技术领域
本发明属于生物制药技术领域,具体为一种RBX1在制备抑制未分化甲状腺癌转移药物中的应用。
背景内容
甲状腺癌是一种高度流行的癌症。***状甲状腺癌(PTC)占所有甲状腺癌病例的85%;可固化90%以上。与之形成鲜明对比的是,未分化甲状腺癌(ATC)是最恶性的甲状腺癌,约占甲状腺癌死亡率的40%。与PTC或其他甲状腺癌不同,ATC生长迅速,侵袭性强。阐明其进展和转移的机制对治疗学的发展具有重要意义。
细胞代谢的重新编程是癌细胞的一个关键特征。有氧糖酵解,也称为沃堡效应,在多种癌症的生长中起着重要作用,包括乳腺癌、肝癌、非小细胞肺癌、膀胱癌和甲状腺癌。丙酮酸激酶是一种限速糖酵解酶,可促进磷酸烯醇式丙酮酸转化为丙酮酸。它有四种亚型:PKL(丙酮酸激酶肝)、PKR(丙酮酸激酶红细胞)、PKM1(丙酮酸激酶M1型)和PKM2(丙酮酸激酶M2型)。最近的报告强调了PKM2在多种癌症的致癌、有氧糖酵解和转移中的作用。众所周知,PKM,特别是PKM2,在某些癌症中过度表达。例如,PKM2在40%以上的急性白血病患者中过度表达,并与不良治疗结果相关。在胃癌中,PKM2过表达与预后相关,表明其可能用作预后生物标志物。通过抑制PKM2和增强PDC活性,癌症代谢已被重新编程,这增加了对几种癌症的抗癌效果。同时,许多研究表明,癌细胞通过调节PKM选择性剪接来促进PKM2的表达,从而达到正常细胞的代谢边缘;因此,靶向这种选择性剪接被证明是消除癌细胞的有效策略。因此,确定PKM替代剪接的潜在因素和机制,促进PKM2介导的Warburg效应,对于克服当前癌症治疗的挑战至关重要。
泛素和泛素样蛋白对蛋白质的修饰是细胞生理学中涉及的主要翻译后调节机制。Skp1/Cullin/F-box(SCF)复合物是最大的E3泛素连接酶和广泛泛素化蛋白。RBX1首先从酵母中分离和纯化,作为人和酵母SCF复合物的必需成分。越来越多的证据表明,RBX1的表达促进了许多癌症类型的肿瘤进展和转移,包括乳腺癌、肝癌、非小细胞肺癌、膀胱癌和卵巢癌。最近,研究表明RBX1是癌症中泛素蛋白酶体***(UPS)的关键调节因子。由于其E3泛素连接酶活性,RBX1有助于目标蛋白的泛素化,并标记其降解。然而,它在ATC进展和潜在信号级联中的确切作用仍不清楚。
迄今为止,关于RBX1是否调控未分化甲状腺癌肿瘤细胞的转移和迁移以及RBX1的细胞内信号调节机制,并没有报道。
发明内容
本发明的目的在于为了解决如上问题,而提供一种抑制肿瘤转移的方法和试剂。
本发明的第一方面,提供了一种RBX1的下调剂,所述RBX1下调剂能够在蛋白或基因水平上下调RBX1的表达或活性;
所述RBX1的下调剂选自:核酸抑制物(如干扰分子)、蛋白抑制剂、蛋白水解酶以及蛋白结合分子(如抗体或配体)中的一种。
优选的,所述RBX1的下调剂为:以RBX1基因或其转录本为靶序列、且能够抑制RBX1基因表达或基因转录的干扰分子;
所述RBX1的下调剂为:shRNA、小干扰RNA、dsRNA、微小RNA、反义核酸,或者能表达或形成所述shRNA、小干扰RNA、dsRNA、微小RNA、反义核酸的构建物,或特异性与RBX1蛋白结合的结合分子(如能够抑制RBX1蛋白活性的抗体或配体)。
优选的,所述RBX1的下调剂具有以下shRNA结构:Seq正向-X-Seq反向;
其中,Seq正向的核苷酸序列具有SEQIDNO:1或SEQIDNO:2所示序列;
Seq反向为与Seq正向完全互补或者非完全互补的序列,非完全互补(包括基本上互补的情形)可以存在部分错配或者突起;
X为位于Seq正向和Seq反向之间的间隔序列,并且所述间隔序列与Seq正向和Seq反向不互补。优选的,所述的间隔序列长度5~50nt;更佳地,7~30nt。
优选的,所述的具有shRNA结构的RBX1的下调剂可形成以下结构:

本发明的第二方面,提供了一种RBX1在制备抑制未分化甲状腺癌转移药物中的应用,将所述的RBX1的下调剂制备成抑制未分化甲状腺癌肿瘤转移的组合物。
本发明的第三方面,提供了一种组合物,包含治疗有效量的所述RBX1的下调剂,以及药学上可接受的载体或辅料。
本发明有如下有益效果:
本发明发现RBX1的表达在ATC组织中显著上调。我们的临床研究表明,RBX1的高表达与生存不良和远处侵袭显著相关。功能分析表明,RBX1通过增强Warburg效应促进ATC细胞转移,PKM2在RBX1介导的有氧糖酵解中起关键作用。此外,我们证实RBX1调节PKM选择性剪接并促进ATC细胞中PKM2介导的Warburg效应。此外,由RBX1介导的PKM选择性剪接诱导的ATC细胞迁移和需氧糖酵解依赖于SMAR1/HDAC6复合物的破坏;RBX1作为E3泛素连接酶,通过泛素蛋白酶体途径降解ATC中的SMAR1。因此,我们的研究首次确定了ATC细胞中PKM选择性剪接调控的机制,并为RBX1在细胞适应代谢应激中的作用提供了线索。

在抑制未分化甲状腺癌转移中的应用研究
一、材料
患者和标本
来自南昌大学第二附属医院普通外科的30名ATC患者的组织样本。病理学家证实,所有标本均来自正常组织。这项研究获得南昌大学附属第二医院临床研究伦理委员会的批准,所有受试者同意提供研究样本。
甲状腺癌细胞株TPC-1、BCPAP、KTC-1、C643(前四类为PTC细胞系)、CAL62、8505C、KMH-2、KMH-5M和BHT101(后五类为ATC细胞系)购自Procell Life Science&TechnologyCo.,Ltd.。细胞系在37°C的含5%CO2的培养箱中,在含有10%胎牛血清FBS(Gibco;ThermoFisher Scientific,Inc.)的DMEM或1640(Gibco;Thermo Fisher Scientific,Inc.)中培养。
二、实验方法
1、实时定量PCR(qRT-PCR)
(1)qRT- PCR 反应体系:
该体系的配制过程应全程冰上操作
(2)按照上述体系内各组分体积配制混合管后,将其准确上样到qRT- PCR仪专用的96孔板中,样品加完后贴上封板膜以密封上样孔,按以下条件设置上机程序并开始qRT-PCR。
(3)分析qRT-PCR得出的实验数据:目的基因和内参基因的差值即ΔCt值,实验组与对照组的差值即为ΔΔCt值,最终实验结果以2—ΔΔ
Ct 值的形式表示mRNA差异表达程度。
2、蛋白质印迹法(western blotting)
根据制造商的说明,使用RIPA缓冲液(Millipore;Sigma Aldrich)裂解ATC细胞。使用BCA蛋白质检测试剂盒(Takara生物技术有限公司)测定总蛋白质浓度。使用10%SDS-PAGE胶分离样品(20µg),然后转移至PVDF膜。在室温下用5%脱脂牛奶封闭膜2小时,然后在4℃下用不同蛋白的抗体抗RBX1(1:100;目录号ab221548,Abcam)、PKM(1:500;目录号ab150377,Abcam)、泛素(1:100,目录号10201-2-AP;Proteintech)、SMAR1(1:500,目录号Pa5-100402;Thermo Fisher Scientific)和HDAC6(1:500;目录号ab133493;Abcam)处理。Tubulin表达(1:20 00;cat.No.11224-1-ap;protentech group,Inc.)用作内参对照。用Tris缓冲盐-0.1%吐温-20洗涤三次后,将膜与HRP标记的山羊抗兔IgG二级抗体(1:5000;猫号ab6728;Abcam)在4℃下孵育。最后,使用ECL***对蛋白质带进行可视化,并使用ImageJ软件进行分析。
3、免疫组化(IHC)染色(免疫组织化学法检测)
(1)ATC组织包埋后将蜡块制成石蜡切片,放入37℃烤箱中烘干;
(2)烘干石蜡切片后将其放入二甲苯和梯度酒精内以达到脱蜡目的;
(3)脱蜡完成后将石蜡玻片用PBS缓冲液冲洗2-3次,5min/次;
(4)石蜡玻片冲洗完成后将其置于装有柠檬酸盐缓冲液的量杯内,放入微波炉内高火蒸煮,加热至沸腾后转中火避免气泡;
(5)上述抗原修复完成后3% H2O2浸泡处理10min;
(6)石蜡玻片经H2O2孵育后用PBS缓冲液冲洗2-3次,5min/次
(7)石蜡玻片冲洗完成后擦干表面液体,向玻片上的组织滴加覆盖TBST缓冲液配制的5%山羊血清以达到封闭目的,室温封闭1h;
(8)封闭完成后擦干表面封闭用血清,向玻片上的组织滴加适量一抗稀释液配制的一抗,置于4°C层析冷柜一抗孵育过夜;
(9)次日将玻片从层析冷柜中取出复温, 用PBS缓冲液冲洗2-3次,5min/次;
(10)石蜡玻片冲洗完成后擦干表面液体,向玻片上的组织滴加适量二抗,置于37°C二抗孵育1h;
(11)二抗孵育完成后用PBS缓冲液冲洗2-3次,5min/次;
(12)石蜡玻片冲洗完成后擦干表面液体,向玻片上的组织滴加适量DAB显色液,将玻片显微镜下观察染色情况,待显色完成后终止染色;
(13)显色完成的玻片浸入苏木素溶液中复染,孵育2-3min后流水冲洗玻片上剩余苏木素溶液。
(14)复染后的玻片再次放入梯度酒精脱水后,二甲苯处理后中性树胶封片。
使用无血清蛋白质阻断缓冲液阻断切片30分钟,之后将其与抗RBX1(1:1000;目录号ab221548,Abcam)孵育。使用奥林巴斯BH-2显微镜(奥林巴斯公司)获得图像并数字化;使用ImageJ 1.51v软件(美国国立卫生研究院)对DAB信号进行量化。
4、shRNA
介导的双链RNA(shRNA),用于沉默RBX1(shRBX1)、SMAR1(shSMAR1)和PKM2(shPKM2),由中国上海的一家公司合成。使用基因药物技术合成全长人RBX1、SMAR1和PKM2cDNA,并连接到pcDNA3.1载体中,分别生成p-RBX1、p-SMAR1和p-PKM2。空白载体用作阴性对照。根据制造商的说明,使用Lipofectamine 3000转染试剂(Invitrogen,USA)转染这些shRNA质粒和构建体。
5、细胞侵袭与迁移实验
Transwell分析用于评估ATC细胞系的迁移和侵袭性,并进行了一些修改。对于侵入测定,在聚碳酸酯膜的上部预先施加一层基质凝胶。具体步骤为:
(1)从-20℃中取出购买到的自带有基质胶(Matrigel)的transwell 小室,在室温条件下复温;
(2)Transwell小室复温后在其上室中加入PBS磷酸缓冲液200µl,放入温度设置为37℃、CO2浓度设置为5%的细胞培养箱中培养1h后吸净加入的PBS磷酸缓冲液,然后在下室中加入含有10%FBS 的DMEM培养基。
(3)按上述细胞传代和细胞计数的方法将要进行实验的细胞以30000个/小室的量接种到小室中,需要注意的是此时接种的细胞是用不含血清的培养基重悬的。接种完成后放入温度设置为37℃、CO2浓度设置为5%的细胞培养箱中培养48h。
(4)培养完成后用移液枪吸去上室中的原有的培养基,然后将小室置于4%多聚甲醛以固定细胞,固定30min。
(5)固定完成后将孔中的多聚甲醛固定液吸去,再用 PBS 磷酸缓冲液对细胞冲洗2-3遍。冲洗完成后将小室置于滤过的0.1%的结晶紫染色液染色,染色 30min。
(6)染色完成后将孔中的结晶紫染色液吸去,用清水反复冲洗并用棉签将小室内部残留的细胞擦去,随后将其放在通风处下晾干。
(7)将晾干的Transwell小室倒置在显微下用不同倍数拍照,保存图片并分析实验结果。
6、体内转移实验
将1×106个细胞(CAL62、KMH-2)在100μL磷酸盐缓冲盐水中的等分试样皮下注射到裸小鼠的侧翼。一旦产生的肿瘤直径达到1-2cm,将其切除并切成约1mm3的块;然后将这些碎片植入裸小鼠(每组6只,雄性BALB/cnu/nu,6-8周)。小鼠在肿瘤植入后六周被安乐死。肺被切除、处理并包埋在石蜡中。所有动物工作均经南昌大学附属第二医院动物实验伦理委员会批准,并根据NIH《实验动物护理和使用指南》进行。
7、耗氧率(OCR)和细胞外酸化率(ECAR)的测定
细胞外通量分析仪XF96(Seahorse Bioscience,Billerica,MA,USA)用于测量细胞糖酵解能力和线粒体呼吸。根据制造商的说明,分别使用XF细胞水压力测试套件和海马生物科学。
8、免疫共沉淀(co-immunoprecipitation, co-IP )和泛素化测定
对于体内泛素化测定,在收获前将RBX1敲低或RBX1过表达的ATC细胞加入MG132处理4小时。随后,细胞裂解物用抗SMAR1抗体免疫沉淀;并且用抗Ub抗体测量SMAR1的泛素化。
免疫共沉淀的具体方法为:
(1)弃去细胞培养基,用预冷的PBS漂洗细胞两遍,彻底去除培养皿中的液体后将培养皿置于冰上;
(2)按照Western及IP裂解液: PMSF=100:1的体积比配制细胞裂解液,充分混匀后均匀滴在步骤(1)中的细胞培养皿中,置于冰上裂解15-30min;
(3)利用干净的刮匙迅速刮取培养皿底物的细胞裂解产物并收集于1.5mlEppendorf管中,随后放入预冷至4℃的高速离心机中以12000r/min的转速离心15min;
(4)离心结束后取60μl的上清液转移至另一个新的1.5ml Eppendorf管中作为Input,取5-10μl蛋白样品用作BCA法蛋白浓度测定,并标记好细胞蛋白名称、日期等信息后置于-20℃冰箱保存备用;在剩余蛋白样品中取500μl的上清液转移至另一个新的1.5mlEppendorf管中,按照抗体说明书加入适量抗体及50μl Protein A琼脂糖珠,置于4℃摇床过夜,使蛋白样品、抗体与磁珠耦联;
(5)从摇床上取出Eppendorf管放入4℃预冷的离心机中,以3000rpm/min的转速离心5分钟,用1ml移液器小心吸取上清弃去,注意不要吸到沉淀中的磁珠,再加入预冷的600μl Western及IP裂解液后置于4℃摇床使之充分混匀5min;
(6)重复步骤(5)三次;
(7)将Eppendorf管置于4℃预冷的离心机中,以3000rpm/min的转速离心5分钟,用1ml移液器小心吸取上清弃去,加入50μl(与磁珠等体积)的2×上样缓冲buffer,水浴沸煮5-10min;最后通过 Western blotting分析实验结果。
9、EdU 实验
(1)取对数生长期的甲状腺癌细胞,制备单细胞悬液。然后细胞计数,记数后计算细胞浓度。将细胞接种 96 孔板。
(2)将各组质粒通过脂质体转染至甲状腺癌细胞中,每组设 6 个复孔,实验重复3 次;
(3)每孔加入稀释后 EdU 的培养基,培养箱内孵育 2 小时,用吸引器吸弃培养基;加入细胞固定液,30min 后,每孔先后加入 2mg/ml 的甘氨酸、100µl PBS,弃上清液,PBS 清洗一次;然后先后加入 100µl Apollo 染色反应液、100µl 0.5% Triton-X 渗透液染色,弃上清液,PBS 清洗一次;再加入 100µl Hoechst 33342 反应液行 DNA 染色,避光、室温下脱色摇床孵育 30min,弃上清液,PBS 清洗一次;最后在荧光显微镜下观察细胞染色情况,统计分析实验结果。
三、实验结果
1、RBX1的过表达与ATC不良预后相关
为了了解RBX1与ATC(未分化甲状腺癌)的临床相关性,我们使用qRT-PCR和western blotting检测了30个ATC组织样本和相应的相邻对照组织(癌旁组织)中RBX1的表达。与癌旁组织相比,RBX1 mRNA在肿瘤样品中过表达(图1A-B)。此外,ATC组织中RBX1蛋白水平显著升高,与qRT-PCR结果一致(图1C-D)。如图1E所示,RBX1在62.6%的ATC组织样本中过度表达。这些结果表明,RBX1蛋白表达水平在ATC组织中显著上调(图1E)。
western blotting结果显示,ATC细胞系中RBX1的表达显著高于甲状腺细胞系和PTC细胞系(图1F-G)。接下来,RBX1蛋白过度表达与ATC临床病理参数之间的相关性测试表明,RBX1水平低的患者的总生存时间显著长于RBX1水平高的患者(图1H)。总之,这些数据支持RBX1在ATC进展中发挥作用的假设,并为ATC诊断提供了潜在的生物标志物。
2、RBX1在体外和体内调节ATC细胞的迁移和侵袭性
我们在两个高侵袭性ATC细胞系CAL62和KMH-5M中用亚型特异性shRNA敲除RBX1。与加扰的shRNA相比,shRBX1#1和shRBX1#2两个不同的干扰片段显著降低了ATC细胞中的RBX1表达(图2A),以及8305C细胞系中RBX1过表达的稳定模型(图2B)。Transwell分析还表明,RBX1敲低显著抑制了CAL62和KMH-5M细胞的转移能力,而RBX1过表达促进了8305C细胞的迁移能力(图2C-D)。与对照细胞相比,RBX1敲低显著抑制了ATC细胞的增殖能力,但RBX1过表达显著促进了ATC细胞增殖能力(图2E-F)。我们通过在包括shNC(对照处理组)和shRBX1组的裸鼠中建立肿瘤模型,进一步研究了RBX1对ATC转移的影响。连续肺切片显示,shRBX1组中ATC肺微转移的数量显著减少(图2G)。相反,RBX1的过度表达增加了肺转移结节的数量(图2H)。总之,这些结果表明RBX1的稳定敲除可以在体外和体内抑制ATC的侵袭和转移,同时在ATC进展和转移过程中也作为癌基因候选。
3、RBX1通过增强Warburg效应促进ATC进展
E3泛素连接酶有能力促进几种类型癌症进展中的重编程代谢。由于Warburg效应是肿瘤细胞(包括ATC)中普遍存在的一种典型的代谢转变,我们探讨了RBX1在ATC葡萄糖代谢中的作用。RBX1敲低显著降低了CAL62细胞中葡萄糖-6-磷酸(G6P)、葡萄糖消耗、乳酸生成和ATP的细胞水平(图3A),而RBX1过表达在8305C细胞中具有相反的作用(图3B)。为了进一步验证RBX1对ATC糖酵解的影响,我们测量了反映总糖酵解通量的ECAR(细胞外酸化检测)。RBX1敲低显著降低了CAL62细胞的糖酵解速率和容量(图3C-D),而RBX1过表达增加了8305C细胞的ECAR(图3E-F)。作为线粒体呼吸指标的OCR(耗氧率)在CAL62/shRBX1细胞中增加(图3G-H),而RBX1过表达降低了8305C细胞中的OCR(图3I-J)。为了研究Warburg效应是否导致ATC细胞的进展,用不同浓度的2-脱氧葡萄糖(2-DG)处理CAL62/shRBX1和8305C/p-RBX1细胞24小时。此外,CAL62/shRBX1和8305C/p-RBX1细胞的迁移和侵袭能力以剂量依赖的方式降低(图3M-N)。为了证明糖酵解调节ATC迁移和侵袭,将细胞培养在含有半乳糖而非葡萄糖的培养基中,降低糖酵解通量,迫使细胞依赖氧化磷酸化。这大大消除了RBX1过表达诱导的8305C细胞迁移和侵袭能力的增加(图3O-Q)。这些发现表明,RBX1抑制ATC细胞中的氧化磷酸化,同时促进有氧糖酵解,但通过增强ATC细胞系中的Warburg效应促进迁移和侵袭。
4、PKM2对RBX1促进Warburg效应至关重要
与正常细胞相比,癌细胞中PKM2比PKM1更高的表达是实现Warburg效应代谢优势的关键因素。PKM2表达的减少可能导致Warburg效应的抑制,抑制致瘤潜能。因此,我们通过最初测量RBX1敲低和过表达ATC细胞中PKM2的表达,来确定RBX1是否调节PKM2表达。Western blotting显示RBX1敲低显著降低了CAL62细胞中PKM2的表达(图4A)。相反,RBX1过表达显著增加了8305C细胞中PKM2的表达(图4B)。此外,PKM2的上调减弱了CAL62/shRBX1细胞中PKM2表达的减少(图4C)。进一步实验发现,PKM2表达的恢复消除了RBX1沉默诱导的ATC细胞转移能力的降低(图4D)。体内肿瘤转移试验显示,PKM2的过度表达挽救了CAL62/shRBX1细胞肝内和肺转移的发生率降低(图4E)。此外,PKM2表达的上调挽救了RBX1介导的ATC细胞中G6P、葡萄糖消耗、乳酸生成和ATP水平的降低(图4F)。同时,RBX1敲低降低了ATC细胞中的ECAR和OCR,而PKM2的同时过表达减弱了糖酵解速率和能力的降低(图4G-H)。接下来,我们评估了PKM2表达降低对RBX1和PKM2蛋白水平的影响,以及对RBX1过表达8305C细胞的细胞迁移和侵袭性的影响。Western blotting显示,RBX1过表达显著增加PKM2表达,而PKM2表达的敲低显著抑制了RBX1诱导的8305C细胞PKM2表达增加(图4I)。同时,PKM2下调显著降低了RBX1增强的细胞迁移和侵袭(图4J)。此外,体内转移试验表明,PKM2下调降低了8305C-RBX1组肝内和肺转移的发生率(图4K)。PKM2的敲除挽救了RBX1介导的ATC细胞中G6P、葡萄糖消耗、乳酸生成和ATP水平的增加(图4L)。RBX1过表达增加了ATC细胞中的ECAR和OCR,而同时敲低PKM2则减弱了糖酵解速率和能力的增加(图4M–N)。这些发现表明PKM2是RBX1调节有氧糖酵解的功能下游靶点,对RBX1介导的肿瘤进展至关重要。
5、RBX1调节PKM选择性剪接以促进PKM2介导的Warburg效应
癌细胞通过调节PKM选择性剪接和促进PKM2的表达,获得了高于正常细胞的代谢优势(31)。为了进一步证明RBX1是否是该现象的原因,我们测量了ATC患者样本中RBX1、PKM2和PKM1的表达水平,并通过western blotting将其与周围非癌组织中的表达水平进行了比较。ATC组织中RBX1和PKM2蛋白水平显著升高;此外,散点图显示RBX1和PKM2表达之间呈正相关(图5A–B)。然而,PKM1在ATC组织中的表达明显较低,RBX1和PKM1表达呈负相关(图5C–D)。基于在ATC中观察到的RBX1和PKM2表达之间的正相关以及RBX1和PKM1之间的负相关,这些数据表明RBX1在调节PKM选择性剪接中起关键作用。
为了研究RBX1在PKM选择性剪接调控中的作用,在CAL62细胞中shRNA介导的RBX1表达缺失后,使用定量qRT-PCR和western blotting测量PKM同种型表达。在mRNA水平上,CAL62细胞中RBX1的敲除导致PKM1上调和PKM2表达降低(图5E)。在蛋白水平上对RBX1缺失细胞中PKM同种型表达的进一步分析表明PKM1的上调和PKM2的表达减少,这与RNA表达数据一致(图5F)。此外,qRT-PCR和western blotting均显示,RBX1的上调显著增加了8305C细胞中PKM2的表达,并降低了PKM1的表达(图5G–H)。然后我们研究了高或低RBX1表达的组织样品中PKM1同种型和PKM2同种型的表达。高RBX1表达的组织样品中的PKM2/PKM1比率显著高于低RBX1表达组织样品(图5I)。为了进一步验证RBX1在PKM选择性剪接调控中的作用,产生了双报告PKM小基因***。与对照相比,RBX1过表达条件下的eGFP(增强型绿色荧光蛋白)/mCherry(单体红色荧光蛋白)比率增加了5.4倍。与对照相比,RBX1敲除条件下eGFP/mCherry比率的倍数差异减少了3.2倍,这进一步验证了其在PKM选择性剪接调控中的作用(图5J-K)。总之,这些研究证实RBX1调节PKM选择性剪接以促进ATC细胞中PKM2介导的Warburg效应。
6、通过SMAR1/HDAC6复合物降解调节PKM选择性剪接
我们最初尝试使用质谱法鉴定ATC细胞中的RBX1相互作用蛋白。如表1所示,我们确定了几个已报道的RBX1相互作用体,包括TLE3、FoxA1和SHP-1,以及一个先前未报道的RBX1相互作用体SMAR1。这些实验验证了SMAR1与HDAC6的直接相互作用以及SMAR1-HDAC6作为复合物的共存。这种复合物的形成进一步表明了SMAR1介导的PKM选择性剪接调控中这些蛋白质之间的分子动力学。因此,我们假设RBX1通过SMAR1/HDAC6复合物降解调节PKM选择性剪接。Co-IP测定证实了RBX1和SMAR1之间的相互作用(图6A)。我们发现,纯化的GST-RBX1(带GST标签的RBX1)在体外与FLAG标记的SMAR1结合(图6B)。对接分析还揭示了RBX1和SMAR1之间的结合相互作用(图6C)。这些发现表明RBX1与ATC细胞中的SMAR1结合。此外,如图6D所示,RBX1敲低上调了CAL62细胞中SMAR1的蛋白水平,而RBX1过表达显著降低了8305C细胞中SMAR2的表达。然而,SMAR1 mRNA表达不受ATC细胞中RBX1改变的影响(图6E-F),表明RBX1在翻译后调节SMAR1的表达。

已有研究表明,SMAR1是多泛素化的,并被UPS降解。RBX1作为一种E3泛素连接酶,在蛋白质降解和循环中发挥作用;因此,我们假设RBX1通过调节SMAR1的泛素化来介导SMAR1的减少。为了验证这一假设,我们首先测量了在使用环己酰亚胺(CHX)抑制蛋白合成后RBX1敲除细胞中SMAR1蛋白的退化。如图6G所示,沉默RBX1显著抑制了ATC细胞中SMAR1的降解。此外,我们发现在用蛋白酶体抑制剂MG132处理后,在RBX1过表达细胞中恢复了SMAR1的蛋白质水平(图6H)。第二,RBX1的异位表达增加了SMAR1的泛素化水平,而RBX1的敲低降低了SMAR1多泛素化(图6I)。最后,数据表明,SMAR1的所有Lys位点的突变消除了RBX1在ATC细胞中诱导的SMAR1多泛素化(图6J)。此外,泛素上Lys48位点的突变几乎完全消除了RBX1介导的SMAR1泛素化,而泛素上K63R突变没有影响(图6K)。这些发现表明RBX1通过ATC中的泛素蛋白酶体途径对SMAR1的降解负责。
最后,我们确定RBX1是否通过破坏的SMAR1/HDAC6复合物影响PKM选择性剪接。在RBX1敲除的CAL62细胞中测量SMAR1、HDAC6、PKM1和PKM2表达以及SMAR1/HDAC6复合物的变化。我们发现,CAL62细胞中RBX1敲除显著增加了SMAR1、SMAR1/HDAC6复合物和PKM1的表达水平,降低了PKM2的表达,但HDAC6蛋白水平没有变化(图6L–M)。相反,8305C细胞中RBX1的过表达显著降低了SMAR1、SMAR1/HDAC6复合物和PKM1的表达,并增加了PKM2的表达(图6N–O)。总之,这些结果表明,由RBX1介导的PKM选择性剪接调控诱导的ATC细胞迁移和需氧糖酵解依赖于SMAR1/HDAC6复合物的破坏。
7、RBX1介导的PKM选择性剪接调控是SMAR1依赖性的
我们将shSMAR1质粒转染到RBX1敲除的ATC细胞中,并测量其对生物功能的影响。RBX1敲除显著增加SMAR1蛋白表达,而SMAR1敲除显著抑制RBX1敲减诱导的ATC细胞SMAR1表达增加(图7A)。进一步实验发现,SMAR1表达的下调消除了RBX1沉默诱导的ATC细胞转移能力的降低(图7B-C)。SMAR1的下调挽救了CAL62/shRBX1细胞增殖能力的降低(图7D–E)。SMAR1表达的敲除挽救了shRBX1介导的ATC细胞中G6P、葡萄糖消耗、乳酸生成和ATP水平的降低(图7F)。同时,shRBX1降低了ATC细胞中的ECAR和OCR,而同时敲低SMAR1减弱了糖酵解速率和容量的降低(图7G-H)。相比之下,SMAR1上调显著降低了RBX1增强的细胞迁移和侵袭(图7I-K),降低了8305C/RBX1组的增殖能力(图7L-M)。此外,SMAR1表达的上调挽救了RBX1介导的ATC细胞中细胞G6P、葡萄糖消耗、乳酸生成和ATP水平的增加(图7N)。同时,RBX1过表达增加了ATC细胞中的ECAR和OCR,而SMAR1的同时上调减弱了糖酵解速率和容量的增加(图7O-P)。总之,这些发现表明RBX1介导的PKM选择性剪接的调节,促进ATC细胞迁移和有氧糖酵解,是SMAR1依赖性的。
四、实验结论
我们的研究证实,RBX1在ATC组织中高度表达,RBX1的表达增加与ATC患者的恶性程度和生存率相关。此外,我们发现RBX1不仅抑制ATC细胞中的氧化磷酸化,同时促进有氧糖酵解,还通过增强ATC细胞系中的Warburg效应促进迁移和侵袭。PKM2是RBX1调节有氧糖酵解的功能下游靶点,对RBX1介导的肿瘤进展至关重要。我们的发现表明,RBX1通过破坏ATC细胞中的SMAR1/HDAC6复合物来调节PKM选择性剪接以促进PKM2介导的Warburg效应。基于这些发现,RBX1应被视为ATC诊断和治疗的候选生物标志物。

Claims (10)
1.一种RBX1在制备抑制未分化甲状腺癌转移药物中的应用,其特征在于:将所述RBX1的下调剂制备成抑制未分化甲状腺癌肿瘤转移的组合物。
2.根据权利要求1所述的应用,其特征在于:所述RBX1下调剂能够在蛋白或基因水平上下调RBX1的表达或活性;
所述RBX1的下调剂选自:核酸抑制物、蛋白抑制剂、蛋白水解酶以及蛋白结合分子中的一种。
3.根据权利要求1所述的应用,其特征在于:所述RBX1的下调剂为:以RBX1基因或其转录本为靶序列、且能够抑制RBX1基因表达或基因转录的干扰分子;
所述RBX1的下调剂为:shRNA、小干扰RNA、dsRNA、微小RNA、反义核酸,或者能表达或形成所述shRNA、小干扰RNA、dsRNA、微小RNA、反义核酸的构建物,或特异性与RBX1蛋白结合的结合分子。
4.根据权利要求1所述的应用,其特征在于:所述RBX1的下调剂具有以下shRNA结构:Seq正向-X-Seq反向;
其中,Seq正向的核苷酸序列具有SEQIDNO:1或SEQIDNO:2所示序列;
Seq反向为与Seq正向完全互补或者非完全互补的序列;
X为位于Seq正向和Seq反向之间的间隔序列,并且所述间隔序列与Seq正向和Seq反向不互补。
5.根据权利要求4所述的应用,其特征在于:所述的具有shRNA结构的RBX1的下调剂可形成以下结构:

其中,||表示在Seq正向和Seq反向之间形成的氢键。
6.根据权利要求4所述的应用,其特征在于:所述的间隔序列长度5~50nt。
7.根据权利要求4所述的应用,其特征在于:所述RBX1的下调剂是包含所述shRNA结构的表达载体,所述表达载体为病毒表达载体。
8.根据权利要求7所述的4所述的应用,其特征在于:所述病毒表达载体中,还包括与所述shRNA结构操作性连接的启动表达元件、该组成型或诱导型的启动子。
9.根据权利要求7所述的4所述的应用,其特征在于:所述病毒表达载体为慢病毒载体,具体为四环素诱导表达慢病毒载体,或者pTRIPz慢病毒载体。
10.一种组合物,其特征在于:包含治疗有效量的如权利要求2或3所述的RBX1的下调剂,以及药学上可接受的载体或辅料。
