背景技术
2011年,Wang课题组(J Exp Med 2011,208:577-592)和陈列平等(JImmunol2011,187:1537–154)分别报道了一种新型免疫检查点蛋白:T细胞激活抑制物免疫球蛋白可变区结构域(V-domain immunoglobulin suppressor of T-cell activation,VISTA)。VISTA是可抑制T细胞免疫应答的负向免疫检查点,其属于B7家族成员。VISTA胞外段免疫球蛋白可变区(immunoglobulin variable region,IgV)结构与PD-1相似,故又称PD-1同源物(PD-1homolog,PD-1H),是Ⅰ型跨膜蛋白,在骨髓来源的细胞上高表达,且VISTA的存在会抑制T细胞活化和增殖,在调节先天和适应性免疫应答中发挥重要作用。在T淋巴细胞中,VISTA在初始CD4+T细胞和CD8+T细胞上表达,但在B细胞上的表达水平较低。VISTA可作为抗原呈提细胞(APC)的抑制配体,通过未知受体调节T细胞反应。此外,VISTA还可以作为T细胞上的抑制性受体。在多种小鼠肿瘤模型和临床前研究表明,VISTA作为负向免疫检查点在肿瘤免疫治疗领域存在的潜在治疗效果。单抗和小分子抑制剂靶向人的VISTA在早期临床实验中用于肿瘤免疫治疗。
除了在肿瘤免疫治疗中发挥重要作用,VISTA在炎症和自身免疫性疾病中也发挥重要作用。2014年,本发明人与合作者报道了VISTA基因缺陷可导致活化的T细胞累积,并伴随着细胞因子和化学因子产生的增加(PNAS 2014,111(41):14846-14851),这表明VISTA基因缺陷可以加速自身免疫性疾病进程。2020年,本发明人的课题组报道了VISTA缺陷可以加重银屑病样皮肤炎症发生,并且T细胞和树突状细胞数量显著增加(Theranostics,2020,10:10483-10497)。在炎症状态下,VISTA在各种免疫细胞类型上的表达可能发生改变。例如,在咪喹莫特诱导的小鼠银屑病模型中,TCRγδT细胞上的VISTA表达高于引流***上的VISTA表达(Sci Rep.2017,7:1485-1487)。在类风湿性关节炎患者滑膜组织中检测到VISTA的表达(Arthritis Res Ther.2017,19:270-274)。在VISTA敲除小鼠中发现CD4+和CD8+T细胞自发激活,细胞因子的产生增加(如IFN-γ,TNF-α和IL-17A),以及多种组织的慢性炎症,这表明VISTA途径受到抑制后外周免疫耐受性丧失(Science.2020,367:6475)。此外,VISTA KO小鼠更易发生自身免疫性疾病,如实验性自身免疫性脑脊髓炎(ArthritisRheumatol.2017,69:814-825)。在小鼠哮喘模型中观察到,VISTA缺失促进嗜酸性粒细胞在肺部大量积累。VISTA激动性单抗则可以改善小鼠对移植物抗宿主病(GVHD)和实验性肝炎的自身免疫反应(Journal of Immunology,2011,187:1537-1541)。在易患***性红斑狼疮的Sle1.Sle3小鼠中,VISTA在炎性单核细胞(F4/80+Gr-1+CD11bhigh)和活化T细胞(CD3+CD4+CD44high)中表达减少(Lupus.2018,27:210-216)。老年雌性VISTA-KO小鼠可发展为严重的自身免疫性疾病,表现为升高的抗核抗体(ANA)样的***性红斑狼疮(SLE)特征。2019年,陈列平等报道了VISTA-/-BALB/c小鼠可自发发展为红斑狼疮,与人类狼疮患者症状相似,研究发现小鼠皮肤狼疮病灶有浆细胞样树突状细胞(plasmacytoid DC,pDC)聚集。VISTA激动性单克隆抗体(MH5A)可减少pDC促炎细胞因子的产生,包括红斑狼疮中的关键致病性细胞因子IFN-α。另外,作者还对临床红斑狼疮患者的活检样本和外周血进行了VISTA表达的研究,发现与正常样本相比,VISTA在狼疮患者免疫细胞中的表达均高于健康对照组(可能是代偿性的)(Sci Transl Med.2019,11:522-536)。综上所述,VISTA在自身免疫性疾病发生发展中发挥重要作用,而激活VISTA信号通路有望成为防治红斑狼疮等自身免疫性疾病的新策略和新途径。
红斑狼疮是(lupus erythematosus,LE)是一种多发于育龄期女性、累及多脏器的病因不明的自身免疫性炎症性***病,全球患病率约为0.12-0.39%。***性红斑狼疮(system lupus erythematosus,SLE)是红斑狼疮各类型中最为严重的一型,其病理机制复杂,受环境因素、遗传易感性和免疫细胞固有功能障碍的影响;以异常活化的T细胞、自身反应性B细胞和大量异常细胞因子为特征,最终导致多器官损害,尤其是造成狼疮性肾炎,是SLE最常见的并发症及死亡原因之一,是影响SLE预后的重要因素,具有潜在的致死性。SLE病机的复杂性和高度异质性使其有效治疗面临巨大困难,目前临床上常用于治疗SLE的类固醇药物***片、抗疟疾药羟氯喹、免疫抑制剂口服环磷酰胺、沙利度胺以及罗氏的生物制剂Rituxan(利妥昔单抗)等都属于超说明书用药,且存在疗效欠佳、不良反应严重等问题。贝利单抗是近60多年来FDA批准的唯一针对SLE的治疗药物,但该药费用昂贵并需注射使用,且只对部分患者有效。盘状红斑狼疮(Discoid lupus erythematosus,DLE)是最常见的慢性皮疹性红斑狼疮,目前尚无FDA批准的治疗药物。因此,红斑狼疮治疗药物的研发成为国际药学界公认的最具挑战性的研究领域之一。
银屑病(psoriasis)俗称“牛皮癣”,是一种由环境因素刺激、多基因遗传控制、免疫介导的皮肤病,典型表现为鳞屑性红斑或者斑块,局限于一处或全身广泛分布。欧美研究提示,银屑病患病率为1%~3%,而我国银屑病的患病率较低,约为0.47%。该病可发生于各个年龄段,无性别差异;北方地区发病多于南方;30%的患者有家族史;多数患者冬季复发或加重,夏季缓解。
伊马替尼(结构如下)是一种酪氨酸激酶抑制剂,通过抑制酪氨酸激酶活性而破坏肿瘤细胞的信号传递,以达***的目的。2001年美国食品药品管理局正式批准伊马替尼用于治疗慢性髓性白血病(CML),之后又批准该药治疗无法切除的或转移性恶性胃肠道间质肿瘤。伊马替尼还可治疗急性淋巴细胞性白血病、骨髓增生异常综合征、进行性***性肥大细胞增生症、嗜酸性粒细胞增多综合征、慢性嗜酸性粒细胞白血病等疾病。
迄今为止,无任何文献报道伊马替尼通过VISTA这个靶点对红斑狼疮以及银屑病等自身免疫性疾病的效应。
发明内容
发明目的:为解决目前临床上缺乏有效的***以及银屑病等自身免疫性疾病的药物这一问题,本发明提供了伊马替尼作为VISTA激动剂在预防或***以及银屑病等自身免疫性疾病中的一种新用途。
本发明的另一个目的是提供一种预防或***以及银屑病等自身免疫性疾的药物组合物。
技术方案:为了实现以上技术目的,本发明提供伊马替尼在制备VISTA激动剂方面的用途。

本发明所述的药物组合物在制备预防或治疗自身免疫性疾病中的用途,所述药物组合物其含有伊马替尼作为活性成分和药学上可接受的载体。
其中,所述自身免疫性疾病包括红斑狼疮、银屑病、自身免疫性脑脊髓炎、自身免疫性肝炎或类风湿性关节炎。。
在某些优选的实施方案中,所述的药物组合物可用于预防或***以及银屑病。
本发明所述的药物组合物的剂型是胶囊剂、散剂、片剂、颗粒剂、丸剂、注射剂、糖浆剂、口服液、吸入剂、软膏剂、栓剂或贴剂。
本发明首先通过表面等离子共振(SPR)技术初步筛选出selleck化合物库中与人VISTA蛋白的亲和能力较好的化合物,接着通过细胞水平实验首次发现伊马替尼可显著地与VISTA蛋白结合,并激动其下游信号通路,进而抑制IFN-γ、TNF-α、IL-1β和IL-2等炎症因子的释放。因此,伊马替尼可用于制备VISTA激动剂。
对于所述的***性红斑狼疮,本发明采用cGVHD样的***性红斑狼疮小鼠为实验对象。在小鼠动物实验水平,伊马替尼的给药量为50mg/kg,所述化合物可通过商业化途径购买。本发明人发现,伊马替尼有效降低了狼疮小鼠尿蛋白、外周血中尿素氮、肌酐以及抗dsDNA抗体、肾脏中炎症因子IL-6/MCP-1/TNF-α/TGF-β/IL-1β、肾脏中免疫复合物IgG的沉积,并减轻了狼疮小鼠肾脏病理损害;对于所述的银屑病,本发明采用咪喹莫特诱导的银屑病小鼠为实验对象。在小鼠动物实验水平,伊马替尼的给药量为50mg/kg,所述化合物可通过商业途径购买。本发明人发现,伊马替尼有效降低了银屑病小鼠耳厚度、皮肤损伤程度、皮肤中炎症因子TNF-α以及IL-1β的mRNA表达含量。
有益效果:与现有技术相比,伊马替尼能够有效改善狼疮小鼠病情,具体表现如下:(1)伊马替尼处理后狼疮小鼠尿蛋白水平明显下降;(2)伊马替尼能够降低狼疮小鼠外周血中尿素氮、肌酐以及抗dsDNA抗体水平;(3)用伊马替尼进行干预后,狼疮小鼠肾脏病理损害明显减轻、肾脏中炎症因子IL-6/MCP-1/TNF-α/TGF-β/IL-1β以及免疫复合物IgG沉积也同样减少。同时伊马替尼能够有效改善银屑病小鼠病情,具体表现如下:(1)伊马替尼处理后银屑病小鼠耳厚度、皮肤损伤程度降低;(2)伊马替尼能够显著降低皮肤中炎症因子TNF-α以及IL-1βmRNA的表达。
本发明首次提供了伊马替尼(Imatinib)作为免疫检查点VISTA激动剂的应用,具体涉及伊马替尼可以激动免疫检查点VISTA,从而发挥***以及银屑病等自身免疫性疾病的用途。此外,伊马替尼是一种已经应用于临床的抗肿瘤药物,其安全性以及毒副作用已经过临床考验,本发明通过体内外实验已经验证其可以作为VISTA激动剂治疗或预防红斑狼疮的进展,从而加速药物的临床研究,推动临床用药,以解决现有***性红斑狼疮以及银屑病等自身免疫性疾病治疗药物匮乏、副作用大、价格昂贵的问题。

下面通过实施例具体地说明本发明的内容。在本发明中,以下所述的实施例是为了更好的阐述本发明,并不是用来限制本发明的范围。在不背离本发明的精神和范围的前提下,可以对本发明进行各种变化和修饰。
伊马替尼购买于上海sellcek公司,其CAS号为152459-95-5。
实施例1
表面等离子共振(SPR)初筛30个老药(selleck化合物库)与human VISTA蛋白的亲和力
表面等离子共振(Surface Plasmon Resonance,SPR)是一种光学现象,当入射光以临界角入射到两种不同折射率的介质界面时,可引起金属自由电子的共振,由于共振致使电子吸收了光能量,从而使反射光在一定角度内大大减弱。当表面等离子波和消逝波共振时检测到的反射光能量大幅度减弱,此时对应的入射光波长为共振波长,对应的入射角为共振角也就是SPR角。SPR随表面折射率的变化而变化,而折射率的变化又和结合在金属表面的生物分子质量成正比。因此可以通过获取生物反应过程中SPR角的动态变化,得到生物分子之间相互作用的特异性信号。SPR实验将人VISTA蛋白(RD system,Cat#7126-B7)通过氨基偶联的方式固定至CM5芯片(GE Healthcare,Cat#BR-1005-30)上,使用BiacoreT200(GE Healthcare)仪器,25℃检测不同浓度的化合物流经CM5芯片表面的响应值。
①VISTA蛋白的pH预富集:实验中所用的芯片为CM5,人VISTA蛋白用醋酸钠pH4.5、5.0、5.5分别稀释到10μg/ml,各准备100μl流速为10μl/min,依次进样180s,50mM的氢氧化钠为洗脱液,1x PBS-P+(GE Healthcare,Cat#28-9950-84)为整个流路***的缓冲溶液。通过预富集实验,确定PH=5.0的条件为最佳偶联条件。故用pH=5.0醋酸钠将VISTA蛋白稀释至10μg/ml,200μl进行正式的偶联操作,结果如图1。
②VISTA蛋白配体偶联:以最适pH=5.0的醋酸钠溶液将人VISTA蛋白稀释至20μg/ml,通过氨基偶联的方式偶联至CM5芯片的Flow cell 2样品通道,其中Flow cell 1为空白通道作为背景扣除,设置VISTA蛋白最大目标偶联量为10000RU。1x PBS-P+为整个流路***的缓冲溶液,将EDC与NHS以1:1比例混合后,注射入***来活化芯片表面。VISTA蛋白以10μl/min流速,流经600s,与CMS芯片表面充分结合。最后注射乙醇胺至流路,封闭芯片表面的剩余酯基团。结果如图1所示,显示芯片表面的偶联最为10375RU。已达到目标偶联量,即蛋白偶联成功结果。
③溶剂校正:小分子样品的运行缓冲液选用含5%DMSO(Sigma,Cat#d8418)的1xPBS-P+。按照如下表1混合4.5%和5.8%DMSO母液,配置5%DMSO浓度校正曲线。

④亲和力测定:用1.05x PBS-P+稀释0.4mM的小分子母液20倍,得到20μM在5%DMSO中的小分子,20μM作为最高进样浓度。之后用配好的5%DMSO运行缓冲液将分析物浓度向下对半稀释至少5个浓度梯度(根据实际样品亲和力强弱进行浓度梯度调整),并增加一个0浓度,作为空白对照。含5%DMSO的1x PBS-P+运行缓冲液以10μl/min的流速进样,设定样品结合时间与解离时间均为60s。通过Biacore T200 Evaluation软件进行检测和数据处理。实验结果表明(如图2所示),化合物伊马替尼与人VISTA蛋白结合能力较强。
实施例2
CCK-8评价化合物对PBMC以及Jurkat细胞的细胞毒性的效应
Cell Counting Kit-8(简称CCK-8)(Beyotime,Cat#C0038),是一种基于WST-8而广泛应用于细胞增殖和细胞毒性的快速、高灵敏度检测的试剂盒。CCK-8试剂中含有WST-8(化学名:2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐)是一种类似于MTT的化合物,它在电子耦合试剂1-甲氧基-5-甲基吩嗪鎓硫酸二甲酯(1-Methoxy PMS)存在条件下,可以被线粒体内的脱氢酶还原为具有高度水溶性的橙黄色甲瓒产物Formazan,生成的Formazan数量与活细胞的数量成正比。因此可利用这一特性直接进行细胞增殖和毒性分析,细胞增殖越多越快,则颜色越深;细胞毒性越大,则颜色越浅。
使用CCK-8试剂盒进行操作,首先将复苏的PBMC细胞(Allcells,Cat#PB003F-C-10M)的密度调整为1x106个/ml,Jurkat细胞的密度调整至2x105个/ml,以100μl/孔接种于96孔板中,同时每孔加入100μl的化合物伊马替尼处理细胞,每个浓度6个副孔,控制化合物的终浓度为0.15625μM、0.3125μM、0.625μM、1.25μM、2.5μM、5μM和10μM,细胞在37℃培养箱中培养48h后,弃去培养基,每孔避光加入含10%CCK-8的新鲜培养基100μl,继续培养4小时,然后在微孔震荡仪上震摇5分钟,酶标仪读取450nm处的吸光度值。结果如图3所示化合物伊马替尼在0~10μM时没有毒性。
实施例3
Jurkat T细胞激动模型的建立,用于鉴定化合物细胞活性
PMA(Phorbol-12-myristate-13-acetate,PMA)作为佛波酯的一种和外源凝集素(Phytohemagglutinin,PHA)可联合激动Jurkat T细胞,刺激T细胞的增殖和白介素2的分泌。在细胞激活的同时,若体系中存在VISTA蛋白产生的抑制信号,则激动效果减弱。在细胞模型的构建过程中,同时给予激动剂与VISTA蛋白,通过分析细胞上清中细胞因子的分泌情况判断T细胞被活化的情况。
①Jurkat细胞激活实验
第一天:在96孔板中包被humanVISTA蛋白溶液。使用PBS将human VISTA蛋白(RDsystem,Cat#7126-B7)分别稀释至5μg/ml、10μg/ml。每孔加入100μl蛋白溶液,4℃包被过夜。
第二天:吸去孔中的包被蛋白溶液,用PBS清洗2次,每孔200μl。每孔加入100μlJurkat细胞(中国科学院细胞库),每孔2x105个细胞。每孔再加入100μl PMA(Beyotime,Cat#S1819)与PHA(Sigma,Cat#L1668)的混合液,控制PMA的终浓度为1ng/ml,PHA的终浓度为6μg/ml。将96孔板放至37℃℃细胞培养箱内,48h后收集细胞上清用ELISA试剂盒(Biolegend,Cat#431804)检测IL-2的分泌量(组别设置如表2所示)。结果如图4所示,5μg/ml和10μg/ml的人VISTA蛋白均能抑制PMA-PHA诱导的Jurkat T细胞IL-2的分泌。

②化合物活性检测实验
第一天:在96孔板中包被human VISTA蛋白溶液。使用PBS将human VISTA蛋白稀释至5μg/ml。每孔加入100μl蛋白溶液,4℃包被过夜。
第二天:吸去孔中的包被蛋白溶液,用PBS清洗2次,每孔200μl。每孔先加入100μlJurkat的细胞,每孔2x105个细胞。化合物检测组加入100μl含有待测化合物的PMA与PHA的混合液,同时控制化合物的终浓度为2.5μM、5μM和10μM。对照组加入100μl PMA与PHA的混合液。空白组加入100μl的完全培养基。组间控制PMA的终浓度为1ng/ml,PHA的终浓度为6μg/ml。将96孔板放至37℃细胞培养箱内,48h后收集细胞上清检测IL-2的分泌量(组别设置如表3所示)。结果如图5所示,化合物伊马替尼在2.5μM、5μM和10μM均能减少PMA-PHA诱导的Jurkat细胞IL-2分泌并且具有剂量依赖性,表明化合物伊马替尼促进了VISTA蛋白的抑制作用,是VISTA蛋白的激动剂。

实施例4
PBMC细胞激动模型的建立,用于鉴定化合物细胞活性
Anti-CD3与T细胞上的T细胞受体(TCR)结合激活了TCR相关信号通路,而CD28是作为共刺激分子,与抗原呈递细胞(APC)的B7结合后可以加强TCR信号刺激,所以anti-CD28就是模拟APC的作用,与CD28结合后而强化TCR信号,进而刺激T细胞的增殖和细胞因子的分泌。在细胞激活的同时,若体系中存在人VISTA蛋白产生的抑制信号,则激动效果减弱。在细胞模型的构建过程中,同时给予激动剂与VISTA蛋白,通过分析细胞上清中细胞因子的分泌情况判断T细胞被活化的情况。
①PBMC细胞激活实验
第一天:在96孔板中用PBS包被空白组,激动组使用PBS将抗anti-human-CD-3抗体(Biolegend,Cat#317326)和anti-human-CD28抗体(Biolegend,Cat#302934)稀释至2.5μg/ml,抑制组保持anti-human-CD-3抗体和anti-human-CD28抗体浓度不变,同时设置humanVISTA蛋白浓度为2.5μg/ml、5μg/ml、10μg/ml,包被时每孔加入100μl蛋白混合溶液(蛋白均用PBS稀释),4℃包被18小时。
第二天:吸去孔中的包被蛋白溶液,用PBS清洗2次,每孔200μl。对照组与空白组均加入100μl完全细胞培养基,37℃孵箱预处理30min。然后每孔加入100μl的已复苏的PBMC(Allcells,Cat#PB003F-C-10M),每孔细胞1x105个(组别设置如表4所示)。将96孔板放至37℃细胞培养箱内,48小时后收集细胞上清用ELISA试剂盒(Biolegend)检测IFN-γ、IL-2和TNF-α的分泌量,结果如图6A、B和C所示。表明humanVISTA蛋白浓度在2.5μg/ml、5μg/ml和10μg/ml均能抑制Anti-human-CD-3抗体和anti-human-CD28抗体诱导的PBMC细胞IFN-γ、IL-2和TNF-α的分泌。

②化合物活性检测实验
第一天:在96孔板中用PBS包被空白组,激动组使用PBS将anti-human-CD-3抗体和anti-human-CD28抗体稀释至2.5μg/ml,抑制组以及化合物组保持anti-human-CD-3抗体和anti-human-CD28抗体浓度不变,humanVISTA蛋白浓度为2.5μg/ml包被时每孔加入100μl蛋白混合溶液,4℃包被18小时。
第二天:吸去孔中的包被蛋白溶液,用PBS清洗2次,每孔200μl。化合物检测组每孔加入100μl待测化合物,同时控制化合物的终浓度为2.5μM、5μM和10μM。对照组与空白组均加入100μl完全细胞培养基,37℃孵箱预处理30min。然后每孔加入100μl的已复苏的PBMC(Allcells,Cat#PB003F-C-10M)细胞,每孔细胞1x105个(组别设置如表5所示)。将96孔板放至37℃细胞培养箱内,48小时后收集细胞上清用ELISA试剂盒(Biolegend)检测IFN-γ、IL-2和TNF-α的分泌量,结果如图7A、B和C所示。伊马替尼在能减少anti-human-CD-3抗体和anti-human-CD28抗体诱导的PBMC细胞IFN-γ、IL-2和TNF-α的分泌并且具有剂量依赖性,表明化合物伊马替尼促进了VISTA蛋白的抑制作用,是VISTA蛋白的激动剂。

实施例5
CFSE实验建立human VISTA蛋白的PBMC激动模型,用于鉴定化合物对PBMC细胞增殖的影响
荧光染料CFSE,是一种可对活细胞进行荧光标记的新型染料,可以标记活体细胞。CFSE进入活细胞后,可与胞内蛋白共价结合,水解后释放出绿色荧光。CFSE随着细胞***而平均分配至子代细胞中,而导致荧光强度逐级递减,依据这一特性,可通过流式细胞仪被用于检测细胞增殖活力。
①CFSE实验建立human VISTA蛋白的PBMC激动模型
第一天:在96孔板中用PBS包被空白组,激动组使用PBS将抗anti-human-CD-3抗体(Biolegend,Cat#317326)稀释至1μg/ml,抑制组保持anti-human-CD-3抗体浓度不变,设置humanVISTA蛋白浓度为2.5μg/ml、5μg/ml、10μg/ml。蛋白均用PBS稀释,包被时每孔加入100μl蛋白溶液,4℃包被18小时(组别设置如表6所示)。
第二天:首先进行CFSE染料母液配制,取一支CFSE固体染料(5mM,Biolegend,Cat#423801)加入36μl的无水DMSO,配制母液浓度为5mM,溶解后混匀,进行分装,每管2μl,-20℃避光干燥保存,备用;接着进行细胞染色,取1μl的CFSE染料母液,加入PBS至1ml,配成终浓度为5μM的CFSE染料工作液。再取已复苏的PBMC,室温200xg,离心15分钟,弃上清。细胞中加入0.2ml的CFSE染料工作液,37℃避光孵育10分钟,室温200xg,离心15分钟,弃上清。沉淀用2ml新鲜的1640完全培养基重悬,室温避光孵育10分钟,计数后将细胞密度调整为1x106个/ml备用。弃去前一天板中的溶液,PBS洗两遍,然后每孔加入100μL CFSE染色的PBMC,再加入100μL的培养基,然后将其放入细胞培养箱中培养120小时。培养120小时后将样品收集于1.5mL离心管中,室温下800xg离心5分钟,弃上清;细胞用200μL流式细胞染色液重悬,室温800xg,离心5分钟,弃上清,重复洗涤两次。加入200μL流式染色液(PBS+2%FBS)重悬细胞后,使用BD C6流式细胞仪进行流式检测,FlowJo-v10进行数据处理。实验结果如图8所示,在0μg/mL、2.5μg/mL、5μg/mL或10μg/mL的人VISTA蛋白都会对PBMC的增殖产生抑制作用

②化合物活性检测实验
第一天:在96孔板中用PBS包被空白组,激动组使用PBS将抗anti-human-CD-3抗体(Biolegend,Cat#317326)稀释至1μg/ml,抑制组保持anti-human-CD-3抗体浓度不变,设置humanVISTA蛋白浓度为2.5μg/ml、5μg/ml、10μg/ml。蛋白均用PBS稀释,包被时每孔加入100μl蛋白混合溶液,4℃包被18小时。
第二天:首先进行CFSE染料母液配制,取一支CFSE固体染料加入36μl的无水DMSO,配制母液浓度为5mM,溶解后混匀,进行分装,每管2μl,-20℃避光干燥保存,备用;接着进行细胞染色,取1μl的CFSE染料母液,加入PBS至1ml,配成终浓度为5μM的CFSE染料工作液。再取已复苏的PBMC,室温200xg,离心15分钟,弃上清。细胞中加入0.2ml的CFSE染料工作液,37℃避光孵育10分钟,室温200xg,离心15分钟,弃上清。沉淀用2mL新鲜的1640完全培养基重悬,室温避光孵育10分钟,计数后将细胞密度调整为1x106个/ml备用。弃去前一天板中的溶液,PBS洗两遍,每孔加入100μl指定浓度的化合物伊马替尼,同时控制化合物的终浓度为2.5μM、5μM和10μM,37℃孵育30分钟(组别设置如表7所示)。然后每孔加入100μlCFSE染色的PBMC,将其放入37℃细胞培养箱中培养120小时。培养120小时后将样品收集于1.5mL离心管中,室温下800xg离心5分钟,弃上清;细胞用200μl流式细胞染色液重悬,室温800xg,离心5分钟,弃上清,重复洗涤两次。加入200μL流式染色液重悬细胞后,使用BD C6流式细胞仪进行流式检测,FlowJo-v10进行数据处理。实验结果如图9所示,伊马替尼会加重2.5μg/mL的human VISTA蛋白对PBMC的增殖产生的抑制作用。表明伊马替尼是VISTA的激动剂。

首先通过慢病毒转染构建了Jurkart过表达VISTA全长(Jurkat-VISTA-FL)以及空载体(Jurkat-EV)细胞(Br J Pharmacol,2021,178:1445-1458)。首先将两种细胞密度调整为2x105个/ml,并以100μl/孔接种于96孔板中,化合物检测组加入100μl含有待测化合物的PMA与PHA的混合液,同时控制化合物的终浓度为2.5μM、5μM和10μM。对照组加入100μl PMA与PHA的混合液。空白组加入100μl的完全培养基。组间控制PMA的终浓度为1ng/ml,PHA的终浓度为6μg/ml(组别设置如表8所示)。将96孔板放至37℃细胞培养箱内,48h后收集细胞上清检测化合物对Jurkat-VISTA-FL以及Jurkat-EV细胞IL-2的抑制率。实验结果如图10所示,表明化合物伊马替尼是靶向VISTA的激动剂。

实施例7
化合物对BALB/c野生型(wild type,WT)和BALB/cVISTA敲除(knockout,KO)小鼠CD4+T细胞IL-2的抑制率的影响,用于鉴定化合物的靶向性
第一天:在96孔板中分别包被anti-mouse-CD-3抗体,激动组与化合物实验组anti-mouse-CD-3抗体浓度为2.5μg/ml,对照组不加anti-mouse-CD-3抗体,只加入等体积的PBS(组别设置如表9所示)。抗体用PBS稀释,包被时每孔加入100μl anti-mouse-CD-3溶液,4℃包被过夜。
第二天:首先使用EasySep小鼠CD4+T细胞分离试剂盒从BALB/C野生型(wildtype,WT,上海南方模式生物科技股份有有限公司)和BALB/C VISTA敲除(knockout,KO,上海南方模式生物科技股份有限公司)小鼠的脾脏中分离出总的小鼠CD4+T细胞。要求CD4+T细胞纯度达到90%以上时才可以用于实验,细胞纯度结果如图11所示。接着吸去孔中的包被anti-mouse-CD-3抗体,用PBS清洗2次,每孔200μl。再将提取出来的CD4+T细胞的密度调整为1x106个/ml,并以100μl/孔接种于96孔板中,然后再将细胞放入37℃细胞培养箱内孵育30min后再加入不同浓度化合物,控制化合物的终浓度为2.5μM、5μM和10μM,,加入化合物后,再将96孔板放入37℃细胞培养箱内,72h后收取上清检测化合物对WT CD4+T以及VISTAKO的CD4+T细胞IL-2的抑制率,结果如图12所示,表明化合物伊马替尼是靶向VISTA的激动剂。

实施例8
①伊马替尼对cGVHD样***性红斑狼疮的小鼠的治疗效应选择试验动物:选择购买自北京维通利华公司CB6F1小鼠以及BALA/c小鼠,雌性,日龄48-56天;于普通环境下饲养,自由摄食饮水。
②药物制备:伊马替尼给药剂量均为50mg/kg,溶解于0.5%羧甲基纤维素钠的水溶液中,并用环磷酰胺(CTX)作为阳性对照组,将CTX溶解于生理盐水中配制成25mg/kg的浓度。
③动物分组、造模和给药:将小鼠随机分为空白对照组(Control)、模型组(Model)以及化合物组。再将模型组以及化合物组的小鼠构建成cGVHD样狼疮小鼠模型。取6至8周BALB/c雌性小鼠和(C57BL/6J×BALB/c)F1杂交雌性鼠,在无菌条件下,取出BALB/c鼠脾脏、***(肠系膜、腹股沟、颈部)和胸腺,制备淋巴细胞悬液。调整细胞密度为1x108个/ml,每次200μl通过尾静脉注射到Fl代鼠体内,注射时间分别为1、4、7、10天。对照组注入生理盐水,诱导红斑狼疮模型。6周后,对照组以及模型组给予0.05%羧甲基纤维素钠的水溶液,化合物组给予化合物伊马替尼每日一次灌胃给药(50mg/kg),给药周期为四周。
④血清anti-dsDNA抗体、尿素氮以及肌酐含量测定:用ELISA试剂盒测定各组小鼠血清中anti-dsDNA抗体;用尿素氮以及肌酐检测试剂盒(南京建成)去测量小鼠血清中尿素氮以及肌酐含量。结果如图13所示,模型组anti-dsDNA抗体、尿素氮以及肌酐水平均高于对照组,伊马替尼组anti-dsDNA抗体、尿素氮以及肌酐水平相对于模型组显著降低。
⑤脾脏重量测量:给药结束后,取小鼠脾脏并称重,如图14所示,模型组小鼠脾脏重量高于对照组,伊马替尼组脾脏重量水平相对于模型组均显著降低。
⑥尿蛋白含量测定:用尿蛋白检测试剂盒测定各组小鼠尿液中尿蛋白含量结果如图15所示,模型组尿蛋白高于对照组,伊马替尼组尿蛋白水平相对于模型组显著降低。
⑦RT-PCR:总RNA用TRIZOL试剂提取,mRNA用Prime Scrip RT Master Mix转录成cDNA。RT-PCR分析测量IL-6、MCP-1、TNF-α、TGF-β以及IL-1βmRNA水平,结果如图16所示,模型组IL-6、MCP-1、TNF-α、TGF-β以及IL-1β水平高于对照组,伊马替尼组IL-6、MCP-1、TNF-α、TGF-β以及IL-1β水平显著低于模型组。
⑧HE染色:小鼠给药结束后,取小鼠的肾脏并将其浸泡于4%多聚甲醛中,石蜡包埋;用苏木精和伊红染色(HE),HE染色流程如下:石蜡切片脱蜡至水:依次将切片放入二甲苯Ⅰ20min-二甲苯Ⅱ20min-无水乙醇Ⅰ5min无水乙醇Ⅱ5min-75%酒精5min,自来水洗。苏木素染色:切片入苏木素染液染3-5min,自来水洗,分化液分化,自来水洗,返蓝液返蓝,流水冲洗。伊红染色:切片依次入85%、95%的梯度酒精脱水各5min,入伊红染液中染色5min。脱水封片:切片依次放入无水乙醇I5min-无水乙醇II5min-无水乙醇Ⅲ5min-二甲苯Ⅰ5min二甲苯Ⅱ5min透明,中性树胶封片。显微镜镜检,图像采集分析。HE结果如图17所示,结果表明模型组的肾脏损伤程度以及炎性细胞浸润程度大于对照组,且有显著性差异,伊马替尼组肾脏损伤程度以及炎性细胞浸润程度相对于模型组有显著改善。
⑨免疫荧光法检测肾脏免疫复合物的沉积情况:小鼠给药结束后,取小鼠的肾脏,将其制作冰冻切片,厚度为5-7μm,丙酮固定。用直接免疫荧光法检测肾脏组织IgG的沉积情况。首先取出冰冻切片,在室温下干燥30min,然后再用5%的BSA封闭1小时,接着再用PBST冲洗3次,每次5min;将切片与Alexa Fluor 488缀合的山羊抗小鼠IgG(1:1000)在4℃下孵育过夜,然后第二天用PBST冲洗3次,每次5min洗去未结合的山羊抗小鼠IgG,将切片用4,6-二脒基-2-苯基吲哚(DAPI)染色10min,再用PBST冲洗,封片后在荧光倒置显微镜下观察。免疫荧光结果如图18所示,结果表明模型组肾脏肾小球以及系膜中具有大量的免疫复合物IgG的沉积,而伊马替尼组显著减轻了免疫复合物IgG的沉积。
⑩统计学处理:体重、尿蛋白等均采用graphpad prism软件进行数据处理,采用多样本均数比较的方差分析(One-WayANOVA)处理。
综上所述,化合物伊马替尼处理后,狼疮小鼠尿蛋白水平明显下降,外周血中尿素氮、肌酐以及抗dsDNA抗体水平液显著性降低,同时狼疮小鼠肾脏病理损害明显减轻、肾脏中炎症因子IL-6/MCP-1/TNF-α/TGF-β/IL-1β以及免疫复合物IgG沉积也同样减少。表明化合物伊马替尼在治疗狼疮性肾炎这类***性红斑狼疮中具有显著效果。
实施例9
化合物伊马替尼对小鼠银屑病模型的治疗效应
①选择试验动物:选择购买自北京维通利华公司的BALB/C小鼠,雌性,日龄48-56天;于普通环境下饲养,自由摄食饮水。
②药物制备:伊马替尼给药剂量均为50mg/kg,溶解于0.5%羧甲基纤维素钠的水溶液中。
③动物分组、造模和给药:小鼠按体重随机分为空白对照组(Control)、模型组(Model)、化合物组。用剃毛器去除背部毛发,露出2cm×3cm的皮肤区域。5%咪喹莫特(Imiquimod,IMQ)乳膏(局部剂量62.5mg)每天对右耳及背部给药,Toll样受体在银屑病发生和发展进程中起重要作用,咪喹莫特是Toll样受体激动剂,可用于银屑病造模。上午造模,下午给药,实验周期为7天。每天对小鼠进行称重、拍照、测量每只小鼠的右耳厚度以及观察小鼠右耳以及背部情况,为了对小鼠皮肤炎症的严重情况进行评分,基于临床牛皮癣面积和严重程度指数开发了客观评分***(Psoriasis Area and Severity Index;PASI),红肿和鳞屑从0到4独立评分(0:无症状;1:轻度;2:中度;3重度;4:极其严重)。结果如图19所示,与模型组耳厚度以及红肿程度相比,对照组以及给药组小鼠耳厚度以及红肿程度显著性降低。
④RT-PCR:总RNA用TRIZOL试剂提取,mRNA用Prime Scrip RT Master Mix转录成cDNA。RT-PCR分析测量TNF-α以及IL-1βmRNA水平,结果如图20所示,模型组TNF-α以及IL-1β水平高于对照组,伊马替尼组TNF-α以及IL-1β的mRNA水平显著低于模型组。
⑤HE染色:从咪喹莫特乳膏涂抹造模开始,每天对小鼠进行拍照并观察小鼠右耳及背部情况,每天测量每只小鼠的右耳厚度,耳厚度测量结果如图21所示。取小鼠背部及右耳皮肤标本浸泡于4%多聚甲醛中,石蜡包埋;用苏木精和伊红染色(HE);HE定量结果如图21所示,结果表明模型组的棘皮厚度大于对照组,且有显著性差异(P<0.05),化合物组棘皮厚度相对于模型组有显著降低。
⑥统计学处理:HE染色情况、右耳厚度等均采用graphpadprism软件进行数据处理,多组间采用单因素方差分析(One-WayANOVA)处理。
综上所述,伊马替尼处理后银屑病小鼠耳厚度、皮肤损伤程度以及皮肤中炎症因子TNF-α以及IL-1β的mRNA表达量均具有显著降低。表明化合物伊马替尼对银屑病具有显著的治疗效果。
Claims (10)
1.伊马替尼在制备VISTA激动剂方面的用途。
2.伊马替尼作为VISTA激动剂在制备预防或治疗由VISTA介导的疾病的药物中的用途。
3.根据权利要求2所述的应用,其特征在于,所述由VISTA介导的疾病为自身免疫性疾病。
4.伊马替尼在制备预防或治疗自身免疫性疾病的药物中的用途。
5.根据权利要求3或者4所述的用途,其特征在于,所述自身免疫性疾病包括红斑狼疮、银屑病、自身免疫性脑脊髓炎、自身免疫性肝炎或类风湿性关节炎。
6.根据权利要求3或者4所述的用途,其特征在于,所述自身免疫性疾病为红斑狼疮或者银屑病。
7.根据权利要求5所述的用途,其特征在于,所述红斑狼疮包括:皮肤型红斑狼疮、***性红斑狼疮或者药物诱导性红斑狼疮;所述银屑病为斑块状银屑病、泛发性脓疱型银屑病、红皮病型牛皮癣或银屑病关节炎。
8.一种药物组合物在制备预防或治疗自身免疫性疾病中的用途,所述药物组合物其含有伊马替尼作为活性成分和药学上可接受的载体。
9.根据权利要求8所述的用途,其特征在于,所述自身免疫性疾病包括红斑狼疮、银屑病、自身免疫性脑脊髓炎、自身免疫性肝炎或类风湿性关节炎。。
10.根据权利要求8所述的用途,其特征在于,所述的药物组合物的剂型优选是胶囊剂、散剂、片剂、颗粒剂、丸剂、注射剂、糖浆剂、口服液、吸入剂、软膏剂、栓剂或贴剂。
