Abstract

本发明公开了一种给水管壁生物膜分级培养装置及其使用方法。该装置包括反应器桶体,反应器上盖,一级样片悬挂转盘、二级样片悬挂转盘、三级样片悬挂转盘、具孔轴、电机、固定栓柱、取样支架;所述的样片悬挂转盘可通过固定栓柱卡入样片悬挂转盘凹槽将其固定在具孔轴某一位置上,实现样片的分级取样;反应器桶体上部设置有两个进水口,下部设置有两个出水口;所述电机输出轴与具孔轴连接;所述电机下部固定于反应器上盖;所述电机通过速度控制器连接电源。本发明适用于给水管壁的大批量,多尺生物膜取样挂片的生物膜培养,具有样片悬挂转盘高度可调,组装简便,制造组装价格低等优点。

Description

一种给水管壁生物膜分级培养装置
技术领域
本发明涉及一种生物膜的培养装置及使用方法,特别是关于一种给水***管壁附生生物膜培养装置及其使用方法。
背景技术
城市给水***是将饮用水从处理厂送到用户端的关键基础设施,原水经过水处理厂相关处理工艺处理后,可以达到饮用水卫生标准,但处理后的水并不是绝对无菌的。进入管网的细菌可以利用残留在管网水中的少量有机物作为营养物质进行增殖,并且附着在给水管道管壁形成生物膜。生物膜脱落后会对管网水中细菌总数及细菌群落结构产生影响,加速余氯衰减,产生嗅和味,生物膜还会导致金属管网的腐蚀,增加管网水的色度,浊度等,并且生物膜还为致病菌提供稳定的增殖环境,在饮用水中会构成严重的健康风险。一般地,直接在给水***中进行生物膜取样分析较为困难,为保证实际的给水***的供水安全,常利用模拟反应器研究生物膜。当前使用比较广泛的如CDC(Centers for Disease Control)和BAR(Biofilm Annular Reactor)反应器。但是每次实验运行前生物膜取样挂片须在反应器内稳定一定时间;并且在不同反应器内部培养的生物膜初始状态也可能存在差异,影响平行实验结果;反应器一旦制作完成后,只能进行某一尺寸的生物膜挂片培养检测。因此,开发一套可悬挂多尺寸生物膜取样挂片,使管壁附生生物膜的快速培养及稳定的装置,对给水***内附生生物膜进行模拟培养是非常必要的。
发明内容
本发明的目的是提供一种给水管壁生物膜分级培养装置及其使用方法。
本发明的技术方案概述如下:一种给水管壁生物膜分级培养装置包括反应器桶体、反应器上盖、一级样片悬挂转盘、二级样片悬挂转盘、三级样片悬挂转盘、具孔轴,电机、固定栓柱、取样支架;所述反应器桶体上部设置有两个进水口,下部设置有两个出水口;所述样片悬挂转盘边缘装有螺柱,若干生物膜取样挂片打孔后固定在所述螺柱上;所述悬挂转盘中心孔直径大于具孔轴直径;所述悬挂转盘中心凹槽设置长度大于所述固定栓柱长度,宽度等于所述固定栓柱直径;所述电机输出轴与具孔轴连接;
所述电机下部固定于反应器上盖;所述电机通过所述速度控制器连接电源。
进一步的,所述进水口位置高于第一级样片悬挂转盘。
进一步的,所述反应器桶体、反应器上盖、固定栓柱均采用有机玻璃材质。
进一步的,取样支架为半弧形结构。
本发明提供的技术方案的有益效果是:
(1)本发明可以在同一水力条件下进行大批量,多尺寸生物膜取样挂片培养;
(2)本发明可以分级、分批次取出挂片,当一部分取样挂片取出时,另一部分取样挂片仍在水中,不对挂片上生物膜产生扰动。
(3)上进下出的供水方式可实现反应器内部水量的控制。
(4)材料易得,组装拆卸简便。

具体实施方式
为使本发明的目的、技术方案和优点更加清楚,下面对本发明实施方式作进一步地详细描述。
如图1所示,本发明所述的一种给水管壁生物膜分级培养装置包括反应器桶体1、所述电机7固定于反应器上盖2,所述具孔轴6与电机7的输出轴连接,所述反应器上盖2可盖于反应器桶体1上;
如图2所示,所述一级样片悬挂转盘3周边通过胶水固定螺柱14,所述螺柱14可以悬挂若干不同尺寸及不同材质的取样挂片;所述反应器上盖2与所述反应器桶体1分离后,所述具孔轴6从下至上***样片悬挂转盘中心孔12,并根据取样挂片尺寸将所述一级样片悬挂转盘3放置在具孔轴6合适位置;将所述固定栓柱15***具孔轴孔16;将所述一级样片悬挂转盘3沿所述具孔轴6下移,使固定栓柱15卡入所述固定凹槽13;同样的,依据所述一级样片悬挂转盘3的实施方式将二级样片悬挂转盘4和三级样片悬挂转盘5固定于具孔轴6上。将连接好的反应器上盖2、样片悬挂转盘3盖于反应器桶体1上;
所述的出水口10和出水口11通过止水阀关闭后,打开进水口8和进水口9,通过水管将自来水引入反应器桶体1中,当自来水液面完全淹没一级样片悬挂转盘3及悬挂的样片时,停止进水;接通所述电机7的速度控制器,调整实验所需的转速;
如图3所示,取样时,所述反应器上盖2抬起,所述取样支架17支撑于反应器上盖2和反应器桶体之间,取下一级样片悬挂转盘3上悬挂的样片时,保证二级样片悬挂转盘4和三级样片悬挂转盘5仍处于水液面以下。

1.一种给水管壁生物膜分级培养装置,它包括装置主体反应器桶体,反应器上盖,一级样片悬挂转盘、二级样片悬挂转盘、三级样片悬挂转盘、具孔轴、电机、固定栓柱、取样支架;所述一级样片悬挂转盘、二级样片悬挂转盘、三级样片悬挂转盘设置有中心孔和凹槽;所述具孔轴有直径与固定栓柱直径相同的通孔;所述电机固定于反应器上盖,所述具孔轴与电机的输出轴连接;样片悬挂转盘周边螺柱悬挂若干样片;所述具孔轴***样片悬挂转盘中心孔,所述固定栓柱***具孔轴孔并卡入所述固定凹槽;所述电机通过螺钉固定于反应器上盖,并连有速度控制器。
2.如权利要求1所述的一种给水管壁生物膜分级培养装置,其特征在于:所述的样片悬挂转盘可通过固定栓柱卡入样片悬挂转盘凹槽将其固定在具孔轴某一位置上。
3.如权利要求1所述的一种给水管壁生物膜分级培养装置,其特征在于:所述具孔轴可固定样片悬挂转盘,所述一级样片悬挂转盘高度低于进水口。
4.如权利要求1所述的一种给水管壁生物膜分级培养装置,其特征在于:所述的样片悬挂转盘边缘固定的螺柱可悬挂不同尺寸的生物膜取样挂片。
5.如权利要求1所述的一种给水管壁生物膜分级培养装置,其特征在于:所述具孔轴固定于电机输出轴,电机固定于反应器上盖。
6.如权利要求1所述的一种给水管壁生物膜分级培养装置,其特征在于:所述反应器桶体上部设置有两个进水口,下部设置有两个出水口。
