Abstract

发明背景
透明质酸是一种天然存在的多糖,包含有以β1-4键连接的交替的N-乙酰基-D-葡糖胺和D-葡糖醛酸单糖单位,以及以β1-3糖苷键连接的双糖单位。它通常是作为钠盐存在,具有大约50000-8×106的分子量范围。
发明概述
本发明提供了一种促进骨生长的组合物,由于它含有透明质酸和生长因子,使该组合物具有粘滞性和生物可降解性,足以使它在所需要的骨生长部位保持足以促进骨生长的一段时间。
提供了几种含有透明质酸和生长因子的,具有必要粘滞性和生物可降解性的组合物。
在此所用的名词“透明质酸”缩写为HA,它的含意包括透明质酸及其盐,如钠盐,钾盐,镁盐,钙盐等。
在此生长因子意指那些被发现对诱导或引导骨、韧带、软骨或者与骨或关节有关的其它组织生长起作用的蛋白质或非蛋白质因子。
这些生长因子特别包括bGFG,aFGF,EGF(表皮生长因子),PDGF(血小板衍生生长因子),IGF(***),TGF-βI至III,包括TGF-β超家族(BMP-1至12,GDF1至12,dpp,60A,BIP,OF)。

优选实施方案描述
将对该组合物的配制及其使用方法作更详细地描述。
优选的HA是未交联的,具有500,000及以上的分子量,典型的分子量范围是104-107。这种骨生长促进组合物一般是在水溶液中含有大约0.1-4%重量的未交联HA,为了保持生长因子的生物活性,并维持组合物的适当pH,此水溶液中还含有其它的溶液赋形剂,如缓冲剂盐,糖,抗氧化剂和防腐剂。优选的组合物含有大约0.1-2%重量的未交联HA。该溶液典型的pH范围是4-9,优选地是大约6.0±1.0,而最优选地是大约5.0。
生长因子在该溶液中存在的典型浓度范围是大约10-6-100mg/ml,特别在用bFGF时,优选地是大约0.1-20mg/ml。该浓度应取决于特定骨的部位和功能,以及注入的体积和生长因子的比活性。
重要的是,用于促进骨生长的溶液应具有一定的粘滞度,使之既能通过注射器或导管注射,但是在发挥骨生长促进作用之前又不会过早地被体液稀释。优选地,该组合物的粘滞度是在10-106cP(厘泊)的范围内,对于含有bFGF的组合物,优选地是大约75,000cP。
对此组合物同样重要的是具有生物可降解性,它足以使该组合物能保留在所需要的骨生长部位,发挥骨生长促进活性。
该组合物通常必须在所需要的骨生长部位保持大约3-30天的时间,一般是3-14天。如果该组合物过早地被散开了,所希望的骨生长促进作用不是还未发生,就是所形成的骨没有理想的强度。
如果该组合物在所需要的骨生长部位保持过长的时间,它在骨该部位的存在就可能会抑制骨的自然形成,有时还会导致完全没有骨形成。
该组合物一般是通过在适量的赋形剂如柠檬酸钠,EDTA和蔗糖中混合HA和生长因子,配制成溶液,以便HA和生长因子能以所需要的浓度保留在溶液中,并且使溶液表现出适当的粘滞性和生物可降解性。可借助于任何便利的方式将此溶液施用于所需要的骨生长部位,一般是通过注射器或导管导入。
给予本发明的骨生长组合物,对于加速创伤愈合,防止在伤害发生之后的进一步组织损伤,消除危害自然愈合过程的处理,以及为愈合创造最佳的物理学和生物学条件,都可能是理想的。所要求的骨生长部位包括如下骨折部位:胫骨/腓骨骨折;股骨/肱骨骨折;前臂骨折;后位移远端桡骨(Colles)骨折;应力性骨折,包括与胫骨夹板有关的运动性骨折,以及脚骨损伤;椎骨压迫性骨折,肋骨骨折,和锁骨骨折。所要求的骨生长部位还包括与下列状态有关的病理性骨缺损部位:骨质疏松,骨软化症,甲状旁腺分泌过多,肾性骨营养不良,以及原发性和转移性骨癌。
将以下述实施例对本发明作更详细的描述,提供这些实施例是为了对本发明进行说明,而不打算对权利要求中列举的发明内容加以限制。
实施例1
将透明质酸钠(Genzyme,MW2×106,无菌,在1%溶液中的粘滞度为6500cP)和bFGF(Scios-Nova,在9%蔗糖,20mM柠檬酸钠和1mM EDTA溶液(pH5)中,4.3mg/ml)混合。通过将过滤除菌的bFGF溶液以及其它赋形剂(柠檬酸钠,水等)与适量无菌的固体HA混合构成制剂。迅速使HA分散开,通过用注射器前后反复中注,防止形成大颗粒凝块。无菌操作配制制剂,用带有21G针头预先充灌的1ml塑料注射器,对以乙酰丙嗪,甲苯噻嗪和***麻醉的雄性Sprague-Dawley大鼠(8-9周龄,160-180克)给药。对颈后皮肤横面作一个5-10mm的切口,用于定位矢状缝和人字形缝的交点。以21G针头在骨膜和顶骨之间注射待测制剂50微升。给药后14天将动物无痛苦处死。
用于组织学分析的组织固定在以中性缓冲液配制的10%甲醛液中。在甲酸(RapidBone Decal)中对组织脱钙处理至少2小时,同时不断地缓慢搅动。再将样品脱水,并用石蜡渗透。然后按横切面方向将样品包埋,并作5μm的切片。用苏木精和曙红对切片染色,用于组织学分析。按照如表1中所示的0-4记分法,对新骨形成进行评分。

以类似于Nada et al.,Endocrinology,124:2991-4,1989中的方法测定总的顶骨厚度。对每块组织学切片,在矢状缝侧面2-3mm的部位(大约是矢状缝与切片边缘之间的中点)拍摄照片。在照片的左,中,右侧取得了整块骨的三个骨厚度测定数据,并进行评分,用于确定总的骨厚度。在此测定中既包括致密的皮层骨,也包括网织状新骨。
所有组对每种处理的反应在相同处理组中各动物之间是一致的。定性测定表明,以各种bFGF/HA凝胶制剂处理的动物组显示出新骨形成,而以安慰剂处理的和生长对照动物显示出极微量的新骨形成或者没有新骨形成(表2)。显然,此项研究所测定的各种bFGF/HA制剂之间仅存在微小的差异。但是,看来确实存在剂量反应性作用(图1A,1B)。

表3显示接受不同制剂骨膜下注射后,大鼠颅盖的总的骨厚度。含有bFGF和HA的所有制剂都表现出有新骨形成。表3中的头二行数据表示平行实验结果。接受配制在2%HA凝胶中的100μg bFGF的二平行试验动物组,总的顶骨厚度在第一试验组是0.49±0.10,在第二试验组是0.59±0.12,有17%的差异。但是,二组动物总的骨厚度与对照相比较,定性和定量测定都有显著的差异。与未接受制剂处理的动物相比较,含有100μg bFGF和HA的所有制剂都使新骨形成至少增加了61%。
图1A和1B显示bFGF和HA的浓度对总的骨厚度的作用。当bFGF的剂量从10μg增加至100μg,总的骨厚度从0.45mm增加至0.54mm,增加了20%。当HA的浓度增加,可见总的骨形成也随之增加,直至接近0.5%HA出现最大的骨形成增加为止,在0.5%以上继续增加HA的浓度,不引起由bFGF在此模型中诱导出的新骨形成进一步增加(图1B)。

因此可见,单次骨膜下注射在HA凝胶中的100μg bFGF,表现出对膜内骨形成显著超过对照的定性和定量作用。给药后14天,对于用配制在HA凝胶中的100μg bFGF处理的动物,在注射位置新骨形成达到11%。所有的安慰剂组和对照组在注射后14天,骨厚度增加都小于18%。当bFGF的剂量从10μg增加至100μg,总的骨厚度从0.45mm增加至0.54mm,增加了20%。在0.5%以上继续增加HA的浓度,不引起由bFGF在此模型中诱导出的新骨形成进一步增加。
实施例2
用8个不同的制剂进行实施例1中所述的试验。bFGF和透明质酸共同使用作为与其它7个组合物的对比,这些组合物中bFGF是与其它载体一起使用,或者这些载体是单独使用作为安慰剂。结果概括显示在如下图2和表4中。

实施例3
如在实施例1中配制透明质酸钠(2%)和bFGF(4mg/ml)的制剂,用于对兔骨折部位给药。还配制了一种制剂,含有4mg/ml bFGF,6mg/ml兔纤维蛋白原,0.2mg/ml抑酶肽,以及用于保持pH和稳定性的其它赋形剂。这种纤维蛋白原制剂类似于以前报导的用于骨折修复的组合物1。对新西兰白兔在腓骨骨干中段通过外科手术制造一个1mm的断裂口,用于构成骨折模型。这种实验方法以前曾被用于检验兔骨折的愈合2。用50μl HA/bFGF制剂,或50μl纤维蛋白原/bFGF制剂对动物进行治疗,或者不给治疗。
在治疗后23天,借助于四点弯曲技术(four point bendingtechnique)对每个治疗组的10根治愈腓骨测定其机械强度。图3表示各组的断裂压力(load at failure):未治疗的腓骨,HA/bFGF治疗的腓骨,和纤维蛋白原/bFGF治疗的腓骨。HA/bFGF治疗的腓骨比未治疗的对照强53%,而纤维蛋白/bFGF治疗的腓骨比未治疗的对照强30%。图4显示全部三个治疗组的冲击韧性(energy to failure)。这种测定表明,HA/bFGF治疗的腓骨比未治疗的对照强43%,而纤维蛋白/bFGF治疗的腓骨比未治疗的对照弱3%。
此外,还在治疗后30天测定了10根未治疗腓骨和10根HA/bFGF治疗腓骨的机械强度。图3显示,HA/bFGF治疗的动物与对照相比较,骨断裂压力高36%,并且这种差异具有统计学显著性(P=0.02)。图4表明,HA/bFGF组与对照相比较,骨冲击韧性高79%,并且这种差异具有统计学显著性(P=0.01)。图3和图4还表明,与未治疗的腓骨相比较,HA/bFGF治疗腓骨的强度能更快地恢复到未损害骨的强度,表明能加速骨愈合。

实施例4
用实施例1中的方法比较如下各制剂总的骨形成作用:实施例1中的HA/bFGF制剂,实施例3中的纤维蛋白/bFGF制剂,以及在蔗糖/柠檬酸盐缓冲水溶液中的bFGF制剂。以含有100μg bFGF的每种制剂50μl作骨膜下注射给药,给药后7天和14天处死动物。另外用不给予治疗的动物作对照。
四组中的每一组动物之间对每种处理的反应非常一致。在第7天,所有bFGF处理的动物显示有对bFGF反应在颅盖骨上形成的膜内骨。对照动物显示出极微量的,或者没有新骨形成。定性测定表明,以配制在HA凝胶中的bFGF制剂处理的动物组,比以任何其它的bFGF制剂处理的动物组,有更多的新骨形成。对于第14天的样品,在bFGF/HA凝胶处理的动物中骨形成量的差异甚至更明显。尽管在所有bFGF处理的动物中都有新骨形成,然而,显而易见的是,bFGF/HA凝胶处理的动物与任何其它处理组动物相比较,形成了厚得多的骨体。
图5显示骨厚度的定量测定结果。处理后第7天,给予配制在2%HA凝胶中的100μg bFGF的动物与未受到处理的动物(即对照)相比较,骨厚度增加了95%。对于用配制在纤维蛋白凝胶中的bFGF和配制在柠檬酸盐缓冲水溶液中的bFGF处理的其它bFGF处理组,骨形成分别增加了86%和55%。

实施例5
通过对大鼠顶骨作骨膜下注射,测定了碱性成纤维细胞生长因子(bFGF)制剂中透明质酸的分子量对膜内骨形成的作用。
材料和方法
将具有分子量760-2300KDa的HA(从Genzyme和LifecoreBiomedical得到)用于配制制剂。bFGF是以冷冻液(4.3mg/ml)的形式获得(Scios-Nova),是被冷冻在调整至pH5.0的9%蔗糖,20mM柠檬酸钠,和1mM EDTA的溶液中。其它试剂(蔗糖,柠檬酸钠,EDTA)购自Sigma。
通过将过滤除菌的bFGF溶液(2mg/ml)与适量的HA(20mg/ml)混合配制制剂。溶液和载体开始是在以截止阀连接的各自的注射器中。通过用注射器反复前后冲注使制剂混合均匀。无菌操作配制制剂,用带有21G针头的预先充灌的1ml塑料注射器给药。
将雄性Sprague-Dewley大鼠(8-9周龄,160-180克)用乙酰丙嗪,甲苯噻嗪和***混合液麻醉。对颈后皮肤横向作一个小切口(5-10mm)。确定矢状缝和人字形缝交点的位置,然后用21G针头在交点左侧骨膜和顶骨之间分别注射50μl每种制剂。处理后14天,通过CO2窒息将动物无痛苦处死。
将用于组织学分析的组织固定在以中性缓冲液配制的10%甲醛溶液中。在甲酸(RapidBone Decal)中对组织脱钙处理至少2小时,同时不断地缓慢搅动。再将样品脱水,并用石蜡渗透。然后按横切面方向将样品包埋,并作5μm的切片。用苏木精和曙红对切片染色,用于组织学分析。按照0-4的记分法对新骨形成进行评分。评分0表示没有网织状新骨,评分1表示有少量或斑片状的网织状骨区域,评分2表示有较大的斑片状骨形成区域,评分3表示有薄的,连续的网织状骨(少于原有顶骨的50%),评分4表示有厚的,连续的网织状骨(大于原有顶骨的50%)。
骨厚度测定
在注射位置测定顶骨的总厚度。对每块组织学切片,在矢状缝侧面2-3mm的部位(大约是矢状缝与切片边缘之间的中点)拍摄照片。在照片的左、中、右侧取得了整块骨的三个骨厚测定数据,并进行了评分,用于确定总的骨厚度。致密的皮层骨和网织状新骨都包括在此测定中。
结果
定性测定表明,用bFGF处理的所有动物组都显示有一些新骨形成,而只用HA凝胶处理的动物和对照动物显示有极微量的,或者没有新骨形成。组织学分析表明,bFGF处理的动物显示有网织状新骨和片层状成熟骨存在。在注射位置,在更成熟的低层骨表面形成了明显的网织状新骨层。偶尔网织状骨也存在于右侧,但是不会达到象在左侧所见到的程度。网状新骨显示有正常的逆转线,骨髓隙和通常的染色特征。这些组的大多数动物获得了骨形成评分3(28/30),并有2只动物获得了最高评分4。在网织状骨的上面,紧靠网织状新骨,存在脂肪和纤维组织区,并且显示具有正常的结构形式。没有任何区域显示有指示HA/bFGF制剂可能具有抗原潜在特性的慢性炎症细胞病灶。
HA凝胶处理的动物没有显示出新骨形成,或者仅显示有非常少量的新骨形成,大多数动物获得了骨形成评分0(26/30),30只动物中的3只获得骨形成评分1,并有1只动物获得骨形成评分3。这种新骨形成可能是外科手术过程对骨膜的刺激结果。组织的任何部分未见异常,也没有征候表明所试验的任何HA具有作为抗原的可能性。
未接受外科手术也未接受处理的动物,没有显示出新骨形成,6只动物全部获得骨形成评分0。这组动物非常类似于接受HA凝胶的动物组,不同的是没有作为骨膜刺激结果的新骨形成。这些样品由成熟的骨组织构成,在腔隙中存在正常的骨细胞,还可见骨髓隙。所有的切片中,在骨组织表面都存在少量微细纤维组织。
关于骨厚度,bFGF处理组比生长对照组骨厚度增加了68%-100%。以配制在最高分子量HA凝胶中的bFGF处理动物,获得了最大的骨厚度增加(100%),此凝胶是由从Lifecore购得的最大分子量HA所形成。HA分子量对骨形成有轻微的作用。当HA分子量增加,新骨形成量也增加。骨形成的这种增加可能是由于制剂粘滞度增加。当粘滞度增加,它将成为较厚的扩散屏障,使bFGF较长时间保持在局部位置。HA较长的保留时间于是将导致较多的骨形成。
实施例6
此实施例将论述骨膜下注射HA+bFGF凝胶后透明质酸的局部分布和存留时间。这项研究测定了骨膜下注射含有碱性成纤维细胞生长因子(bFGF)的HA凝胶之后骨膜的增生,新骨形成,以及透明质酸(HA)的局部分布和存留时间。通过组织学评定法测定了第3天骨膜的厚度,第10天的骨厚度。
材料和方法
材料:
透明质酸钠(HA)从Lifecore Biomedical购得(Chaske,MN,1300KDa)。bFGF是在被校正至pH5的9%蔗糖,20mM柠檬酸钠,和1mMEDTA溶液中,以冷冻液(4.3mg/ml)形式由Scios-Nova提供的。制剂缓冲试剂(蔗糖,柠檬酸钠,EDTA,BSA)购自Sigma。己二酰二酰肼(AD)和1-乙基-3[3-(二甲胺基)丙基]碳化二亚胺(EDC)购自Aldrich。磺基-NHS-生物素(SNB),2-(4′羟基苯偶氮基)苯甲酸(HABA),3,3′-二胺基联苯胺四氢氯化物(DAB)金属增强的底物试剂盒,以及亲合素-辣根过氧化物酶(Av-HRP)结合物购自Pierce,吐温20购自Baker。
生物素化
通过二步反应制备HA-生物素(HA-Bi)结合物。按照Pouyaniand Prestwich,Bioconjugate Chem.5:370-372(1994)的方法,先合成酰肼基-HA,随后制备HA-Bi。可通过将200mg HA溶解于50ml水中来制备酰肼基-HA。将AD(3.5g)加入此HA溶液,并用0.1N HCl校准pH至4.75。溶液中再加入EDC(382mg)开始反应。定时监测pH,加入0.1N HCl使pH维持在4.75。4小时之后(此时未发现pH进一步增加),通过以1N NaOH中和至pH7使反应停止。将此产物透析72小时(Specta/Por,截止分子量为6000-8000),然后冷冻干燥48小时。
可通过将15mg酰肼基-HA溶解于1.5ml0.1M NaHCO3来制备HA-Bi结合物。加入SNB(50mg)开始反应。此溶液在室温下用小磁性搅拌棒搅拌20小时。再将此溶液透析72小时,然后冷冻干燥48小时。按照厂商的试验方案(Pierce)借助于取代测定试验确定其取代程度。简单地说,是将900μl亲合素-HABA试剂置于1ml的比色杯中。将它在500nm的吸光度与900μl亲合素-HABA溶液加100μl 1mg/mlHA-生物素溶液的吸光度相比较。平均取代度是每摩尔生物素30摩尔重复的二糖单位。
制剂
如表5中所述,通过将过滤除菌的bFGF溶液与固体HA混合配制制剂。通过反复前后移动以截止阀连接的二个注射器使制剂混合均匀。无菌操作配制制剂,用带有21G针头的预先充灌的1ml塑料注射器给药。

动物模型
将雄性Sprague-Dewley大鼠(6-7周龄,160-180克,每组5只)用乙酰丙嗪,甲基噻嗪和***混合液麻醉。对颈后皮肤横向作一个小切口(5-10mm)。确定矢状缝和人字形缝交点的位置,然后用21G针头在交点左侧骨膜和顶骨之间分别注射50μl每种制剂。处理后14天,通过CO2窒息将动物无痛苦处死。
组织学
将用于组织学评定的组织固定在以中性缓冲液配制的10%甲醛溶液中,然后在13-15%EDTA溶液中脱钙,同时不断地缓慢搅动。再将样品脱水,并用石蜡渗透。然后按横切面方向将样品包埋,并作4μm的切片。每个样品制备二块切片,用苏木精和曙红(H&E)染色,或者按照如下的方法,用Bi:Av-HRP对HA作组织化学染色,先将组织切片在封闭液(在PBS中的1%BSA和0.05%吐温)中孵育30分钟,随后在检测结合物溶液(1μg/ml亲合素-HRP,在1%BSA和0.05%吐温PBS液中)中孵育60分钟。然后将这些组织切片置于漂洗液(0.05%吐温PBS液)中5分钟。用新鲜的PBS/吐温液重复漂洗5次。用金属增强的DAB试剂盒对HA-Bi:Av-HRP复合物染色。施加DAB底物之后5分钟,将切片在水中漂洗。存在此复合物则形成黑色沉淀。最后,将这些切片用苏木精(H)对细胞的细节部分进行复染。
骨膜和骨厚度测定
在注射位置测定骨膜和顶骨的总厚度。对每块组织学切片,在矢状缝侧面2-3mm的部位(大约是矢状缝与切片边缘之间的中点)拍摄照片。在照片的左,中,右侧取得了三个厚度测定数据,并进行评分,用于确定总的骨厚度或总的骨膜厚度。在此骨膜厚度范围内包含有类似于正常骨膜染色特征和细胞形态的组织。致密的皮层骨和网织状新骨都包括在骨厚度测定中。
结果:
以配制在HA凝胶中的bFGF处理的动物显示在第3天有骨膜厚度增加,在第10天有显著的网织状骨形成。以不含有bFGF的制剂处理的动物显示出有限的骨膜增厚和骨形成。发现HA-Bi第3天在紧靠增厚的骨膜组织中,而第10天在紧靠新形成的骨组织中。
HA-Bi,给药后第3天:
在注射位置,清晰的HA团块存在于骨膜上方,存在一部分由于外科创伤从片层状骨***的骨膜。在对HA染色的区域中,存在局部的纤维组织区和非特异性细胞浸润,其中淋巴细胞和退化的细胞清晰可见。周围组织由微细纤维组织构成。
HA-Bi,给药后第10天:
HA-Bi处理的动物显示出带有非特异性纤维组织区的正常片层状骨,这种纤维组织类似于在骨上面的颗粒状组织。此区域含有淋巴细胞,微小血管,脂肪细胞和少量未染色的物质碎片。在左侧颅盖表面致密的纤维组织内有棕黑色的过氧化物酶染色。HA非特异性地分布在纤维组织内。
HA-Bi+bFGF,给药后第3天:
在注射位置,覆盖在原先存在的片层状骨上面的骨膜层有增生。定量测定表明,HA-Bi+bFGF处理动物的骨膜与只用HA-Bi凝胶处理的动物相比较,厚度增加了403%。在增厚的骨膜上面,存在大量纤维化的,过度增生的纤维组织。在这种纤维组织内,存在脂肪细胞,并且存在有非特异性炎症细胞浸润,包含有一些多形核白细胞,巨噬细胞和浆细胞。残留的HA延伸跨越了中线缝,并且看来是在遭受包裹。这些样品显示出浓缩的棕黑色染色物质(即HA),主要集中在被包裹组织的范围内。这种物质多数看来是非特异性地保留在纤维网内,并且有一些看来是非特异性地聚积在局部巨噬细胞的细胞质内。
HA-Bi+bFGF,给药后第10天:
在注射位置,预先存在的颅盖片层状骨已被一厚层具有正常结构和染色特性的,正在成熟的网织状骨覆盖了。用HA-Bi+FGF处理的动物与接受HA-Bi凝胶处理的动物相比较,总的骨厚度增加了70%。这种新骨一般恰好延伸超过中线缝,到达颅盖右侧。在环绕新形成网织状骨的纤维增生组织表层,可见DAB染色的HA。过氧化物酶染色表明,HA一般存在于靠近新形成骨的组织中。在网织状骨的上面,存在纤维-骨膜层。在它的表面存在一片广阔的已形成血管并含有脂肪细胞的微细纤维组织区。在此被微细纤维组织层限定的良好发育的区域内,还存在一些淋巴细胞,浆细胞,以及组织细胞。
HA+bFGF,给药后第3天和第10天:
定性和定量试验都表明,生物素结合了HA不影响对该制剂的生物反应(表6)。HA+FGF处理组与HA-Bi+FGF处理组相比较,骨膜和骨厚度没有显著差别(P>0.05),但是与HA-Bi处理的对照组相比较有显著差别(P<0.001)。组织学检验表明,用不带有生物素的HA+bFGF处理的动物,类似于用HA-Bi+bFGF处理的动物,不同的是前者不存在来自DAB底物的棕黑色染色区域。没有预期这些区域会被染色,因为不存在生物素。少数细胞确实显示染色阳性,是因为存在内源性过氧化物酶活性。

通过骨膜下注射给予HA+bFGF凝胶,对骨膜增生和促进主动的骨形成有显著的作用。给药后3天,骨膜比对照增厚将近5倍。此外,对于HA/bFGF处理的大鼠,给药后10天顶骨厚度比对照增加了70%。在此所试验的制剂中,HA载体通过使原材料定位而引导新骨形成,发现HA存在于主动骨形成区。注射HA+bFGF之后,HA提供了一个靠近新骨形成位置的bFGF储存场所。
除了提供引导bFGF释放的位置之外,看来HA还具有维持促进骨形成环境的生物特性。HA与FGF可能具有协同作用。
对本发明上述的公开内容和描述是其例证性的说明,按照本发明的精髓原理,有可能对其大小、形状和材料,以及对其优选实施方案的细节作出各种改变。
Claims (11)
1.一种含有透明质酸和以下定义的生长因子的骨生长促进组合物,所述生长因子能够诱导或引导骨、韧带、软骨或者与骨或关节有关的其它组织生长,其中该组合物具有粘滞性和生物可降解性,足以使它在所需要的骨生长部位保持足以促进骨生长速率和幅度的一段时间。
2.根据权利要求1的组合物,其中的透明质酸是未交联的。
3.根据权利要求1的组合物,其中该组合物溶液中含有0.1-4%重量的透明质酸。
4.根据权利要求1的组合物,其中的生长因子包括碱性成纤维细胞生长因子。
5.根据权利要求4的组合物,其中所述的碱性成纤维细胞生长因子是以大约10-6-100mg/ml的浓度范围存在于组合物中。
6.含有透明质酸和以下定义的生长因子的组合物在制备在需要骨生长的病患、损伤或异常部位促进骨生长速率和幅度的药物中的用途,所述生长因子能够诱导或引导骨、韧带、软骨或者与骨或关节有关的其它组织生长,其中该组合物具有粘滞性和生物可降解性,足以使它在该部位保持足以促进骨生长的一段时间。
7.根据权利要求6的用途,其中的透明质酸是未交联的。
8.根据权利要求6的用途,其中所述组合物中透明质酸占组合物重量的大约0.1-4%。
9.根据权利要求6的用途,其中的生长因子包括碱性成纤维细胞生长因子。
10.根据权利要求9的用途,其中所述的碱性成纤维细胞生长因子是以大约10-6-100mg/ml的浓度范围存在于组合物中。
11.权利要求6的用途,其中所述部位是原位或骨内。
