Description

Traditional Chinese medicine composition for dispelling effects of alcohol and protecting liver as well as preparation method and application thereof
Technical Field
The invention relates to the field of traditional Chinese medicines and health-care foods, in particular to a traditional Chinese medicine composition for dispelling the effects of alcohol and protecting liver as well as a preparation method and application thereof.
Background
The liver is mainly used for organs of human metabolism, and toxic substances, toxins and metabolism and decomposition products of medicaments which are exotic and harmful wastes generated in the human metabolism process are detoxified in the liver. The liver is the largest reticuloendothelial phagocytic system, and can engulf, sequester and eliminate various antigens invading and endogenous, and meanwhile, almost all coagulation factors are produced by the liver, and the liver plays an important role in regulating the dynamic balance of two systems, namely human coagulation and anticoagulation.
The liver is a main organ for metabolizing alcohol, but the metabolism process of the liver causes irreparable damage to the liver, and long-term drinking of the liver can damage liver cell membranes and cause the liver cells to swell, necrose and the like. Alcohol is the first factor causing liver injury, long-term drinking causes adipogenesis of the liver, cannot be metabolized in liver cells, and is increasingly accumulated into alcoholic fatty liver; during the process of alcohol metabolism, the generated metabolites, ethanol and metabolic derivatives can cause inflammatory reaction to the liver, and the inflammation causes damage to liver cells and even necrosis, thereby leading to alcoholic hepatitis; if the damage to fatty liver and alcoholic hepatitis is not controlled in time, the damage can develop, then the damage can gradually develop into nodules, fibroplasia and liver fibrosis, and finally liver cirrhosis occurs, and finally liver cancer is formed.
The traditional Chinese medicine considers that alcoholism is caused by excessive drinking by external factors, and is caused by weakness of spleen and stomach, abnormal operation capability, alcoholic damp-heat and spleen and stomach imbalance by internal factors. The general treatment principle of the traditional Chinese medicine treatment method is that the traditional Chinese medicine treatment method treats both principal and secondary aspect of disease: in the early stage, alcohol dispelling and sweating and diuresis are the main treatment methods. The principle of "eliminating dampness by upper and lower parts" is proposed by Li Donghuan of one of four families of Jinyuan, which considers that "what is known as strong heat and toxic, smell is strong, and what is known as intangible, if injury occurs, the most wonderful method is to stop the heat and sweat, and secondly, the principle is to eliminate dampness by upper and lower parts, what is known as what is the wine disease? The middle stage disease is caused by the damage of spleen and stomach to liver and gallbladder, and this stage treatment is mainly to treat the disease and takes into account the healthy qi of the human body.
The prior anti-hangover medicine mainly comprises chemical medicines, traditional Chinese medicine preparations, health care products and the like. Although the chemical and the traditional Chinese medicine preparation take effect, the chemical and the traditional Chinese medicine preparation can generate great side effects on liver, kidney and the like, and the health care product produced by the raw materials with homology of medicine and food basically does not generate side effects on human bodies, and can realize quick alcohol effect under the condition of reasonable formula.
Disclosure of Invention
Aiming at the prior art, the invention aims to provide a traditional Chinese medicine composition for dispelling the effects of alcohol and protecting liver as well as a preparation method and application thereof. The composition has effects of relieving hangover and protecting liver.

Preferably, the traditional Chinese medicine extract bag comprises the following raw materials in parts by weight: 6-7 parts of cordyceps militaris, 13-14 parts of flos puerariae lobatae, 13-14 parts of semen hoveniae, 13-14 parts of chrysanthemum, 13-14 parts of silybum marianum, 9-11 parts of dark plum, 9-11 parts of mulberry leaf, 9-11 parts of dried orange peel and 9-11 parts of honeysuckle.
In the anti-alcoholic beverage, various raw material medicines contain active ingredients beneficial to anti-alcoholic, and after reasonable compatibility, the active ingredients produce a synergistic effect, so that the anti-alcoholic beverage can be used for rapidly and effectively dispelling the effects of alcohol. Wherein:
the Cordyceps militaris (Cordyceps militaris (L.ex Fr.) Link) belongs to Cordyceps genus of Clavipitaceae family, and also called Cordyceps militaris, cordyceps militaris. Cordyceps militaris contains Cordyceps polysaccharide, cordycepin, cordycepic acid, proteins, amino acids, superoxide dismutase, etc. Has uric acid reducing, tumor inhibiting, antiviral, antioxidant, liver function improving, immunity regulating, sexual function protecting, and radioprotective effects.
The flos Puerariae Lobatae is dried flower of Pueraria lobata (willd.) Ohwi of Leguminosae, is rich in volatile oil, and contains quercetin, ge Huadai, triterpene saponin, beta-sitosterol, flos Puerariae Lobatae isoflavone, etc. The Chinese medicine considers that the flos puerariae lobatae has the functions of dispelling the effects of alcohol, activating spleen, clearing heat, detoxicating, protecting liver, nourishing skin, tonifying kidney and the like, and is used for treating the symptoms of excessive drinking, headache, dizziness, polydipsia, chest and diaphragm fullness, acid regurgitation and other gastric qi injuries. Modern researches suggest that saponin and isoflavone contained in flos Puerariae Lobatae have oxidation-reduction effect, and can accelerate oxidation of alcohol to make alcohol lose toxicity, shrink and protect gastrointestinal mucosa, slow absorption of alcohol, and prevent alcohol from rapidly entering blood circulation, thereby improving metabolism abnormality caused by alcohol. Has effects in relieving hangover, activating spleen, clearing away heat and toxic materials, protecting liver, caring skin, and invigorating kidney, and can be used for treating excessive drinking, headache, dizziness, polydipsia, fullness in chest and diaphragm, and gastric qi injury such as emesis.
The semen Hoveniae is fruit of Hovenia dulcis Thunb (Hovenia acerba lindi.) of Hovenia of Rhamnaceae, and is rich in glucose, potassium nitrate and potassium malate; the seeds contain loline, hovenin C, D, G, G' and H. In traditional Chinese medicine, the fruits and seeds can be used as medicines, and have the effects of clearing heat, promoting urination and dispelling alcohol toxicity; the glucose and organic acid contained in the beverage can not only expand blood volume of human body, but also relieve alcoholism, and has the effects of sobering up and tranquillizing. Modern researches have found that hovenia dulcis thunb can significantly reduce the concentration of ethanol in blood and promote the removal of ethanol.

The above-mentioned mouse sobering-up experiment refers to an anti-hangover beverage anti-hangover effect experiment in Chinese patent document CN108271963A (application No. 201810365814.3).
As can be seen from the above table, there was a significant difference in sober-up in mice compared to the control group. The anti-alcoholic beverage can shorten the drunk time of mice and has better drunk effect. Meanwhile, the effect of rapidly dispelling the effects of alcohol can be exerted under the synergistic effect of the principal drug, the ministerial drug, the adjuvant drug and the conductant drug.
Test example 2:
the beverage prepared by adding water to the anti-hangover and liver-protecting compositions obtained in example 1, comparative example 2, comparative example 3 and comparative example 4 was subjected to a mouse anti-hangover test.
60 mice (half of each female and male mice, each group has mass error of not more than 10 g) are taken, and the mice are fasted and forbidden late before the experiment. Mice were randomly divided into 6 groups of 10 mice each, wherein the control group was filled with 0.3ml of physiological saline per day, 0.3ml of white spirit after 30min, and the other 5 groups were each filled with 0.3ml of the compositions of example 1 and comparative examples 1 to 4, and 0.3ml of white spirit after 30 min. Each group of mice was continuously perfused with stomach for 5 weeks. After the last day of gastric lavage is finished for 30min, separating serum from blood of the small ball eyeballs, and measuring the activity of ALT and AST in the serum of each group of mice according to the specification of the glutamic pyruvic transaminase (ALT) test kit and the dark oxaloacetic transaminase (AST) test kit by using a spectrophotometer. The data are shown in table 1.
After eyeball blood is collected from each group of mice, dissecting, taking out liver, accurately weighing the liver mass, removing a proper amount of liver tissue, taking 0.9% physiological saline as a homogenate medium, preparing 10% liver homogenate, centrifuging, taking supernatant, and detecting Glutamic Pyruvic Transaminase (GPT) for later use.
(the above mouse experiment refers to the functional test in Chinese patent document CN108271963A (application No. 202011407436.4)
Liver index = liver mass/body weight x 100%.
The level of GPT in the liver homogenate of mice was determined according to the GPT kit instructions and the data are shown in table 2.

Comparative example 4 6.61±1.15 176.82±1.91
Control group 7.03±1.02 186.13±4.58
As can be seen from the data in Table 1, both the products of example 1 and comparative examples 1-4 effectively reduced ALT and AST, but it is evident that the magnitude of ALT and AST reduction in example 1 is greater than that in comparative examples 1-4, mainly because the composition of the present invention has various antioxidant components, has excellent antioxidant ability, can raise superoxide dismutase activity in vivo, scavenge a large amount of oxygen free radicals generated by tissues and cells after alcoholism, reduce damage to cells, protect cell membranes from stability, and promote decomposition and metabolism of alcohol.
According to the data in Table 2, the anti-alcoholic liver-protecting drink can obviously reduce the serum glutamic-pyruvic transaminase and glutamic-oxaloacetic transaminase levels and repair the pathological damage of the liver.
Meanwhile, the effect of rapidly protecting liver can be achieved under the synergistic effect of the principal drug, the ministerial drug, the adjuvant drug and the conductant drug.
Test example 3:
the anti-hangover and liver-protecting composition prepared in example 1 was subjected to a human efficacy test.
The efficacy of the anti-hangover and liver-protecting beverage in example 1 was observed by taking 2 bags of the beverage after drinking, which was frequently taken by a large number of drunk volunteers.
