背景技术
胃癌是源自胃黏膜上皮的恶性肿瘤,其在全球范围内的癌症发病率中位居第五,在癌症死亡原因中位居第三,我国是胃癌的高发高死亡地区,占全球发病人数的47%,因此探究胃癌发生发展的分子机制,找到其有效的治疗手段和治疗靶点至关重要。胃癌的发病机制涉及环境和遗传等多种因素,其中某些氨基酸在胃癌细胞代谢中起着关键作用,对这些氨基酸的生物学功能的研究和探讨有助于为临床提供有效精准的诊断方法、治疗靶点等,从而提高胃癌的早诊早治,延长患者的生存时间。
大多数癌症包括胃癌、肺癌和结肠癌等都表现出对谷氨酰胺的异常依赖,此现象也被称为“谷氨酰胺成瘾”。而在神经元中,谷氨酰胺在谷氨酸脱羧酶(GAD)的作用下可以转化为非蛋白氨基酸——γ-氨基丁酸(GABA),在哺乳动物中枢神经***中作为主要的抑制性神经递质发挥作用。除神经***外,GABA在某些实体肿瘤例如结肠癌、乳腺癌等癌症中存在含量升高的现象。且有研究表明,GABA主要与GABAA受体以及GABAB受体结合发挥作用。但在胃癌中,GABA水平的变化以及GABA与GABAA受体、GABAB受体结合后介导胃癌细胞增殖、迁移以及凋亡的作用以及机制并不明确。
拮抗剂与受体结合后发挥作用,不仅需要结构上匹配,还需要较高的受体亲和力。受体亲和力是药物分子与受体的结合能力,它可以用放射性配体示踪来定量地分析,达到正好拮抗掉50%放射性配体的待测药物浓度被称为该药物的IC50。IC50值越小,说明药物的受体亲和性越强,药效也就越好。作为同一种受体的拮抗剂,因其与受体结合能力不同,对受体产生的抑制强度也不相同。
本发明通过阐明GABA及其受体在正常胃细胞以及胃癌细胞中水平差异以及对胃癌细胞增殖、迁移的影响,为分析GABA与胃癌发生发展的关系,探索以GABA受体为靶点进行靶向治疗的策略提供理论基础,进一步为临床上基于GABA受体,使用GABA受体拮抗剂组合物进行胃癌治疗以及改善胃癌患者的预后提供新的诊疗方案。
发明内容
本发明的目的在于提供γ-氨基丁酸受体拮抗剂组合物及其在制备治疗胃癌药物中的应用,本发明的受体拮抗剂组合,不仅可以与GABAA受体、GABAB受体结合发挥作用,而且抑制能力显著,便于及早对患者进行有效的治疗,减低患者的死亡率;该拮抗剂组合物作为以GABA受体为靶点和核心的治疗胃癌的药物,有望成为肿瘤靶向治疗的一种新手段。
本发明提供了如下技术方案之一:
一种γ-氨基丁酸受体拮抗剂组合物,所述拮抗剂组合物由GABAA型受体拮抗剂和GABAB型受体拮抗剂组成,用于阻断γ-氨基丁酸与GABAA型受体和GABAB型受体结合;其中,GABAA型受体拮抗剂为SR95531,GABAB型受体拮抗剂包括β-(氨基甲基)-4氯代苯乙烷磺酸、CGP 35348、CGP 52432。
进一步地,所述GABAB型受体拮抗剂与GABAA型受体拮抗剂的质量比为2-2.5:1。
进一步地,100μM上述GABA受体拮抗剂组合物中各有效成分的用量分别为:β-(氨基甲基)-4氯代苯乙烷磺酸2.5μg、CGP 35348为2.25μg、CGP 52432为3.84μg、SR95531为3.7μg。
进一步地,GABA受体拮抗剂组合物的作用靶点为GABAA受体以及GABAB受体,通过阻断GABA与两受体的表达而抑制GABA的积累对胃癌细胞增殖、迁移的促进作用,起到治疗胃癌的作用。

一种非诊疗目的检测胃癌细胞凋亡的方法,包括采用如权利要求1或2的GABA受体拮抗剂组合物阻断GABA与受体的结合导致胃癌细胞凋亡进行检测的步骤。

(1)采用体外培养方法培养胃癌细胞MKN28和MGC803;

(3)随后收集上述药物刺激后细胞,上流式细胞仪检测;
(4)釆用Annexin V-FITC染色检测胃癌细胞MKN28和MGC803的细胞凋亡率。
进一步地,胃癌细胞MKN28和MGC803接种量为2.0×106个/孔;贴壁后加入100μMGABA受体拮抗剂组合物药物。
本发明的有益效果:
本发明通过对GABA水平在胃癌组织及癌旁组织中进行比较分析,发现GABA在胃癌中异常积累并促进胃癌的增殖、迁移,利用GABA受体拮抗剂组合物特异性地阻断GABA与GABA受体在胃癌细胞中的结合,起到了促进胃癌细胞凋亡的作用,在胃癌靶向治疗中提供了有效治疗手段,为减低胃癌患者死亡率提供更及时的临床策略。
本发明GABA受体拮抗剂组合物选用可同时阻断GABAA受体和GABAB受体的有效成分组合,对胃癌细胞增殖、迁移及侵袭抑制明显,对治疗胃癌药物提供了靶向治疗的新手段。

具体实施方式
为能清楚说明本方案的技术特点,下面通过具体实施方式,结合附图,对本发明进行详细阐述。本发明的范围并不限于下述实施例。本领域的专业人员能够理解,在不背离本发明的精神和范围的前提下,可以对本发明进行各种变化和修饰。
下述实施例中所涉及的仪器、试剂、材料等,若无特别说明,均为现有技术中已有的常规仪器、试剂、材料等,可通过正规商业途径获得。下述实施例中所涉及的实验方法,检测方法等,若无特别说明,均为现有技术中已有的常规实验方法,检测方法等。下述实施例中所涉及的份、百分比等,若无特别说明,均为重量单位。
发明人研究发现,GABA在胃癌细胞中的水平明显高于胃黏膜上皮细胞,并且对胃癌细胞的增殖、迁移以及侵袭有一定的影响,因此以GABA为靶点,对其在胃癌细胞的积累含量进行检测,有望成为胃癌早期诊断的一种新方法和手段,进一步使用GABA受体拮抗剂组合物相关药物阻断GABA与受体的结合,为胃癌的治疗和预后提供新思路。
一、GABA在正常胃粘膜上皮细胞和胃癌细胞中表达水平的分析1.实验方法
1.1微量法检测正常胃粘膜上皮细胞和胃癌细胞中GABA水平
(1)取24h培养后的细胞培养基1mL,1000转离心5min;
(2)去沉淀,取30μL上清,使用GABA含量检测试剂盒(上海酶联生物)进行GABA含量检测,苯酚和次氯酸钠与GABA反应,产生蓝绿色产物;
(3)反应产物在640nm有最大吸光值,使用酶标仪检测分光光度值,对应标准曲线计算上清中细胞分泌的GABA含量。
1.2Western blot检测GABA合成的关键酶GAD1以及分解的关键酶ABAT在人胃粘膜上皮细胞和多种胃癌细胞中的蛋白表达
1.2.1提取细胞蛋白
(1)将正常胃黏膜上皮细胞及胃癌细胞进行研磨裂解后收集细胞然后放至冰上;
(2)超声破碎细胞,一般选用30%超声强度、25kHz频率,超声25次,每次超声持续1s,每次超声间隔1s;
(3)13,500rpm,4℃离心10分钟,将离心得到的上清转移至新EP管并置于冰上;
(4)G250法测定蛋白浓度:
利用BSA(牛血清白蛋白,0.5μg/μL)制作蛋白标准曲线。取5个EP管分别加入下表所示的体积,配制成0、0.05、0.1、0.15、0.2μg/μL五个蛋白浓度(见下表1)。

加入1mLG250染液,颠倒混匀,按每孔200μL加入96孔板中,设置4个重复;在595nm吸收波长处测量样本吸光度值;待测样本稀释50倍后加入1mL G250染液,颠倒混匀,按每孔200μL加入96孔板中,同样设置4个重复,依据标曲公式计算样本浓度;
(5)用新鲜的细胞裂解液将所有的样本稀释成相同的蛋白浓度,根据样本总体积加入相应体积的5×SDS蛋白上样缓冲液,使其最终稀释成1×SDS;
(6)金属浴100℃煮蛋白,10分钟;-20℃保存备用。
1.2.2蛋白质印迹
(1)配制SDS-PAGE胶(上层胶浓度为4%,下层胶浓度为10%);
(2)每孔上样30μg目标蛋白,电泳后将胶上的蛋白转至经甲醇活化过的PVDF膜上(Roche,3010040001);
(3)使用5%脱脂牛奶于37℃恒温箱中封闭2小时后,使用PBST缓冲液漂洗,每次5分钟,漂洗3次;
(4)使用稀释后的一抗孵育,在摇床上4℃过夜;
(5)使用PBST缓冲液漂洗,每次5分钟,漂洗3次;
(6)加入合适比例的辣根过氧化合物酶标二抗(Beyotime),在摇床上孵育2小时;
(7)使用PBST缓冲液漂洗,每次5分钟,漂洗3次;
(8)使用ECL增强化学发光试剂(Bio-rad,1705061)曝光;
(9)使用Image J软件进行灰度分析与统计。
2.结果分析
发明人利用一系列胃癌细胞系和胃黏膜上皮细胞GES-1,采用微量法检测GABA水平,发现胃癌细胞MKN28、MGC803的GABA水平高于GES-1。参见图1。发明人进一步发现在胃癌细胞MKN28、MGC803中参与GABA合成的关键酶GAD1蛋白表达水平上调,参与分解的关键酶ABAT蛋白表达水平下调,具体参见图2,证明GABA在胃癌细胞中的积累是由于GAD1表达增多而ABAT表达减少,谷氨酰胺分解产生的GABA无法及时降解而导致的。
二、不同浓度GABA对胃癌细胞的增殖功能影响
1.实验方法
1.1细胞计数测定
在37℃加湿的5% CO2培养箱中,1×105胃癌细胞在含10% FBS的RPMI 1640培养基中培养,培养于35mm细胞培养皿中。在设计的时间内使用BioTech Aμtomated Cell Coμnter对胃癌细胞进行计数,组进行3次测试。
2.结果分析
发明人利用一系列胃癌细胞系和胃黏膜上皮细胞GES-1,采用细胞计数实验检测GABA对胃癌细胞增殖的影响,发现给予相同浓度GABA试剂,对比于加等体积超纯水的对照组,GES-1、MKN28和MGC803细胞系随着天数增加,细胞数量显著增加。如图3所示。
三、不同浓度GABA对胃癌细胞的迁移功能影响
1.实验方法
1.1划痕实验检测不同浓度GABA对人胃粘膜上皮细胞GES-1和胃癌细胞MKN28和MGC803迁移功能的影响
(1)培养板划线,首先使用马克笔在6孔板背后,用直尺均匀的划横线,大约每隔0.5~1cm一道,横穿过孔。每孔至少穿过3条线;
(2)在孔中加入约5×105个细胞,接种原则为过夜后融合率达到80%;
(3)第二天用枪头垂直于前一天划在平板背面的线在细胞层上进行划痕,不同孔之间使用同一只枪头;
(4)划痕完成后,使用无菌PBS洗细胞3次,洗去不贴壁的细胞,即划线时划线的细胞,是划线后留下的间隙清晰可见,然后更换新鲜无血清培养基;
(5)将细胞放入37℃,5% CO2培养箱,培养。然后0h、24h取出细胞,显微镜线观察并测量划痕的宽度,并拍照;
(6)用Image J软件分析统计细胞迁移率。
2.结果分析
发明人利用一系列胃癌细胞系和胃黏膜上皮细胞GES-1,采用细胞划痕实验检测加入不同浓度GABA后对GES-1、MKN28、MGC803细胞迁移功能的影响。发现给予不同浓度GABA试剂(0、10、50、100μM),对比于加等体积超纯水的对照组,GES-1、MKN28和MGC803细胞系细胞迁移率随着GABA浓度的增加,细胞迁移率增加,具体参见图4,提示GABA会促进胃癌细胞的迁移。
四、GABA受体拮抗剂及其组合物对人胃黏膜上皮细胞和胃癌细胞凋亡的影响
1.实验方法
(1)取GABA受体拮抗剂及其组合物分别刺激后的细胞培养基上清液,加入流式管;用PBS洗一遍培养皿再将PBS加入流式管;
(2)之后用1mL胰酶将细胞消化后,用1mL培养基终止消化,再将细胞悬液加入上述流式管;
(3)将流式管离心,1300r,5min;
(4)弃上清后加入1mL PBS重悬沉淀;
(5)再离心,1300r,5min;
(6)弃上清后,加入100μL Bindng Buffer、5μLAnnexin V-FITC、10μL PIStaingSolμtion混合液重悬,避光孵育15min;
(7)之后加入400μL Bindng Buffer,过滤至流式细胞仪上样管,上流式细胞仪检测。
上述方法提及的GABA受体拮抗剂分别为:0-100μM的GABAB型受体拮抗剂——β-(氨基甲基)-4氯代苯乙烷磺酸、GABAB型受体拮抗剂——CGP35348、CGP 52432、GABAA型受体拮抗剂——SR95531。
上述方法提及的GABA受体拮抗剂组合物浓度为0-100μM,其中配置100μM拮抗剂组合物各有效成分的用量分别为:β-(氨基甲基)-4氯代苯乙烷磺酸2.5μg、CGP 35348为2.25μg、CGP 52432为3.84μg、SR95531为3.7μg。
2.结果分析
发明人利用一系列胃癌细胞系MKN28和MGC803,采用细胞凋亡实验检测加入不同浓度的单一GABA受体拮抗剂及本发明GABA受体拮抗剂组合物后对MKN28、MGC803细胞凋亡的影响。
如图5-7,分别为加入0-100μM不同的GABA受体拮抗剂后,胃癌细胞系MKN28和MGC803细胞凋亡情况。由图5可以看出,给予不同浓度GABAB型受体拮抗剂——β-(氨基甲基)-4氯代苯乙烷磺酸,对比于不给予任何药物的对照组,MKN28和MGC803细胞系随着GABAB受体拮抗剂浓度增加,细胞凋亡率增加,表明β-(氨基甲基)-4氯代苯乙烷磺酸促进细胞凋亡,随着浓度增加细胞凋亡的比例也在上升,起到抑癌的作用。由图6看出,给予不同浓度的另一种GABAB型受体拮抗剂——CGP35348,对细胞凋亡并无影响,表明只有CGP35348单一拮抗剂的条件下不能起到抑癌的作用;而CGP 52432为与CGP35348相似的结构构型受体拮抗剂,与GABA结合及作用机制基本一致,可推断CGP 52432具备与CGP 35348相似的效果,并不能较好的起到抑癌作用;由图7可以看出,给予不同浓度的GABAA型受体拮抗剂——SR95531,随着GABAA受体拮抗剂浓度增加,细胞凋亡率增加,表明SR95531促进细胞凋亡,随着浓度增加细胞凋亡的比例也在上升,起到抑癌的作用。由图8可以看出,当给予不同浓度的GABAA型受体拮抗剂——SR95531和GABAB型受体拮抗剂——β-(氨基甲基)-4氯代苯乙烷磺酸两种组合成分时,随着受体拮抗剂浓度增加,细胞凋亡率增加,表明上述两种拮抗剂组合物会促进细胞凋亡,随着浓度增加细胞凋亡的比例也在上升,起到抑癌的作用。然而,当将上述具有不同拮抗效果的各成分以特定配比制成全新的拮抗剂组合物后,继续用于细胞凋亡实验检测,发现,该拮抗剂组合物能够更明显促进胃癌细胞系凋亡,抑癌作用相比前述具有抑癌效果的不同类型的单一GABA拮抗剂(β-(氨基甲基)-4氯代苯乙烷磺酸、SR95531)及组合更明显。浓度为100μM的两种拮抗剂组合物包含β-(氨基甲基)-4氯代苯乙烷磺酸和SR95531作用于胃癌细胞MKN28和MGC803时,存活率分为72%和54.5%,而当浓度为100μM的四种拮抗剂组合物包含β-(氨基甲基)-4氯代苯乙烷磺酸、CGP 35348、CGP 52432、SR95531作用于胃癌细胞MKN28和MGC803时,存活率分为37.5%和11.3%,明显低于两种拮抗剂组合物,提示本发明GABA受体拮抗剂组合物会起到显著抑癌作用,组合使用的上述拮抗剂产生了意料不到的技术效果。
综合上述结果表明:GABA在胃癌中水平升高,促进胃癌细胞增殖、迁移,抑制GABA与GABA受体的结合会导致胃癌细胞凋亡。进一步研究发现,相比于使用单一的受体拮抗剂-β-(氨基甲基)-4氯代苯乙烷磺酸、SR95531,使用包含β-(氨基甲基)-4氯代苯乙烷磺酸、CGP35348、CGP 52432、SR95531(又名Gabazine)的GABA受体拮抗剂组合物,所产生的治疗效果显著提升,意味着本发明的拮抗剂组合物作为一种新的拮抗剂产品可以为临床上基于GABA受体,使用GABA受体拮抗剂组合物进行胃癌治疗以及改善胃癌患者的预后提供新的诊疗方案,为临床上基于GABA受体进行胃癌治疗提供新的靶点。

Claims (7)
1.一种γ-氨基丁酸受体拮抗剂组合物,其特征在于,所述拮抗剂组合物由GABAA型受体拮抗剂和GABAB型受体拮抗剂组成,用于阻断γ-氨基丁酸与GABAA型受体和GABAB型受体结合;其中,GABAA型受体拮抗剂为SR95531,GABAB型受体拮抗剂包括β-(氨基甲基)-4氯代苯乙烷磺酸、CGP 35348、CGP 52432。
2.根据权利要求1所述的γ-氨基丁酸受体拮抗剂组合物,其特征在于,所述GABAB型受体拮抗剂与GABAA型受体拮抗剂的质量比为2-2.5:1。
3.如权利要求1或2所述的γ-氨基丁酸受体拮抗剂组合物在制备治疗胃癌药物中的应用。
4.如权利要求1或2所述的γ-氨基丁酸受体拮抗剂组合物在制备胃癌浸润靶点药物中的应用,其特征在于,所述靶点为GABAA型受体和GABAB型受体。
5.一种非诊疗目的检测胃癌细胞凋亡的方法,其特征在于,包括采用如权利要求1或2的GABA受体拮抗剂组合物阻断GABA与受体的结合导致胃癌细胞凋亡进行检测的步骤。
6.根据权利要求5的检测方法,其特征在于,包括如下操作步骤:
(1)采用体外培养方法培养胃癌细胞MKN28和MGC803;
(2)将胃癌细胞MKN28和MGC803置于培养皿中培养,贴壁后,加入GABA受体拮抗剂组合物药物刺激24h;
(3)随后收集上述药物刺激后细胞,上流式细胞仪检测;
(4)釆用Annexin V-FITC染色检测胃癌细胞MKN28和MGC803的细胞凋亡率。
7.根据权利要求6的检测方法,其特征在于,胃癌细胞MKN28和MGC803接种量为2.0×106个/孔;贴壁后加入100μM GABA受体拮抗剂组合物药物。
