Abstract
本发明属于生物技术领域,具体涉及一种Caerin1.1/1.9联合抗CD47抗体在制备治疗黑色素瘤药物中的应用。本发明中Caerin1.1的氨基酸序列如SEQ ID NO.1所示;Caerin1.9的氨基酸序列如SEQ ID NO.2所示。本发明的Caerin1.1/1.9可以使B16细胞凋亡的同时促使B16细胞高表达CD47,在抗CD47抗体的存在下,促使巨噬细胞吞噬肿瘤细胞,提高caerin1.1/1.9的疗效,也优于单独使用抗CD47抗体,为扩大CD47抑制/阻断药物的应用奠定基础。本发明提供的Caerin1.1/1.9还可以与anti‑CD47抗体,治疗型疫苗联用,使50%荷瘤小鼠完全清除肿瘤。

背景技术
恶性黑色素瘤是由皮肤和其他器官黑素细胞产生的肿瘤。皮肤黑色素瘤表现为色素性皮损在数月或数年中发生明显改变。虽其发病率低,但其恶性度高,转移发生早,死亡率高,因此早期诊断、早期治疗很重要。恶性黑素瘤大多发生于成人,巨大性先天性色素痣继发癌变的病例多见于儿童。由于黑色素瘤对人体危害极大,因此,其治疗一直是备受各界关注的。
免疫治疗是肿瘤治疗的新方法。免疫检查点抑制剂如抗PD1,CTLA4已广泛在临床使用。随着科学研究的不断发展进步,新的免疫检查点也不断被发现,如anti-CD47,但是这些免疫检查点在实际使用时,存在一个共性问题,由于这些免疫检查点在诱导肿瘤细胞凋亡的同时,还会影响巨噬细胞,使其无法吞噬肿瘤细胞,最终破坏肿瘤免疫微环境,限制了免疫治疗效率。
Caerin1多肽家族是澳大利亚树蛙皮肤分泌的抗菌多肽,包括Caerin1.1、Caerin1.9等,具有抗菌和抗肿瘤细胞生长的活性。研究发现,Caerin1.1、Caerin1.9在体外可以抑制人和小鼠***细胞,人甲状腺癌细胞,人乳腺癌细胞生长,通过TNFalpha信号传导通路诱导Hela细胞凋亡;偶联核素,可以提高其抗肿瘤效率;体内可以抑制TC-1细胞生长,并吸引血细胞如T细胞,NK细胞等到肿瘤组织。但是这些仅证明了Caerin1.1、Caerin1.9可以抑制肿瘤,并不会改变上述问题。而且,现有技术中并没有关于Caerin1.1、Caerin1.9用于黑色素瘤的报道。
综上可知,现有技术中普遍具有在治疗过程中,破坏肿瘤免疫微环境,影响免疫治疗效率,Caerin1.1、Caerin1.9应用范围窄等技术问题。
发明内容
针对现有技术普遍具有的问题,本发明提供了一种Caerin1.1/1.9联合抗CD47抗体在制备治疗黑色素瘤药物中的应用。三者联合用于制备治疗黑色素瘤的药物,提高了免疫治疗效果。
为了达到上述目的,本发明采用的技术效果为:
一种Caerin1.1/1.9联合抗CD47抗体在制备治疗黑色素瘤药物中的应用。
优选地,所述Caerin1.1/1.9与抗CD47抗体(clone:MIAP301)的质量比为3:10~25(即60μg:200~500μg/小鼠)。
小鼠添加的抗CD47抗体的剂量为200~500μg /小鼠,Caerin1.1/1.9的添加剂量为60μg/小鼠,其中,Caerin1.1与Caerin1.9的质量比为1:1,即各添加30μg/小鼠。
优选地,所述Caerin1.1的氨基酸序列如SEQ ID NO.1所示;所述Caerin1.9的氨基酸序列如SEQ ID NO.2所示。

机理研究发现,Caerin1.1/1.9改善肿瘤免疫微环境,重新极化肿瘤浸润巨噬细胞,使其从M2型向M1型转化。分泌更多的白介素12,更少的白介素10。如果治疗型疫苗含黑色素瘤治疗性疫苗,白介素10抑制抗体,caerin1.1/1.9与之联合使用,抑制TC-1细胞生长的效率更好。Caerin1.1/1.9与anti-CD47抗体,治疗型疫苗三者联合使用,可以使20-50%的TC-1荷瘤小鼠完全清除肿瘤。
发明人发现,Caerin1.1/1.9 体外抑制黑色素瘤B16细胞生长,诱导黑色素瘤B16细胞凋亡,并可以进入B16细胞,在体内抑制B16细胞生长。使B16细胞高表达CD47,在B16模型中提高抗CD47抗体效率,吸引血细胞到肿瘤,特别是巨噬细胞,CD4+T细胞到肿瘤,使巨噬细胞吞噬肿瘤细胞。与anti-CD47抗体,治疗型疫苗联用,可以使50%荷瘤小鼠完全清除肿瘤。
与现有技术相比,本发明技术方案具有如下优势:本发明解决了抗CD47在黑色素瘤的疗效问题,诱导B16细胞高表达CD47,抗CD47与Caerin1.1/1.9联用提高了其治疗B16肿瘤的疗效。抗CD47与治疗性疫苗和Caerin1.1/Caerin1.9进行三联治疗能达到较使用α-PD-1时更好的治疗B16肿瘤的效果。

具体实施方式
下面结合具体实施例对本发明作进一步解释,但是应当注意的是,以下实施例仅用以解释本发明,而不能用来限制本发明,所有与本发明相同或相近的技术方案均在本发明的保护范围之内。本实施例中未注明具体技术或条件者,按照本领域常规技术方法和仪器说明书内容进行操作;所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规产品。
试验例1 体外检测试验
一、试验过程:
1.MTT法检测细胞存活情况:取每孔7×104个B16分别与不同浓度(0,6,8,10,12,14,16,18,20µg/mL)的F1/F3培养过夜,各浓度4个复孔,利用MTT试剂盒(产品编号:C0009M)检测细胞存活情况,并作图计算IC50值。两次独立实验取最新一次结果展示。
2.细胞凋亡检测:分为试验组与对照组,用完全培养基(组分由1640基础培养基+南美血清+双抗(青霉素、链霉素按1:1混合)按体积比90:9:1组成)培养B16细胞,其中,试验组中加入Caerin1.1/Caerin1.9,终浓度为10µg/mL,对照组中加入P3,终浓度同样为10µg/mL,于37℃,5%的CO2培养箱中培养过夜,利用细胞凋亡试剂盒(Annexin V-EGFP 细胞凋亡检测试剂盒,产品编号:C1067M)检测其凋亡情况。
二、试验结果:
1.细胞存活情况如图1所示,图1中,A代表不同浓度下的细胞存活情况,B代表计算的存活率。由图1可知,IC50(IC50半数致死量)=11.76µg/mL,Caerin1.1/Caerin1.9能明显抑制B16细胞的增殖。
2.细胞凋亡情况如图2所示,图2中,A为细胞凋亡情况分布图,B为统计学分析结果,由此可知,在10µg/mL时,其对B16有着明显的诱导凋亡的作用。
试验例2 体内检测试验
1.试验对象:C57BL/6小鼠共12只,平均分成3组,每组4只;
2.试验方法:
2.1瘤重检测:A.每只C57BL/6小鼠皮下种植4×105个B16细胞,记荷瘤时为DAY 0,在DAY3时,三组分别瘤内注射100μL含30μg F1/F3(即Caerin1.1/Caerin1.9)的PBS溶液;100μL含30μg P3的PBS溶液;阴性对照组每天注射100μL PBS,连续7天给药;B.造模方法同上,每组8只,于DAY13时处死,取肿瘤进行称重。
2.2细胞活性检测:试验过程同2.1,于DAY13取肿瘤组织处理成单细胞悬液进行流式染色,利用流式细胞仪进行检测;选用的流式抗体如下:FITC-CD45.2,APC-cy7-CD3e,Percp-cy5.5-CD4,PE-cy7-CD8a,APC-cy7-B220,PE-NK1.1,PE-F4/80,Percp-cy5.5-CD11b,FVS510死活染料;同时检测巨噬细胞亚型的改变,DAY13取肿瘤组织处理成单细胞悬液进行流式染色,利用流式细胞仪进行检测,选用的流式抗体如下:FITC-CD45.2,PE-F4/80,Percp-cy5.5-CD11b,BV421-Ly6C,FVS510死活染料。
3.试验结果:
3.1试验结果如图3所示,图3中,A为Caerin1.1/Caerin1.9联合治疗黑色素瘤荷瘤小鼠的生存周期,B为其各组肿瘤大小称重数据图,由图可知,单用Caerin1.1/Caerin1.9能在体内有效抑制肿瘤生长。
3.2细胞活性检测试验结果如图4所示,图4中A~G含义如下:A代表CD45+细胞占活细胞的比例;B代表CD3+细胞占CD45+细胞的比例;C代表CD4+细胞占CD45+细胞的比例;D代表CD8+细胞占CD3+细胞的比例;E代表NK1.1+细胞占CD45.2+细胞的比例;F代表巨噬细胞占CD45.2+细胞的比例;G代表巨噬细胞亚型占巨噬细胞的比例不同抗体下的细胞活性。由此可知,CD8+T细胞和CD4+T细胞数量上升;巨噬细胞数量上升;促进肿瘤免疫的炎性巨噬细胞上升,抑制肿瘤免疫的组织驻留巨噬细胞下降,巨噬细胞的INFα receptor表达上升,亚型巨噬细胞内receptor表达上升,说明:在经过多肽治疗后,“冷肿瘤”转变为“热肿瘤”,而且提示多肽发挥抑制肿瘤生长的作用与巨噬细胞及INFα有着很重要的关系。
试验例3 肿瘤涂抹治疗检测
1.试验对象:C57BL/6小鼠12只,分为3组,每组4只。
2.试验过程:每只C57BL/6小鼠皮下种植4×105个B16细胞,记荷瘤时为DAY 0,于DAY3时将小鼠分为三组:UN组(不做任何处理),imquimod组,F1/F3 Cream1-2组,于DAY 3开始涂抹实验,涂抹开始前腹腔注射1%的戊巴比妥进行麻醉,imquimod组:将imquimod软膏均匀涂抹于肿瘤表面至刚好覆盖肿瘤,隔天涂抹一次,涂抹至DAY11;F1/F3 cream1-2组:将F1/F3软膏均匀的涂抹于肿瘤表面至刚好覆盖肿瘤,一天两次,连续涂抹9天。于DAY12时取肿瘤组织处理成单细胞悬液进行流式染色,利用流式细胞仪进行检测。选用的流式抗体如下: FITC-CD45.2,PE-F4/80,Percp-cy5.5-CD11b,BV421-Ly6c,FVS510死活染料。
3.试验结果:具体试验结果如图5所示。图5中,左图为各组组织驻留巨噬细胞的数量变化结果,右图为炎性巨噬细胞的数量变化结果,由图5可知,经过不同剂型的多肽治疗后,巨噬细胞亚型的变化基本保持一致,说明了F1/F3治疗黑色素瘤与巨噬细胞相关的结论具有可靠性。
试验例4 F1/F3联用治疗MDSC向巨噬细胞转化
1.实验对象:C57BL/6小鼠6只,分为2组,每组3只。
2.试验过程:每只C57BL/6小鼠皮下种植4×105个B16细胞,记荷瘤时为DAY 0,在DAY3时,二组分别瘤内注射100μL含30μg F1/F3(即Caerin1.1/Caerin1.9)的PBS溶液;阴性对照组每天注射100μL PBS,连续7天给药。于DAY13取B16肿瘤组织处理成单细胞悬液进行流式染色,并利用流式细胞仪进行检测。选用的抗体如下:FITC-CD45.2,Percp-cy5.5-CD11b,APC-Ly6G,BV421-Ly6c,FVS510死活染料,PE-F4/80;造模方法同上,于DAY13取小鼠的脾脏处理成单细胞悬液,利用流式细胞仪对m-MDSC进行分选,设置4个组:PBS组,PBS+GMCSF组,F(F1/F3)组,F(F1/F3)+GMCSF组,对分选的细胞进行细胞计数,每组设置2个复孔,每孔100000个细胞,并按照组别加入10ng/mL的GMCSF,刺激培养三天后利用流式细胞仪进行检测。选用的抗体如下:FITC-CD45.2,Percp-cy5.5-CD11b,APC-Ly6G,BV421-Ly6c,FVS510死活染料,PE-F4/80。
3.实验结果:此为两次独立实验,取最新一次实验结果进行展示。具体实验结果如图6所示,图6中,A代表巨噬细胞占CD45.2+细胞的比例;B代表MDSC占CD45.2+细胞的比例;C代表m-MDSC占MDSC的比例;D代表PMN-MDSC占MDSC的比例。由此可知:经过多肽治疗后的肿瘤巨噬细胞有升高的趋势,和前面结果保持一致,此外MDSC、m-MDSC及PMN-MDSC占比有减少的趋势,说明多肽同时也会通过降低髓源性抑制细胞的占比来达到促进肿瘤免疫的目的,如后续分选体外刺激培养实验结果(图7)所示,图7中,AB两图表示未添加或添加刺激剂的情况下,两组巨噬细胞占CD45.2阳性细胞比例的典型流式图,C图代表加入刺激剂后巨噬细胞占CD45.2+细胞的比例的柱状统计图。结合A图未加GMCSF刺激剂m-MDSC未向巨噬细胞分化的现象可知:相较于PBS组,F1/F3治疗组的脾脏分选所得的m-MDSC在GMSCF刺激分化后的巨噬细胞更多,在体外初步证明经过多肽治疗后,m-MDSC分化为巨噬细胞。
试验例5 F1/F3联合α-PD-1治疗黑色素瘤
1.试验过程:在C57BL/6小鼠皮下接种4×105 B16细胞到侧腹部的时间记为DAY0,通常在能明显观察到肿瘤时(DAY 3)将其分为4组PBS+PBS,PBS+α-PD-1,F(F1/F3)+PBS,F(F1/F3)+α-PD-1,每组各10只,按照分组要求开始连续7天瘤内注射100μL含30μg F1/F3的PBS溶液和PBS溶液,然后分别于DAY 9和DAY 15按组别腹腔注射含200µg α-PD-1的PBS溶液和PBS溶液。于DAY 43对已经治愈的小鼠,在另一边的侧腹部皮下接种4×105 B16细胞进行肿瘤再刺激,并同时设置相同数量的C57小鼠在侧腹部接种相同数目B16细胞作为control组。
2.试验结果:具体试验结果如图8所示,相较于单用F1/F3治疗策略,F1/F3与α-PD-1免疫联合治疗并未延长小鼠的生存周期,但经过免疫联合疗法治疗后的小鼠肿瘤预后要优于单用F1/F3多肽。
试验例6 F1/F3联用治疗模型增加CD47的改变
1.试验对象:C57BL/6小鼠21只,分为3组,每组7只。
2.试验方法:造模方法同试验例3,试验分为PBS组、P3组及F1/F3组,于DAY13取肿瘤组织处理成单细胞悬液进行流式染色,利用流式细胞仪进行检测,连续进行2次试验,流式抗体选用如下:FITC-CD45.2,FVS510死活染料,BV421-CD47。
3.试验结果:此为两次独立实验结果的总结,具体试验结果如图9所示;图9中,
经过多肽治疗后的肿瘤的生长虽然得到抑制,但是治疗组肿瘤细胞上CD47的表达明显增加,说明在治疗过程中,肿瘤细胞仍然发出“Don’t eat me”信号,实现免疫逃逸,这为后续Caerin1.1/Caerin1.9联合Anti-CD47治疗黑色素瘤提供一定的理论基础与可行性,与此同时我们发现巨噬细胞上的SIRPα表达降低,预示着在经过多肽治疗后的免疫微环境在抑制肿瘤细胞免疫逃逸的发生,此结果为后续F1/F3,治疗性疫苗,anti-CD47三联治疗黑色素瘤提供一定的理论基础。
试验例7 F1/F3联合CD47治疗黑色素瘤
1.试验过程:在C57BL/6小鼠皮下接种4×105 B16细胞到侧腹部的时间记为DAY0,通常在能明显观察到肿瘤时(DAY 3)将其分为4组:PBS+PBS组,PBS+anti-CD47组,F+PBS组,F+anti-CD47组,每组6只,并按照分组要求于DAY3开始连续7天分别瘤内注射含F1/F3各30μg的PBS溶液和PBS溶液,同时分别腹腔注射含anti-CD47 200μg的PBS溶液和PBS,此后每隔一天腹腔注射anti-CD47 100μg直到实验结束,体积均为100μL。
2.试验结果:具体试验结果如图10所示,此为两次独立实验的综合结果,相较于单用F1/F3或单用anti-CD47,F1/F3与anti-CD47免疫联合治疗明显延长小鼠的生存周期。
试验例8 F1/F3联合α-PD-1与治疗性疫苗治疗黑色素瘤
1.试验过程:在C57小鼠皮下接种4×105 B16细胞到侧腹部的时间记为DAY 0,通常在能明显观察到肿瘤时(DAY 3)将其分为三组:PBS+PBS+PBS组,P3+V+α-PD-1组,F1/F3+V+α-PD-1组,并开始连续7天肿瘤内分别予以PBS,含F1/F3各30μg的PBS溶液或含P3(阴性对照)30μg的PBS溶液,体积都为100μL。分别于DAY 9和DAY 21腹腔注射300μg α-PD-1,于DAY3,9,18肌肉注射治疗性疫苗(V,由三个组分组成:黑色素瘤治疗性疫苗50μg/只,MPLA15μg/只,α-IL10r 300μg/只,平均每只C57小鼠重量为16-20g),连续重复3次。
2.试验结果:具体试验结果如图11所示,此为三次独立实验的总和。肿瘤体积变化结果如图12所示,三次独立实验取最新一次实验结果展示。由图12可知,经过三联治疗后的小鼠生存周期得到了极大的延长,并有接近半数小鼠的肿瘤彻底被清除,相较于PBS+PBS+PBS组与P3+V +α-PD-1组有着明显的显著性差异,而P3+V +α-PD-1组较PBS+PBS+PBS组的生存时间也明显增加,说明基于F1/F3,α-PD-1及治疗性疫苗的三联疗法可行性,并为后续anti-CD47的三联免疫治疗提供理论基础。
试验例9 三联疗法免疫微环境变化研究
1.试验过程:造模方法同试验例8,于DAY23取肿瘤组织处理成单细胞悬液进行流式染色,利用流式细胞仪进行检测。选用的流式抗体如下:FITC-CD45.2,APC-cy7-CD3e,Percp-cy5.5-CD4,PE-cy7-CD8a,PE-NK1.1,PE-F4/80,Percp-cy5.5-CD11b,APC-Ly6C,BV421-CD56,BV421-MHCII,PE-CY7-CD86,FVS510死活染料。
2.试验结果:试验结果如图13~图14所示F1/F3+V+α-PD-1组较另外两组,CD4+T细胞、DP T细胞、CD56+NK细胞的占比升高,代表着三联疗法较二联疗法能更好的促进肿瘤免疫。同时结果还表示不管是CD86+的巨噬细胞还是MHC II+的巨噬细胞其Ly6c+亚群占比升高,这与单用多肽治疗时的结果保持一致,初步判断三联疗法同样也是通过巨噬细胞发挥主要作用。
试验例10 F1/F3治疗裸鼠荷瘤模型
1.试验过程:造模方法同试验例8,将小鼠分为三组:PBS组,P3组,F1/F3组,,在裸鼠皮下接种5×105 B16细胞到侧腹部的时间记为DAY 0,于DAY3按照组别分别瘤内注射100ul含F1/F3多肽各30μg的PBS溶液,含P3 30μg的PBS溶液及PBS溶液,连续给药7天,于DAY13将小鼠处死并取肿瘤进行肿瘤称重。
2.试验结果:具体试验结果如图15所述。F1/F3治疗后,荷瘤裸鼠的生存周期明显延长。
试验例11 F1/F3联合anti-CD47与治疗性疫苗治疗黑色素瘤
1.试验过程:在C57小鼠皮下接种4×105 B16细胞到侧腹部的时间记为DAY 0,通常在能明显观察到肿瘤时(DAY 3)将其分为四组:PBS+PBS+PBS组,P3+V+anti-CD47组,F1/F3+V+anti-CD47组,F1/F3+V+α-PD-1组,并开始连续7天肿瘤内分别予以PBS,含F1/F3各30μg的PBS溶液或含P3(阴性对照)30μg的PBS溶液,体积都为100μL。F1/F3+V+α-PD-1组分别于DAY 9和DAY 21腹腔注射含300μg α-PD-1的PBS溶液,体积为100μL,P3+V+anti-CD47组、F1/F3+V+anti-CD47组、F1/F3+V+α-PD-1组于DAY3,9,18肌肉注射100μL治疗性疫苗(V,由三个组分组成:黑色素瘤治疗性疫苗50μg/只,MPLA 15μg/只,α-IL10r 300μg/只,平均每只C57小鼠重量为16-20g),连续重复3次,其余组注射100μL PBS。P3+V+anti-CD47组,F1/F3+V+anti-CD47组于DAY 3开始每只老鼠腹腔注射含200μg anti-CD47的PBS溶液,体积为100μL,每隔一天给药一次,其余组腹腔注射100μl PBS组作为对照,直至实验结束。
2.试验结果:具体实验结果如图16所示。由图可知,基于F1/F3联合anti-CD47与治疗性疫苗的三联疗法治疗黑色素瘤的效果优于基于F1/F3联合α-PD-1与治疗性疫苗的疗法,二联疗法也能抑制肿瘤的生长,但是其治疗效果不如三联疗法。以上说明将α-PD-1替换为anti-CD47能更好的抑制肿瘤生长并改善三联疗法的治疗效果。
最后,需要说明的是,上述实施例仅例示性说明本发明的原理、性能及功效,而非用于限制本发明。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通常知识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。。

1. 一种Caerin1.1/1.9联合抗CD47抗体在制备治疗黑色素瘤药物中的应用;所述Caerin1.1/1.9与抗CD47抗体的质量比为3:10~25;所述Caerin1.1/1.9由Caerin1.1与Caerin1.9按质量比1:1组成;所述Caerin1.1的氨基酸序列如SEQ ID NO.1所示;所述Caerin1.9的氨基酸序列如SEQ ID NO.2所示。
2.如权利要求1所述的应用,其特征在于,所述药物还包括药学上可接受的载体。
3. 一种Caerin1.1/1.9在诱导黑色瘤细胞B16大量表达CD47中的应用;所述Caerin1.1/1.9由Caerin1.1与Caerin1.9按质量比1:1组成;所述Caerin1.1的氨基酸序列如SEQ ID NO.1所示;所述Caerin1.9的氨基酸序列如SEQ ID NO.2所示。
