Abstract
The invention discloses a traditional Chinese medicine composition for dispelling the effects of alcohol and protecting liver, and a preparation method and application thereof, and relates to the fields of traditional Chinese medicines and health-care foods. The composition of the invention comprises a traditional Chinese medicine extract, vitamin B, vitamin C, erythritol and sucralose, wherein the traditional Chinese medicine extract comprises Cordyceps militaris, flos puerariae lobatae, semen hoveniae, chrysanthemum, silybum marianum, dark plum, mulberry leaf, dried orange peel and honeysuckle. The invention is prepared by reasonably proportioning the materials with homology of medicine and food, and the composition has the functions of dispelling the effects of alcohol and protecting liver through the synergistic effect of the components.

Technical Field
The invention relates to the field of traditional Chinese medicines and health-care foods, in particular to a traditional Chinese medicine composition for dispelling the effects of alcohol and protecting liver as well as a preparation method and application thereof.
Background
The liver is mainly used for organs of human metabolism, and toxic substances, toxins and metabolism and decomposition products of medicaments which are exotic and harmful wastes generated in the human metabolism process are detoxified in the liver. The chrysanthemum has the effects of dispelling wind, clearing heat, improving eyesight and detoxifying, and is mainly used for treating headache, dizziness, conjunctival congestion, dysphoria with smothery sensation in the chest, furuncle, toxic swelling and the like.
The silybum marianum is a silybum marianum plant of the Compositae, and can play a role in protecting liver, resisting oxidization, promoting regeneration of liver cells and the like, and is a flavonoid compound extracted from silybum marianum seeds, namely silymarin. The main components of the silymarin comprise silybin, silydianin and the like, wherein the silybin has the highest biological activity and the strongest liver protection effect. According to the research, the silybin has the effects of protecting and stabilizing liver cell membranes, promoting liver metabolism, resisting oxidization, diminishing inflammation, reducing blood fat and the like, so that the liver protecting effect can be achieved. The silybin has an antioxidant effect, and mainly protects liver cell membranes through lipid peroxidation. Its lipid-lowering effect is mainly due to the fact that silybin can lower triglyceride content in liver and serum total cholesterol level.

The honeysuckle has the effects of clearing heat and detoxicating, resisting inflammation, tonifying deficiency and treating wind, and has certain curative effect on dizziness, dry mouth, thirsty and other symptoms.
In the anti-alcoholic liver-protecting beverage, all raw materials are substances with homology of medicine and food, wherein cordyceps militaris plays a main role as a monarch drug, kudzuvine flower and hovenia dulcis seed play an auxiliary role as ministerial drugs, chrysanthemum, silybum marianum and dark plum play an auxiliary role as adjuvant drugs, and mulberry leaf, dried orange peel and honeysuckle play an auxiliary role as messenger drugs. The above-mentioned mouse sobering-up experiment refers to an anti-hangover beverage anti-hangover effect experiment in Chinese patent document CN108271963A (application No. 201810365814.3).
As can be seen from the above table, there was a significant difference in sober-up in mice compared to the control group. The anti-alcoholic beverage can shorten the drunk time of mice and has better drunk effect. Meanwhile, the effect of rapidly dispelling the effects of alcohol can be exerted under the synergistic effect of the principal drug, the ministerial drug, the adjuvant drug and the conductant drug.
Test example 2:

Mice were randomly divided into 6 groups of 10 mice each, wherein the control group was filled with 0.3ml of physiological saline per day, 0.3ml of white spirit after 30min, and the other 5 groups were each filled with 0.3ml of the compositions of example 1 and comparative examples 1 to 4, and 0.3ml of white spirit after 30 min. Each group of mice was continuously perfused with stomach for 5 weeks. After the last day of gastric lavage is finished for 30min, separating serum from blood of the small ball eyeballs, and measuring the activity of ALT and AST in the serum of each group of mice according to the specification of the glutamic pyruvic transaminase (ALT) test kit and the dark oxaloacetic transaminase (AST) test kit by using a spectrophotometer. The data are shown in table 1.
After eyeball blood is collected from each group of mice, dissecting, taking out liver, accurately weighing the liver mass, removing a proper amount of liver tissue, taking 0.9% physiological saline as a homogenate medium, preparing 10% liver homogenate, centrifuging, taking supernatant, and detecting Glutamic Pyruvic Transaminase (GPT) for later use.

Liver index = liver mass/body weight x 100%.
The level of GPT in the liver homogenate of mice was determined according to the GPT kit instructions and the data are shown in table 2.

As can be seen from the data in Table 1, both the products of example 1 and comparative examples 1-4 effectively reduced ALT and AST, but it is evident that the magnitude of ALT and AST reduction in example 1 is greater than that in comparative examples 1-4, mainly because the composition of the present invention has various antioxidant components, has excellent antioxidant ability, can raise superoxide dismutase activity in vivo, scavenge a large amount of oxygen free radicals generated by tissues and cells after alcoholism, reduce damage to cells, protect cell membranes from stability, and promote decomposition and metabolism of alcohol.
According to the data in Table 2, the anti-alcoholic liver-protecting drink can obviously reduce the serum glutamic-pyruvic transaminase and glutamic-oxaloacetic transaminase levels and repair the pathological damage of the liver.
Meanwhile, the effect of rapidly protecting liver can be achieved under the synergistic effect of the principal drug, the ministerial drug, the adjuvant drug and the conductant drug.
Test example 3:
the anti-hangover and liver-protecting composition prepared in example 1 was subjected to a human efficacy test.
The efficacy of the anti-hangover and liver-protecting beverage in example 1 was observed by taking 2 bags of the beverage after drinking, which was frequently taken by a large number of drunk volunteers.

the traditional Chinese medicine extract is prepared from the following raw materials in parts by weight: 5 to 8 parts of cordyceps militaris, 12 to 15 parts of flos puerariae lobatae, 12 to 15 parts of semen hoveniae, 12 to 15 parts of chrysanthemum, 12 to 15 parts of silybum marianum, 8 to 12 parts of dark plum, 8 to 12 parts of mulberry leaf, 8 to 12 parts of dried orange peel and 8 to 12 parts of honeysuckle;
the vitamin B consists of the following raw materials in parts by weight: 0.001-0.002 parts of vitamin B1, 0.001-0.002 parts of vitamin B2 and 0.001-0.002 parts of vitamin B6;
the composition for dispelling the effects of alcohol and protecting liver is prepared by the following method:
(1) Mixing Cordyceps militaris, flos Puerariae Lobatae, semen Hoveniae, flos Chrysanthemi, herba Silybi Mariani, mume fructus, folium Mori, pericarpium Citri Tangerinae and flos Lonicerae at a certain weight ratio, soaking in purified water, heating to obtain extractive solution, centrifuging to obtain centrifugate, vacuum-compressing the centrifugate to obtain extract, vacuum-drying, pulverizing, and sieving to obtain Chinese medicinal extract;
(2) Mixing the Chinese medicinal extract, vitamin B1, vitamin B2, vitamin B6, vitamin C, erythritol and sucralose according to weight ratio.
