技术领域
本发明属于生物技术领域,涉及利用化学材料或生物膜对细胞表面进行修饰的方法、其制备的细胞及应用,特别地,是涉及一种基于多巴胺聚合的活细胞表面功能化和应用。
背景技术
细胞生物学和微生物学方面的最新发现突出了基于细胞的疗法的重要性,这种疗法提供了改变和治疗现有药物无法充分解决的疾病进程的潜力。活细胞的使用包括造血干细胞的输血,嵌合抗原受体T细胞疗法和粪便微生物群移植,已经成功地治疗了许多顽固性疾病,例如先天性缺陷,癌症和炎性肠病。不幸的是,细胞在处理过程中易受到不良环境的影响,例如在操作过程中遇到的大幅度温度波动和离心力作用,以及移植后遭受的宿主免疫***和代谢微环境的损害,从而导致细胞死亡和治疗效果下降。此外,单形式的疗法易导致治疗效果不足,这主要是由于复杂的细胞相互作用的特性和细胞靶标的多样性。因此,迫切需要一种能够同时具有细胞保护作用并赋予细胞多种功能的策略来工程化改造预想的细胞来增强细胞疗法的疗效。
活细胞的表面修饰是将功能性基团附加到细胞表面,这样是提供了一种独特的工具,能够以可编辑的方式生成工程细胞,使细胞具有本身所不具有的外源特性。目前,已经开发出多种方法来用化学合成细胞保护涂层来包被细胞,形成核壳结构。例如,给哺乳动物细胞包被一层聚合物、二氧化硅或磷酸钙的外衣,保护细胞在操作、处理和储存过程中免受外部侵害,从而增强存活率。作为真菌的代表,给酵母细胞穿上一层Fe(III)-鞣酸的复合物外衣来保护其免受有害环境的侵害。最近,申请人用生物膜包被了益生菌,以保护细菌免受现实环境中的致死因素的影响。例如,经红细胞膜伪装的益细菌具有免疫原性屏蔽作用,可减少巨噬细胞对它们的清除。然而,传统的表面修饰涉及复杂而繁琐的制造过程,并且对各种细胞的可及性和通用性有限。更重要的是,大多数已报道的方法都涉及单一的细胞保护功能,缺乏多功能性,而这对于增强细胞疗法的疗效至关重要。
聚多巴胺(PDA)是一种受贻贝中的粘附蛋白启发的合成材料,由于其邻苯二酚基团具有很强的粘附性,因此已被广泛用作各种表面的包被层。多巴胺的聚合反应是在细胞相容性条件下发生的,由此产生的PDA表现出微不足道的细胞毒性。由于细胞膜上存在胺基、硫醇和羟基,PDA可以通过形成共价键或氢键沉积在细胞表面。据此,通过多巴胺和功能性分子共沉积的方法有望在细胞表面形成多功能化包被层。
发明内容
本发明涉及了一种利用多巴胺的聚合改性或修饰的细胞,具体地,本发明涉及了多巴胺聚合介导的活细胞表面多功能化修饰方法以及多巴胺壳聚糖共修饰的益生菌制剂用于肠炎的治疗。所述的聚多巴胺多功能化细胞外衣或包被层是在细胞相容条件下制得,该细胞外衣依据共沉积的分子种类能够赋予活细胞额外的功能。
优选地,所述多巴胺在细胞相容性的条件下聚合时,加入功能性分子以使得细胞表面同时包被多巴胺聚合物和功能性分子。
优选地,多巴胺和氨基末端的聚环氧乙烷和/或壳聚糖共沉积在细胞表面,以形成包含氨基末端的聚环氧乙烷和/或壳聚糖的多巴胺聚合物;优选地,所述壳聚糖为脱乙酰壳聚糖。
所述多巴胺与荧光素共沉积在细胞表面。
优选地,所述细胞为细菌细胞、真菌细胞或哺乳动物细胞。
本发明另一方面涉及一种基于多巴胺聚合的表面功能化细胞,细胞表面包被有壳聚糖与多巴胺反应形成的包被层;所述包被层通过壳聚糖与多巴胺共沉淀形成;优选地,所述壳聚糖为脱乙酰壳聚糖。
本发明还涉及基于多巴胺聚合对活细胞表面功能化的方法,在细胞相容性条件下,多巴胺和功能性分子通过共价键、氢键或π-π堆积共沉积到细胞表面,以在细胞表面聚合形成功能化聚多巴胺包被层。

具体实施方式
下面结合附图和实施方式对本申请作进一步的详细说明。可以理解的是,此处描述的具体实施方式仅用于解释本发明的技术方案、发明构思,而非对本发明所做的限制性说明。
生物膜修饰的细胞或经生物膜修饰的细胞,是利用分隔细胞器或外界的膜***,隔绝外界周围环境,能够让细胞独立于外部环境而存在。一般的生物膜由脂质组成,或者具有生物相容性的化合物聚合形成等。
本申请涉及的PDA包被层,其多巴胺在细胞相容性条件下简单共沉积形成,同时可以包含各种功能性小分子,从而在细胞表面形成多功能包被层。申请人证明了这种方法可以包被各种细胞(包括细菌、真菌和哺乳动物细胞)的可及性和多功能性。借助氢键、π-π堆积、迈克尔加成和席夫碱反应,可以简单地将各种功能性小分子和多巴胺共聚合以形成多形态PDA包被层。多功能PDA可以包被在微生物和哺乳动物细胞(包括细菌,真菌和人类哺乳动物细胞)的表面,并且包被的细胞在活力和生物活性方面的变化可忽略不计。
本发明的具体实施例中,基于多巴胺聚合物的表面功能化的细胞,其细胞表面包被多巴胺聚合物;参照如上所述,多巴胺聚合物,是通过多巴胺在细胞相容性条件下形成并聚合在细胞表面,或沉积到细胞表面而形成。本发明的某些实施例中,将其他功能性分子和多巴胺共沉淀到细胞表面,例如聚环氧乙烷(例如经过修饰),荧光素等。
本发明的其他实施例中,本发明涉及的多巴胺聚合的表面功能化的细胞,多巴胺和壳聚糖共沉积在细胞表面;申请人发现与氨基末端的聚环氧乙烷、壳聚糖共沉积的细胞,相对于单多巴胺聚合,进一步增强细胞对胃肠道应激源的存活率,提高其体内存活率。
包被后的细胞,例如与配体共沉积的肠道菌群,作为靶向治疗结肠炎的口服疗法。由于包被层增加了对胃酸和胆汁盐的耐受性,与未包被的细菌相比,修饰的细胞在小鼠胃中的存活率高出6倍,在肠道中的保留率高出30倍以上。通过对发炎的结肠粘膜的靶向能力,被包被的细胞在病理组织中进一步显示出4倍更高的积累。值得注意的是,与一线药物氨基水杨酸(ASA)相比,这些多形态细胞在硫酸葡聚糖硫酸钠(DSS)诱导的小鼠结肠炎模型中显示出极大的治疗功效。
实验材料和准备
EcN菌株购自中国普通微生物培养物收集中心(GMCC,中国),并在37℃的含有抗生素的Luria Bertani(LB)培养基培养。
HeLa细胞系获自美国典型培养物保藏中心(ATCC),并在Roswell Park MemorialInstitute(RPMI)1640培养基(美国Sigma)中培养,其培养基添加了10%(v/v)灭活的FBS(美国Sigma)和1%的抗生素/抗真菌溶液(v/v),在37℃,5%CO2的培养箱中的。多巴胺、三(羟甲基)氨基甲烷盐酸盐(Tris-HCl)、低粘度虾壳中的壳聚糖、异硫氰酸荧光素(FITC)、罗丹明B、碘化丙啶(PI)、二脒基苯基吲哚(DAPI)购自Sigma。
PEO-NH2(2kDa)和Rho-PEO购自Ponsure Biotechnology(中国上海),DSS购自Sangon Biotech(中国上海)。质粒pBBR1MCS2-Tac-mCherry,pBBR1MCS2-Tac-GFP和所有其他试剂均从国内供应商处购买并按原样使用。
实施例1表面修饰的大肠杆菌
选取大肠杆菌的一个菌落(EcN)置于含有适当抗生素的LB培养基中于37℃孵育5小时。通过在4200×g下离心5分钟收集细菌,并将其重悬于冰冷的PBS中。然后将EcN添加到1ml的10mM Tris-HCl缓冲液(pH 8.5)中,10mM Tris-HCl缓冲液中包含1mg/ml的多巴胺和3.0×108CFU/ml的细菌。在室温下振动半小时后,加入0.5mg PEO-NH2(氨基末端的聚环氧乙烷(PEO-NH2))。反应45分钟后,加入额外量的0.5mg PEO-NH2继续反应45分钟。将包被有PEO-NH2和PDA的EcN于4000×g离心5分钟收集底部样品,并通过PBS洗涤3次进行纯化。然后将产物重悬在无菌PBS中并在4℃下保存。在不添加PEO-NH2的情况下制备包被PDA的大肠杆菌(EcN@P)。通过改变反应混合物中的多巴胺和PEO-NH2的总量为0.5mg,以相同的方法制备EcN@P0.5和EcN@P0.5E0.5。通过分别将0.1mg/ml的FITC和罗丹明B与1mg/ml的多巴胺混合,可分别制得EcN@P/FITC和EcN@P/罗丹明B。将100微升稀释后的菌液均匀涂抹于含有适当抗生素的LB琼脂平板上,于37℃下培养24小时,记录细菌的菌落数(CFU)。
本实施例中,氨基末端的聚环氧乙烷(PEO-NH2)、小分子异硫氰酸荧光素(FITC)和罗丹明B与多巴胺共沉积,以验证包被这种方法的普适性(如图1a所示)。参照图1b和1c,透射电子显微镜(TEM)图像表明PDA包被的EcN(简称为EcN@P)具有不透明的外壳,该外壳由大量的纳米粒子所构成。不透明性是由于PDA独特分子结构所致,该结构由高度共轭的苯环组成。相比于EcN的透明光滑边缘,EcN@P具有粗糙的表面结构,PEO、FITC或罗丹明B与多巴胺在细胞表面的共沉积形成的包被层(分别定义为EcN@PE,EcN@P/FITC和EcN@P/Rho)显得较为完整(图1c和图2)。在聚合过程中,包被层可以通过PDA和膜糖蛋白的亲核基团之间的共价键或氢键形成。图1d的UV-Vis光谱显示,包被的细胞显示出罗丹明B(556nm)和FITC(492nm)典型吸收峰。如图1e和1f所示,多巴胺与FITC或罗丹明B共沉积的细胞呈现出绿色或红色荧光信号。此外,如图1i所示,与罗丹明B标记的PEO-NH2(Rho-PEO)的共沉积涂层同时显示出绿色和红色的荧光信号。流式细胞仪分析显示包被细胞上的荧光强度明显增加,进一步证实了PEO、FITC和罗丹明B成功掺入包被层中(如图1g,h和k)。另外,可以通过调节包被层的结构组成来调节EcN的表面物理化学性质。如图1j所示,用PDA和PEO-NH2共沉积包被细胞后,水接触角分别从48±4°降至25±3°和0°。EcN@PE的表面水接触角降低归因于包被层中亲水性PEO的存在。
然后,申请人测试了包被细胞的活力和生长状况。如图1l所示,当多巴胺和PEO-NH2的浓度均为0.5mg/ml时,观察到近90%的细胞存活率。即使浓度增加到1mg/ml,存活率也可以维持高达70%。与PEO、FITC或罗丹明B的共沉积可以稍微改善细胞存活率(图3a和3b)。OD600的记录显示了包被与未包被EcN具有相似的生长曲线(图1m和图3a和3b),进一步表明修饰的EcN细胞能够保持活力。两个结果证实了在细胞相容性条件下使用多巴胺辅助共沉积来包被活细菌的简单性和可行性。
实施例2表面修饰的酵母
从酵母提取蛋白胨-葡萄糖(YPD)肉汤琼脂平板中选出一个酿酒酵母菌落,置于在YPD肉汤中后,将酵母在37℃的振荡培养箱中培养24小时。通过以4000×g离心5分钟收集细胞,并通过用PBS洗涤3次来纯化。将2×107CFUs/ml的酵母细胞加入1ml含1mg/ml多巴胺的Tris-HCl缓冲溶液(pH8.5,10mM)中震动半小时。然后,加入0.5mg PEO-NH2进行45分钟反应,然后再加入0.5mg PEO-NH2进行另外45分钟反应。通过以4000×g离心5分钟并用PBS洗涤3次获得所得的酵母(Yeast@PE)。在不添加PEO-NH2的情况下制备PDA包被的酵母(Yeast@P)。通过在反应混合物中使用0.5mg多巴胺或PEO-NH2可分别制备出[email protected]和[email protected]。通过分别与0.1mg/ml的FITC和罗丹明B共沉积共同制备Yeast@P/FITC和Yeast@P/罗丹明B。取100微升稀释后的细胞悬浮液,均匀涂抹在YPD琼脂平板上,于37℃下培养48小时并计数CFU来确定细胞数目。
作为真菌的代表,酵母细胞可用于评估包被层的可及性和多功能性。通过在弱碱性溶液中与多巴胺简单混合并在室温下振荡,即可制备PDA包被的酵母细胞(Yeast@P)。以同样的方法,在聚合过程中,加入功能性分子,可制得PEO-NH2(Yeast@PE)、FITC(Yeast@P/FITC)或罗丹明B(Yeast@P/Rho)共沉积包被的酵母细胞(如图4a所示)。如图4b所示,与未包被的细胞的透明性相反,发现包封的酵母是不透明的。与PDA包被的酵母不同,与PEO共沉积可以改善细胞的分散性。如图4c和图5所示,TEM成像显示天然酵母的细胞膜光滑透明,而所有包被的细胞上都出现了粗糙和不透明的边缘。如图4d所示,对于包被的酵母细胞,观察到了共沉积功能分子的特征吸收峰。如图4e和4f所示,Yeast@P/FITC和Yeast@P/Rho的荧光图像进一步表明FITC和罗丹明B存在于包被层中。如图4g和图6所示,共聚焦成像证明了PEO成功掺入包被层中,PDA和Rho-PEO发出的荧光信号重叠。如图4h-4j,共沉积细胞的流式谱图显示荧光强度发生了显着变化。如图4m所示,天然细胞的水接触角为41±3°,在分别用PDA和PEO共沉积包被后,其接触角降至21±3°和0°。为了检查包被层是否对细胞活性有影响,使用平板计数和OD600记录评估了细胞活力和生长状况。图4k表示对于用0.5mg/ml多巴胺处理的细胞存活率在80%左右。尽管用1mg/ml的多巴胺包被会降低细胞存活率至50%,但与PEO-NH2的共沉积可恢复到80%。如图4l和图7所示,用PDA或共沉积PDA包被后,观察到与未修饰细胞相似的生长曲线,这表明包被的酵母在培养基中保持了原有的活力并能正常增殖。
实施例3表面修饰的哺乳细胞
在含有10%胎牛血清(FBS),200U/ml青霉素和100U/ml链霉素的RPMI 1640培养基中培养HeLa细胞。通过胰蛋白酶处理收集细胞,并用PBS洗涤3次。将HeLa细胞(2×105)与0.5mg/ml的多巴胺和0.5mg/ml的PEO-NH2在培养基(pH 8.5)中轻轻摇动40分钟。以800×g离心10分钟后,用PBS洗涤两次来收集得到的细胞(HeLa@PE)。
在确认了该方法包被微生物细胞(大肠杆菌和酵母菌)的能力之后,申请人检测了包被哺乳动物细胞的可行性。将人HeLa细胞与多巴胺和PEO-NH2于轻微震荡的条件下反应40分钟,可制得共沉积多巴胺涂层包被的HeLa细胞(如图8a所示)。包被细胞的表面形态可通过扫描电子显微镜(SEM)表征,结果显示出粗糙并布满纳米颗粒的细胞表面,这与未包被细胞的相对光滑表面形成鲜明对比(图8b)。共聚焦显微镜图片显示出清晰的红色环装荧光图片(图8c和图9)。通过流式细胞仪分析了PEO共沉积包被层,结果显示包被细胞的荧光强度明显增强(如图8d所示)。为了检测包膜的生物相容性,通过碘化丙啶(PI)染色和细胞计数试剂盒-8(CCK-8)分析评估了细胞活力(如图8e和图10)。用PDA(HeLa@P)或PEO共沉聚多巴胺(HeLa@PE)包被后,细胞活力几乎达到80%。CCK-8分析进一步证实,孵育24小时后,未修饰的细胞和包被细胞之间没有发现明显差异(如图8f所示)。因此,聚合介导的表面多功能化方法可用于装饰多种细胞。
实施例4壳聚糖和多巴胺共沉积包被细菌
将携带pBBR1MCS2-Tac-mCherry的EcN(卡那霉素抗性)置于含有50μg/ml卡那霉素的LB培养基中于37℃培养过夜。将过夜培养液在新鲜的LB培养基中按1:50稀释,并在37℃下孵育5小时。通过以4000×g离心5分钟来收集细菌。然后将EcN添加到15ml的10mM Tris-HCl缓冲液(pH 8.5)中,该缓冲液含有0.5mg/ml的多巴胺和2×108CFU/ml的EcN。在2小时的反应过程中,每30分钟将5μl的脱乙酰壳多糖溶液(5mg/ml)添加到反应混合物中。以4000×g离心5分钟后,将细菌重悬于15ml Tris-HCl缓冲液中,再添加5μl壳聚糖溶液(5mg/ml)进行另外30分钟反应。离心得到壳聚糖和聚多巴胺共沉积包被细菌(EcN@PCS),并保存在4℃下以备后用。在不添加壳聚糖的情况下可制备EcN@P。通过使用FITC标记的壳聚糖可在EcN表面实现FITC标记的PCS包被层。
通过PDA与壳聚糖上的氨基之间的迈克尔加成反应以及席夫碱反应,壳聚糖可成功掺入包被层中(图11a)。如图11b所示,用PDA和脱乙酰壳多糖共沉积聚多巴胺(EcN@PCS)装饰后的EcN悬浮液分别呈现出浅灰色和深灰色。与PDA包被的EcN相比,EcN@PCS具有较为光滑外壳,该外壳由许多均匀而紧凑的纳米颗粒组成(图11c和图12)。这是由于壳聚糖上存在的大量氨基与PDA之间可发生交联作用。通过与FITC标记的壳聚糖(FITC-CS)共沉积可标记PCS外壳,并在共聚焦成像下显示绿色荧光(图11d和图13)。细胞流式图显示出荧光强度明显增强,进一步证实了壳聚糖成功掺入到包被层中(图11e)。Zeta电势测量显示分别用PDA和PCS包被后,细胞表面电位从-24mV升高到-21和-16mV(图11f)。EcN@PCS的Zeta电位的增加主要归因于包被层中存在带正电荷的壳聚糖。
实施例5益生菌口服利用率的提高
申请人进一步研究了包被层保护细菌抵抗胃肠道(GI)环境损害的作用。PDA包被层增加了EcN在含有胃蛋白酶的模拟胃液(SGF,pH 2)中的存活率(图11g)。值得注意的是,与壳聚糖的共沉积进一步增强了它们对SGF的抵抗力,这归因于共沉淀后能够在EcN表面形成紧密而均匀的外壳。与SGF接触3小时后,EcN@P的存活率是未包被的EcN的2.5倍,EcN@PCS的存活率是未包被的EcN的15倍。图11h显示了在0.3mg/ml胆汁酸中EcN存活率,存活的细菌数量随时间的增加而减少,与未包被的EcN和EcN@P相比,EcN@PCS保持最高的存活率。TEM图像从视觉上证明PCS包被层在胃液和胆汁酸中依然能够保持其完整性(图14)。在含有胰蛋白酶的模拟肠液(SIF,pH 6.8)中的生长情况表明,用PDA或PCS包被后,细菌保留了其增殖能力(图11i)。综上,PCS包被层极大地提高了细菌在胃肠道中的存活率,并且对其生物活性的影响有限。
为了检查PCS包被层是否可以提高小鼠体内EcN存活率,口服给药后评估了EcN在胃肠道中的保留情况。值得注意的是,用PDA或PCS包被层对携带pBBR1MCS2-Tac-mCherry的EcN的荧光强度的影响微不足道(图15a)。在给小鼠灌胃2×108CFU细菌4小时后,通过体内成像***(IVIS)对小鼠进行成像。EcN@PCS在小鼠的腹部和胃肠中表现出最强的荧光强度(图15b-d)。为了量化细菌保留的差异,通过平板计数分别计算了胃、肠、结肠和盲肠中EcN的数量。与未包衣或包被的EcN相比,EcN@PCS口服后的存活率最高,在胃中高6倍,在肠道中高30倍以上(图15e和图16)。用EcN@PCS给药后,肠道,结肠和盲肠中的所有EcN计数均高于未包被的EcN和EcN@P。这些结果表明,用PCS包被益生菌可以大大提高其口服生物利用度。
实施例6靶向传输到病理组织
考虑到壳聚糖靶向结肠的能力,申请人研究了PCS包被层是否可以增强DSS诱导的小鼠结肠炎模型中EcN在炎症性结肠组织中的积累。根据IVIS分析显示,与未包被的EcN和EcN@P相比,在2×108CFU的EcN@PCS灌胃4小时后,集中在DSS小鼠腹部的荧光强度显着增加(图15f)。胃肠组织离体成像显示,从用EcN@PCS给药的小鼠身上切下的结肠中荧光信号比未包被的EcN组高出5倍以上(图15g和h)。然而,肠道中的整体荧光强度仅高2倍,这进一步说明与壳聚糖共沉积可改善EcN在病理性结肠中的积累。为了计算EcN在胃肠组织中的分布,通过平板计数对在胃、小肠、盲肠、结肠和直肠中定殖的细菌数量进行定量分析。如图15i,j和图18所示,通过包被PCS壳层,靶向结肠的EcN百分比从4%增加到16%,这说明EcN@PCS具有靶向小鼠结肠炎部位的能力。
实施例7增强结肠炎的治疗功效
申请人研究了EcN@PCS对治疗DSS诱导的小鼠结肠炎的功效。给小鼠喂食4%DSS,持续7天以产生炎症,然后每天给小鼠灌胃EcN@PCS,并持续五天(图17a)。用PBS、未包被的EcN和ASA灌胃的炎症性老鼠组以及健康的小鼠组作对照组。用DSS喂养可引起小鼠体重持续下降,而用EcN@PCS治疗可有效恢复体重(图17c)。在治疗5天后,测量肠道的长度,在所有治疗组中,EcN@PCS治疗的小鼠显示出最大的平均长度,这近似于健康小鼠的平均长度(图17b和17d)。此外,进行了双盲酶联免疫吸附测定(ELISA)和组织病理学分析以评估结肠的炎症。与PBS,未包被的EcN和ASA组相比,EcN@PCS处理的小鼠可显着减轻炎症,血清中白介素6(IL-6)处于最低水平,该水平与健康的小鼠接近(图17e)。组织病理学分析结果显示,与PBS(平均评分8.7),未包被的EcN(平均评分6.3)和ASA(平均评分7.0)组的小鼠相比,从接受EcN@PCS处理的小鼠中收获的结肠组织的病理评分显着下降(平均评分3.0)(图17f)。图17h中的苏木精和曙红(H&E)染色的代表性图像显示,与健康结肠相似,从用EcN@PCS处理的小鼠中采样的结肠组织中未观察到明显的组织学损伤和炎症区域。另外,髓过氧化物酶(MPO)染色表明用EcN@PCS处理的小鼠的MPO阳性细胞平均数最低。如图17g和17i所示,在炎症性结肠中EcN的积累明显增加。增加的EcN能够通过调节菌群平衡和抑制促炎细胞因子和趋化因子的表达来改善结肠炎的治疗效果。
本申请中,通过在细胞相容性条件下与多巴胺简单混合,已将各种功能性小分子和聚合物合并到包被层中。细胞表面的物理化学性质和相关功能都可以通过调节包被层的结构组成来调控。申请人已经证明了这种策略装饰多种细胞(包括细菌,真菌和人类哺乳动物细胞)的多功能性和可行性。同时,申请人设计并制备了多巴胺与壳聚糖共沉积包被的益生菌,作为增强结肠炎治疗的口服药剂。由于包被层具有双重的细胞保护和结肠炎部位靶向作用,与未包被的细菌相比,经修饰的细菌表现出明显提高的口服生物利用度和在发炎组织中的靶向积累。临床前研究已经证实,即使在结肠炎的小鼠模型中,与现有药物相比,治疗效果也大大提高。
Claims (5)
1.基于多巴胺聚合的表面功能化细胞,其特征在于,细胞表面包被多巴胺聚合物,以赋予细胞额外的功能;所述多巴胺聚合物在细胞相容性的条件下聚合形成;
多巴胺和氨基末端的聚环氧乙烷和/或壳聚糖共沉积在细胞表面,以形成包含氨基末端的聚环氧乙烷和/或壳聚糖的多巴胺聚合物;
所述细胞为EcN、酵母细胞、Hela细胞。
2.根据权利要求1所述的多巴胺聚合的表面功能化细胞,其特征在于,多巴胺和壳聚糖共沉积在细胞表面以形成包含壳聚糖的多巴胺聚合物,所述细胞为EcN。
3.基于多巴胺聚合的表面功能化细胞,其特征在于,细胞表面包被多巴胺聚合物,以赋予细胞额外的功能;所述多巴胺聚合物在细胞相容性的条件下聚合形成;
所述多巴胺与荧光素共沉积在细胞表面;
所述细胞为EcN、酵母细胞、Hela细胞。
4.基于多巴胺聚合对活细胞表面功能化的方法,其特征在于,在细胞相容性条件下,多巴胺和功能性分子通过共价键、氢键或π-π堆积共沉积到细胞表面,以在细胞表面聚合形成功能化聚多巴胺包被层,所述功能性分子为氨基末端的聚环氧乙烷、FITC、罗丹明B、壳聚糖中的任意一种;
所述细胞为EcN、酵母细胞、Hela细胞。
5.改善胃肠道功能和/或治疗结肠炎的药物,其特征在于,包括如权利要求2所述的基于多巴胺聚合的表面功能化的细胞。
