Abstract
本发明提供一种可即时分离的缓释微针贴片及其制备方法。该微针贴片包括基板层和由微针组成的微针阵列,并在两者之间添加一可溶性聚合物基质作为连接层,所述连接层的主要成分为透明质酸钠。该聚合物基质溶解速率快、可实现刺入皮肤浅层进行给药。敷贴后即时去除基板层,仅留下载药针体在皮肤毛囊周围发挥缓释作用,在有效地降低患者的用药频率下增加治疗效率,并保持用药者的良好形象。

技术领域
本发明属于生物医用材料领域,更具体地,涉及一种可即时分离的微针贴片及其制备方法。
背景技术
雄激素源性脱发(AGA)是临床上最常见的进行性毛发生长障碍。雄激素源性脱发的病因是多因素和多基因的,包括激素失衡和头皮血流不足。然而,雄激素依赖是男性雄激素源性脱发的主要特征。
目前用于雄激素源性脱发治疗的方案繁多,包括物理治疗的中医针灸和低频激光治疗、手术治疗植发和药物治疗如米诺地尔和非那雄胺。但是现有的治疗方案均存在一定的弊端,如针灸疗法疼痛感强烈、植发价格昂贵、疗程漫长,并且均需要专业医疗人员进行操作。目前FDA批准用于治疗雄激素源性脱发的药物是米诺地尔和非那雄胺。非那雄胺能够有效阻断睾酮向二氢睾酮的转化,用于男性雄激素源性脱发患者的治疗,但为维持治疗效果,需要每天长期使用,且副作用比较难以接受。米诺地尔已有的剂型为外用涂抹溶液,其代谢产物米诺地尔硫酸盐在治疗脱发中起主要作用。局部涂抹米诺地尔溶液可能的不良反应包括接触性皮炎、皮肤干燥瘙痒和无选择性多毛症等,但发生率低,且产生皮肤刺激主要与溶剂丙二醇有关。另外,一般需要3~6个月连续每天用药才能观察到一定的临床效果,且因给药中断易发生毛发脱落现象,米诺地尔溶液频繁用药极大地影响了患者依从性。
针对目前雄激素源性脱发治疗中涂抹米诺地尔外用制剂递送药物效率低、给药繁琐导致患者依从性低的问题,微针贴片是一种新型的治疗方法。
根据目前检索到的中国专利文件(参见CN111544573A,CN112153957A,CN113786394A,CN113750250A),已有的用于治疗脱发的微针贴片在使用时,需按压一定时间(5-10分钟)并且贴敷至微针针尖溶解后取下。而本发明在基板层与微针阵列间添加一可溶性聚合物基质作为连接层,当本微针贴片贴于脱发皮肤上后,用指部轻微按压不足一分钟,即可去除基板层,实现了即时分离的效果,仅留下载药针体在皮肤毛囊周围发挥缓释作用。
本发明在有效降低患者用药频率的前提下,增加了治疗效率,并保持用药者的良好形象。另外,本发明采用可生物降解材料与促毛发生长药物共溶于溶剂的形式,将该药物荷载于微针针体中。因此微针中的药物分布较集中,主要集中于微针针体尖端部。与通过药物增溶制备的可溶性微针相比,本发明实现了微针载药量高的效果。
发明内容
针对目前雄激素源性脱发治疗中涂抹米诺地尔外用制剂递送药物效率低,给药繁琐导致患者依从性低的问题,本发明提供一种可即时分离的缓释微针贴片及其制备方法,以解决现有技术中存在的缺陷。
本发明的一个发明点为提供一种可即时分离的微针贴片,其特征在于:包括基板层和由微针组成的微针阵列,并在两者之间添加一可溶性聚合物基质作为连接层,所述连接层的主要成分为透明质酸钠。
可选地,微针基板层采用的材料为聚乙烯吡咯烷酮(PVP),溶液浓度为0.1-0.2g/mL。
可选地,所述微针由可生物降解的材料和促进毛发生长的物质组成,所述可生物降解的材料和所述促进毛发生长的物质以共聚物的形式共溶于溶剂中。
可选地,所述可生物降解的材料是聚乳酸-羟基乙酸共聚物(PLGA),所述促进毛发生长物质是米诺地尔,所述溶剂是二甘醇二甲醚。
可选地,根据共溶性能选择PLGA,米诺地尔与二甘醇二甲醚的优化配比,其中PLGA与米诺地尔的质量比为6:2-6:4,PLGA和米诺地尔的固体质量与二甘醇二甲醚液体质量比为1:10-1:12。
可选地,所述米诺地尔的含量为每片微针贴片中含有1.695~2.001mg。
本发明的另一个发明点在于提供一种制备上述可即时分离的微针贴片的方法,该方法包括以下步骤:以生物可降解材料和难溶性药物为原料,采用微针模具并利用干燥法制备微针针体,制得的针体集中填充于微针模具腔体尖端部分;继续在装载有针体的模具中添加透明质酸钠溶液,形成连接于针体下部的连接层;采用PVP材料制作空白基板,得到最终的微针贴片。
可选地,制作微针的模具由聚二甲基硅氧烷材料(PDMS)材料制成,模具底座设有一橡胶圈以防止制备微针过程中药液的流失,便于药液回收,节省制备成本。
可选地,制作微针针体的过程如下:形成促毛发生长物质与可生物降解材料的共溶物,以所述生物可降解材料和难溶性药物为原料,将两者粉末加入溶剂后搅拌,搅拌溶解后制得模液;将模液加入模具后离心,刮去模具表面多余的模液并回收,滴加溶剂后静置离心,将模液冲洗到微针针体腔尖端,反复冲洗后放置于真空干燥箱中,干燥并去除溶剂。
可选地,添加连接层的过程如下:将透明质酸钠加入纯水中,室温搅拌后取透明质酸钠水溶液加入已经装载有制备好的微针针体的模具中,离心干燥并得到具有连接层的完整针体部分。
可选地,制作空白基板层的过程如下:将PVP加入纯水中,室温搅拌后将PVP溶液加入带有微针阵列的模具上,离心干燥并得到具有基板层的完整微针贴片。
本发明的另一个发明点在于,使用上述可即时分离的微针贴片或利用上述制备方法制得的微针贴片在治疗脱发和/或促进毛发生长方面的应用。
综上所述,本发明微针基板层和针体的连接层采用的材料为透明质酸钠,在微针刺入皮肤后,接触皮肤组织间液就能够快速溶解,给药后即时去除基板层,仅留下载药针体在皮肤毛囊周围发挥缓释作用;选用快速溶解的聚合物材料PVP作为微针基板层制备材料,PVP能够提供微针阵列支撑能力,所得微针机械强度合适,让载药微针阵列能够刺入皮肤浅层发挥药效;选用PLGA共聚物作为载药针体材料,PLGA共聚物通过与皮肤组织间液接触缓慢降解,释放其中的药物米诺地尔,药物完全释放所需时间久,达到缓释效果。
因此,本发明在实现给药时基板层即时分离的同时,利用多种因素控制微针的机械强度保证了其透皮效果,结合微针的长度和底部直径,可以将药物递送直达毛囊附近,提高药物靶向性以及透皮给药的治疗效果。

具体实施方式
为了让基板层与针体连接层能够断裂,基板层与针体之间添加溶解速率快的可溶性聚合物基质作为连接层,实现给药后即时去除基板层,仅留下载药针体在皮肤毛囊周围发挥缓释作用,连接层采用的材料为透明质酸钠。本发明中采用的透明质酸钠水溶液浓度为0.2-0.8g/mL,优选为0.04g/mL。透明质酸钠分子量为200-400kDa,优选为340kDa。通过采取该优选的透明质酸钠水溶液浓度和透明质酸钠分子量,在微针刺入皮肤后,该连接层接触皮肤组织间液就能够快速溶解。
本发明的微针贴片中,可生物降解材料是PLGA共聚物,促进毛发生长物质为难溶性的米诺地尔,采用的溶剂为二甘醇二甲醚。PLGA共聚物作为制备本发明的载药针体材料,其通过与皮肤组织间液接触缓慢降解,释放其中的药物米诺地尔。优选地,PLGA共聚物分子量为10kDa,乳酸单体和羟基乙酸单体的比例为50:50。根据共溶特点选择优化的PLGA共聚物、米诺地尔和溶剂配比,制备微针时溶剂挥发,PLGA共聚物和米诺地尔填充进模具微孔腔体中,为了制备中溶剂更快挥发,PLGA共聚物与米诺地尔的质量比为6:2-6:4,PLGA共聚物和米诺地尔的固体质量与溶剂液体质量比为1:10-1:12。优选地,PLGA共聚物与米诺地尔的质量比为6:4,PLGA共聚物和米诺地尔的固体质量与溶剂液体质量比为1:12,三者共溶可以稳定存在。
米诺地尔主要的药效学发挥作用的位置处于毛囊,毛囊从皮下组织向上延伸至真皮层,而人体的皮肤中神经末梢主要分布于真皮层,为了避免触及神经产生痛感,同时保障药物递送效率和发挥药效,需要对微针形态、长度以及阵列分布等进行合理设计。所述的微针针体呈圆锥形,高度为850μm,针体底部直径为350μm,尖端直径小于10μm,相邻微针针尖间距为600μm,透皮递送药物效果佳。微针阵列采用10*10方形阵列。微针阵列的三维尺寸和采用的微针数量可以依据给药部位和所需剂量进行调整以达到最佳递送药物的目的。
微针阵列中药物分布较集中,主要偏针体尖部,与通过药物增溶法制备的可溶性微针相比,本发明中药物集中分布的聚合物微针载药量高。作为优选,米诺地尔在所述微针生成的微针阵列中的量为每片微针中含量为1.695~2.001mg。
微针模具由柔性PDMS制成,保证制备出的微针能够完整地脱模,和具有足够的力学性能。
制备出的微针利用力传感器来检测其能够承受的外力即微针给药时刺入皮肤的力,以此来检测微针是否能顺利地刺入皮肤、实施给药。
模具底座为直径22mm的圆形PDMS材料,底座有一圈高2mm的橡胶圈圈用以防止制备微针过程中药液的流失,节省成本。
微针基板层采用的材料为PVP,厚度约为0.5mm。PVP的分子量为44kDa,在高浓度时模液粘稠、不便于操作,且所得微针基板层脆性太强,在剥离和给药时容易碎裂,本发明中采用的PVP用纯水溶解,溶液浓度为0.1-0.2g/mL,所得微针机械强度合适,易于协助支撑微针刺入皮肤。
微针贴片可通过人工指部按压或机械轻敲的方式,使得针体刺透皮肤并实现释药。所述药物米诺地尔荷载在微针针体内,基底无需荷载药物,所需原料药物减少,降低了成本及提高药品的生物利用度。
1.一种荷载米诺地尔的微针贴片的制备
1)制作微针针体
在40mg PLGA和26.7mg粉末中加入760mg二甘醇二甲醚,以300rpm,室温25℃±2℃下搅拌4h,再加入纯水继续搅拌1h。取50μL模液加入洗净烘干的模具中,40℃,3500rpm离心20min,随后补充50μL模液,40℃,3500rpm离心20min。用盖玻片刮去模具表面多余的模液并回收,滴加50μL二甘醇二甲醚放置5min,再离心40℃,3500rpm,20min使针体模液中溶剂挥发,而含药聚合物充分填充于模具,再用溶剂二甘醇二甲醚洗涤离心,重复三次,将含药聚合物集中紧密填充于针体尖端。离心过程中挥发溶剂的方式让含药聚合物的填充量最大化,米诺地尔负载的含量也最大化。离心填充后,将微针模具放于真空干燥箱中,真空气压为0.08MPa,45℃下干燥12小时去除溶剂。
2)添加连接层:
400mg透明质酸钠加入10mL纯水中,300rpm室温搅拌7h,取50μL透明质酸钠溶液加入第一步中制得微针针体的模具中(由于溶剂被去除,第一步制得微针针体的模具有一部分是未填充状态),离心25℃,3500rpm,15min,35℃下干燥2h。
3)制作空白基板层
500mg PVP加入5mL纯水中,400rpm室温搅拌7h,取200μL PVP溶液加入带有微针阵列的模具上,离心15℃,3500rpm,15min,烘箱35℃下干燥7h,剥离出微针贴片,所述基底平面优选尺寸为1*1cm,基底与微针阵列垂直,使按压时方便可行。
4)形成微针贴片
得到的微针贴片贴敷于医用胶带上,封装备用。
米诺地尔微针制备流程如图1,负载米诺地尔的微针体显微镜图像在显微镜下成像如图2。
2.微针贴片的使用方法:
微针阵列和其基底由透气防过敏的软性医用胶布支撑,使得微针能够贴敷于皮肤表面。
将合适尺寸和形状的荷载米诺地尔的聚合物微针贴片贴于脱发皮肤上,用指部按压使微针阵列刺入皮肤,当微针针体与基板层连接处溶解后,快速去除基板层,PLGA共聚物降解释放米诺地尔,以到达治疗雄激素源性脱发的目的。
实施例1
1)实验目的:
载药微针和空白微针的力学性能检测
2)实验操作:
使用推拉力计与数显标尺配套设备测试负载米诺地尔的PLGA共聚物聚合物、以及空白PLGA共聚物的微针机械特性。
3)实验结果:
结果如图3所示,载药和空白聚合物微针的机械性能都足以刺入并穿透皮肤。
实施例2:
1)实验目的:
载药微针含药量均一性检测
2)实验操作:
取同一批次载米诺地尔微针贴片5片,将米诺地尔微针贴片在乙腈(含0.1%甲酸)中充分溶解,再加入甲醇析出PLGA共聚物,离心去除PLGA共聚物,吸取上清溶液烘干,再溶解于乙腈(含0.1%甲酸)中制得测试样本溶液,将所得样本进行HPLC分析。并与标品溶液进行比较,计算每片微针载药量。
HPLC条件如下:色谱柱为Agilent ZORBAX SB-C8;流动相乙腈(含0.1%甲酸)-水(80:20);体积流量为1mL/min;检测波长为281nm;柱温35℃;进样量10μL。
3)实验结果:
图4是负载米诺地尔的微针体显微镜图像,该结果表明,米诺地尔的含量范围为1.695~2.001mg/每片微针,且均一性较好。
实施例3:
1)实验目的:
绘制米诺地尔体外释药曲线
2)实验操作:
在装有1L释放介质的锥形瓶中放置一片微针贴片,37℃的水浴,摇床以80rpm/min转动;在预定的时间点0、1、3、6、9、12、15、18、21、24、27、30天收集释放介质溶液1mL,并用等量的PBST释放介质溶液替换。
PBST释放解质溶液的配置如下:137mM NaCl,2.68mM KCl,10.14mM Na2HPO4,1.76mM KH2PO4,和0.02%(w/v)的Tween 80,以0、15、20、25%(v/v)的配比加入乙醇。
液质联用分析释放介质中米诺地尔的含量,并与标品溶液进行比较,计算累计释放药物含量。其中6500+LC-MS/MS***检测条件如下:色谱柱Agilent ZorbaxEclipse Plus C18(2.1*100mm,3.5μm);流动相:以0.2%甲酸水溶液为流动相A,以含0.2%甲酸的甲醇为流动相B,柱温30℃,流速0.3ml/min,梯度洗脱程序:0-2min:5%-25%B,2-4min:25%-55%B,4-11min:55%-80%B,11-18min:80%-90%B。以三重四极杆串联质谱仪检测;电喷雾离子源(ESI),扫描方式:正离子扫描,检测方式:核磁共振功能成像MRI。
3)实验结果:
米诺地尔微针体外释药曲线如图5,该图是显示37℃下载含乙醇的磷酸盐缓冲盐水中米诺地尔从本发明的微针贴片中随时间(0至96小时)累积释放的图表。该结果表明,能实现持续给药,药物完释放所需时间久。
实施例4
1)实验目的:
验证米诺地尔微针贴片药效的治疗效果
2)实验材料:
SPF级雄性C57BL/6J小鼠购自江苏南京集萃药康公司;
米诺地尔(纯度98%)天津希恩思生化科技有限公司;
医用级HA-EP120-40中低分子透明质酸钠购自华熙福瑞达生物医药有限公司;
PLGA共聚物购自上海麦克林生化科技有限公司。

小鼠雄激素源性脱发模型的制备方法:在小鼠背部区域用脱毛膏脱毛,脱毛区域面积约为2*2cm,每天在脱毛区域涂抹8mg/mL睾酮溶液,涂抹剂量为0.1mL。建立小鼠雄激素源性模型,仅脱毛的健康组小鼠作为对照。
整个造模周期为28天,处理方案如图6。
观察给药后小鼠发毛生长情况的变化,对结果的统计学分析应用Graph PadPrism软件,包括t检验以及Two-way ANOVA。实验平行数据至少由3个不同个体所得。*代表P值小于等于0.05,**代表P值小于等于0.01,***代表P值小于等于0.001。
5)实验结果:
图7A结果表明,健康组小鼠在脱毛后第9天出现明显黑色素沉积,第16天脱毛区域的毛发已完全生长呈正常毛发状态;模型组直至第28天仅出现一些黑色素沉积,未生长出毛发;米诺地尔酊剂组,在第28天长出毛发但并未完全覆盖脱毛区域;空白微针组仅长出一部分毛发;载药微针组在第28天长出毛发并几乎完全覆盖脱毛区域。
图7B是通过在造模以及进行不同处理治疗方案时统计各组小鼠毛发生长转变情况,由图可以看出直至第21天,模型组毛发一直处于休止期,健康组毛发在第12天已明显生长,米诺地尔微针组毛发生长迟于健康组,但与米诺地尔酊剂组相比较早。
图7C不同处理方案14天后,H&E染色对照的结果与图7A一致,负载米诺地尔的微针贴片处理的雄激素源性脱发小鼠毛囊大多数已迁移至真皮下层,说明毛囊处于生长后期,相较于米诺地尔酊剂组毛囊周期更加靠后,说明载药微针能够更加快速治疗毛囊微型化症状。
实施例5
1)实验目的:
通过测试可溶性微针的机械力度,选取最优透明质酸钠分子量
2)实验操作:
采取不同分子量的透明质酸钠制备可溶性微针,测试对应可溶性微针的机械力度。
3)实验结果:
结果如图8所示,该图为微针的力-位移曲线图,横坐标的距离表示使用力传感器往下压微针时,该微针在力传感器的作用下,被下压的高度。
不同分子量透明质酸钠对应的可溶性微针的机械力度表明,低分子量透明质酸钠微针机械力度不足以刺入皮肤,高分子量透明质酸钠刺入能力足够但是溶液粘度过大,制备过程难以操作,因此本发明透明质酸钠分子量优选为340kDa。
实施例6
1)实验目的:
采取四种不同浓度的透明质酸钠水溶液,测试每种浓度下微针所能承受的最大外力以及实现即时分离效果所需的时间
2)实验操作:
采用340kDa的透明质酸钠配置四种不同浓度的透明质酸钠水溶液,分别为0.02g/mL,0.04g/mL,0.06g/mL和0.08g/mL;利用力传感器,检测制备出的微针所能够承受的最大外力,即微针给药时刺入皮肤的力,以此来检测微针是否能顺利地刺入皮肤、实施给药;同时利用秒表记录每种浓度透明质酸钠水溶液制得微针的即时分离效果。

当透明质酸钠浓度为0.02g/mL时,每根微针所能承受的最大外力为0.08N,微针机械力度小,该力度不足以使微针完全刺入皮肤,针体刺入皮肤成功率低。而当透明质酸钠溶液浓度为0.04g/mL,每根微针所能承受的最大外力增至0.28N,即时分离时间只增加三秒。因此采用的透明质酸钠溶液浓度为0.04g/mL,即时分离效果最好,又能保证微针能刺破皮肤。
实施例7
1)实验目的:
根据共溶特点选择优化的PLGA共聚物、米诺地尔和溶剂二甘醇二甲醚的配比。
2)实验结果:
PLGA共聚物与米诺地尔的质量比为6:2和6:4时,且PLGA共聚物和米诺地尔的固体质量与溶剂液体质量比为1:12时,三者共溶可以稳定存在。
本发明在基板层与微针阵列间添加由透明质酸钠制成的可溶性聚合物基质作为连接层,当本微针贴片贴于脱发皮肤上后,用指部轻微按压不足一分钟,即可去除基板层,实现即时分离的效果,仅留下载药针体在皮肤毛囊周围发挥药物缓释作用。而目前已知的微针贴片,均需按压一定时间(5-10分钟)至微针针尖溶解后方可取下。因此,本发明大大缩短了按压的过程,方便了使用者。另外,本发明提供的微针贴片每次使用可以达到72小时左右的有效成分均匀释放,大大降低使用频率。
在本发明中,除非另有说明,否则本文中使用的科学和技术名词具有本领域技术人员所通常理解的含义。并且,本文中所用的试剂、材料和操作步骤均为相应领域内广泛使用的试剂、材料和常规步骤。
以上所述仅为本发明的优选实施例而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

1.一种可即时分离的微针贴片,其特征在于,包括基板层和由微针组成的微针阵列,并在两者之间添加一可溶性聚合物基质作为连接层,所述连接层的主要成分为透明质酸钠。
2.根据权利要求1所述的微针贴片,其特征在于,微针基板层采用的材料为聚乙烯吡咯烷酮,溶液浓度为0.1-0.2g/mL。
3.根据权利要求1所述的微针贴片,其特征在于,所述微针由可生物降解的材料和促进毛发生长的物质组成,所述可生物降解的材料和所述促进毛发生长的物质共溶于溶剂中。
4.根据权利要求3所述的微针贴片,其特征在于,所述可生物降解的材料是聚乳酸-羟基乙酸共聚物,所述促进毛发生长物质是米诺地尔,所述溶剂是二甘醇二甲醚。
5.根据权利要求4所述的微针贴片,其特征在于,根据共溶性能选择聚乳酸-羟基乙酸共聚物,米诺地尔和二甘醇二甲醚的优化配比,其中聚乳酸-羟基乙酸共聚物与米诺地尔的质量比为6:2-6:4,聚乳酸-羟基乙酸共聚物和米诺地尔的固体质量与溶剂液体质量比为1:10-1:12。
6.根据权利要求4所述的微针贴片,其特征在于,所述米诺地尔的含量为每片微针中含有1.695~2.001mg。
7.一种可即时分离的微针贴片的制备方法,其特征在于,所述方法包括以下步骤:
(1)以生物可降解材料和难溶性药物为原料,采用微针模具并利用干燥法制备微针针体,制得的针体集中填充于微针模具腔体尖端部分;
(2)继续在装载有针体的模具中添加透明质酸钠溶液,形成连接于针体下部的连接层;
(3)采用聚乙烯吡咯烷酮材料制作空白基板,得到最终的微针贴片。
8.根据权利要求7所述的制备方法,其特征在于,制作微针的模具由聚二甲基硅氧烷材料制成,模具底座设有一圆圈以防止制备微针过程中药液的流失,便于药液回收,降低制备成本。
9.根据权利要求7所述的制备方法,其特征在于,制作微针针体的过程如下:
1)形成促毛发生长物质与可生物降解材料的共溶物:以所述生物可降解材料和难溶性药物为原料,将两者粉末加入溶剂后搅拌,搅拌溶解后制得模液;
2)将模液加入模具后离心,刮去模具表面多余的模液并回收,滴加溶剂后静置离心,将模液冲洗到微针针体腔尖端,反复冲洗后放置于真空干燥箱中,干燥并去除溶剂。
10.根据权利要求7所述的制备方法,其特征在于,添加连接层的过程如下:将透明质酸钠加入纯水中,室温搅拌后取透明质酸钠水溶液加入已经装载有制备好的微针针体的模具中,离心干燥并得到具有连接层的完整针体部分。
11.根据权利要求7所述的制备方法,其特征在于,制作空白基板层的过程如下:将聚乙烯吡咯烷酮加入纯水中,室温搅拌后将聚乙烯吡咯烷酮溶液加入带有微针阵列的模具上,离心干燥并得到具有基板层的完整微针贴片。
12.如权利要求1所述的微针贴片或如权利要求7所述的制备方法制得的微针贴片在治疗脱发和/或促进毛发生长方面的应用。
