Abstract
本发明提供了一种人工关节假体材料LP01‑PS及其制备方法,属于人工关节假体材料技术领域。本发明提供的人工关节假体材料LP01‑PS,由聚醚醚酮、LP01和聚多巴胺合成。本发明的富载LP01的LP01‑PS,具有良好的弹性模量、细胞和动物生物相容性,对大肠杆菌和金黄色葡萄球菌的增殖、代谢、扩散等有直接的抑制作用,增强巨噬细胞对S.aureus和E.coli增殖、代谢、扩散等的抑制作用。另外,本发明的LP01‑PS具有促骨髓间充质干细胞成骨分化的作用,能够提高大鼠骨髓间充质干细胞矿化能力、成骨相关基因表达。
Description
技术领域
本发明属于人工关节假体材料技术领域,尤其涉及一种人工关节假体材料LP01-PS及其制备方法。
背景技术
近百年来,使用内植物是治疗骨科疾病最基本的方法,但内植松动和细菌感染是骨科手术的灾难性并发症,给患者带来极大的痛苦和经济负担。细菌感染引起局部炎症反应,产生的细胞毒素抑制了局部的成骨反应,是导致骨科假体松动的重要原因;同时,假体松动产生的磨损颗粒会引起生物免疫反应,大量的巨噬细胞聚集在界面上吞噬各类假体材料颗粒,诱导破骨细胞的生成,致使发生骨溶解等恶性循环,且造成的局部免疫环境恶化也容易引起细菌感染。因此,开发兼具抗菌和成骨作用的骨科假体材料具有重要的临床转化价值。
聚醚醚酮(Polyetheretherketone,PEEK)作为最接近骨组织弹性模量的常见内植物材料之一,在力学性能方面是最接近人体骨组织的常见可用材料之一,然而,其生物惰性限制了其临床广泛应用,目前临床上仅在脊柱脊髓外科的椎间融合器方面广泛使用,在人工关节假体和接骨板等方面的研究很少报道。因此,如何通过材料表面改性,既能保留PEEK材料固有的力学优势,又能提高其表面活性,是目前PEEK材料在人工关节假体等应用方面亟需解决的步骤和从基础研究走向临床的关键。
皮肤作为第一道防线,与环境接触,暴露在无数的微生物面前。在这些微生物中,表皮葡萄球菌是驻留在皮肤上最丰富的细菌,并且通常与其宿主具有良性关系,通过产生脂肽调节宿主对有害病原体入侵的免疫反应,在宿主防御中发挥重要作用。而现有技术中并未有来自表皮葡萄球菌的脂多肽LP01是否能够用于人工关节内植物中的相关研究。
发明内容
有鉴于此,本发明的目的在于提供一种兼具抗菌和成骨性能的人工关节假体材料LP01-PS。
为了实现上述发明目的,本发明提供了以下技术方案:
本发明提供了一种人工关节假体材料LP01-PS,所述LP01-PS的原料包括聚醚醚酮、LP01和聚多巴胺。
本发明还提供了一种上述人工关节假体材料LP01-PS的制备方法,包括如下步骤:将聚醚醚酮进行磺化和水热处理后,得表面多孔的磺化聚醚醚酮;将磺化聚醚醚酮、聚多巴胺粉末、LP01粉末和Tris溶液混合,得混合物,摇动合成,得LP01-PS。
优选的,所述聚醚醚酮为聚醚醚酮棒或聚醚醚酮圆盘。
优选的,所述聚醚醚酮棒的直径为2mm,长度为10mm。
优选的,所述聚醚醚酮圆盘的直径为10mm,厚度为1mm。
优选的,所述磺化和水热处理的步骤包括:将聚醚醚酮在室温下用98%的浓硫酸磺化处理8min,清洗后,120℃水浴4h,得到表面多孔的磺化聚醚醚酮。
优选的,所述混合物中聚多巴胺的浓度为1-25mg/ml,所述混合物中LP01的浓度为1-25mg/ml。
优选的,所述Tris溶液的摩尔浓度为0.01M,所述Tris溶液的pH值为8.5。
优选的,所述摇动合成的转速为5rpm,时间为1小时。
优选的,摇动合成后,去除未结合和松散结合的颗粒。
本发明的有益效果:
本发明成功制备获得的富载LP01的LP01-PS,具有良好的弹性模量、细胞和动物生物相容性,对大肠杆菌(E.coli)和金黄色葡萄球菌(S.aureus)的增殖、代谢、扩散等有直接的抑制作用,促进巨噬细胞中Cathelicidin LL-37、mBD-2、mBD-3、mBD-4的表达,增强巨噬细胞对S.aureus和E.coli增殖、代谢、扩散等的抑制作用。另外,本发明制备获得的LP01-PS具有促骨髓间充质干细胞成骨分化的作用,能够提高大鼠骨髓间充质干细胞(rBMSCs)矿化能力、成骨相关基因ALP、OCN、COL-1、BMP-2、OPN、RUNX2、BSP、OC表达,促进SD大鼠股骨髁间植入材料周围新生骨的生长。

本发明提供了一种人工关节假体材料LP01-PS,所述LP01-PS的原料包括聚醚醚酮、LP01和聚多巴胺。
在本发明中,所述LP01为人表皮葡萄球菌来源的脂肽LP01,其分子式如下所示:
其蛋白结构如图1A左所示,本发明对于脂肽LP01的具体来源以及具体制备方法没有特殊限定,只要是合成的分子式如上所述即可,在本发明具体实施例中,人表皮葡萄球菌来源的脂肽LP01由上海吉尔生化有限公司合成。本发明对于聚醚醚酮和聚多巴胺的具体来源没有特殊限定,采用本领域常规市售产品均可。
本发明还提供了一种上述人工关节假体材料LP01-PS的制备方法,包括如下步骤:将聚醚醚酮进行磺化和水热处理后,得表面多孔的磺化聚醚醚酮;将磺化聚醚醚酮、聚多巴胺粉末、LP01粉末和Tris溶液混合,得混合物,摇动合成,得LP01-PS。
在本发明中,所述聚醚醚酮优选的为聚醚醚酮棒或聚醚醚酮圆盘,所述聚醚醚酮棒的直径优选为2mm,长度优选为10mm;所述聚醚醚酮圆盘的直径优选为10mm,厚度优选为1mm。在本发明中,所述磺化和水热处理的步骤优选的包括:将聚醚醚酮在室温下使用98%的浓硫酸磺化处理8min,清洗后,120℃水浴4h,以去除材料表面残余酸性物质,得到表面三维多孔的磺化聚醚醚酮;所述清洗优选的用去离子水反复冲洗,所述冲洗的次数优选为3次,所述水浴优选的在水热釜中进行。在本发明中,将磺化聚醚醚酮、聚多巴胺粉末、LP01粉末和Tris溶液混合得混合物后,所述混合物中聚多巴胺的浓度优选为1-25mg/ml,更优选为5-25mg/ml,所述混合物中LP01的浓度优选为1-25mg/ml,更优选为5-25mg/ml。在本发明中,所述Tris溶液的摩尔浓度优选为0.01M,所述Tris溶液的pH值优选为8.5;所述摇动合成的转速优选为5rpm,所述摇动合成的时间优选为1小时;摇动合成后,优选的需去除未结合和松散结合的颗粒,所述去除未结合和松散结合颗粒的方法优选的为用双蒸水轻柔冲洗,所述冲洗的次数优选为5次。

PEEK的原材料购自德国盖尔公司,然后切割成直径2mm、长度10mm的棒(rod)或直径10mm、厚度1mm的圆盘(disc)。用98%的浓硫酸在室温下磺化PEEK 8分钟,去离子水反复冲洗3次后,在水热釜中120℃水浴4h,以去除材料表面残余酸性物质,得到表面多孔磺化PEEK(SPEEK)。将SPEEK棒放入烧杯中,加入0.01M Tris pH 8.5和25mg/ml聚多巴胺粉末(PDA,中国上海阿拉丁生物化学技术有限公司)。然后加入人工合成的LP01粉末(99%,中国上海吉尔生化生物有限公司),使烧杯中LP01粉末含量为25mg/mL。将这些混合物以5rpm的速度摇动1小时,然后用双蒸水轻柔冲洗去除任何松散结合的颗粒得LP01-PS,记为25LP01-PS。上述合成过程见图1A右。
实施例2
与实施例1的区别在于烧杯中聚多巴胺粉末的浓度为5mg/ml,LP01粉末的浓度为5mg/ml,其余均同实施例1,得LP01-PS,记为5LP01-PS。
实施例3
与实施例1的区别在于烧杯中聚多巴胺粉末的浓度为1mg/ml,LP01粉末的浓度为1mg/ml,其余均同实施例1,得LP01-PS,记为1LP01-PS。
对比例1
与实施例3的区别在于未加入LP01粉末,其余均同实施例3,记为PS。
实施例4
以未添加聚多巴胺粉末和LP01粉末的纯SPEEK为对照组,记为Con组。
使用液体接触角测量仪(LSAMOB-L,LAUDA Scientific,Hazi,Germany)测试实施例1、实施例2、实施例3、对比例1以及Con组各组材料的接触角,并使用酶标仪(FC,Thermo,New York,USA)检测LP01-PS在PBS中的药物释放曲线。另外,使用FEI NovaNano450扫描电子显微镜(JEOL,JSM-6310LV,Tokyo,Japan)观察各组材料表面。结果分别如图1B、C、D所示。
由图1B可以看出,LP01粉末添加量越多,在多孔表面富集的脂蛋白球越密集,LP01-PS的液体接触角越大。disc和rod在PBS中的药物释放曲线显示,同等富载浓度的情况下,disc比rod释放LP01更快更多(图1C)。这与disc表面积更大有关。LP01在PBS中溶解度较差,扫描电镜显示,大量LP01聚集成葡萄串样小球体,嵌在磺化PEEK的表面三维孔中(图1D)。由于LP01绝大部分富集到磺化PEEK的表面孔隙内部,而非磺化PEEK表面平坦处。因此,25LP01-PS组表面的孔隙最细小、最分散,液体接触角等理化特性最佳。
实施例5
LP01-PS的细胞和动物生物相容性评估
将RAW246.7(CL-0190,Wuhan Zishan Biotechnology Co.LTD)和rBMSCs(STCC5011,Wuhan Zishan Biotechnology Co.LTD)用于细胞相容性测试。RAW246.7和rBMSCs在添加10%胎牛血清(SH3010902HI,Gibco,Grand Island,USA)的DMEM(MT15013LX,Corning,NewYork,USA)培养基中培养。将实施例1-3、对比例1以及实施例4(Con组)共5组圆盘分别水平放置于24孔板中。用细胞免疫荧光(LSM 510meta,Zeiss,Oberkochen,Germany)检测细胞骨架,使用扫描电镜(JEOL,JSM-6310LV,Tokyo,Japan)观察细胞形态、粘附、细胞伪足。采用CCK-8法(C0038,Beyotime Bio-Tech)检测不同时间点细胞增殖情况,采用ELISA(C0017,Beyotime Bio-Tech)检测培养基中乳酸脱氢酶(LDH)的变化。其中25LP01-PS与巨噬细胞RAW246.7和大鼠rBMSCs共培养3天后,所得的结果分别如图1G、H、I、J所示。结果表明,各组LDH释放量(图1G)和细胞数量(图1H)无明显差异。两类细胞可以很好地粘附在不同组的材料表面,并且都可以产生足够多的伪足(图1I)。细胞免疫荧光显示各组细胞表面细胞骨架生长良好、分布均匀(图1J)。表明本发明LP01-PS材料在细胞水平的生物安全性良好
在小鼠BABL/c小鼠(Shanghai SLAC Laboratory Animal Co.,Ltd)的背部皮下植入25LP01-PS,然后记录不同时间点小鼠的体重变化。使用全自动血生化分析仪(ADVIA2400,Zeiss,Oberkochen,Germany)检测外周静脉血生化变化,使用全自动血细胞分析仪(BC6800,Mindray,Shenzhen)检测外周血细胞变化。1、4周后取出小鼠的心、肝、脾、肺、肾进行HE染色,使用光镜显微镜(LSM 510meta,Zeiss,Oberkochen,Germany)观察器官组织是否发生变性等变化。结果如图1E、F和图2所示。LP01-PS皮下植入小鼠背部后0天、7天、28天,处死小鼠采集其主要脏器进行HE染色观察。心、肝、脾、肺、肾,HE染色结果显示各组脏器状态良好(图1E)。在不同时间点,小鼠体重变化无统计学差异(图1F)。由图2可知,不同时间点,各组小鼠外周血细胞和外周血生化指标,差异无统计学意义。表明本发明LP01-PS材料在动物水平的生物安全性良好。
实施例6
体外抗菌实验
将S.aureus(SF8300,Sangon Biotech,Shanghai)和E.coli(ATCC35218,SangonBiotech,Shanghai)用PBS缓冲液重悬,调整细菌浓度至107CFU/ml。将细菌悬液分为5组,分别为对照组(Con,实施例4)、含有PDA的培养基组(PS,对比例1)、含有1mg/ml药物的LP01-PS的培养基组(1LP01-PS实施例3)、含有5mg/ml药物的LP01-PS的培养基组(5LP01-PS,实施例2)和含有25mg/ml药物的LP01-PS的培养基组(25LP01-PS,实施例1)。
(1)LP01-PS直接抗菌的体外研究
首先,将10mm×10mm×1mm圆片分别放置于24孔板中,每孔放置1个圆片。然后在每个孔中接种500ul浓度为~106CFU/ml的菌液,37℃共培养24小时。使用SPM法(110704011,BKMAN,Changde,China)对原始TSB培养基中的浮游细菌进行定量分析。无菌钳取出培养的标本,用新鲜的PBS轻轻清洗,去除非粘附细菌。将圆片放入1ml新鲜PBS中,使用超声波清洗器(150W,B3500S-MT,Branson Ultrasonics Co,Shanghai)振动10分钟,并使用混合涡旋器(Vortex Genie 2,Scientific Industries,Bohemia,USA)混合2分钟,去除附着的细菌。然后使用SPM法计算菌悬液中的活菌数,各组浮游菌和材料表面粘附菌的数量使用log10 CFU/ml表示。
随后,将圆片放置于新的24孔板中,经过PBS洗涤后用LIVE/DEAD BacLight混合染料(L7007,Invitrogen,Carlsbad,USA)染色30分钟,并在荧光显微镜(LSM 510meta,Zeiss,Oberkochen,Germany)下观察和拍照。死亡的细菌被染成红色,而活的细菌被染成绿色。接下来,使用2.5%戊二醛在4℃下固定48小时,然后用梯度酒精溶液(50、60、70、80、90、95和100%v/v)干燥。干燥后,在这些圆片上喷金,使用扫描电镜(JEOL,JSM-6310LV,Toky,Japan)观察各组材料表面细菌分布。
针对LP01-PS对S.aureus直接抗菌性能的研究,使用rt-PCR方法(T100,Bio-Rad,Shanghai,China)检测以下基因的表达水平:生物膜形成特异性基因fnbA和fnbB mRNA的表达水平,了解S.aureus生物膜形成的情况。TCA/PTS能量代谢途径相关基因sucA和sucB的表达水平,评估S.aureus在代谢水平的变化。agr群体应激感应***中主要基因agrA和agrC的表达水平,评估S.aureus的应激反应能力。促进S.aureus生物膜降解的基因sak和HtrA的表达水平,了解S.aureus生物膜分解的情况。
针对LP01-PS对E.coli直接抗菌性能的研究,使用rt-PCR方法(T100,Bio-Rad,Shanghai,China)检测以下基因的表达水平:检测材料表面E.coli的TCA/PTS能量代谢途径相关基因lacD、lacB mRNA表达水平,了解E.coli的代谢途径。同时检测活力相关基因csgA、fliA、fimA和毒力相关基因stx2、ehaA的mRNA表达水平,评估E.coli的生长状态和潜在毒力。同时,检测E.coli生物膜合成相关基因ompA、pgaA、luxS的mRNA表达水平,评估E.coli的生物膜形成能力,以及与锌稳态相关的基因ykgM、zinT、ZunA和ZunB的mRNA表达水平,评估E.coli的适应环境的能力。
结果如图3所示。LP01-PS和细菌共培养体系中,在材料表面负载的LP01越多,扫描电镜显示LP01-PS组材料表面的S.aureus和E.coli数量逐渐减少(图3A),细菌活死染色显示LP01-PS组材料表面E.coli的死亡比例逐渐增多(图3B)。材料表面粘附菌mRNA定量分析结果显示S.aureus菌生物膜形成特异性基因fnbA和fnbB、TCA/PTS能量代谢途径相关基因sucA和sucB表达减少,S.aureus的agr群体应激传感***主要基因agrA和agrC、促进生物膜降解的S.aureus sak和HtrA基因表达增加(图3C),E.coli TCA/PTS能量代谢途径相关基因lacD和lacB、运动相关基因csgA、fliA和fimA、毒力相关基因stx2和ehaA、生物膜合成相关基因ompA、pgaA和luxS、锌稳态相关基因ykgM、zinT、ZunA和ZunB mRNA表达量下降,组间差异有统计学意义(图3D)。稀释涂板法测得的浮游菌和材料表面粘附菌的数量就越少,组间差异有统计学意义(图3E)。
(2)LP01-PS增强巨噬细胞抗菌性能的体外研究
为探讨LP01-PS对巨噬细胞抗菌性能的影响,在24孔板中分别放置不同组材料,分为Con组、PS组、1LP01-PS组、5LP01-PS组和25LP01-PS组。每组均有3个重复孔,每个孔内放置一个10mm×10mm×1mm圆片。将含有2.5×105RAW264.7的细胞接种于每个孔内,然后加入含有100U/ml青霉素、100U/ml链霉素和10%胎牛血清(SH3010902HI,Gibco,Grand Island,USA)的DMEM细胞培养基(MT15013LX,Corning,NewYork,USA)。将板置于37℃、5%CO2的细胞培养箱中进行培养。经过1天的培养,大部分细胞成功粘附生长,然后更换新鲜的细胞培养基,继续培养3天。
使用rt-PCR、Westernblot和免疫荧光法检测巨噬细胞TLR2/P65信号通路mRNA和蛋白水平的变化,并通过Elisa法检测巨噬细胞分泌Cathelicidin LL-37、小鼠β-defensin2、小鼠β-defensin3和小鼠β-defensin4的水平。使用rt-PCR检测RAW246.7中LL-37、mBD2、mBD3和mBD4 mRNA水平的变化。在细胞培养3天后,当细胞融合度达到90%时,更换新的细胞培养基,并加入S.aureus和E.coli(500ul,1×106CFUs/mL),共同培养24小时。最后,检测各组的细菌增殖和代谢指标。
LP01-PS通过TLR2/P65信号通路增强巨噬细胞抗菌的体外研究
随机将25LP01-PS的12个圆盘平均分成4组,装入24孔板中,每孔1个圆盘。当RAW246.7达到90%融合时,为其替换为新鲜培养基。一共分为4组,25LP01-PS组不加任何化学阻断剂,25LP01-PS+OxPAPC组加入OxPAPC 30ul/ml,25LP01-PS+Bay11组加入Bay11-70825uM,25LP01-PS+OxPAPC+Bay11组同时加入OxPAPC 30ul/ml和Bay11-70825uM。3天后,加入S.aureus或E.coli(500ul,1×106CFUs/mL),37℃共培养24小时。然后,检测以上四组的细菌增殖、代谢指标。
为了进行巨噬细胞TLR2/P65信号通路基因沉默,将小鼠TLR2或NF-κB/p65特异性短发夹RNA克隆到pLL3.7载体(Addgene,Shanghai,China)中。制备出含特异性shRNA的pLL3.7构建物4mg,包装质粒psPAX24g,包膜质粒pMD2.G 2g的混合物,然后通过磷酸钙沉淀法转染293T细胞。48小时后,收集含有小鼠TLR2或NF-κB/p65特异性shRNA的慢病毒,转染RAW246.7。用微量注射器向10cm培养皿的RAW246.7周围注入1012/L的沉默TLR2或NF-κB/p65基因的慢病毒50ul,采用rt-PCR和Western blot检测shRNA阻断巨噬细胞TLR2或NF-κB/p65表达的效率,制备出符合沉默要求的各组RAW246.7。随机将25-LP01-PS的12个圆盘平均分成4组,装入24孔板中,每孔1个圆盘。
25LP01-PS+Scramble组加入不表达shRNA的慢病毒空载体转染的RAW246.7,25LP01-PS+TLR2 shRNA组加入表达TLR2 shRNA的慢病毒转染的RAW246.7,25LP01-PS+p65shRNA组加入表达p65 shRNA的慢病毒转染的RAW246.7,25LP01-PS+TLR2/p65 shRNA组同时加入表达TLR2和p65 shRNA的慢病毒转染的RAW246.7。当RAW246.7达到90%融合时,加入S.aureus或E.coli(500ul,1×106CFUs/mL),37℃共培养24小时。然后,检测以上四组的细菌增殖、代谢指标。
为了进行巨噬细胞TLR2/P65信号通路基因高表达,将TLR2或NF-κB/p65过表达质粒(zorin,shanghai)、PsPAX、pmD2G质粒共转染293T细胞。约48小时后收集慢病毒上清,经膜过滤和超离心纯化浓缩病毒。流式细胞术检测TLR2或NF-κB/p65基因高表达慢病毒滴度。用微量注射器向10cm培养皿的RAW246.7周围注入1012/L的高表达TLR2或NF-κB/p65基因的慢病毒50ul,采用rt-PCR和Western blot检测巨噬细胞TLR2或NF-κB/p65高表达的效率,制备出符合高表达要求的各组RAW246.7。随机将1-LP01-PS的12个圆盘平均分成4组,装入24孔板中,每孔1个圆盘。1LP01-PS+Mock组加入不表达mRNA的慢病毒空载体转染的RAW246.7,1LP01-PS+TLR2 over组加入表达TLR2 mRNA的慢病毒转染的RAW246.7,1LP01-PS+p65 over组加入表达p65 mRNA的慢病毒转染的RAW246.7,1LP01-PS+TLR2/p65 over组同时加入表达TLR2和p65 mRNA的慢病毒转染的RAW246.7。当RAW246.7达到90%融合时,加入S.aureus或E.coli(500ul,1×106CFUs/mL),37℃共培养24小时。然后,检测以上四组的细菌增殖、代谢指标。
上述实验结果如图4-图6所示。
LP01-PS与巨噬细胞共培养3天后,在材料表面负载的LP01越多,巨噬细胞迁移速度越快(图4A);巨噬细胞迁移数量越多(图4B)。同时,LP01-PS促进巨噬细胞LL-37、mBD2、mBD3、mBD4的细胞外分泌(图4C);促进巨噬细胞LL-37、mBD2、mBD3、mBD4的mRNA表达(图4D)。以上结果提示,LP01-PS增强巨噬细胞本身的抗菌能力,且巨噬细胞分泌的抗菌肽也可增强周围其他组织细胞的抗菌能力。
在LP01-PS、巨噬细胞和细菌共培养体系中,负载的LP01越多,稀释涂板法测得的浮游菌和LP01-PS表面粘附菌的数量就越少(图4E)。LP01-PS表面粘附菌mRNA定量分析结果显示,S.aureus生物膜形成特异性基因fnbA和fnbB、TCA/PTS能量代谢途径相关基因sucA和sucB表达减少。然而,S.aureus的agr群体应激传感***主要基因agrA和agrC、促进生物膜降解的S.aureus sak和HtrA基因表达增加(图4F)。E.coli TCA/PTS能量代谢途径相关基因lacD和lacB、运动相关基因csgA、fliA和fimA、毒力相关基因stx2和ehaA、生物膜合成相关基因ompA、pgaA和luxS、锌稳态相关基因ykgM、zinT、ZunA和ZunB mRNA表达量下降(图4G),组间差异有统计学意义。扫描电镜显示LP01-PS组表面的S.aureus和E.coli数量逐渐减少(图4H)。细菌活死染色显示LP01-PS组表面E.coli的死亡比例逐渐增多(图4I)。以上结果提示,LP01-PS增强巨噬细胞体外杀灭S.aureus和E.coli的能力。
经过LP01-PS与巨噬细胞共培养3天后,rt-PCR结果表明,在LP01-PS组中,巨噬细胞TLR2和P65的mRNA表达量明显增加(图5B)。同时,随着表面负载的LP01越多,巨噬细胞的免疫荧光显示TLR2和P65蛋白信号量也随之增加,并且P65蛋白在细胞核内的部分含量也增加(图5A)。WB及其定量分析结果也呈现相同趋势(图5C)。当使用TLR2信号通路阻断剂OxPAPC和P65信号通路阻断剂BAY11-7082处理巨噬细胞后,巨噬细胞的抗菌肽LL-37、mBD2、mBD3和mBD4的细胞外分泌和mRNA表达量明显降低(图5E),同时巨噬细胞的迁移速度也显著减缓(图5G)。这些结果表明,LP01-PS能够通过TLR2/P65信号通路增强巨噬细胞的抗菌能力。
在LP01-PS、巨噬细胞和细菌的共培养体系中,扫描电镜观察表明,在阻断剂处理后的巨噬细胞组中,表面的S.aureus和E.coli数量逐渐增加。细菌活死染色显示E.coli的死亡比例逐渐下降(图5D)。同时,阻断剂处理后的巨噬细胞组中,粘附在LP01-PS表面的细菌(mRNA定量分析)中的E.coli TCA/PTS能量代谢途径相关基因lacD和lacB、运动相关基因csgA、fliA和fimA、毒力相关基因stx2和ehaA、生物膜合成相关基因ompA、pgaA和luxS、锌稳态相关基因ykgM、zinT、ZunA和ZunB的mRNA表达量增加(图5F)。在粘附在LP01-PS表面的S.aureus中,生物膜形成特异性基因fnbA和fnbB、TCA/PTS能量代谢途径相关基因sucA和sucB的表达量也增加。而S.aureus的agr群体应激传感***主要基因agrA和agrC、促进生物膜降解的基因sak和HtrA的表达量下降(图5H)。另外,稀释涂板法测得的阻断剂组浮游菌和材料表面粘附菌的数量增加(图5I),组间差异有统计学意义。综上结果提示,TLR2/P65信号通路化学阻断剂能够降低巨噬细胞的抗菌能力。
慢病毒沉默或高表达巨噬细胞TLR2/p65信号通路后,巨噬细胞的抗菌能力的体外变化结果如图6所示。LP01-PS与巨噬细胞共培养3天,加入等量的TLR2/p65信号通路沉默或高表达的慢病毒,TLR2/p65信号通路沉默或高表达的慢病毒处理后,巨噬细胞的抗菌肽LL-37、mBD2、mBD3、mBD4的细胞外分泌(图6A)和mRNA表达(图6B)表现出不同的趋势。具体而言,沉默时这些指标下降,而高表达时则上升。综上所述,这些结果提示,TLR2/p65信号通路沉默慢病毒能够降低巨噬细胞的抗菌能力,而TLR2/p65信号通路高表达慢病毒则可以增强巨噬细胞的抗菌能力。
在LP01-PS、巨噬细胞和细菌共同培养的体系中,通过涂板稀释检测发现,E.coli和S.aureus的浮游菌和材料表面粘附菌数量变化趋势是,当TLR2/p65信号通路沉默时数量增加,当TLR2/p65信号通路高表达时数量减少(图6C)。同时,扫描电镜显示材料表面的S.aureus和E.coli数量变化趋势也是相同的,即沉默时数量增加,高表达时数量减少(图6D)。通过细菌活死染色,发现当TLR2/p65信号通路沉默时,E.coli的死亡比例下降,而高表达时则上升(见图6E)。这表明,TLR2/p65信号通路沉默慢病毒能够降低巨噬细胞抑制细菌增殖的能力,而TLR2/p65信号通路高表达慢病毒则能够提高其抑制细菌增殖的能力。
材料表面粘附菌mRNA定量分析结果表明,E.coli在TCA/PTS能量代谢途径相关基因lacD和lacB、运动相关基因csgA、fliA和fimA、毒力相关基因stx2和ehaA、生物膜合成相关基因ompA、pgaA和luxS、锌稳态相关基因ykgM、zinT、ZunA和ZunB的mRNA表达量沉默时增加,高表达时下降(图6F)。而S.aureus的生物膜形成特异性基因fnbA和fnbB、TCA/PTS能量代谢途径相关基因sucA和sucB的表达变化趋势是,沉默时增加,高表达时下降。另外,S.aureus的agr群体应激传感***主要基因agrA和agrC、促进生物膜降解的S.aureus sak和HtrA基因的表达变化趋势是,沉默时下降,高表达时增加(图6G)。以上结果表明,TLR2/p65信号通路沉默慢病毒处理的巨噬细胞抑制细菌代谢的能力下降,而TLR2/p65信号通路高表达慢病毒处理的巨噬细胞则能够提高对细菌代谢的抑制能力。
实施例7
体内抗菌实验
小鼠背部皮下植入物材料相关细菌感染模型的建立
在BALB/c小鼠背部建立皮下植入物材料相关感染模型,具体步骤如下:经腹腔注射3%戊巴比妥钠,待小鼠麻醉后,背部剃毛,消毒,正中部切开1cm切口,一共分五组,分别向每组小鼠背部植入Con(实施例4)、PS(对比例1)、1LP01-PS(实施例3)、5LP01-PS(实施例2)和25LP01-PS(实施例1)不同材料,每组3只,同时向植入假体材料背部周围注入100ul的自带荧光的金黄色葡萄球菌S.aureus(1×107CFU/ml),然后仔细缝合伤口。另外选择5只小鼠作为同源背景样品,分别在背部植入Con组圆片,并将100ul PBS注射到圆片背部正中皮肤下。在无特定病原体(SPF)条件下让这些小鼠自由获取水和食物,饲养14天。
小鼠背部材料周围皮肤的生物荧光检测
为了观察5组小鼠抗感染效果的时间过程,首先吸入2%异氟醚麻醉BALB/c小鼠,然后通过IVIS Lumina XRMS成像***在术后0、1、3、7、14天进行背部拍照。每只小鼠成像60s,获取小鼠胡光子信号,生成伪彩色图像。相应的生物发光光子强度(PI)表示为生物发光通量:photons/second/cm2/steradian(sr).此外,在小鼠背部的生物发光区域指定为感兴趣区域(ROIs),用以量化细菌含量,然后通过瑞金医院上海市伤骨科研究所提供的IVIS软件(Caliper Life Sciences,PerkinElmer)计算ROI的生物发光通量。
小鼠背部材料周围皮肤病理组织分析
收集各组内置物材料周围软组织,4℃固定于10%***缓冲液中3天,再用新鲜PBS冲洗3遍,在梯度浓度酒精溶液中脱水,并包埋在石蜡中。使用切片机(Leica,Hamburg,Germany)将样品切片。最后在二甲苯溶液中脱蜡,并利用May-Grünwald Giemsaand hematoxylin and eosin(H&E)stainings,通过光学显微镜检查组织学变化。同期,收集各组材料周围软组织,经Trizol法提取RNA,通过rt-PCR法检测各组周围组织中小鼠Cathelicidin LL-37、β-防御素2(mBD-2)、β-防御素3(mBD-3)、β-防御素4(mBD-4)在mRNA水平变化;取材料周围皮下软组织,放入液氮中冻存,再在高速研磨机中研磨成粉末状、称重,溶解于PBS中,用Elisa法检测Cathelicidin LL-37、β-防御素2(mBD-2)、β-防御素3(mBD-3)、β-防御素4(mBD-4)在组织蛋白水平变化,并做统计分析。
小鼠背部材料周围皮肤中细菌数量检测
为了定量评价动物模型体内细菌量,无菌条件下取出各组皮下内置物垫片,首先用新鲜的PBS轻轻清洗样品两次以去除游动细菌。然后,将它们浸入1mLPBS中,以150kHz超声处理10分钟,然后涡旋2分钟以去除粘附的细菌,在PBS溶液中连续稀释,取100μL稀释剂均匀分布在SBA上,一式三份。此外,获取植入物周围的软组织,称重,并使用高速匀浆机(中国上海晶鑫实业有限公司)在装有1mL PBS的无菌管中匀浆,软组织匀浆在PBS溶液中连续稀释,取100μL稀释剂均匀分布在SBA上,一式三份。最后,这些平板在37℃下培养过夜,并计算细菌菌落的数量,并分别对材料周围组织和材料表面做定量分析。
小鼠背部材料周围皮肤中细菌代谢能力检测
搜集上述实验中各组皮下内置物圆片,首先用新鲜的PBS轻轻清洗样品两次以去除游动细菌。然后,将它们浸入1mLPBS中,以150kHz超声处理10分钟,然后涡旋2分钟以去除粘附的细菌。再经过离心、PBS漂洗3次,Trizol法提取各组细菌RNA,通过rt-PCR方法检测金黄色葡萄球菌S.aureus的生物膜形成特异基因fnbA、fnbB的mRNA水平变化,金黄色葡萄球菌TCA/PTS能量代谢路径相关基因sucA、sucB的变化;金黄色葡萄球菌的agr群体感应***主要基因agrA、agrC的变化,金黄色葡萄球菌控制生物膜降解的基因sak、HtrA的变化。
上述结果如图7所示。在小鼠背部皮下感染模型建立后,随着LP01-PS表面负载的LP01数量增加,活体荧光检测***在0、1、3、7、10、14天显示小鼠背部荧光强度逐渐降低(图7A和B)。在第14天,取出小鼠背部各组材料并超声震洗LP01-PS表面,分离出各组的S.aureus。LP01-PS表面粘附菌mRNA定量分析结果显示,S.aureus生物膜形成特异性基因fnbA和fnbB、TCA/PTS能量代谢途径相关基因sucA和sucB表达减少,而S.aureus的agr群体应激传感***主要基因agrA和agrC、促进生物膜降解的S.aureus sak和HtrA基因表达增加(图7C)。组间差异具有统计学意义。此外,随着LP01-PS表面负载的LP01数量增加,稀释涂板法测得的材料表面粘附菌和材料周围皮肤组织中S.aureus数量均减少(图7D)。以上结果提示,LP01-PS在体内具有明显的抗S.aureus作用。
在LP01-PS表面负载的LP01数量越多的情况下,材料周围皮肤组织抗菌肽LL-37、mBD2、mBD3、mBD4的蛋白分泌和mRNA表达均增多,差异具有统计学意义(图7E,图7F)。此外,扫描电镜结果显示LP01-PS组表面的S.aureus数量减少(图7G)。HE染色、MPO免疫组化、Giemsa染色,结果表明,在LP01-PS表面负载的LP01数量越多的情况下,周围皮肤浸润的炎症细胞越少,炎症渗出物越少,细菌数量也越少。在25LP01-PS组的材料周围皮肤中,几乎没有炎症反应和细菌残留(图7H)。这些结果提示,LP01-PS能够提高其周围皮肤的抗菌能力。
以上结果表明,本发明LP01-PS在体内外具有显著的双重抗菌能力。在体外,LP01-PS不但对S.aureus和E.coli的增殖、代谢、扩散和毒力具有直接的抑制作用,而且间接提高巨噬细胞对S.aureus和E.coli的杀灭作用。在体内,LP01-PS能够促使周围皮肤组织增加LL-37、mBD-2、mBD-3、mBD-4的分泌。可见,LP01-PS不仅在表面直接抗菌,还能提高巨噬细胞或周围组织的免疫抗菌能力,从而有效杀灭隐藏在假体周围组织中的细菌。
实施例8
体外成骨实验
LP01-PS体外促进rBMSCs成骨分化
一共分5组,分别将Con、PS、1LP01-PS、5LP01-PS和25LP01-PS装入24孔板中,每孔1枚10mm×10mm×1mm圆片,每组3个复孔。按照每孔5×105rBMSCs进行接种,且每孔含青霉素100U/ml、链霉素100U/ml和10%胎牛血清的DMEM细胞培养液500ul,放入37℃、5%CO2的细胞培养箱培养。接种1天后,大部分细胞成功贴壁生长,然后更换新鲜细胞培养液。继续培养细胞,每2-3天更换一次新鲜细胞培养基,搜集各组骨髓间充质干细胞,在第14天的时间点收集细胞,通过rt-PCR检测成骨相关基因ALP、OCN、COL-1、BMP-2、OPN、RUNX2、BSP、OC在mRNA水平的变化。通过ALP染色检测各组细胞矿化能力。进行茜素红(ARS)染色以评估细胞外钙沉积形成。使用rt-PCR、Western blot和免疫荧光法检测rBMSCs的Wnt/β-catenin信号通路相关基因LRP-5、LRP-6、Axin2、β-catenin的mRNA和蛋白水平的变化。
LP01-PS通过Wnt/β-catenin信号通路体外促进rBMSCs成骨分化
将25LP01-PS装入24孔板中,共计分为3组,每孔1枚10mm×10mm×1mm圆片,每组3个复孔。按照每孔5×105rBMSCs进行接种,且每孔含青霉素100U/ml、链霉素100U/ml和10%胎牛血清的DMEM细胞培养液500ul,放入37℃、5%CO2的细胞培养箱培养。接种1天后,大部分细胞成功贴壁生长,然后更换新鲜细胞培养液。25LP01-PS组不加任何化学阻断剂,25LP01-PS+20uM FH535组加入20uM FH535,25LP01-PS+40uM FH535组加入40uM FH535。在建立共培养***后的第14天收集细胞,通过rt-PCR检测成骨相关基因ALP、OCN、COL-1、BMP-2、OPN、RUNX2、BSP、OC在mRNA水平的变化。通过ALP染色检测各组细胞矿化能力。进行茜素红(ARS)染色以评估细胞外钙沉积形成。
体内成骨实验
LP01-PS增强rBMSCs成骨分化的体内研究
SD大鼠股骨髁间植入LP01-PS材料建立动物成骨模型
根据前人研究,在SD大鼠股骨髁间建立动物成骨模型。具体步骤如下:经腹腔注射3%戊巴比妥钠,待小鼠麻醉后,右下肢剃毛、彻底消毒,右膝内侧正中部切开1cm切口,通过电钻和克氏针向右膝股骨髁间打直径2mm小孔,然后向每组股骨髁间分别植Con、PS、1LP01-PS、5LP01-PS和25LP01-PS不同材料,材料为2mm×2mm×10mm圆柱体,每组3只,然后仔细缝合伤口。在无特定病原体(SPF)条件下让这些大鼠自由获取水和食物,饲养8周。
多色连续荧光标记法和Van Gieson染色测定SD大鼠成骨情况
应用多色连续荧光标记方法标记那些新形成的骨组织。术后第4和6周,依次腹腔注射茜素红S(30mg/kg,AL,Sigma-Aldrich,USA),钙黄绿素(20mg/kg,CA,Sigma-Aldrich,USA)。手术后8周,将这些大鼠安乐死,取下带棒的右股骨,锯成合适的大小,然后将这些股骨固定在10%的甲醛缓冲液中用于后续分析。接下来,将这些外置股骨用乙醇脱水并嵌入甲基丙烯酸酯中,无需脱钙。然后在锯切切片机(Leica SP1600,Munich,Germany)上制作切片并抛光至约50um的厚度,然后通过共聚焦激光扫描显微镜(CLSM)观察。然后,切片用vanGieson's picrofuchsin染色进行组织学观察
显微CT评估
使用Micro-CT(Skyscan 1172,Bruker Micro-CT,Munich,Germany)扫描上述准备好的股骨,以研究各组内置入材料周围的骨组织变化。扫描是在最大X射线能量和360°范围、50kV下进行的,分辨率为18um。三维(3D)图像由Nrecon(Bruker micro CT,Skyscan公司,Munich,Germany)重建,并使用CTAn程序进行分析(Bruker micro CT,Skyscan公司,Munich,Germany)。选择骨髓腔内半径为0.5mm的棒的周围区域来量化新形成的骨。然后是对新骨的骨-材料接触面积百分比(Bone-implant contact,%)、骨矿物质密度(BMD)、骨体积分数(骨体积/总体积,BV/TV,%)、骨小梁数量(Tb.N,mm-1)、骨小梁厚度(Tb.Th,μm)、骨小梁分离度(Tb.Sp)进行统计分析。
拔出测试(Pull-out test)
术后8周每组取3个样品,用钢丝固定在钢制装置上,试验以1mm/min的加载速率进行,然后记录拔出期间的载荷-挠度曲线。我们将失效载荷定义为最大载荷。
实时荧光定量PCR
分别使用Trizol(12183555,Invitrogen,California,USA)从实验小鼠背部皮肤、RAW246.7、细菌或rBMSCs中分离总RNA,根据制造商的说明逆转录cDNA(A48571,ThermoFisher Scientific Inc.,Waltham,USA)。RT-PCR在96孔板上进行,使用SYBR Premix ExTaq试剂盒(RR001B,TaKaRa,Dalian,China),最终体积为20uL,应用快速RT-PCR***(ABI7500,Applied Biosystems,Carlsbad,USA)。PCR条件为:94℃初始变性5min,36个循环的94℃变性30s,退火40s,72℃延伸40s,最后的72℃延伸5min。得到循环阈值,将数据通过GAPDH归一化为处理,并根据2-ΔΔCt法计算相对表达量。每个实验重复三次。所有引物均从Sangon(Shanghai Shengong Biological Engineering Co.,LTD.Shanghai,China)公司获得。
Western blot分析
不同处理的RAW246.7或rBMSCs细胞在放射免疫沉淀法(RIPA)缓冲液和PhosSTOP(4906837001,Roche,Basel,Switzerland)中被裂解。蛋白浓度通过BCA蛋白测定法(P0010S,Beyotime,Shanghai,China)进行测定,使用10%十二烷基硫酸钠-聚丙烯酰胺凝胶电泳分离等量蛋白(20μg),随后转移到PVDF膜(GVHP04700,Millipore,Massachusetts,USA)。在5%脱脂奶粉封闭2小时后,使用特异性抗体进行免疫染色,包括兔源NF-kB/p65多克隆抗体(1:100dilution,ab209216,Abcam,San Francisco,USA)、兔源TLR2单克隆抗体(1:100dilution,ab32536,Abcam,San Francisco,USA、兔源β-catenin单克隆抗体(1:100稀释,Abcam,旧金山,美国)、兔源Actin多克隆抗体(1:100稀释,Abcam,旧金山,美国)和兔源GAPDH多克隆抗体(1:500稀释,Abcam,旧金山,美国),在4℃下轻轻摇晃过夜。洗涤后,分别使用特异性二抗(1:100dilution,ab205718,Abcam,San Francisco,USA)在37℃下进行处理2小时。通过Thermo Scientific公司的增强化学发光试剂进行免疫标记检测,并使用Scion Image Beta4.02(Scion Corporation,NIH,Maryland,USA)进行信号强度的定量分析。
上述结果见图8。LP01-PS与rBMSCs共培养14天后,随着材料表面PDA和LP01富集量的增加,其成骨分化主要基因ALP、OCN、COL-1、BMP-2、OPN、RUNX2、BSP、OC等明显增加(图8A),rBMSCs向LP01-PS迁移数量增加(图8B),ALP和ARS染色也明显加深(图8C)。LP01-PS植入大鼠股骨后8周,最大拔出力增加(图8D),micro-CT显示材料周围新生骨增加(图8E),新生骨的序列荧光染色显示不同时间点钙盐沉积增加(图8F),材料周围Van Gieson染色显示新生的胶原纤维增加(图8G)。进一步分析显示,新生骨的BMD、骨材料接触面积百分比、BV/TV、Tb.N、Tb.Th均增加、Tb.Sp下降(图8H)。以上结果提示LP01-PS促进rBMSCs体外成骨分化和体内新生骨的生成。
LP01-PS与rBMSCs共培养7天后,大鼠rBMSCs的β-catenin的细胞免疫荧光强度增加(图8I),WB蛋白表达及其定量增加(图8J),WNT/β-catenin信号通路的主要亚基LRP5、LRP6、Axin2、β-catenin的mRNA表达增加(图8K)。WNT/β-catenin信号通路化学阻断剂处理rBMSCs后,rBMSCs的ALP和细胞外钙盐沉积的ARS染色变浅(图8L),成骨基因的表达下降(图8M)。以上结果提示LP01-PS能通过rBMSCs的WNT/β-catenin信号通路促进其体外成骨分化。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种人工关节假体材料LP01-PS,其特征在于,所述LP01-PS的原料包括聚醚醚酮、LP01和聚多巴胺。
2.权利要求1所述人工关节假体材料LP01-PS的制备方法,其特征在于,包括如下步骤:将聚醚醚酮进行磺化和水热处理后,得表面多孔的磺化聚醚醚酮;将磺化聚醚醚酮、聚多巴胺粉末、LP01粉末和Tris溶液混合,得混合物,摇动合成,得LP01-PS。
3.根据权利要求2所述制备方法,其特征在于,所述聚醚醚酮为聚醚醚酮棒或聚醚醚酮圆盘。
4.根据权利要求3所述制备方法,其特征在于,所述聚醚醚酮棒的直径为2mm,长度为10mm。
5.根据权利要求3所述制备方法,其特征在于,所述聚醚醚酮圆盘的直径为10mm,厚度为1mm。
6.根据权利要求2所述制备方法,其特征在于,所述磺化和水热处理的步骤包括:将聚醚醚酮在室温下用98%的浓硫酸磺化处理8min,清洗后,120℃水浴4h,得到表面多孔的磺化聚醚醚酮。
7.根据权利要求2所述制备方法,其特征在于,所述混合物中聚多巴胺的浓度为1-25mg/ml,所述混合物中LP01的浓度为1-25mg/ml。
8.根据权利要求2所述制备方法,其特征在于,所述Tris溶液的摩尔浓度为0.01M,所述Tris溶液的pH值为8.5。
9.根据权利要求2所述制备方法,其特征在于,所述摇动合成的转速为5rpm,时间为1小时。
10.根据权利要求2所述制备方法,其特征在于,摇动合成后,去除未结合和松散结合的颗粒。
