本发明公开了一种堆肥木质纤维素的降解方法,属于固体废弃有机物肥料利用技术领域,本发明联合光照降解堆肥木质纤维素,在林业剩余物中加入复合光催化剂,完成堆肥反应。通过特制滤光膜过滤UV‑A波段,在无UV‑A波段下添加香豆素和骨炭后进行好氧堆肥反应,有效加速了堆肥木质纤维素的降解与腐殖质的形成,堆肥产品各项腐熟指标表现优异。

技术领域
本发明属于固体废弃有机物肥料利用技术领域,特别是涉及一种堆肥木质纤维素的降解方法。
背景技术
近年来,随着人口、粮食需求的提高以及城镇绿化的增加,北京市每年产生约500万吨的林业剩余物。林业剩余物主要为园林绿化维护过程中产生的枯枝、落叶、杂草、败花等,还包括枝桠、梢头、灌木、树桩(伐根)、枯倒木、遗弃材及截头等木质物质,由于其具有大量难降解的木质纤维素已经成为继城市生活垃圾之后的第二大城市固体废弃物。
目前,林业剩余物处理方式仍以堆放、填埋、焚烧为主,这些方法不仅生物质转化率很低,同时还会造成诸多环境污染,如占用土地资源、破坏土壤内部结构、产生有毒有害气体(CO、CO2、二噁英、H2S)、加重温室效应。因此,实现林业剩余物低成本、高效率和稳定的转化,使大量林业剩余物转化为高价值产品的同时,还能够减小对环境的危害已经成为当前的重要任务。
林业剩余物堆肥处理在有效降低填埋及焚烧处理对生态环境造成危害的同时,还能高效降解林业剩余物中的木质纤维素。此外,堆肥产品中含有丰富的有机质及一定量的营养元素,可以用作土壤改良剂或栽培基质,改善土壤环境的同时还可以减少不可再生资源(如泥炭)的开采,保护湿地,经济方面和生态方面获得双重收益。尽管如此,传统堆肥仍存在效率低、周期长、污染环境等问题。林文琪等指出木质纤维素含量高的原材料难以被微生物分解,导致堆肥周期增长,堆肥产品质量降低。刘晓林和刘军也指明氨气等有毒有害气体的产生降低了堆肥质量和堆肥产品的品质,由此造成的环保问题较为突出。
因此,本领域亟需一种高效降解堆肥木质纤维素的方法。
发明内容
针对我国林业剩余物堆肥处理周期过长,堆肥材料木质纤维素含量高、结构复杂难以降解,常用外源添加剂效果低、成本高以及堆肥光降解作用不易引发等问题,本发明提出了一种堆肥木质纤维素的降解方法,以解决上述问题。本发明通过特制的无UV-A滤光膜,在无UV-A波段下添加香豆素和骨炭,有效加速了堆肥木质纤维素的降解与腐殖质的形成,所得堆肥产品各项腐熟指标表现优异。
本发明提供的技术方案之一:
一种复合光催化剂,包括香豆素和骨炭。
香豆素的光催化作用:作为光敏感性单体来合成一些易被降解的高聚物,其能够与生物的各种活性位点产生非共价的相互作用,促进微生物生长及其代谢,同时香豆素的光量子产率较高、光稳定性非常好。
骨炭的光催化作用:以其自身疏松多孔的结构为光催化下的磷酰化反应顺利进行和微生物生长发育提供空间,同时为微生物提供钾、钙、磷等元素供其生长发育。
将香豆素和骨炭联合应用能够发挥协调增效的机理:香豆素与骨炭中的磷元素发生磷酰化反应生成香豆素磷酸酯类衍生物,加速降解木质纤维素的微生物快速生长发育,从而促进绿化废弃物光降解;二者结合形成的疏松多孔结构,也可以在一定程度上为微生物提供依附生存环境。
本发明提供的技术方案之二:
一种堆肥木质纤维素的降解方法,在林业剩余物中加入上述复合光催化剂,联合光照降解堆肥木质纤维素。
所述林业剩余物包括林业生产和经营过程中产生的未商品化利用的有机物质、绿化维护过程中产生的枯枝、落叶、杂草、败花以及林业管理中产生的枯倒木、遗弃材及截头等木质物质。
光照作为一种可再生、取之不尽用之不竭的“添加剂”,与传统堆肥结合有助于降低木质纤维素降解成本,提高降解效率,实现无污染降解。在特定波段的光辐射下,木质素的发色基团会吸收光能,断裂醚键及碳碳键,使得木质素等难降解物质加速降解;此外,光降解也可以提高微生物对林业剩余物的降解效率,木质素等组分形成的生物质可通过光降解直接转化为芳烃原料。
具体的降解方法为:将所述复合光催化剂加入林业剩余物中,调节含水率和C/N比,加入微生物菌剂,混合均匀后堆成圆锥形堆体,将圆锥形堆体置于由滤光膜搭制的反应器中,进行好氧堆肥反应,当堆体温度等于或低于室温时,堆肥完成。
将骨炭与香豆素混合于60℃条件下放置10-12 h会促进药渣中的香豆素类化合物发生磷酰化,生成香豆素磷酸酯类衍生物(也叫做酰基膦氧化物光催化剂),这类物质具有极高的引发聚合活性、高感光度以及宽感光度范围,可加速矿化大分子物质或者加速其他大分子物质降解,且对微生物活性也有一定的促进作用。而在绿化废弃物堆肥过程中,高温期(50-60℃)一般要维持3天以上,这一温度条件为药渣中的香豆素类化合物发生磷酰化提供了保障,也为使用骨炭与香豆素作为复合光催化剂提高绿化废弃物木质纤维素降解作用提供了基础。
但该反应也存在一定限度,磷酰化反应后过多游离的香豆素对光降解微生物生长与产酶具有一定的抑制作用;骨炭含量过高,其复杂多孔的结构会减缓微生物的移动,同时会吸附游离香豆素,从而减缓纤维素、半纤维素及木质素的降解效率,因此本发明中,所述复合光催化剂中香豆素的加入量为林业剩余物干重的15~25wt.%,骨炭的加入量为林业剩余物干重的15~25wt.%。
更为优选的,所述香豆素的加入量为林业剩余物干重的20wt.%,所述骨炭的加入量为林业剩余物干重的20wt.%。
优选的,所述C/N比为25-30%;所述含水率为25-30wt.%。
优选的,以干重计,每1kg林业剩余物中加入5mL微生物菌剂。
更为优选的,所述微生物菌剂包括乳酸菌、酵母菌和放线菌,所述乳酸菌、酵母菌和放线菌的浓度均大于20×108CFU/mL,总浓度在60×108 - 90×108中间。
本发明的有益效果:在无UV-A波段下,将香豆素和骨炭作为复合光催化剂加入林业剩余物堆肥中,相比未添加复合光催化剂的处理,能够在更短时间内产生品质更优的堆肥产品。
与未添加复合光催化剂的堆肥反应相比,香豆素加入量为20wt.%和骨炭加入量为20wt.%的堆肥反应中,30天即获得了完全腐熟的堆肥产品,且堆肥过程中最高温度达到了57.6℃,嗜热期达9天,堆肥产品的腐殖化系数提高了24.38%,堆肥产品的有机质含量减少了35.29%,氮元素流转更通顺,总氮含量更高,脱氢酶、纤维素酶和漆酶的活性均更高,堆肥产品的纤维素含量降低了1.81%,半纤维素含量降低了4.90%,木质素含量降低了16.00%,最大持水量提高了19.92%、总孔隙度提高了27.13%,堆肥产品位于最佳粒径范围内的百分比(容量)下降了14.29%,粗糙度指数改善了4.27%,有效磷含量提高了29.31%,速效钾含量提高了36.37%,铵态氮含量降低了28.17%,硝态氮含量提高了103.82%、全磷含量提高了28.69%;发芽实验中种子根长提高了23.36%、发芽率提高了20.00%、发芽势提高了58.16%。

本发明的室温指的是25±2℃。
本发明实施例中所用滤光膜为特制的无UV-A滤光膜,购自广东海悦光学技术有限公司,阻挡波长在320-380 nm之间的UV-A辐射,为0.23 mm厚聚烯烃薄膜(14S);骨炭购买于广州市孜雅萱批发贸易商行;香豆素购买于天津信盈科技有限公司;微生物菌剂的菌剂原液购自广州真微生物科技有限公司,用水稀释制备得到堆肥添加微生物菌剂。
本发明实施例中的林业剩余物为北京市国家植物园2021年秋季及2022年春季景观维护期间产生的枯枝落叶。
实施例
于2022年7-9月在北京市国家植物园(40°00′64″N- 116°20′22″E)进行实验。该地区属半湿润半干旱季风气候,平均气温23.5℃,日平均日照时间为14.3h。
将林业剩余物压碎为5-20mm长的细段,以林业剩余物的干重计按照表1加入香豆素和骨炭,加水调节含水率为60±5wt.%,添加尿素调节C/N比至25-30%,同时为了提高堆体内微生物含量及堆肥初期微生物活性,以每1kg林业剩余物(干重)加入5mL微生物菌剂(微生物菌剂包括乳酸菌、酵母菌和放线菌,其中乳酸菌、酵母菌和放线菌的浓度均大于20×108CFU/mL,总浓度在60×108 - 90×108中间),混合均匀后,堆成直径为1m,高为0.5m的圆锥形堆体;用木棍在圆锥形堆体周围支起特制的无UV-A滤光膜搭制的反应器中,包裹整个堆体,以便所有通过反应器的光都通过无UV-A滤光膜,减少光谱辐射的能量损失,在无UV-A滤光膜上扎随机、等大的膜孔使空气和水汽流通,实现好氧堆肥。

堆肥过程中每天利用探杆***式温度计在堆体上中下三个不同的位置对堆体温度进行监测,记录后取平均值,见图1 T1-T9以及空白组(CK)在堆肥反应中温度的变化,并利用同一温度计监测环境温度并记录,当堆体温度等于或低于室温时,堆肥完成。
由图1可知复合光催化剂的添加缩短了堆肥进程,使堆肥过程中最高温更高,嗜热期更长,堆肥周期更短;其中T5(20wt.%香豆素+20wt.%骨炭)30天即获得了完全腐熟的堆肥产品,且堆肥过程中最高温达到了57.6℃,嗜热期达9天,CK(未添加复合光催化剂)堆肥周期则长达43天。
堆肥反应过程中,分别于第0、5、10、15、20、25、30、33、34、36、37、39、40、43天取200±20g的样品,所有样品被分成两部分,分别在冰箱4℃和室温下储存。
测定室温下自然风干的样品的腐殖化系数、有机质、总氮、pH、电导率(EC)、纤维素、半纤维素、木质素以及速效养分,并与未加入复合光催化剂的空白组(CK)进行比对。
腐殖化系数变化见图2,腐殖质的测定方法为重铬酸钾氧化-容量法;由图2可知复合光催化剂的添加促进了堆肥产品腐熟,堆肥过程中腐殖化系数增长更快,堆肥产品腐殖化系数更高更加稳定,其中T5(20wt.%香豆素+20wt.%骨炭)堆肥产品的腐殖化系数相较于空白组(CK)提高了24.38%。
有机质变化见图3,有机质的测定方法为重铬酸钾容量-稀释热法;由图3可知复合光催化剂的添加促进了绿化废弃物堆肥过程中有机质的降解,各处理有机质含量相较于空白组(CK)更少,其中T5(20wt.%香豆素+20wt.%骨炭)堆肥产品的有机质含量相较于空白组(CK)的有机质含量减少了35.29%。
总氮变化见图4,总氮的测定方法为凯氏定氮法;由图4可知复合光催化剂的添加对于堆肥过程中氮元素的循环及氮元素的重新固定具有一定的调节作用,T5(20wt.%香豆素+20wt.%骨炭)在堆肥过程中相较于氮元素流失更少,重新固定更多,堆肥产品总含氮量更多。
pH变化见图5,电导率(EC)变化见图6,pH和导电率的测定方法: 通过pH400防水型笔式pH计和EC400防水型笔式电导率/TDS/盐度计测定样品与去离子水滤液(体积比为1:10)的pH值和电导率(EC);由图5和6可知,复合光催化剂的添加改善了堆肥过程中pH值及EC值,其中T5(20wt.%香豆素+20wt.%骨炭)在堆肥过程中堆体内部酸碱度及无机盐含量在所有处理中处于最优水平。
纤维素、半纤维素以及木质素变化分别见图7、8和9,纤维素、半纤维素以及木质素的测定方法为NREL法;由图7-9可知,复合光催化剂提高了堆肥过程中纤维素、半纤维素和木质素的降解效率,其中T5的堆肥产品(20wt.%香豆素+20wt.%骨炭)与空白组(CK)的堆肥产品相比,纤维素含量降低了1.81%,半纤维素含量降低了4.90%,木质素含量降低了16.00%。
速效养分中有效磷、速效钾、全磷、铵态氮、硝态氮、全钾的含量见表2,其中有效磷、速效钾、全磷、铵态氮、硝态氮、全钾的测定方法为采用752紫外光栅分光光度计测定全磷;采用FP640火焰光度计测定全钾;采用钼睇抗比色法测定有效磷;采用火焰光度法测定速效钾,采用比色法测定铵态氮和硝态氮。

通过表2可以看出:本发明通过在林业剩余物中加入复合光催化剂,提高了堆肥产品的速效养分含量,相较于空白组(CK),T5(20wt.%香豆素+20wt.%骨炭)的有效磷含量提高了29.31%、速效钾含量提高了36.37%、铵态氮含量降低了21.98%、硝态氮含量提高了103.82%、全磷含量提高了28.69%。
对室温下自然风干的样品的最大持水量、总空隙度、容重以及粗糙度进行测定,并与空白组(CK)进行比对,测定方法为环刀法,结果见表3。

由表3可知,复合光催化剂的添加提高了堆肥产品的持水量以及总空隙度,改善了粗糙度;其中T5(20wt.%香豆素+20wt.%骨炭)堆肥产品的腐殖化系数相较于空白组(CK),最大持水量提高了19.92%、总孔隙度提高了27.13%,容重下降了14.29%,粗糙度指数改善了4.27%。
测定4℃下储存的样品中脱氢酶、木质素过氧化物酶和漆酶的活性,同空白组(CK)进行比对,测定方法分别为氯化三苯基四氮唑(TTC)比色法、藜芦醇法以及ABTS法;其中脱氢酶、纤维素酶和漆酶的变化分别见图10、11和12;由图10-12可知复合光催化剂的添加提高了堆肥过程中酶活性,对光降解微生物的生长繁殖有促进作用,T5(20wt.%香豆素+20wt.%骨炭)在堆肥过程中相较于其他处理脱氢酶、纤维素酶和漆酶的活性均更高,这一结果可以反映出T5(20wt.%香豆素+20wt.%骨炭)的微生物活性更强。
应用例
将实施例中T1-T9以及空白组(CK)所得堆肥产品用于种子发芽实验,即:将5 kg堆肥样品(干重)加入50 mL蒸馏水中,剧烈震荡20min后过滤。然后,将滤液放在无菌培养皿中的一张滤纸上,用滤液湿润滤纸。蒸馏水作为对照组。最后,在每个培养皿的滤纸上均匀放置20颗大白菜种子(Brassica parachinensis),培养皿放入25℃黑暗环境中。培养48 h后,计算种子根长、发芽率和发芽势(GI)。发芽率=种子发芽数/20×100%,发芽势(%) =(处理组平均发芽种子数×处理组平均根长)/(对照组平均发芽种子数×对照组平均根长)×100%,测定结果见表4。

由表4可知,本发明在联合光照降解堆肥木质素过程中,加入复合光催化剂,能够促进堆肥产品腐熟,降低了其植物毒性,T5(20wt.%香豆素+20wt.%骨炭)相较于空白组(CK)的堆肥产品在发芽实验中种子根长提高了23.36%、发芽率提高了20.00%、发芽势提高了58.16%。
以上所述的实施例仅是对本发明的优选方式进行描述,并非对本发明的范围进行限定,在不脱离本发明设计精神的前提下,本领域普通技术人员对本发明的技术方案做出的各种变形和改进,均应落入本发明权利要求书确定的保护范围内。
Claims (5)
1.一种堆肥木质纤维素的降解方法,其特征在于,将复合光催化剂加入林业剩余物中,调节含水率和C/N比,加入微生物菌剂,混合均匀后堆成圆锥形堆体,将圆锥形堆体置于由滤光膜搭制的反应器中,进行好氧堆肥反应,当堆体温度等于或低于室温时,堆肥完成;所述复合光催化剂由香豆素和骨炭组成;
所述复合光催化剂中香豆素的加入量为林业剩余物干重的15~25wt.%,骨炭的加入量为林业剩余物干重的15~25wt.%。
2.根据权利要求1所述的一种堆肥木质纤维素的降解方法,其特征在于,所述复合光催化剂中香豆素的加入量为林业剩余物干重的20wt.%,骨炭的加入量为林业剩余物干重的20wt.%。
3.根据权利要求1所述的一种堆肥木质纤维素的降解方法,其特征在于,所述C/N比为25~30%。
4.根据权利要求1所述的一种堆肥木质纤维素的降解方法,其特征在于,以干重计,每1kg林业剩余物中加入5mL微生物菌剂。
5.根据权利要求1所述的一种堆肥木质纤维素的降解方法,其特征在于,所述微生物菌剂包括乳酸菌、酵母菌和放线菌,所述乳酸菌、酵母菌和放线菌的浓度均大于20×108CFU/mL,总浓度为60×108-90×108CFU/mL。
