在一个实施方案中,本公开提供了用于对富含淋巴细胞的样品中的CD3-细胞杂质进行流式细胞术定量的方法和组合物。在一个实施方案中,本公开提供了符合目的的2-8色T细胞杂质流式细胞术组(panel),该组检测多达七种不同的血细胞表面标志物并且还包括活力染料。在一个实施方案中,本公开提供了使用该组来检测和定量在用于免疫疗法的T细胞产物的不同制造阶段获得的样品中的CD3-细胞的方法。本文所公开的组和方法具有几种促进效率的性质,包括但不限于易于使用、消除滴定不同抗体批次的需要、包含批次匹配的同种型对照、批次间一致性、室温下的长期稳定性、使易于出错的移液步骤和混合最小化、流线型工作流程和改善的数据可靠性。在一个实施方案中,该组或方法可用于表征用于免疫疗法的T细胞群体中的杂质。以下是本公开的示例性、非限制性实施方案。

在一些实施方案中,通过裂解红血细胞并耗竭单核细胞(例如通过使用通过PERCOLLTM梯度的离心)来从PBMC分离T细胞。在一些实施方案中,通过本领域已知的阳性或阴性选择技术来进一步分离T细胞的特定亚群,诸如CD4+、CD8+、CD28+、CD45RA+和CD45RO+T细胞。例如,可使用针对阴性选择的细胞特有的表面标记物的抗体组合来完成通过阴性选择的T细胞群的富集。在一些实施方案中,可使用经由负磁性免疫粘附或流式细胞术进行的细胞分选和/或选择,其使用针对存在于阴性选择的细胞上的细胞表面标记物的单克隆抗体的混合物。例如,为了通过阴性选择来富集CD4+细胞,单克隆抗体混合物通常包含针对CD8、CD11b、CD14、CD16、CD20和HLA-DR的抗体。在一些实施方案中,使用流式细胞术和细胞分选来分离用于本公开中的感兴趣的细胞群。
在一个实施方案中,使用涂覆有抗CD3抗体的Dynabeads从PBMC中分离CD3+T细胞。使用CD8微珠(例如,德国天美旎生物技术公司(Miltenyi Biotec)和/或CD4微珠(例如,德国天美旎生物技术公司)通过阳性选择进一步单独分离CD8+和CD4+T细胞。
PBMC可以直接用于免疫细胞的基因修饰(诸如CAR)。在分离PBMC之后,进一步分离T淋巴细胞,并且在基因修饰和/或扩增之前或之后,将细胞毒性和辅助T淋巴细胞两者分选为幼稚、记忆和效应T细胞亚群。在一个实施方案中,通过鉴定与这些类型的CD8+细胞的每一种相关的细胞表面抗原,可以将CD8+细胞进一步分选为幼稚、中央记忆和效应细胞。在其他实施方案中,中央记忆T细胞的表型标志物的表达包括CCR7、CD3、CD28、CD45RO、CD62L和CD127并且对颗粒酶B是阴性的。在一些实施方案中,中央记忆T细胞是CD8+、CD45RO+和CD62L+T细胞。在某个实施方案中,效应T细胞对CCR7、CD28、CD62L和CD127是阴性的并且对颗粒酶B和穿孔素是阳性的。在附加实施方案中,可以将CD4+T细胞进一步分选为亚群。例如,可通过鉴定具有细胞表面抗原的细胞群来将CD4+T辅助细胞分选为幼稚、中央记忆和效应细胞。

在一个实施方案中,本文所述的T细胞制备物可用于工程改造自体细胞疗法。术语“工程改造的自体细胞疗法”(可缩写为“eACTTM”,也称为过继细胞转移)是通过其收集患者自身的T细胞并随后对这些细胞进行基因改造以识别和靶向在一种或多种特异性肿瘤细胞或恶性肿瘤的细胞表面上表达的一种或多种抗原的过程。可以将T细胞工程改造以表达例如嵌合抗原受体(CAR)或T细胞受体(TCR)。对CAR阳性(+)T细胞进行工程改造以表达对某种肿瘤抗原具有特异性的细胞外单链可变区片段(scFv),该scFv连接到包含共刺激结构域和激活结构域的细胞内信号传导部分。

实施例1
设计用于CD3-杂质的符合目的的流式细胞术组
实验地开发了符合目的的8色T细胞杂质流式细胞术组,以评估T细胞样品或产物中的CD3+细胞纯度,以及活细胞。该组可用于表征血细胞样品中的CD3-杂质(NK-T细胞、NK细胞、B细胞、单核细胞),包含通过单采血液成分术获得的那些、PBMC、和在用于免疫疗法的T细胞产物的制造过程中制备的那些。
首先选择一组细胞表面标志物以鉴定血液样品中的不同细胞群体。这些标志物如表8所示。

然后按照多种标准开发了七种抗体和荧光团组合(加上远红外活力染料)的特定组。具有较高丰度的抗原与较暗的荧光染料匹配,而具有较低丰度的那些抗原与较亮的荧光染料匹配。这使得该组能够进行更灵敏的检测。评估各种抗体与荧光染料的组合以最小化光谱溢出到仪器中的相邻通道中,并且实现所有群体的最佳分辨率。补偿设置也确保了频谱溢出的校正。所选择的组描述于表9.1、表9.2、表9.3、表9.4和表9.5中。

为了优化方法组,滴定所有七种抗体以确定给出稳健信噪比、最小背景、具有一致%阳性信号的染色强度的使用体积/浓度。参见表9.1、表9.2、表9.3、表9.4和表9.5。
通过用处于所滴定的体积/浓度或制造商推荐体积的抗体混合物对样品进行染色来验证所选抗体滴定的性能。
用于该方法的试剂如表10中所指定。

为了确定用于染色的最佳抗体浓度,将抗体连续稀释并一式两份地测量。使用以下等式计算染色指数(SI):
其中MFIp是阳性群体的中值荧光强度(MFI),MFIn是阴性群体的MFI,并且rSDn是阴性群体的稳健标准偏差。
创建SI曲线以选择给出显著SI值的抗体的稳健质量。过量的抗体体积可人为地增加整个细胞群体的阳性信号和阴性信号两者。因为不能通过单独的SI确定最佳抗体浓度,所以检查阳性和阴性靶群体的MFI以及阳性群体的频率。例如,针对以下抗体比较两个不同的抗体克隆,并且选择具有较高SI的克隆以包括在示例性组中的一个组中:
PE-Cy7 CD19抗体:滴定并比较克隆SJ25C1和HIB19。两个克隆显示出相似的特异性,但克隆HIB19具有更高的SI。
PerCP-Cy5.5 CD14抗体:克隆MφP9和M5E2经测试并显示出相似的特异性。克隆MφP9具有更高的SI。
PE CD56抗体:滴定并比较克隆NCAM16.2和HCD56。NCAM16.2显示出更好的特异性和分辨率。
在该优化努力期间滴定该方法中所使用的所有抗体。作为示例,图1A和图1B中分别指示了针对抗CD3 APC和抗CD14 PerCP-Cy5.5的优化抗体体积(μL)和浓度/质量(ng或μg)。CD14 PerCP-Cy5.5抗体显示没有CD14+MFI饱和。对流式曲线图的详细检查表明,1.3μL的抗体足以产生阳性信号,并随后优化用于FACSCanto II***中的样品获取/分析的对数参数。1×106个细胞的每次反应的抗体体积如表12中最终确定。
将(A)从每种不同抗体的液体原液为每次运行新鲜制备的上述抗体的新鲜制备的液体混合物的性能与(B)从所有七种抗体的冻干混合物新鲜制备的或在重悬后使用几天的相同抗体的液体混合物的性能进行比较。

实施例2
(A)液体试剂和(B)冻干试剂的染色可比性
测试该方法的可再现性。用以下两种类型的混合物一式两份地测试五个健康供体单采血液成分术样品:(A)通过将根据表5和表6的液体形式的单独抗体组合而新鲜制备的混合物,或(B)冻干的抗体混合物(试验试剂)。在同一天由一名分析者并行进行测试,以理解冻干试剂和液体试剂之间的可比性并评估冻干试剂的功能性。测定每种感兴趣的参数CD45+、T细胞、NK细胞、单核细胞和B细胞的%变化。
从五个健康供体收获单采血液成分术样品。将悬浮在细胞染色缓冲液(BSA)中的含有1百万个血细胞的总共200微升与100微升的(A)液体混合物或(B)从冻干混合物重悬的混合物混合。将CYTO-TROL对照细胞()(贝克曼库尔特公司(Beckman Coulter))用作阳性对照。它们是表现出可用所选单克隆抗体检测的表面抗原的人淋巴细胞的冻干制备物。这些细胞是从外周血中分离的并且表达代表在正常淋巴细胞上发现的那些抗原的抗原。
将样品在2℃至8℃避光孵育大约25分钟。一旦孵育完成,就将100微升的细胞染色缓冲液(BSA)添加到各个样品中并将样品离心。将沉淀的细胞用每次200微升的细胞染色缓冲液(BSA)洗涤三次,然后用活力染料染色。然后将样品与适当的单色补偿对照一起通过FACS进行处理。结果示出于图2A和图2B(图3)和表14中。使用FlowJo软件,在BD FACSCantoII流式细胞仪上分析CD3+T细胞和CD3-非T细胞杂质的频率。

实施例3
在重悬于染色缓冲液中后冻干试剂的10天稳定性
冻干的抗体试剂经测定为在室温下稳定18个月。在将冻干试剂重悬于每小瓶400μL的染色缓冲液中(这可用于20次测试/小瓶)后,预计产物具有3个月稳定性。还测试了重悬于2000μL的染色缓冲液中后产物的10天稳定性。
在第1天,将冻干的抗体混合物重悬于2000μL的染色缓冲液中。将一部分用于第1天,并将其余部分保存到第10天。单独制备新鲜液体抗体试剂以用于比较。测试来自四名癌症患者和一名健康供体的单采血液成分术样品。将CytoTrol用作阳性对照测试材料(PCTM)。CytoTrol由冻干的富含人淋巴细胞的细胞组成,这些细胞具有批次特异性参照范围的表面标志物。结果示于表15中。

实施例4
中间体精确度-分析者间变异性
分析者间变异性测试评估分析方法在由多个分析者执行时精确操作的能力。两名分析者一式两份地独立测试了两个单采血液成分术样品和CYTO-TROL。两名分析者在同一天使用相同批次的CytoTrol(PCTM)和2个健康供体样品独立地制备和分析样品。结果示于表17中。

方法稳健性
上述实施例中描述的方法也是稳健的。稳健性评估分析方法的可靠性,并且是该方法在方法参数的小但故意的变化下产生类似数据的能力的量度。这种方法的方法稳健性可例如在以下方面评估:
抗体染色/孵育时间:测试抗体染色的孵育持续时间从10分钟至45分钟。任何孵育时间≥45分钟对于表面染色都不是必需的,并且被认为在测试工作流程中是无效的。
所接种的总TVC/孔和获取事件的下限:以0.5×106个或1×106个TVC接种细胞数/孔以用于染色。
为了确定当检测小的或稀少的群体时将提供给定精确度的样品的大小,可以使用如Maecker HT等人细胞计量术部分A(Cytometry Part A)2006(69A):1037-1042;ICSH/ICCS工作组(ICSH/ICCS Workgroup).细胞计量术B临床细胞计量术(Cytometry B ClinCytometry).2013;84:279-357;或Allan AL等人,《肿瘤学杂志(Journal of Oncology)》2010(2010:426218中所述的方法。
在10分钟、20分钟、30分钟和45分钟的抗体染色孵育时间测试这种方法的稳健性。在每个时间点运行两个样品,并比较以下靶细胞群体的检测频率:白细胞中的%CD45+,CD45+细胞中的%亚群(CD3+T细胞、CD56+CD3+NKT细胞、CD56+CD3-NK细胞、CD14+CD3-单核细胞和CD19+CD3-B细胞)。图6和表21显示所有靶细胞群体在所测试的孵育时间范围内是可比的。所有靶细胞群体的%CV小于25%。因此,建立了10分钟至45分钟的动态孵育时间范围。将100μL的染色体积用于所有样品。

基于这种计算,5%的期望%CV提供了良好水平的置信度和精确度。因此,并且,所建立的关键报告参数的LLOQ值中的这种可检测性质量的下限是0.2%。通过外推法,获得400个此类残留细胞需要获取最少0.04×106个TVC。使用这种方法,如果可接受的%CV是15%,则应当获取最少50,000个TVC,因为使用2种起始患者材料执行基础研究,并用全抗体组对10,000个、20,000个、50,000个、100,000个和200,000个事件的CD45+获取事件进行染色。如表22和表23所示,通过%CV将各种获取事件相互比较。当在50,000个、100,000个和200,000个获取事件中比较时,发现表22中所列的数据没有显著影响。这是当收集50,000个事件时具有小于1%的值的潜在变量。结果列于表23中,其中较低的%B细胞数据(仅等于或大于LLOQ值)受到影响,有10,000个和20,000个收集的CD45+事件,但这对B-母细胞参数没有影响。
在同一天,使用3个不同的样品,用1×106个和1×105个TVC执行简单的TVC接种实验,以进行粗略的接种极限估计评估。表24中显示的数据提供了两个TVC接种/孔测试之间所有报告参数频率的可比性。表24中还显示了样品单采血液成分术批次AC25809161中%B细胞低于LLOQ的发现。

实施例11
单色冻干补偿对照的稳定性
根据制造商,补偿对照稳定性是在用染色缓冲液(BSA)重构后一天。在Kite,我们评估了补偿对照的14天稳定性。在冻干补偿对照试剂盒(BD目录号625812)中包括七个冻干补偿对照CD3-APC、CD10-FITC、CD14-PerCPCy5.5、CD19-PECy7、CD34-BV421、CD45-V500和CD56-PE。将300μL的染色缓冲液(BSA)和一滴UltroComp eBeads(补偿珠粒)添加到每个冻干的补偿对照管中并孵育15分钟以制备用于测定的补偿对照。重构后将冻干的补偿对照管在4℃保存14天,并计算MFI以评估稳定性。

表25显示从第0天和第14天重构的冻干单色补偿对照的MFI是可比的。来自第14天冻干的补偿对照的数据显示与第0天新鲜单色补偿对照的差异百分比<25%。这显示了在4℃下储存14天的补偿对照的稳定性。
Claims (17)
1.一种在细胞群体中同时鉴定淋巴细胞、NK-T细胞、NK细胞、单核细胞、早期B细胞祖细胞或它们的组合中的两者或更多者的方法,所述方法包括使用如表2中所述的这些细胞的表面上的标志物中的两种或更多种标志物,任选地以及如表3、表4和表5中所述的荧光标记的抗体中的一种或多种荧光标记的抗体,使用荧光检测方法,来同时检测淋巴细胞、NK-T细胞、NK细胞、单核细胞和/或早期B细胞祖细胞中的两者或更多者的存在或不存在。
2.一种评估主要包括CD4+和/或CD8+T细胞的细胞群体中的非CD3+污染物的方法,所述方法包括使所述细胞群体与针对淋巴细胞、NK-T细胞、NK细胞、单核细胞和/或早期B细胞祖细胞的特异性表面标志物的一种或多种抗体接触以产生混合物,其中所述特异性细胞表面标志物中的两种或更多种特异性细胞表面标志物描述于表2中,任选地,其中所述一种或多种抗体选自表3、表4和表5;以及通过荧光检测方法分析所述混合物中具有特异性细胞表面标志物的细胞的分布。

7.一种用于确定T细胞产物是否适合于免疫疗法的方法,所述方法包括使用表3、表4和表5中所述的抗体或抗体混合物中的一者表征所述T细胞产物中的CD3-细胞杂质(例如,NK-T细胞、NK细胞、单核细胞、早期B细胞祖细胞或它们的组合)中的一种或多种CD3-细胞杂质;以及基于所述T细胞产物中CD3-细胞杂质的水平确定所述T细胞产物是否适合。
8.根据权利要求7所述的方法,其中能够接受的水平由管理机构(例如,
FDA、EMEA等)设定。
9.一种用于使用表3、表4和表5中所述的抗体或抗体混合物中的一者或多者在血细胞群体中鉴定T淋巴细胞、NK-T细胞、NK细胞、单核细胞总淋巴细胞、早期B细胞祖细胞或它们的组合中的至少一者的测定或试剂盒。
10.根据权利要求9所述的测定或试剂盒,其中所述测定或试剂盒用于表征用于免疫疗法的T细胞产物中CD3-细胞的存在。
