本发明涉及植物提取物技术领域,尤其涉及一种含有正十六烷酸的螺旋藻提取物及其制备方法。该制备方法包括:采用超临界CO2萃取方法提取钝顶螺旋藻。本发明提供了一种含有较高含量正十六烷酸的螺旋藻提取物,可作为原料直接应用于化妆品、食品等领域,本发明的提取方法成本较低、步骤简单、易于工业化大规模推广应用。

一种含有正十六烷酸的螺旋藻提取物及其制备方法
技术领域
本发明涉及植物提取物技术领域,尤其涉及一种含有正十六烷酸的螺旋藻提取物及其制备方法。
背景技术
正十六烷酸,又名软脂酸、棕榈酸,是一种有机物,化学式是C16H32O2,是一种饱和高级脂肪酸,白色带珠光的磷片。
其主要的工业用途为用作沉淀剂、化学试剂及防水剂;或者用于制造无味氯霉素及各种棕榈酸金属盐。此外,其钠盐或钾盐可用作乳液聚合时的乳化剂,例如其钠盐是肥皂的主要成分之一,其铝盐和锌盐等用于润滑剂、涂料、油墨和增塑剂中。而且软脂酸与硬脂酸按照一定的比例混合可以得到结晶的硬脂酸产品,可以运用到化妆品行业中。
目前该化合物主要的工业生产方式是通过化工合成的方法,但是化工合成所需要的生产工艺复杂、所需的原料、催化剂等化工原料用品的使用成本较高,且不同的合成反应中会生成不同的副产物,导致其纯度有限,应用于后续化妆品、食品等领域时,会存在不安全的风险。
发明内容
为解决上述技术难题,本发明通过对大量不同种类的植物、不同植物的不同部位进行筛选,同时针对不同植物及不同部位的特性,选择不同的提取方法与植物及其不同部位进行组合尝试,发现通过采用超临界CO2萃取方法提取钝顶螺旋藻时,获得的提取物中所富含的正十六烷酸的含量较高,而且获得的螺旋藻提取物较为安全,可作为原料直接应用于后续化妆品、食品等领域产品的生产过程。
优选地,所述提取的压力为18~22MPa。
优选地,所述提取的温度为45~55℃。
优选地,所述提取的CO2流速为2~4L/min。
优选地,所述提取的夹带剂为无水乙醇。
优选地,所述夹带剂的灌注速度为2~4mL/min。
优选地,所述提取的时间为60min以上。
优选地,制得的提取物在无水乙醇中储存。
优选地,提取物的储存温度为18~22℃。
本发明进一步对超临界CO2萃取方法提取钝顶螺旋藻的工艺参数进行了组合优化,发现在上述各优选条件的组合下,所制备的钝顶螺旋藻提取物中正十六烷酸的含量大幅提升。
本领域技术人员可以进一步通过对上述优选方案进行组合,以得到其它的较优实施方案。
进一步,本发明提供了一种螺旋藻提取物,其由上述任一方案中的制备方法制得。
优选地,螺旋藻提取物中含有的正十六烷酸占所述螺旋藻提取物的重量百分比为4.2%以上;更优选为4.2%~5.0%。
进一步,本发明还提供了一种产品,其中含有上述任一方案中的螺旋藻提取物。
在一些实施方案中,所述产品为药品、食品、食品添加剂、饲料、饲料添加剂、化妆品中的至少一种。

实施例
本实施例提供了一种含有正十六烷酸的提取物,其制备方法如下:
将钝顶螺旋藻制备成粉体,称取80g钝顶螺旋藻粉放入超临界CO2萃取容器中,设置萃取条件为:20MPa压力,50℃的温度,CO2流速为3L/min,夹带剂采用无水乙醇,其灌注速度3mL/min,提取时间为60min,旋转蒸发夹带剂无水乙醇后制得螺旋藻提取物,提取率为5.9%w/w;将制得的螺旋藻提取物在无水乙醇中储存,储存温度为20℃。
试验例
对上述实施例制得的螺旋藻提取物中的正十六烷酸含量进行检测,检测方法如下:
使用Agilent 7890C/5975A-GC/MSD(美国)进行GC/MS分析,该仪器内配有PB-Wax色谱柱,其规格为(30mm×250mm内径;0.25μm)。GC/MS检测采用电离能量为70eV的电离***。仪器中初始温度为50℃,保持3min,随后温度以6℃/min的速度升到280℃。进样器和MS温度分别设定在230℃和250℃,氦气的流速为1.0mL/min,样品注入量为1μL。
分析结果显示,螺旋藻提取物中正十六烷酸含量为4.20%。

1.一种含有正十六烷酸的提取物的制备方法,其特征在于,包括:采用超临界CO2萃取方法提取钝顶螺旋藻。
2.根据权利要求1所述的制备方法,其特征在于,所述提取的压力为18~22MPa。
3.根据权利要求1所述的制备方法,其特征在于,所述提取的温度为45~55℃。
4.根据权利要求1所述的制备方法,其特征在于,所述提取的CO2流速为2~4L/min。
5.根据权利要求1所述的制备方法,其特征在于,所述提取的夹带剂为无水乙醇。
6.根据权利要求5所述的制备方法,其特征在于,所述夹带剂的灌注速度为2~4mL/min。
7.根据权利要求1所述的制备方法,其特征在于,所述提取的时间为60min以上。
8.一种螺旋藻提取物,其特征在于,其由权利要求1~7中任一项所述的制备方法制得。
9.根据权利要求8所述的螺旋藻提取物,其特征在于,其中含有的正十六烷酸占所述螺旋藻提取物的重量百分比为4.2%以上。
10.一种产品,其特征在于,其中含有权利要求8或9所述的螺旋藻提取物。
