Abstract
根据本发明,提供芽孢形成抑制剂,其是含有使产气荚膜梭菌(Clostridium perfringens)在生物体内沉降的浓度的至少1种无机化合物的产气荚膜梭菌的芽孢形成抑制剂,其中,上述无机化合物为选自K、Na、Mg、Ca和Fe的至少1种的盐,上述无机化合物由包含植物氢化油的保护层包覆。
Description
技术领域
本发明涉及产气荚膜梭菌(Clostridium perfringens)的芽孢形成抑制剂。
背景技术
家禽界的坏死性肠炎(necrotic enteritis:NE)是造成重大经济损失(20亿US$/年)的重要疾病(非专利文献1)。坏死性肠炎导致肉鸡(broiler)的发育大幅下降,最坏的情况是导致死亡。在NE中,小肠粘膜处于坏死状态,形成黄褐色或胆汁色的假膜,在潜在性NE中,在粘膜表面形成像凹陷一样的溃疡(非专利文献2)。
已知坏死性肠炎是由作为革兰氏阳性菌的产气荚膜梭菌引起的。已知产气荚膜梭菌是家禽类的肠道常驻菌(非专利文献3)。产气荚膜梭菌的营养细胞是杆菌(1×10μm),但若环境恶化,则形成芽孢(直径1μm的球形)。已知芽孢发芽时分泌NetB毒素(NetB等),其是肠炎的主要原因(非专利文献4、图1)。因此,为了控制产气荚膜梭菌的毒素生产,需要先掌握产气荚膜梭菌的生命周期。
产气荚膜梭菌根据产生的毒素种类进行分型(非专利文献5)。2008年由A型新定义了2种,产生NetB(Necrotic Enteritis Toxin B-like:坏死性肠炎B样毒素)毒素的产气荚膜梭菌被定义为F型,产生CPE(Clostridium Perfringens Enterotoxin:产气荚膜梭菌肠毒素)毒素的产气荚膜梭菌被定义为G型。NetB被报道是NE的主要原因(非专利文献6)。
另外,已知产气荚膜梭菌不存在鞭毛,通过使纤毛(IV型支柱:T4P)伸缩来移动,粘附于宿主动物的肠道等(非专利文献7)。另外,还报道了包括产气荚膜梭菌在内的革兰氏阳性菌的T4P纤毛的结构和机理,已知利用ATP的能量使T4P的顶端伸缩(非专利文献8、图1)。
关于梭菌属在肠道内的作用机制也有报道。梭菌属分泌粘蛋白降解酶,降解粘液层的粘蛋白,从薄化的部位粘附于上皮细胞而诱导细胞死亡,另外,通过破坏紧密连接(tight junction)来诱导炎症(非专利文献9、图1)。
控制家禽的坏死性肠炎的方法之一是使用生长促进用抗生素(AGP:Anti-bioticgrowth promoter(抗生素生长促进剂))的方法。在家禽类中利用亚甲基双水杨酸杆菌肽和阿维拉霉素(Avilamycin)作为AGP(非专利文献10、11)。然而,由于对环境造成的负荷或耐药菌的出现等问题,在世界范围内对AGP强化了使用限制,欧州于2006年全面禁止,美国也禁止使用作为人畜共用的抗生素的泰乐菌素(Tylosin)或粘菌素(Colistin)等。寻求解决这个课题的环境友好型AGP替代品。
作为不利用抗生素的方法,有使用纳米氧化铁作为艰难梭菌(Clostridiumdifficle)的芽孢形成抑制剂的方法(专利文献1)。这里公开的方法是通过使纳米氧化铁与艰难梭菌的芽孢直接作用来抑制发芽的方法,但是,在该文献中没有关于对艰难梭菌的营养细胞的效果的记载。
专利文献1中记载的艰难梭菌在表型、化学分类学、***发育学上也不同于产气荚膜梭菌,还存在鞭毛,因此2016年将其命名为艰难梭菌(Clostridioides difficile)(非专利文献12)。

发明内容
发明所要解决的课题
因此,本发明以提供可预防或治疗产气荚膜梭菌感染症、例如坏死性肠炎的抗生素的替代品为课题。本发明还以提供含有这种替代品的补剂和饲料为课题。本发明还以提供给予这种替代品的方法为课题。
用于解决课题的手段
本发明人发现,在对鸟类给予可使产气荚膜梭菌以营养细胞的状态沉降的无机化合物时,可抑制芽孢的形成。在该方法中,在产生毒素之前可将产气荚膜梭菌以粪便的形式排出到体外,因此可抑制产气荚膜梭菌感染症。产气荚膜梭菌也是人肠道的常驻菌,因此上述无机化合物不仅用于鸟类等动物,还可用作用于预防或治疗人的产气荚膜梭菌感染症或用于减轻感染症的症状的补剂。即,通过本申请提供以下的各发明。
1.芽孢形成抑制剂,其是含有使产气荚膜梭菌(Clostridium perfringens)在生物体内沉降的浓度的至少1种无机化合物的产气荚膜梭菌的芽孢形成抑制剂,其中,上述无机化合物为选自K、Na、Mg、Ca和Fe的至少1种的盐,上述无机化合物被含有植物氢化油的保护层包覆。
2.上述1所述的芽孢形成抑制剂,其中,上述无机化合物为与由选自盐酸、硫酸、磷酸、碳酸、木质素磺酸、硅酸、乳酸、柠檬酸、葡萄糖酸、琥珀酸、富马酸、碘和碘酸的酸形成的阴离子的盐。
3.上述1或2所述的芽孢形成抑制剂,其中,上述无机化合物为选自KCl、NaCl、MgCl2、CaCl2和FeCl2的至少1种。
4.上述1~3中任一项所述的芽孢形成抑制剂,其中,上述无机化合物为KCl。
5.上述1~4中任一项所述的芽孢形成抑制剂,其中进一步包含多糖。

12.上述10或11所述的补剂,其包含可给予1mg/kg体重/天以上的剂量的无机化合物的浓度的无机化合物。
13.上述10所述的补剂,其中,产气荚膜梭菌感染症为坏死性肠炎。
14.饲料,其含有上述1~8中任一项所述的芽孢形成抑制剂。
15.上述14所述的饲料,其进一步含有惯用的饲料,以惯用的饲料的质量为基准,上述无机化合物的浓度为0.013mmol/kg以上。
16.非人动物的产气荚膜梭菌感染症的预防或治疗方法,该方法包括:对非人动物的肠道给予选自K、Na、Mg、Ca和Fe的至少1种的盐的无机化合物。
17.抑制芽孢形成的方法,其是在体外抑制产气荚膜梭菌的芽孢形成的方法,其中,在选自K、Na、Ca和Fe的至少1种的盐的无机化合物的浓度为500mM以上的有效浓度以上温育。
发明效果
根据本发明,可阻止产气荚膜梭菌的营养细胞变成芽孢。根据本发明,还可预防或治疗来自产气荚膜梭菌的坏死性炎症。根据本发明,还可边预防或治疗产气荚膜梭菌感染症,边发挥家畜的体重增加效果。

[无机化合物]
本发明中使用的无机化合物是选自K、Na、Mg、Ca和Fe的至少1种的盐。对构成盐的阴离子没有特别限定,可列举:由选自盐酸、硫酸、磷酸、碳酸、木质素磺酸、硅酸、乳酸、柠檬酸、葡萄糖酸、琥珀酸、富马酸、碘、碘酸等的酸形成的阴离子。从经济观点来看,优选Cl-。
具体而言,优选无机化合物为选自KCl、NaCl、MgCl2、CaCl2和FeCl2的至少1种。更优选无机化合物包括KCl。进一步优选无机化合物为KCl。
对于本发明的芽孢形成抑制剂中的无机化合物的浓度,只要是足以使产气荚膜梭菌在生物体内不凝聚也会沉降的浓度即可,没有特别限定。沉降所需的浓度根据无机化合物的种类或菌体浓度而不同,例如在菌体浓度以波长660nm下的光密度计为2.0左右时,KCl浓度为100mM以上、NaCl浓度为500mM以上、MgCl2浓度超过500mM、FeCl2浓度为10mM以上。为了发挥沉降效果,可提高无机化合物浓度,但在一定程度的浓度下效果已到顶,因此从经济的观点来看,例如希望是1,000mM以下。
[多糖]
据报道,多糖对革兰氏阴性菌发挥凝聚效果(国际公开第2019/177172号),但根据本发明人进行的实验,既没有对作为革兰氏阳性菌的产气荚膜梭菌发挥沉降效果,也没发挥凝聚效果(参照后述的例1和例5)。然而,可知:若与上述无机化合物组合,则增强无机化合物的产气荚膜梭菌的沉降效果和凝聚效果。特别是可知:在本发明的芽孢形成抑制剂对革兰氏阳性菌发挥凝聚效果的情况下,也使家畜的体重增加效果提高。因此,本发明的芽孢形成抑制剂可进一步包含多糖。
作为本发明中可使用的多糖,可列举:选自普鲁兰多糖、黄原胶、瓜尔胶、角叉菜胶、***树胶、果胶、羧甲基纤维素、软骨素、塔拉胶、刺槐豆胶、海藻酸盐(钠盐、钾盐、钙盐或铵盐)、海藻酸酯和它们的混合物的至少1种。其中,从成本效益和饲料登记状况的观点来看,优选***树胶、羧甲基纤维素、瓜尔胶、角叉菜胶、刺槐豆胶、普鲁兰多糖,更优选***树胶。
可将多糖与无机化合物一起或分别给予至对象。例如,如下所述,在对无机化合物和多糖施加包覆剂以形成芽孢形成抑制剂的情况下,可对无机化合物和多糖一起应用包覆剂,制成在一个芽孢形成抑制剂中无机化合物和多糖共存的制剂后给予至对象,也可对无机化合物应用包覆剂以形成芽孢形成抑制剂,与对多糖应用包覆剂而得到的制剂一起给予至对象。
本发明的芽孢形成抑制剂中的多糖浓度可适当确定,从经济的观点来看,优选为0.5~3质量%。更优选为1~2质量%。需要说明的是,只要没有特别记载,则在本说明书中单位“%”表示质量%。
关于使产气荚膜梭菌在生物体内沉降所需的无机化合物的浓度,若与多糖并用,则可较不与多糖并用的情况低。例如,在与***树胶并用时,包含KCl的本发明的芽孢形成抑制剂中的KCl的浓度为50mM以上,可发挥沉降效果。可优选设为100mM以上(例如,100~1,000mM)、更优选设为200mM以上(例如,200~700mM)。为了发挥沉降效果,可提高无机化合物浓度,但在一定程度的浓度下效果已到顶,因此从经济的观点来看,例如希望是500mM以下。
本发明的芽孢形成抑制剂可包含赋形剂。作为赋形剂,只要是通常用于提高成型的、药理学上可接受的物质即可,没有特别限定,例如可列举:碳酸钙、二氧化硅、硅酸钙、沸石、山梨糖醇、玉米淀粉、滑石、酵母膨润土、稻壳、液体石蜡、除具有使产气荚膜梭菌凝聚的性质的多糖以外的多糖、单糖、二糖等。在本发明的芽孢形成抑制剂包含赋形剂的情况下,相对于100质量份的芽孢形成抑制剂,赋形剂的量通常优选为0.1~100质量份。
本发明的芽孢形成抑制剂还可含有可被包含在补剂或饲料中的任选添加剂。作为添加剂,例如可列举:氨基酸、有机酸、维生素、色调强化剂(类胡萝卜素)、香料、活菌剂等。在芽孢形成抑制剂包含任选添加剂的情况下,相对于100质量份的芽孢形成抑制剂,任选添加剂的量通常优选为0.1~100质量份。
[包覆剂]
包覆剂形成上述无机化合物的保护层。包覆剂只要是可形成肠溶性被膜、且即使家畜或人摄取也安全的物质即可,可无特别限定地使用。包覆剂可单独使用一种,也可组合两种以上使用。从操作的容易性和经济性的观点来看,包覆剂优选为植物氢化油或作为片剂包覆剂而惯用的物质的苯甲酸树脂、紫胶(虫胶)、玉米蛋白、羟丙基甲基纤维素和麦芽糖醇等。作为植物氢化油,优选菜籽油、亚麻籽油、红花油、葵花籽油、大豆油、玉米油、花生油、棉籽油、芝麻油、米糠油、橄榄油、棕榈油、棕榈仁油或椰子油的氢化油。上述植物氢化油优选为菜籽油、亚麻籽油、红花油、葵花籽油、大豆油、玉米油、花生油、棉籽油、芝麻油、米糠油、橄榄油、棕榈油、棕榈仁油或椰子油的氢化油。作为包覆剂,其中优选氢化菜籽油和苯甲酸树脂。氢化菜籽油的层因可使芯在短时间内溶出而优选。苯甲酸树脂的层因可使芽孢形成抑制剂在(通过胃后的)中性或碱性下溶出而优选。
以本发明的芽孢形成抑制剂的总质量为基准,包覆剂优选为5~90质量%的量、更优选为20~30质量%的量。包覆剂还可含有可被包含在饲料或人用药物中的任选添加剂。
涂层可以是单层,也可以是2层以上的多层。多层涂层因容易控制在体内的溶出率而优选。特别是,若以氢化菜籽油的层为最外侧的层、以苯甲酸树脂层为与无机化合物接触的最内侧的层,则包覆剂在胃内不溶而在肠内溶解,故优选。
希望本发明的芽孢形成抑制剂在胃液中的溶出率小于60%,另外,希望在肠液中的溶出率为70%以上。为了达到这样的溶出率,可通过制成双层膜或多层膜、或者控制各层的被膜剂的种类或被膜厚度来调整。
特别优选无机化合物为K盐(特别是KCl)、多糖为***树胶、包覆剂为选自氢化菜籽油和苯甲酸树脂的至少1种。其中,特别优选无机化合物为K盐(特别是KCl)、多糖为***树胶、包覆剂为氢化菜籽油和苯甲酸树脂的双层。这种情况下,进一步特别优选由苯甲酸树脂形成的层与无机化合物接触、且在其上形成由氢化菜籽油形成的层。特别是,最优选芽孢形成抑制剂中的KCl浓度为500mM、***树胶浓度为1%、由苯甲酸树脂形成的层与无机化合物接触,在其上形成由氢化菜籽油形成的层。
[包覆方法]
对包覆上述芽孢形成抑制剂的方法没有特别限定,例如可使用市售的流化床型喷雾造粒机,边使粉末或颗粒状的芯流动边喷洒加热至高于熔点的温度而呈液态的包覆剂,从而可得到包覆型芽孢形成抑制剂。若使包覆型芽孢形成抑制剂达到0.05~5mm左右的大小,则操作变得容易,故优选。另外,对加热包覆剂时的温度没有特别限定,只要是包覆剂的熔点以上即可,优选比包覆剂的熔点高5℃~15℃左右。
本发明的芽孢形成抑制剂可由人或非人动物口服摄取,或者对非人动物的肠道直接给药。在对肠道直接给药的情况下,无需考虑因胃酸而失活或对胃粘膜造成的刺激等,因此无需用包含植物氢化油的保护层包覆。在口服摄取和肠道给药的任一种情况下,使对象摄取的无机化合物的量和给药次数均与后述的补剂相同。
[补剂]
本发明的芽孢形成抑制剂还可作为人或非人动物用的补剂。对补剂的形态没有特别限定,可列举:片剂、颗粒剂、散剂、饮剂等。本发明的补剂除包含本发明的芽孢形成抑制剂以外,还可包含作为补剂用成分而已知的成分、例如抗氧化剂或蛋白质。本发明的补剂以每天连续摄取为宜。本发明的补剂的摄取量例如根据给药对象的体重而变化。例如,可调制成可对给予本发明补剂的对象给予1mg/kg体重/天以上、优选1~100mg/kg体重/天的剂量的无机化合物。典型的是,本发明的补剂每天给予1~3次。
作为摄取本发明补剂的对象,可列举:人、牛、绵羊、山羊等反刍动物,马、猪、鸡、狗、鱼等单胃动物。
[饲料]
本发明的芽孢形成抑制剂可直接给予至家畜等非人动物,也可与玉米、大豆粉、米糠、鱼粉、啤酒酵母等赋形剂或稀释剂一起制成饲料。
本发明的饲料还可含有可被包含在惯用的饲料中的任选添加剂。本发明的饲料以每天连续摄取为宜。饲料的摄取量根据家畜的大小而不同,例如,在鸡的情况下,作为上述芽孢形成抑制剂的每天的摄取量,希望相对于除芽孢形成抑制剂以外的惯用的饲料,以无机化合物达到0.013~2.7mmol/kg(1~340ppm)左右、优选0.14~1.4mmol/kg(10~170ppm)的方式进行给予。另外,在惯用的饲料中含有包含多糖的芽孢形成抑制剂的情况下,摄取量相应地增加。例如,在包含133ppm的***树胶作为多糖时,希望相对于惯用的饲料,以无机化合物优选为0.013~1.4mmol/kg(1~170ppm)左右、更优选为0.14~0.7mmol/kg(10~85ppm)、进一步优选为0.54~0.8mmol/kg(40~60ppm)的方式给予。需要说明的是,在本说明书中,“ppm”是指“质量ppm”。
在本说明书中,“家畜”是指人所饲养的生物。具体而言,包括牛、绵羊、山羊等反刍动物,马、猪、鸡、狗、鱼等单胃动物。特别优选对单胃动物给予本发明的饲料。
对本发明的芽孢形成抑制剂的喂食方法没有特别限定。
实施例
例1.菌体沉降实验
在37℃、厌氧条件下,将产气荚膜梭菌ATCCl0873在GAM板(日水制药株式会社制造、改良GAM肉汤“Nissui”)上培养24小时。厌氧培养使用三菱瓦斯化学公司制造的厌氧培养试剂盒“AnaeroPack”来进行。
将所得到的培养物悬浮于4mL使用Merck Millipore公司制造的纯水制造装置制造的纯水中,以将悬浮液的光密度(OD)调整成在波长660nm下为2.0左右。
向投入了0.5mL该悬浮液的小瓶内,以表1中记载的浓度添加表1中记载的沉降化物质,使产气荚膜梭菌与各物质接触。需要说明的是,使用***树胶(“AG”)作为阳性对照。
将小瓶在37℃下静置8小时。其间自动连续测量样品的光密度。
<评价>
将在添加沉降化物质8小时后的样品的光密度的下降量达到未添加沉降化物质的样品的8小时后的光密度的下降量(即由菌体的自然沉降导致的光密度的下降量)的2倍以上的沉降化物质判断为具有沉降效果。
<结果和考察>
结果见表1和图1。对革兰氏阴性菌发挥沉降效果的AG对作为革兰氏阳性菌的产气荚膜梭菌没有发挥沉降效果。在AG的不存在下,在100mM以上的KCl、500mM以上的NaCl或10mM以上的FeCl2下确认到沉降性。对此,认为是通过阳离子改变产气荚膜梭菌菌体的表面电位,减小了斥力。在AG的不存在下未发挥沉降效果的MgCl2(500mM)通过与AG并用而发挥了沉降效果。

例2.凝聚和菌体形状观察实验
使用3株产气荚膜梭菌[ATCCl0873(产毒素A型)、SM101(产毒素F型:CPE毒素生产者)、CNEOP004(产毒素G型:netB毒素生产者)、均为杆菌]实施与例1同样的实验。但没有测量样品的光密度。
在添加沉降化物质8小时后回收样品,使用立式显微镜(OLYMPUS公司制造、型号:BX50)观察各菌株是否凝聚和菌体形状。
<结果>
结果见表2和图2。
对革兰氏阴性菌发挥凝聚效果的AG对产气荚膜梭菌不发挥凝聚效果。在向样品中添加了KCl或NaCl的情况下,在AG的不存在下,菌体的形状由杆菌变成球菌,但菌体自身没有凝聚。在KCl或NaCl与AG的共存下,菌体以球菌的状态凝聚。
在向样品中添加MgCl2的情况下,在AG的不存在下,形状保持杆菌不变,也没有观察到凝聚,但在AG的共存下,观察到了产气荚膜梭菌的凝聚。
在向样品中分别添加了CaCl2和FeCl2的情况下,在AG的不存在下,产气荚膜梭菌以杆菌的状态凝聚。

例3.产气荚膜梭菌菌体表面电位的测定
使用3株产气荚膜梭菌(ATCCl0873、SM101、CNEOP004)实施与例1相同条件的实验。但没有测量样品的光密度。
在添加沉降化物质8小时后回收样品,稀释100倍后,使用Malvern公司制造的Zetasizer Nano(型号:Nano-ZS)测定产气荚膜梭菌菌体的表面电位。还测定了添加沉降化物质前的沉降化物质的表面电位和沉降化物质的表面电位(需要说明的是,测定时的浓度与上述样品的表面电位测定时的浓度一致)。
<结果和考察>
结果见表3和表A。即使单独添加***树胶或KCl,产气荚膜梭菌也没有凝聚,若两者共存则凝聚。对此,认为当菌体表面在水中带负电荷时,通过添加KCl,菌体表面电位的负电荷增加,再通过使***树胶共存,表面电位变为中性,因此菌体和KCl与***树胶相互干扰而凝聚(表3)。在ATCCl0873株的情况下,在添加了MgCl2的情况下,菌体表面电位暂时变为中性,通过进一步添加***树胶,再次观察到菌体表面电位的变化(带负电)(表3-1)。由此认为:菌体和MgCl2与***树胶相互干扰而凝聚。另一方面,对于SM101、CNEOP004,认为在添加MgCl2时,菌体表面电位向中性侧大幅变化而凝聚(表3-2、表3-3)。在分别添加CaCl2(20mM以上)和FeCl2(10mM以上)的情况下,认为在AG的不存在下菌体表面电位向中性侧大幅变化,菌体和CaCl2、菌体和FeCl2相互干扰而凝聚(表3)。

例4.添加KCl时的产气荚膜梭菌的球菌的相位差显微镜观察
有多个报道称:大肠杆菌或弯曲杆菌(Campylobacter)、幽门螺杆菌(Helicobacter pylori)通常在应力环境下将形状变为被称作“球菌形态(coccoid-form)”的球菌,关于梭菌属(Clostridium)也有球形化的报道(International Journal ofCurrent Microbiology and Applied Sciences(2016),5(7),210-223)。在芽孢的形成步骤中,需要伴有细胞***,其所需的营养素是必需的。据报道,产气荚膜梭菌的芽孢与艰难梭菌同样,在从相亮孢子(Phase-bright spore)转变成相暗孢子(Phase-dark spore)后成为营养细胞(J.Am.Chem.Soc.(2014)第136卷,第14498-14504页)。
在例2中,使用不含营养素的纯水,由此预测:例2中观察到的球菌不是芽孢,而是“球菌形态”。
因此,研究了在向产气荚膜梭菌中添加KCl(0.5M)时产生的球菌(直径1μm)是否是营养细胞(营养的(vegetative))或芽孢(spore)中的任一种。芽孢的含水量减少,DNA等被浓缩,所以在相位差显微镜下发白光(相亮孢子)。在发芽成营养细胞之前观察到相暗孢子,但其在相位差显微镜下色调深。营养细胞在相位差显微镜下色调浅,在细胞内观察到黑色斑点。
作为参照,在相位差显微镜下观察到产气荚膜梭菌SM101株的芽孢(图3)。图3中,发白光的椭圆形是相亮孢子,相同形状且看起来发暗的是相暗孢子。此外,观察到了营养型菌的碎片。
另一方面,例2中,将向产气荚膜梭菌SM101株中添加KCl(500mM)时产生的球形菌在相位差显微镜下观察时(图4),与作为营养细胞的杆菌同样色调浅,在细胞内观察到了黑色斑点,因此推测不是芽孢。
因此,认为通过添加KCl等而引起的产气荚膜梭菌菌体的形状变化不会形成芽孢,而是在营养细胞的状态下发生的。
例5.肠道细菌的凝聚观察实验
对于除产气荚膜梭菌以外的肠道细菌,也与例2同样地操作,观察了各菌株的凝聚。选择作为革兰氏阳性菌的干酪乳杆菌(Lactobacillus casei)ATCC393、动物双歧杆菌(Bifidobacterium animalis)JCM1190、作为革兰氏阴性菌的肠道沙门氏菌IAM1648、大肠杆菌MG1655作为肠道细菌。
在37℃下,在MRS板(Difco公司制造)上培养干酪乳杆菌ATCC393和动物双歧杆菌JCM1190,在LB板(BD公司制造)上培养大肠杆菌MG1655,在NB板(Difco公司制造)上培养肠道沙门氏菌IAM1648。动物双歧杆菌JCM1190与产气荚膜梭菌同样在厌氧条件下培养,使用三菱瓦斯化学公司制造的厌氧培养试剂盒“AnaeroPack”。其他的菌在好氧条件下培养。
结果见表4。仅供参考,还一并记载了例2中进行的产气荚膜梭菌ATCCl0873的结果。
对于***树胶共存下的KCl的凝聚效果,在革兰氏阳性菌中尤其是产气荚膜梭菌特异性的,对干酪乳杆菌、动物双歧杆菌没有显示凝聚作用。作为革兰氏阴性菌的肠道沙门氏菌和大肠杆菌通过***树胶进行凝聚。其结果与WO2019/177172中报道的结果相同。革兰氏阴性菌在单独的KCl下均未凝聚,若与***树胶并用则凝聚。因此,暗示了:通过KCl和***树胶的并用,特异性地使称为坏细菌的产气荚膜梭菌、肠道沙门氏菌和大肠杆菌凝聚化。
另外,FeCl2对作为革兰氏阳性菌的产气荚膜梭菌和干酪乳杆菌、动物双歧杆菌显示出凝聚作用。

例6.3种肠道坏细菌的凝聚观察实验
与例5同样地培养产气荚膜梭菌ATCCl0873、肠道沙门氏菌IAM1648、大肠杆菌MG1655的3种菌株。按照与例1同样的方法调制了各菌株的悬浮液。调整成各悬浮液在波长660nm下的光密度(OD)为0.6~0.7左右,之后混合3种菌体的悬浮液,使用离心机回收混合菌体,在小瓶内最终调整成光密度为2.0左右(@波长660nm)。向小瓶中分别添加500mM KCl和1%***树胶的混合溶液、以及500mM NaCl和1%***树胶的混合溶液,与上述的混合菌体接触。将各小瓶在37℃下静置,自动连续测量光密度。
结果见图5。确认到500mM KCl与1%***树胶的组合、以及500mM NaCl与1%***树胶的组合可使3种肠道坏细菌快速凝聚化。例7.包覆型饲料用芽孢形成抑制剂的调制
使用***树胶(和光纯药公司制造)和KCl(和光纯药公司制造)作为芯材,使用氢化菜籽油(熔点67℃)和安息香树脂(中央香料公司制造)作为包覆剂。向呈粉末或颗粒状的芯喷洒规定量的通过加热至高于熔点的温度而呈液态的包覆剂,从而得到了包覆型饲料用芽孢形成抑制剂。
***树胶作为一层涂层,相对于75质量份的芯材,包覆25质量份氢化菜籽油。以下,将该添加剂表示为“包衣***树胶”。另一方面,KCl作为二层涂层,相对于84.77质量份的芯材,在第1层(内侧层)包覆2.23质量份安息香树脂、在第2层(外侧层)包覆13质量份氢化菜籽油。以下,将该添加剂表示为“包衣KCl”。
例8.包衣***树胶的肠溶性试验
(人工胃液处理)向使用Merck Millipore公司制造的纯水制造装置制造的纯水中加入0.2% NaCl和0.2%胃蛋白酶(来自猪胃粘膜、1:5,000、2,500单位/mg),调整至pH2后,投入例7中调制的包衣***树胶,在37℃下进行2小时的酶处理。通过自动连续测量其间的光密度,评价肠溶性。需要说明的是,“2小时”被设想是饲料从达到鸡的胃到通过的时间。
(人工肠液处理)人工胃液处理后,加入0.2%胰蛋白酶(来自猪胰腺、1:5,000、4,500单位/mg),调整至pH6后,在37℃下进行2小时的酶处理。通过自动连续测量其间的光密度,评价肠溶性。需要说明的是,“2小时”被设想是饲料从到达鸡的肠到通过的时间。
需要说明的是,两个处理中的光密度是使用岛津制作所公司制造的分光光度计UVmini-1240测定浊度(光密度[OD]、波长190nm)。另外,使用盐酸和氢氧化钠作为两个处理中的pH调节剂。
结果见图6。胃液处理开始后2小时的溶出率抑制在约60%以下,另一方面,在肠液处理中超过90%。由该结果可知:在施加了作为包覆剂的氢化菜籽油的样品中可进行良好的控释。
例9.包衣KCl的溶出试验
向使用Merck Millipore公司制造的纯水制造装置制造的纯水中投入例7中调制的包衣KCl,在37℃下实施溶出试验。通过自动连续测量其间的光密度,测定溶出率。两个处理中的光密度是使用岛津制作所公司制造的分光光度计UVmini-1240测定浊度(光密度[OD]、波长190nm)。
结果见图7。开始后2小时的溶出率抑制在约50%以下,另一方面,在肠液处理中达到了80%。由该结果可知:在施行了作为包覆剂的安息香树脂和氢化菜籽油的2层包覆的样品中可进行良好的控释。
例10.产气荚膜梭菌感染试验
将例7中调制的包衣KCl添加到Hofacre,C.L.等人,Avian Dis.1998;42(3):579-84所记载的饲料中使芯材的量为40ppm,得到了饲料组合物。需要说明的是,实验委托给SPRG公司在美国进行。
在平养鸡舍中,1个区域使用25只肉鸡鸡苗,给予饲料组合物,重复20次共计500只,从喂养时起饲养6周。试验条件是在第0天给予商用基础上使用的球虫减毒疫苗(0.007mL疫苗/鸟)。之后,在第14、15和16天进行4小时的禁食以及2-3小时的禁水,之后在饮水器中装入产气荚膜梭菌使其达到1×108cfu/mL,进行供水。
评价饲养期间来自坏死性肠炎(NE)的死亡率和来自NE的肠道损伤评分。来自NE的肠道损伤评分设定如下:0=无、1=轻度、2=中度、3=重度,算出评分的平均值。
需要说明的是,以无添加区作为对照(表5中,攻击对照(Challenge Control))。使用亚甲基双水杨酸杆菌肽(BMD)作为阳性对照。BMD作为对由产气荚膜梭菌引起的坏死性肠炎有效的物质而已知,还被广泛用作抗菌性发育促进物质(AGP),是美国使用最多的AGP之一。将BMD添加到上述Hofacre文献所记载的饲料中使其达到55ppm体积,制作阳性对照的饲料组合物。BMD直接使用市售品(Zoetis公司的BMD(注册商标)、未包覆)。
结果见表5。与攻击对照相比,BMD使死亡率下降至13.2%。与攻击对照相比,包衣KCl使死亡率下降了3.8%。与攻击对照相比,N.E.损伤评分得到显著改善(对照:0.75,BMD:0.30,包衣KCl:0.52)。

临床NE结果:数字表示平均值。进行了具有事后Tukey检验的单因素ANOVA以比较各处理。NE损伤评分:每个围栏随机选择3只鸟,在第17天进行评分。(0=无、1=轻度、2=中度、3=重度)
例11.体重增加效果试验
向表6的未添加饲料添加剂的基础饲料中添加例7中调制的包衣KCl和包衣***树胶,使芯材的量分别达到40ppm体积、100ppm体积,得到了饲料组合物。

供试小鸡为肉鸡专用种(UK Chunky)公鸡鸡苗,挑选体重为38~46g的个体用于试验。在平养鸡舍中,所有试验组均给予表6的未添加饲料添加剂的基础饲料,供试小鸡以50只为1组使体重分布基本均匀,重复3次,从喂养时起饲养3周。作为试验条件,为了对环境条件施加负荷,在饲养期间按1天1次、每组约1L的水喷洒在地板前面,在湿地板上饲养。试验区如下实施:1)对照(无处理),2)阳性对照(BMD 55ppm),3)包衣***树胶(133ppm),4)包衣***树胶(133ppm)+包衣KCl(47ppm)。在饲养期间评价鸟的体重增加效果(BodyWeight Gain;BWG)。结果见表7。结果是以阴性对照为100来显示。
关于BMD,与对照(无处理)相比,在整个区间(0-21日龄)体重增加均有所提高。另一方面,在组合了包衣***树胶和包衣KCl的区与对照相比没有发现显著差异,但随着生长,体重增加效果变得明显,在区间(15-21日龄)观察到了体重增加效果。通常是在6~7周龄出栏,因此可期待充分的体重增加效果。

Claims (17)
1.芽孢形成抑制剂,其是含有使产气荚膜梭菌(Clostridium perfringens)在生物体内沉降的浓度的至少1种无机化合物的产气荚膜梭菌的芽孢形成抑制剂,其中,上述无机化合物为选自K、Na、Mg、Ca和Fe的至少1种的盐,上述无机化合物由包含植物氢化油的保护层包覆。
2.根据权利要求1所述的芽孢形成抑制剂,其中,上述无机化合物为与由酸形成的阴离子的盐,上述酸选自盐酸、硫酸、磷酸、碳酸、木质素磺酸、硅酸、乳酸、柠檬酸、葡萄糖酸、琥珀酸、富马酸、碘和碘酸。
3.根据权利要求1或2所述的芽孢形成抑制剂,其中,上述无机化合物为选自KCl、NaCl、MgCl2、CaCl2和FeCl2的至少1种。
4.根据权利要求1~3中任一项所述的芽孢形成抑制剂,其中,上述无机化合物为KCl。
5.根据权利要求1~4中任一项所述的芽孢形成抑制剂,其中,进一步包含多糖。
6.根据权利要求5所述的芽孢形成抑制剂,其中,上述多糖为选自普鲁兰多糖、黄原胶、瓜尔胶、角叉菜胶、***树胶、果胶、羧甲基纤维素、软骨素、塔拉胶、刺槐豆胶、海藻酸盐(钠盐、钾盐、钙盐或铵盐)、海藻酸酯和它们的混合物的至少1种。
7.根据权利要求5所述的芽孢形成抑制剂,其中,上述多糖为***树胶。
8.根据权利要求1~7中任一项所述的芽孢形成抑制剂,其中,上述无机化合物和根据需要而含有的多糖由包含植物氢化油的保护层包覆。
9.根据权利要求8所述的芽孢形成抑制剂,其中,上述植物氢化油为菜籽油、亚麻籽油、红花油、葵花籽油、大豆油、玉米油、花生油、棉籽油、芝麻油、米糠油、橄榄油、棕榈油、棕榈仁油或椰子油的氢化油。
10.根据权利要求1~7中任一项所述的芽孢形成抑制剂,其中,上述无机化合物和根据需要而含有的多糖由氢化菜籽油和苯甲酸树脂这两层保护。
11.用于预防或治疗产气荚膜梭菌感染症的补剂,其含有权利要求1~10中任一项所述的芽孢形成抑制剂。
12.根据权利要求10或11所述的补剂,其包含可给予1mg/kg体重/天以上的剂量的无机化合物的浓度的无机化合物。
13.根据权利要求10所述的补剂,其中,产气荚膜梭菌感染症为坏死性肠炎。
14.饲料,其含有权利要求1~8中任一项所述的芽孢形成抑制剂。
15.根据权利要求14所述的饲料,其进一步含有惯用的饲料,以惯用的饲料的质量为基准,上述无机化合物的浓度为0.013mmol/kg以上。
16.非人动物的产气荚膜梭菌感染症的预防或治疗方法,该方法包括:对非人动物的肠道给予选自K、Na、Mg、Ca和Fe的至少1种的盐的无机化合物。
17.抑制芽孢形成的方法,其是在体外抑制产气荚膜梭菌的芽孢形成的方法,其中,在选自K、Na、Ca和Fe的至少1种的盐的无机化合物的浓度为500mM以上的有效浓度以上温育。
