和防腐剂,与由右旋糖、一种辅助薄膜形成物和二氧化钛在水中形成的本发明的涂层分散体混合。
本发明还包括经涂覆的基底,例如经涂覆的维生素、钙/牡蛎壳片剂以及药品片剂;由本发明的固体薄膜涂层组合物或所说的组合物组份制成的含水涂层分散体;制备该固体薄膜涂层组合物和本发明的涂层分散体的方法。
本发明的固体薄膜涂层组合物和本发明的含水涂层分散体的基本薄膜-形成物是右旋糖。
右旋糖在本发明的固体薄膜涂层组合物和本发明的含水涂层分散体非水组份中的重量百分比的优选范围大约是20%到50%。更优选范围大约是25%到35%。
辅助薄膜形成物可以是聚乙烯吡咯烷酮(PVP)、羟丙基甲基纤维素、甲基纤维素、羟丙基纤维素(HPC)、羧基甲基纤维素钠(CMC钠)、麦芽糖糊精、多右旋糖、改良的食物淀粉、用玉米糖浆颗粒改良的食物淀粉、淀粉(例如,STARCH 1500淀粉)、木薯糊精、乳糖、藻酸钠、PG藻酸盐、***树胶、聚乙烯醇(PVA)、被本发明引为参考的专利号为4,543,370的Colorcon美国专利中例举的任何一种薄膜涂层聚合物以及它们的组合。辅助薄膜形成物被用作薄膜涂层的强度增强剂。它们在涂层中的存在有助于防止薄膜涂层的破裂,还有助于防止涂有涂层片剂边缘的磨损。一种优选的辅助薄膜形成物是CMC钠。辅助薄膜形成物在本发明的固体薄膜涂层组合物和本发明的含水涂层分散体中的非水组份中的重量百分比的范围大约是1%到50%(优选范围)和10%到20%(更优选范围)。不同的典型辅助薄膜-形成物的优选和更优选的范围在下面列出。
二氧化钛(TiO2)在本发明的固体薄膜涂层组合物和本发明的含水涂层分散体中的非水组份中的重量百分比的范围大约是25%到50%(优选的范围)和30%到40%(更优选范围)。
典型的增塑剂为矿物油、分子量在200到8000的聚乙二醇、丙二醇、甘油、甘油三乙酸酯、柠檬酸乙酰三乙酯、柠檬酸三乙酯(CitroflexA2)、柠檬酸三丁酯(Citroflex 4)和柠檬酸乙酰三丁酯(Citroflex A4)。增塑剂的优选范围,以及典型增塑剂的优选和更优选范围在下面给出。

下面的实施例用来描述本发明,所有组份均为重量。实施例1
将下面配方中的固体组份在PK混合器(Paterson Kelly)里混合五分钟形成混合物。然后通过混合器的I-棒将液体组份加到固体组份混合物中再继续混合五分钟生成本发明的固体薄膜涂层组合物。
组合物可选用研磨机来粉碎,比如:锤式研磨机(Apex Machinery,Dartford,England)。
也可选用齿轮混合机将固体薄膜涂层组合物制成颗粒状,例如Hobart齿轮混合机。使用时将固体薄膜涂层组合物放入混合机中接通电源,慢慢加入足够的水直到组合物成为紧密相连的颗粒。这些颗粒用一个1-2毫米的滤网过滤后在30℃的炉中干燥直到水分降到5%以下。组合物再用一个1-2毫米的滤网过滤就制成可以使用的非粉末状的颗粒。也可使用其它制备颗粒的方法,例如喷雾制粒和滚动压制。
将450克制成的薄膜涂层组合物分散到1800克的蒸馏水中(大约30分钟)形成本发明的涂层分散体(20%d的固体),使用24”Accela Cota涂覆机(Thomas Engineering)将分散体涂到15000克维生素片剂上形成本发明的涂层,理论上重量增加3%。

可选择地,将涂层组合物制成颗粒或粉末。
将9.0克制成的薄膜涂层组合物分散到36.0克的蒸馏水中搅拌直到形成发明的涂层分散体(20%的固体),使用Aeromatic Strea 1涂覆机(Niro)将分散体涂到300克3/8”凹的白色placebos上形成本发明的涂层,理论上重量增加3%。

虽然用本发明的固体薄膜涂层组合物制备本发明中所用的涂层分散体比较好,但将固体薄膜涂层组合物的各个组份加入水中制成涂层分散体也可以。
优点
本发明提供了一种具有较好薄膜粘附性和光滑表面的薄膜涂层。
而且,用本发明方法制备的光亮白色薄膜涂层比以HPMC为基础的白色涂层组合物所产生的涂层更加明亮。

Claims (36)

1.一种固体薄膜涂层组合物,该组合物用于形成涂层分散体以对营养滋补品和药品片剂等进行薄膜涂覆,该组合物含有右旋糖,辅助薄膜形成物和二氧化钛。
2.权利要求1的组合物,其中所述右旋糖在组合物中的重量百分比为20%到50%。
3.权利要求1的组合物,其中所述右旋糖在组合物中的重量百分比为25%到35%。
4.权利要求1的组合物,其中所述辅助薄膜形成物为聚乙烯吡咯烷酮(PVP)、羟丙基甲基纤维素、甲基纤维素、羟丙基纤维素(HPC)、羧甲基纤维素钠(CMC钠)、多右旋糖、淀粉、改良的食物淀粉、用玉米糖浆颗粒改良的食物淀粉、木薯糊精、乳糖、麦芽糖糊精、藻酸钠、PG藻酸盐、***树胶、聚乙烯醇(PVA)以及它们的组合。
5.权利要求1的组合物,其中所述辅助薄膜形成物在组合物中的重量百分比的范围是1%到大约50%。
6.权利要求1的组合物,其中所述辅助薄膜形成物在组合物中的重量百分比的范围是10%到大约20%。
7.权利要求1的组合物,其中所述二氧化钛在组合物中的重量百分比的范围是25%到大约50%。
8.权利要求1的组合物,其中所述二氧化钛在组合物中的重量百分比的范围是30%到大约40%。
9.权利要求1的组合物,其中还含有增塑剂。
10.权利要求9的组合物,其中所述增塑剂为矿物油、聚乙二醇、丙二醇、甘油、甘油三乙酸酯、柠檬酸乙酰三乙酯、柠檬酸三乙酯、柠檬酸三丁酯或柠檬酸乙酰三丁酯。
11.权利要求9的组合物,其中所述增塑剂在组合物中的重量百分比的范围是大于0%到大约20%。
12.权利要求9的组合物,其中所述增塑剂在组合物中的重量百分比的范围是5%到大约10%。
13.权利要求9的组合物,其中所述增塑剂在组合物中的重量百分比的范围是3%到大约5%。
14.权利要求1的组合物,其中还含有表面活性剂。
15.权利要求14的组合物,其中所述表面活性剂为吐温80。
16.权利要求14的组合物,其中所述表面活性剂在组合物中的重量百分比的范围是大于0%到大约5%。
17.权利要求14的组合物,其中所述表面活性剂在组合物中的重量百分比的范围是大于0.5%到大约1%。
18.权利要求1的组合物,其中还含有助流剂。
19.权利要求18的组合物,其中所述助流剂为硬酯酸。
20.权利要求18的组合物,其中所述助流剂在组合物中的重量百分比的范围是大于0%到大约15%。
21.权利要求18的组合物,其中所述助流剂在组合物中的重量百分比的范围是大于5%到大约10%。
22.权利要求1的组合物,其中还含有防腐剂。
23.权利要求22的组合物,其中所述防腐剂为柠檬酸钠。
24.权利要求22的组合物,其中所述防腐剂在组合物中的重量百分比的范围是大于0%到大约5%。
25.权利要求22的组合物,其中所述防腐剂在组合物中的重量百分比的范围为大约2%到大约3%。
26.权利要求1的组合物,其中还含有增塑剂,
所述增塑剂为矿物油、聚乙二醇、丙二醇、甘油、甘油三乙酸酯、柠檬酸乙酰三乙酯、柠檬酸三乙酯、柠檬酸三丁酯、或柠檬酸乙酰三丁酯;所述增塑剂在组合物中的重量百分比的范围是大于0%到大约20%;
所述右旋糖在组合物中的重量百分比为20%到50%;
所述辅助薄膜形成物为聚乙烯吡咯烷酮(PVP)、羟丙基甲基纤维素、甲基纤维素、羟丙基纤维素(HPC)、羧甲基纤维素钠(CMC钠)、多右旋糖、淀粉、改良的食物淀粉,用玉米糖浆颗粒改良的食物淀粉、木薯糊精、乳糖、麦芽糖糊精、藻酸钠、PG藻酸盐、***树胶、聚乙烯醇(PVA)以及它们的组合;
所述辅助薄膜形成物在组合物中的重量百分比的范围是1%到大约50%;
所述二氧化钛在组合物中的重量百分比的范围是25%到大约50%。
27.权利要求26的组合物,其中还含有表面活性剂、助流剂和防腐剂;
所述表面活性剂为吐温80,且表面活性剂在组合物中的重量百分比的范围是大于0%到大约5%;
所述助流剂为硬酯酸,且助流剂在组合物中的重量百分比的范围是大于0%到大约15%;
所述防腐剂为柠檬酸钠,且防腐剂在组合物中的重量百分比的范围是大于0%到大约5%。
28.用白色的薄膜涂层在营养滋补品和药品片剂等基底上涂覆的方法,该方法包括的步骤:
在水中混合右旋糖,辅助薄膜形成物和二氧化钛生成含水涂层分散体;
将有效量的所说的涂层分散体涂在所说的基底上形成白色薄膜涂层;且
干燥所说的基底上的该白色薄膜涂层。
29.权利要求28的方法,该方法包括将增塑剂分散到含水涂层分散体中。
30.权利要求28的方法,该方法包括将表面活性剂分散到含水涂层分散体中。
31.权利要求28的方法,该方法包括将助流剂分散到含水涂层分散体中。
32.权利要求28的方法,该方法包括将防腐剂分散到含水涂层分散体中。
33.权利要求28的方法,其中
所述右旋糖在含水涂层分散体非水组份中的重量百分比的范围大约是20%到50%;
所述辅助薄膜形成物在含水涂层分散体非水组份中的重量百分比的范围是1%到大约50%;
所述二氧化钛在含水涂层分散体非水组份中的重量百分比的范围是25%到大约50%。
34.权利要求28的方法,其中
所述右旋糖在含水涂层分散体非水组份中的重量百分比的范围大约是25%到35%;
所述辅助薄膜形成物在含水涂层分散体非水组份中的重量百分比的范围是10%到大约20%;

35.权利要求33的方法,其中还包括使用增塑剂,
所述增塑剂为矿物油、聚乙二醇、丙二醇、甘油、甘油三乙酸酯、柠檬酸乙酰三乙酯、柠檬酸三乙酯、柠檬酸三丁酯或柠檬酸乙酰三丁酯;
所述增塑剂在含水涂层分散体非水组份中的重量百分比的范围是大于0%到大约20%。
36.权利要求35的方法,其中还包括使用表面活性剂、助流剂和防腐剂;
所述表面活性剂为吐温80,且表面活性剂在含水涂层分散体非水组份中的重量百分比的范围是大于0%到大约5%;
