HaCaT cells were seeded in 12-well plates at 37℃with 5% CO 2 Culturing in a maintenance medium for 24h, placing an insert cell-forming scratch model in each well of culture plate, culturing in the culture plate for 24h, setting a normal control group (serum-free DMEM medium) and a positive control group (EGF solution concentration of EGF is 10000 IU/mL), and observing the cell growth state at scratches of 16 hours, 24 hours and 48 hours in a test substance treatment group (120 μg/mL of bletilla striata extracted from different groups) under a microscope, and recording by photographing separately, as shown in FIG. 2, and using ImageJ software calculates the healing area. The healing rate calculation formula is as follows: healing rate = (initial scratch area-scratch area to be measured time)/initial scratch area. The test results are shown in table 3:

As can be seen from the data in table 3: compared with the extraction process adopted in the comparative example, even though the polysaccharide content level in the extract is equivalent, the bletilla striata extract prepared by adopting the specific extraction process has remarkably excellent efficacy of promoting the healing and repairing of the epidermal cells. Meanwhile, the technological parameters of the ultra-high pressure pretreatment obviously influence the effects.
The applicant states that the present invention is described by way of the above examples as a method for preparing a bletilla striata extract of the present invention, and products and applications thereof, but the present invention is not limited to the above examples, i.e. it is not meant that the present invention must be practiced in dependence upon the above examples. It should be apparent to those skilled in the art that any modification of the present invention, equivalent substitution of raw materials for the product of the present invention, addition of auxiliary components, selection of specific modes, etc., falls within the scope of the present invention and the scope of disclosure.
The preferred embodiments of the present invention have been described in detail above, but the present invention is not limited to the specific details of the above embodiments, and various simple modifications can be made to the technical solution of the present invention within the scope of the technical concept of the present invention, and all the simple modifications belong to the protection scope of the present invention.
In addition, the specific features described in the above embodiments may be combined in any suitable manner, and in order to avoid unnecessary repetition, various possible combinations are not described further.

Claims (8)

1. The preparation method of the bletilla striata extract is characterized by comprising the following steps of:
(1) Mixing rhizoma bletilla raw material with water, performing ultrahigh pressure treatment, and performing vacuum reduced pressure reflux extraction treatment on the treated rhizoma bletilla to obtain an extract; the pressure of the ultrahigh pressure treatment is 200-350 MPa, and the pressure maintaining time is 4-8 min;
(2) Filtering the extracting solution by using a filter cloth and performing vacuum filtration to obtain a filtrate;
(3) And freeze-drying the filtered solution to obtain the bletilla striata extract.
2. The method for preparing rhizoma bletillae extract according to claim 1, wherein the rhizoma bletillae raw material is rhizoma bletillae decoction pieces, and the feed liquid ratio of the rhizoma bletillae decoction pieces to water is 1 (10-20) g/mL.
3. The method for preparing rhizoma bletilla extract according to claim 1, wherein the vacuum degree of the vacuum reflux extraction treatment is 0.04-0.08 MPa, the heating temperature is 45-65 ℃, the extraction time is 30-60 min, and the number of times of extraction is 1-4.
4. The method for preparing rhizoma bletillae extract according to claim 1, wherein the filter cloth has a pore size of 150-250 mesh;
the decompression suction filtration uses a filter aid, wherein the filter aid comprises diatomite and/or activated carbon, and the dosage of the filter aid is 0.1-1% of the mass of the extracting solution.
5. The bletilla striata extract prepared by the preparation method according to any one of claims 1-4.
6. Use of the bletilla striata extract according to claim 5 for the preparation of cosmetics with skin moisturizing and/or skin repairing effects.
7. Use of the bletilla striata extract according to claim 5 for the preparation of aquaporin-3 expression promoters.
8. Use of the bletilla striata extract according to claim 5 for preparing a product having an effect of promoting healing of epidermal cell scratches.
