Test example 1
In order to further verify the effectiveness of the traditional Chinese medicine composition in treating atopic dermatitis, the following animal experiments are provided for illustration.
1 materials and methods
1.1 test animals and groups
Selecting 24 SPF-grade BALB/c mice, male mice, 6 weeks old mice and 18-22g of body mass, randomly dividing the mice into 4 groups, wherein each group comprises 6 mice, and the groups respectively comprise a blank group, a model group, a low-dose traditional Chinese medicine group and a high-dose traditional Chinese medicine group; wherein, the AD model is made from 18 mice of the model group, the low-dose traditional Chinese medicine and the high-dose traditional Chinese medicine, and the model making method adopts a DNCB external coating method to make the model (the concrete method refers to the establishment of a model of atopic dermatitis of BALB/c mice induced by sensitization of DNCB [ J ]. 1.2 herbs are dried and collected
The low-dose traditional Chinese medicine and the high-dose traditional Chinese medicine are administered to the decoction prepared in the example 5 for intragastric administration, wherein the low-dose traditional Chinese medicine is administered according to the body mass of 0.1mL/10g, and the high-dose traditional Chinese medicine is administered according to the body mass of 0.3mL/10 g; the model group and the blank group were gazed with 0.9% physiological saline. The administration is 1 time per day, and the administration is continuously performed by gavage for 3 weeks. After the last administration, the patient is sacrificed and the material is taken within 24h (local skin lesion and blood is taken after the eyeball is picked), the skin tissue is compared with pathological sections and is subjected to immunohistochemical staining, and the peripheral blood is taken and is subjected to enzyme linked immunosorbent assay (ELISA) to determine the content of IgE, IL-1 beta, IL-4 and IFN-gamma in the blood serum.
1.3 statistical analysis
Data analysis statistical tests were performed on all data using SPSS20.0 software One-way ANOVA with results expressed as mean ± standard deviation and as (P <0.05) significant differences.

2.1 histopathological changes in skin lesions
As can be seen from the histopathological results of the skin lesions of the mice, the blank group showed no hyperplasia of squamous epithelium and normal dermis (as shown in fig. 1A); the model group showed hyperkeratosis of the epidermis, hyperhypertrophy of the spinous layer, marked proliferation of the squamous epithelium, and infiltration of multiple inflammatory cells in the dermal layer (as shown in fig. 1B); compared with the model group, the low-dose and high-dose traditional Chinese medicines have the advantages that the low-dose traditional Chinese medicine composition and high-dose traditional Chinese medicine group has reduced acanthosis, squamous hyperplasia and inflammatory cell infiltration or no inflammatory cell infiltration (as shown in figures 1C and 1D); the high dose traditional Chinese medicine group was similar to the blank control group.
2.2 serological index Change
The serological inflammation index of the mouse is changed after 3 weeks of intervention after administration, LSD test shows that the expression levels of IgE, IL-1 beta and IL-4 of the model group are obviously increased compared with the low and high dose traditional Chinese medicines and the blank group, and the difference analysis has statistical significance (P is less than 0.05); compared with the blank group, the difference of the traditional Chinese medicine group has no statistical significance. Compared with the blank group and the high-dose traditional Chinese medicine group, the IFN-gamma expression level in the serum of the model group is increased, and the difference has statistical significance; the model group and the low-dose traditional Chinese medicine group have no statistical significance (P >0.05), and the results are detailed in table 1 and figure 2.

3 conclusion
According to the test results, the traditional Chinese medicine composition can obviously improve the skin injury degree of a dermatitis mouse, and obviously reduce inflammatory cell infiltration and squamous epithelial hyperplasia; the high-dose traditional Chinese medicine composition can obviously reduce the content of inflammatory factors in the blood serum of a dermatitis mouse, and can restore the content of the inflammatory factors to a normal level, and the results can fully show that the traditional Chinese medicine composition has a good treatment effect on the aspect of treating atopic dermatitis.
Test example 2
Observation of clinical efficacy of atopic dermatitis
1 general data
150 patients are outpatients, all patients are diagnosed as atopic dermatitis by a doctor of a dermatologist of the hospital, the diagnosis standard refers to Chinese diagnosis and treatment guidelines for atopic dermatitis (2020 edition), development and evolution of the diagnosis standard of atopic dermatitis, and development and evaluation of the diagnosis standard of atopic dermatitis, and all patients are randomly divided into three groups.
Control group 1: 32 men, 20 women, 16-55 years old, average age (38.62 + -3.97);
control group 2: 29 men and 19 women, the age is 20-47 years, and the average age is (31.54 +/-5.03);
treatment groups: 30 men and 20 women, the age is 18-53 years, and the average age (36.52 +/-6.21);
the general data of three groups of patients are compared, and the difference is not statistically significant (P >0.05) and is comparable.
Inclusion criteria were: the diagnosis was confirmed to be allergic dermatitis, skin pruritus and relapse. Exclusion criteria: (1) patients with other complications and contraindications; (2) those who do not take the drug according to the regulations; (3) patients with infectious skin disease were excluded.
2 method
Control group 1: levocetirizine hydrochloride tablets (Chongqing Huabang pharmaceutical Co., Ltd., national drug standard H20040249) are adopted, and the specification is 5 mg/tablet. The use method comprises the following steps: orally administered 1 time/1 tablet/d. The treatment course of the patient is 2 weeks.
Control group 2: the traditional Chinese medicine decoction for invigorating spleen, nourishing blood and dispelling wind is used for treatment, and the formula comprises the following medicines: 30g of tribulus terrestris, 30g of prepared fleece-flower root, 30g of astragalus root, 20g of divaricate saposhnikovia root, 15g of largehead atractylodes rhizome, 15g of Chinese angelica and 10g of centipede are decocted with water, 1 dose is taken every day in the morning and at night for 2 times, and the treatment lasts for 2 weeks.
Treatment groups: the decoction prepared in example 5 was used twice daily for 2 weeks.
3 determination of therapeutic Effect
And (3) healing: the skin lesions subside and the symptoms disappear;
the effect is shown: the skin lesion is faded by 60-90%, and the symptoms are obviously improved;
improvement: the skin lesion is faded by 20-59%, and the symptoms are improved;

According to the results of clinical efficacy observation of atopic dermatitis, the traditional Chinese medicine composition has a curative effect on the atopic dermatitis which is obviously superior to that of levocetirizine hydrochloride tablets and spleen-tonifying blood-nourishing wind-dispelling decoction, the curative rate can reach 68.0 percent, and the total effective rate can reach 88.0 percent. The traditional Chinese medicine composition has the effects of clearing heat and removing toxicity, dispelling wind and removing dampness, relieving itching and killing parasites, can realize higher cure rate of atopic dermatitis, reduce the recurrence of diseases, shorten the treatment period, play a role in treatment fundamentally and greatly relieve the pain of patients.
