Abstract
本发明涉及一种防治新冠病毒肺炎变异或复阳的药食同源组合物和应用,属于中药技术领域。本发明提供了一种防治新冠病毒肺炎变异或复阳的药食同源组合物,各中药组分共同发挥高温熔浸的药理作用,将呼吸道上皮细胞深处的新冠病毒全部熔出,使新冠病毒囊膜内压升高、体壳热涨膨大、裂解、破碎、凋亡,从而达到治疗和预防的目的,实现新冠病毒肺炎的治疗,防止其变异和复阳。同时,本发明所述药食同源组合物可作为食品原料用于各种类型食品的制备,不含任何添加剂和防腐剂,适宜各年龄段人群长期食用,有利于人体的健康。

技术领域
本发明涉及中药技术领域,特别是涉及一种防治新冠病毒肺炎变异或复阳的药物组合物和应用。
背景技术
新型冠状病毒肺炎(CoronaVirus Disease 2019,COVID-19),简称“新冠肺炎”,世界卫生组织命名为“2019冠状病毒病”,是指2019新型冠状病毒感染导致的肺炎,新冠肺炎的致病源与急性呼吸综合征冠状病毒(SARS-CoV)、中东呼吸综合征冠状病毒(MERS-CoV)结构相似,为同一种属的新型冠状病毒(SevereAcute Respiratory Syndrome Coronavirus-2,SARS-CoV-2)。该病传染性强,以对症支持治疗为主。新冠肺炎患者临床表现以发烧、呼吸困难等病毒性肺炎症状为主,并有双肺浸润等影像学特征。但新冠肺炎患者较少出现流鼻涕、打喷嚏、喉咙痛等上呼吸道症状,表明SARS-CoV-2的目标细胞可能位于下呼吸道。体外实验发现瑞德西韦(Remdesivir)、氯喹对SARS-CoV-2感染有治疗作用”,这为新冠肺炎的治疗带来希望。但目前因新冠肺炎的具体致病机制尚不清楚,且尚无经过临床验证的特效药物。
中医药是中国医疗体系的重要组成部分,外感疫病在中医治疗中具有重要地位。《伤寒论》六经辨证理论阐述了外感寒证由表及里的传变规律,并遣方用药,开创外感病之先河。金元至明清时期温病学派形成,对外感热病辨证采用三焦辨证、卫气营血辨证方法,侧重采用清热解表药,进一步丰富了外感病的中医诊治体系。既往SARS、H7N9型禽流感疫情中,在病原体不明、疫苗未研发成功时,中医药发挥了因时、因地、因人治宜、病症结合及辨证论治等优势,为疫情提供了可行的治疗方式。然而,新冠病毒COVID-19具有病情变化骤然,传染性强、死亡率高、变异、复阳等特点,属于中医寒湿(瘟)疫范畴,新冠病毒肺炎所谓的难治及变异、复阳,其根本病因是病毒侵入三焦盘踞膜原,其药物难达其位,很难将病毒驱除杀灭。
发明内容
本发明的目的在于提供了一种药物组合物,各中药组分具备产生高温熔浸的药理作用,能够将体内深处的新冠病毒熔出,使新冠病毒囊膜内压升高,体壳热涨膨大、裂解、破碎、凋亡,从而实现新冠病毒肺炎治疗、防止其变异或复阳的目的。

更优选的,所述葛根为葛根提取物,所述乌梅为醋泡乌梅,所述附子为炮附子,所述甘草为炙甘草。
本发明提供了上述药物组合物的制备方法,所述方法包括如下步骤:
将大枣、鸡血藤、紫苏、柴胡、大黄、肉桂、干姜、细辛、麻黄、附子、夏枯草、天花粉、甘草、半夏、白芷、车前子、石膏、桑白皮、黄芩、黄连、葶苈子、枳实、土茯苓、白术混合水提得到第一混合物;
将蜀椒、五味子、月桂子超微粉碎得到第二混合物;
将乌梅、杏仁、涡虫、花生芽、萝卜、党参、山药、枸杞蒸制、研磨得到第三混合物;
将乌贼墨粉、泥鳅肽、牡蛎肽、葛根、木糖醇、食盐混合得到第四混合物;
将第一混合物与加热后的黑芝麻油混合,搅拌均匀后与第三混合物混合,持续升温后加入第四混合物,经搅拌后加入第二混合物,混匀后即得所述药物组合物。
优选的,所述大枣在浸泡前经过炒制,所述炒制的温度为50-70℃。
优选的,所述水提的料液比为1g:(7-9)mL;所述水提的次数为1-3次,每次水提的时间为1-1.5h。
优选的,所述蜀椒在超微粉碎前经过炒制,所述超微粉碎后第二混合物的粒径≤74μm。
优选的,所述加热后的黑芝麻油的温度为40-60℃,所述升温后的温度为70-85℃。

本发明提供了一种药物组合物,所述组合物中:干姜配大枣调和营卫,调和脾胃升胃气,干姜配五味子化痰涤饮,敛肺平喘,干姜配黄连辛开苦降,降阳和阴;麻黄配石膏辛凉疏泄,清肺平喘,麻黄配杏仁宣降肺气,止咳平喘;细辛配五味子宣收肺气,化饮平喘;柴胡配黄芩和解少阳,调畅枢机;黄连配肉桂交通心肾,水火既济;葶苈子配大枣泻肺行水,甘缓补中;枳实配白术,健脾益气,消积导滞;半夏配天花粉、荡热涤痰,平喘;桑白皮配黄芩,泄热消痰喘;上述诸药可利新冠肺炎引起的小便不通。枸杞配大枣提高血红蛋白,提高耐力;车前子止咳化痰,疏泄畅通新冠肺炎引起的上焦火热,大便秘结。土茯苓、鸡血藤、葛根、白芷、夏枯草,可搜剔新冠肺炎湿热之蕴毒深入百络的血络壅塞,升清降浊,祛瘀生新。夏枯草与黄芩、黄连、天花粉共同疏通厥阴气滞、破癓坚、消寒热、祛湿痹,可使新冠肺炎痰浊自消。紫苏、细辛、麻黄、蜀椒共用可将入侵的新冠肺炎的痰浊湿毒从毛缝里和深入凝痰停血之中的,凡药理所不能到达的地方和无孔不入的瘀毒浊水从肺中清除出去。乌梅、柴胡、五味子、杏仁,统领三焦五脏六腑膜***,刺激半表半里、肌筋膜、细胞膜、肺泡膜、淋巴组织及口腔、舌粘膜分泌水液代谢通道,解决少阳体系水液代谢、淋巴组织水液代谢功能的不足,分泌出大量的新鲜水液、使淋巴、肌筋膜、三焦水液代谢通道畅通,将少阳、淋巴、肌筋膜、三焦整体瘀浊去除,清除肺中浊水,推陈致新,让新鲜水液畅通无阻,上焦得通,胃气阴和,津液得下,从而清除新冠肺炎病毒,达到清肺排毒目的。
本发明所述药物组合物共同发挥高温熔浸的药理作用,将呼吸道上皮细胞深处的新冠病毒全部熔出,使新冠病毒囊膜内压升高、体壳热涨膨大、裂解、破碎、凋亡,从而达到治疗和预防的目的,实现新冠病毒肺炎的治疗,防止其变异和复阳。

本发明中,所述药物组合物中各成分的药理作用如下:
鸡血藤:性苦、甘、温,归肝、肾经。活血调经,舒筋活血,补血,去瘀血,生新血,流利经脉,风血痹症,致新推陈。可使纤维蛋白原减少,纤维蛋白裂决产物增加,鸡血藤水提物的聚酰胺柱层析成分具有极强的抑制Hlv-RT活性作用。当浓度为21μg/mL时,可完全抑制HIV-RT的活性。
紫苏:性辛,温,归肺、脾经。疏表解肌,祛散外邪,辛香达郁,辟秽化浊,善治时疫温病,邪陷少阳,善治气滞气逆,气滞经络,肝郁气滞,胸闷呕吐,气逆咳喘,痰诞凝结。辛温能散,以治抑郁之气,停滞胸膈,痰气交阻,肺逆暴嗽咳声不绝,致新推陈,放邪气出路之要药。
柴胡:性辛、苦,微寒,归肝、胆、肺经。疏散退热、疏肝解郁、升举阳气,疏散少阳半表半里,和解少阳,疏肝解郁,疏肝胆二经之郁,解血室之结,痰热、热毒郁结,痰湿痹阻,火毒壅滞,入少阳、厥阴引经药,去脏腑间邪气,推陈致新,刺激淋巴水液代谢,细胞壁水液代谢。
大黄:味苦,寒,归、脾、大肠、肝、心包经。泻下攻积,清热泻火,凉血解毒,逐瘀通经,利湿退黄,荡涤肠胃积滞,治肠胃湿热积滞,痰涎壅盛,
毒热炽盛,推陈致新。
涡虫:性甘,含有成熟干细胞,可修复受伤组织,有强大的器官再生能力。
肉桂:性辛、甘,大热,归肾、脾、心、肝经。补火助阳,引火归原,散寒止痛,温经通脉。辛甘性热,外散寒邪,内温阳气,温中散寒,治寒邪直中三阴,治风湿痹痛,偏于温暖下焦,补命门不足,益火消阴,为治命门半表半里要药。去沉寒、下行走里,使阳长阴自消,辛行温通,偏走血分,温经通脉功胜。能够将入侵在脂膜原半表半里深处的病毒通过大辛大热的高温从脂类膜原中溶化浸润出来,进而实现消除病毒的目的。
干姜:味辛,热,归脾、胃、肾、心、肺经。温中散寒,回阳通脉,温肺化饮。辛热燥烈,通心助阳,去脏脏沉寒痼冷,发诸之寒气,治经络阻塞,冷痹寒痢,反胃隔绝,元阳欲绝,四肢厥逆,增加细胞机能。
细辛:味辛,温,归心、肺、肾经。祛风散寒,大热,搜风剔骨,达表入里,无孔不入。通窍,温肺化饮。辛温走窜,宣泄郁滞,上达巅顶,通利九窍,散风邪,化湿浊,通鼻窍,辛散温通,下气破痰,咳喘,甚则不得卧,散邪消痰涤饮,活血通脉,细辛,佐姜、桂、能驱脏腑之寒,佐芩、连解诸郁热,细辛,芳香最烈,善开结气,上达颜顶,通利耳目,旁达百骸,内之宣络脉而疏通百节,外之行孔窍而直达肌肤,护扶美容。
麻黄:味辛、微苦,温,归肺、膀胱经。味辛发散温通,能够透出皮肤毛孔之外,又能深入凝痰停血之中,凡药理所不能到的地方,麻黄都能无孔不入。上宣肺气,下利膀胱,通调水道,利尿消肿,温以宣通郁结,邪入半表半里,疏肺郁,虽曰解表,实为泄邪。温热病亦无不赖以宣通,凡寒邪郁肺,鼻塞咽哑,热邪窒肺,浊涕鼻渊,水饮渍肺,面浮喘促,火气灼肺,气热息粗,燥火内燔,无不以清肃疏达肺金。
蜀椒:味辛,温。归脾、胃,肾经。温中止痛,辛散温通,入脾除湿,温中燥湿,除寒痹痛,入肺经以散寒止嗽,下达肾经以纳气平喘,主风邪气,坚齿发、明目。
炮附子:味辛、甘,大热,归心、肾,脾经。辛甘热,为纯阳燥烈之品,效力强大,能上助心阳以通脉,中温脾阳而散寒,下补肾阳以益火。能复散失之元阳,能回阳于顷刻之间,入手少阳三焦、命门,无所不至,为阳中之阳,行而不止,命门要药,能入其窟穴而招之,引火归原,则浮游之火自熄。味辛气温,大辛大热,火性迅发,行而不止,无所不至,回阳救逆。
乌梅:醋泡,味酸、涩,平,归肝、脾、肺、大肠经。乌梅统领三焦五脏六腑膜***,刺激半表半里、肌筋膜、淋巴组织及口腔、舌粘膜分泌水液代谢通道,解决膜***、少阳体系水液代谢,淋巴细胞组织水液代谢功能不足,分泌新鲜水液、祛除物理病理的代谢产物。益精开胃,敛肺,涩肠,止呕定喘安蛔。盖血得酸则敛,得寒则止,得苦则涩,其蚀恶疮胬肉,乌梅酸涩而温,入肺则收,入肠则涩,入筋与骨则软,入虫则伏,入于死肌、恶肉、恶痣则除,刺入肉中则拨,气逆烦满,反胃骨蒸,下脱上逆皆治。
五味子:味酸、甘,温,归肺、心,肾经。入手太阴、足少阴经。专补肾、养五脏,生阴中肌者,肾藏精,精盛则阴强,收摄则真气归元,而暖丹田,腐熟水谷,蒸糟粕而化精微,则精自生,精生阴长,增强能量贮备(细胞能量库)的能量生产,激活一氧化氮的产生,治喘嗽要药。
月桂子:味辛,温,祛风除湿,解毒,杀虫,主治风湿痹痛,河豚中毒,疥癣,耳后疮,降糖、降脂、防腐,可抑制艾滋病病毒。
夏枯草,味苦、辛,寒,归肝、胆经。入足厥阴、少阳经。辛能散结、苦寒下泄除热,补厥阴血脉,虚者尽可倚仗。专散痰核鼠疮,尤通心气,头目之火可祛,胸膈之痞可降,治肺气为邪所壅,则清肃之令不行,而痰既结于胸膈之间而不散,破癓坚、消寒热、祛湿痹,疏通厥阴气滞之功。抑制艾滋病病毒,全草含具有抗人免疫缺陷病毒(HIV)的酸性多糖-夏枯草多糖,夏枯草皂苷具有明显抗艾滋病毒作用。夏枯草所含熊果酸及衍生物对细胞P388、L1210均有显著的细胞毒作用。
天花粉、味甘、微苦,寒,归肺、胃经。清热泻火,生津止渴,消肿排脓,退五脏郁热,降隔上热痰,润心中烦渴,除时疾狂热,治痈疡解毒排脓,用于痰浊壅塞胸膈,痰热壅肺咳嗽胸闷,喘满气急,荡涤胸中郁热垢腻,用于热毒壅滞,导痰浊下行,镇咳祛痰止喘。具有免疫刺激作用:天花粉对脾脏免疫细胞形成和分化有促进作用,可使小鼠脾脏的白髓成分增值,生发中心增大,边缘增宽,1gM、B淋巴细胞及1gG浆细胞的数量明显增多;巨噬分布区扩大,以天花粉饲喂小鼠7天后,免疫球蛋白IgM明显增多。能增强机体免疫功能。天花粉蛋白(TCS)具有增强红细胞免疫功能的作用,可提高红细胞C3b受体及SOD活性水平。增强小鼠PMN吞噬作用。天花粉多糖对人外周血单核细胞有明显促增殖和活化作用,可不同程度地上调T淋巴细胞亚群CD3+、CD4+、CD8+T细胞的含量,并可诱导人外周血单核细胞高水平分泌产生TNF-q、IL-6,以增强机体免疫力。TCS可抑制艾滋病毒(HIV)在感染的免疫细胞内的复制繁衍,减少在免疫细胞中受病毒感染的活细胞数。TCS能使感染的T细胞表面迅速丧失病毒的活性膜蛋白gP120,或使感染的单核白细胞/巨噬细胞内的病毒迅速丧失其中心蛋白P24。天花粉的有效抑制浓度为0.05~10μg/mL。
灸甘草:味甘,平,归心、肺、脾、胃经。补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药。补三焦元气,主五脏六腑寒热邪气,坚筋骨,长肌肉,倍力、金疮肿,解毒。孙思邈论云:有人中乌头、巴豆毒,甘草入腹即定。甘草酸可促进产生IL-2、lFN-r、抑制IL-4、IL10的生成。甘草酸单铵提高IL-2产生和IL-2R表达。促进IL-2刺激自然细胞生长并增强细胞活性。甘草酸抑制溶酶通路,从而阻止了膜攻击复合体的形成。甘草酸可抑制艾滋病毒的复制,具有诱导干扰素增强自然杀伤细胞活动功能。甘草酸能抑制病毒DNA复制而产生抗病毒作用,还可干扰EB病毒向细胞内的穿透,阻断进一步的复制循环。
半夏:味辛,温,归脾、胃、肺经。降逆止呕,消痞散结。辛温而燥,燥湿化痰,为治湿痰、寒痰要药。辛开散结,消痰涎,开胃健脾止呕吐,去胸中痰满,下肺气,治寒痰及形寒饮冷伤肺而咳,大和胃气,除胃寒,进饮食,脾无留湿不生痰,故脾为生痰之源,肺为贮痰之器。涎滑能润,辛温能散亦能润。故行湿而通大便,利窍而泄小便,刺激胃肠粘膜半表半里之邪毒,使病理产物随大便而出,放邪气出路。
葛根:味辛、甘,凉,归脾、胃,肺经,解肌退热,生津止咳,通经活络,辛甘性凉,辛能透散,凉而入寒。升清降浊,解散阳明温病热邪之要药。开腠里,解温热时行疫疾,清透热邪为治疗阳明之要药。
枳实:味苦、辛、酸、微寒。归脾、胃、大肠经。破气消积,化痰除痞,辛行苦降,入脾胃大肠作用强烈,善破气除痞,消积导滞而治胃肠积滞诸症。破气除满。除寒热结,除胸胁痰癖,逐停水,破结实,消胀满。泻痰能冲墙倒壁。
乌贼墨粉:主治血刺心痛,可显著提高干扰素-V(IFN-V)水平,增强NK细胞杀伤活性,充分激活单核一巨噬细胞对淋巴因子激活杀伤细胞(LAK)细胞活性的激活作用。促进骨髓造血干细胞,诱导产生多种细胞因子,促进造血干细胞粒单系祖细胞的增殖,对妇科,外科、内科,等多种出血效果显著,对子宫出血,肺结核咳血,尿血,肺癌咳血,消化道溃疡及炎症等引起的出血效果良好。
白芷:味辛,温,归胃、大肠、肺经。解表散寒,祛风止痛,宣肺通窍,燥湿止带,消肿排脓,其香气烈,辛香温散,入手足阳明、足太阴,走气分,亦走血分,辛以升散,温以畅通气机,调和肠胃,上行头目,下抵肠胃,中达肢体,遍通肌肤以至毛窍,而利泄邪气。
车前子:味甘,微寒,归肝、肾、肺、小肠经。清肺化痰,清上焦火热,开中焦畅通,通利二便,杏仁:味苦,微温,归肺、大肠经。味苦而降,止咳平喘,破壅降逆。
石膏:味辛、甘,大寒,归肺、胃经。入手太阴经,少阳经,足阳明经。辛散透达,解肌透热,邪热郁肺,气***促,高热,神昏,发狂。
桑白皮:味甘,寒,归肺经。泻肺平喘,利尿消肿。主治喘满咳嗽,热痰唾血,皆由实邪郁遏,肺窍不得畅通,泻肺之余,非桑皮不可。泻肺中伏火而补正气,泻邪所以补正也。桑白皮含黄酮类化合物、桑根白皮素、环桑根皮素、桑素、桑色烯、环桑素等;香豆素类化合物、5,7-羟基香豆素、伞形花内脂、东莨菪素、东莨菪内脂(6-甲氧基-7羟基-香豆素);多糖类、黏液素、桑多糖甲壳素、壳聚糖;还含有丁醇、桑辛素、桑皮呋喃、谷固醇、鞣质和挥发油等,具有良好的抗多种病毒的作用。
黄芩:味苦,寒,归肺经、胆经、脾经、大肠经、小肠经。清热燥湿,泻火解毒,止血,安胎。用于湿温、暑温胸闷呕恶,湿热痞满,泻痢,黄疸,肺热咳嗽,高热烦渴,血热吐衄,痈肿疮毒,胎动不安。
黄连:性苦,寒,归心、肝、胃、大肠经。清热燥湿,泻火解毒。入心与胞络,泻心火,去中焦湿热,治诸疮、去风湿,赤眼暴发,止中部见血。凡药能去湿者必增热,能除热者,必不能去湿。惟黄连能以苦燥湿,以寒除热。黄连大苦大寒,苦燥湿,寒胜热,能降一切有余之湿火,而心、脾、肝、肾之热,胆胃大小肠之火,无不治之,上中风火之目病,中平肝胃呕吐,下通腹痛之滞下,皆燥湿清热之效,清涤血热,吐衄溲血,便血淋浊及痛肠斑疹丹毒。抗炎及对免疫***的作用,黄连甲醇提取物有抑制肉芽组织增生的作用,可增强非特异性免疫功能,小檗碱能抑制IL-1或TNF诱导的多形核白细胞与血管内皮细胞的黏附及黏附分子ICAM-1和CD18的表达,黄连素对T细胞早期活化抗原CD69和中期活化抗原CD25的表达有明显抑制效应,黄连素对PKC或其下游的信号途径有影响,并可以通过下调黏附分子表达而抑制淋巴细胞与血管内皮细胞间的黏附作用。从而影响淋巴细胞再循环。证明黄连素对T细胞的活化和增殖均有明显的作用。
葶苈子:性辛、苦,大寒,归肺、膀胱经。泻肺平喘、行水消肿。泄肺气之壅闭而通调水道。为手太阴经正药,亦入手阳明、足太阳经。苦降辛散,而性寒凉,故能破滞开结,定逆止喘。主癜瘢积聚结气,饮食寒热,破坚逐邪,通利水道。
土茯苓:味甘、淡,平,归肝、胃经。解毒,除湿,通利关节。为治梅毒的要药,利湿去热,能入络,搜剔湿热之蕴毒。深入百络,关节疼痛,甚至腐烂,又毒火上行,咽喉痛溃,一切恶症。土茯苓含有落新妇苷、异黄杞苷、土茯苓苷、槲皮素、薯蓣皂苷、胡萝卜苷、生物碱、甾醇、有机酸、挥发油、淀粉、鞣质等。具有良好的抗炎作用和抗人巨细胞毒的作用。
白术:味苦、甘,温,归脾、胃经。健脾益气,治疗新冠肺炎泄泻,痰饮内盛,眩晕心悸胸胁胀满,咳而气短。主风寒湿痹,白术苦温,健脾燥湿,可治风温痹痛诸症,健脾以助肠运,以复肠腑下行之机,故亦可用治便秘。土旺则能旺湿,故患痰饮者,肿满者、湿痹者,皆赖之也。土旺则清气善升,而精微上奉,浊气善降,而糟粕下输,为后天资生之要药。对免疫***的作用:提高TH/TS的比值,纠正T细胞亚群分布紊乱状态,可使低下的IL-2水平显著提高,并能增加T淋巴细胞表面IL-2R的表达,清除自由基,提高红细胞SOD活性。
泥鳅肽:利尿通淋,解毒消肿,抗菌消炎。泥鳅多糖能明显增强ConA诱导小鼠脾淋巴细胞增殖能力,并升高小鼠血清溶血素含量。
牡蛎肽:不仅含有牡蛎原有的维生素、微量元素和牛磺酸等营养成份,而且相对分子量小于1000Dalton的小分子低聚肽占比超过80%。牡蛎肽的主要成分是牡蛎,俗称海蛎子、蚝,据《本草纲目》记载:“生蚝,治虚损,壮阳,解毒,补男女气血,令肌肤细嫩,防衰老”,主要成分糖原能迅速补充恢复体力,并可以提高机体免疫力。同时还具有提高男性血清睾酮水平、调节血脂、抑制血小板聚集、改善高血糖症状、促进新陈代谢等功能,对抗癌和防止癌细胞扩散也有一定效果。
山药:味甘,平,归脾、肺、肾经。益肾气,健脾胃,止泄痢,化痰涎。山药多糖(RDPS-I)可不同程度提高T淋巴细胞增殖能力、NK细胞和血清溶血溶血素活性,以及血清lgG含量,也能增强巨噬细胞的吞噬能力,既具有非特异免疫功能,又能提高特异性细胞免疫和体液免疫功能,山药多糖RDPS-I可显著增强荷瘤小鼠T淋巴细胞增殖能力、NK细胞活性,小鼠脾脏细胞产生IL-2的能力及腹腔巨噬细胞产生TNF-α。
枸杞:味甘,平,归肝、肾、肺经。入足少阴、足厥阴经。养肝、滋肾、润肺。枸杞多糖可显著增强小鼠腹腔静止巨细胞以及硫代乙醇酸钠(TC)活化的巨噬细胞的NO诱生量及胞内溶菌酶与SOD活性,枸杞多糖超过50mg/(bg/d)剂量灌胃可明显增强巨细胞的吞噬指数,对实验小鼠血清溶血素活性呈显著地增强作用,多糖能显著增加小鼠胸腺、脾脏的重量,以10mg/Kg灌胃效果最好,能增强正常小鼠和环磷酰胺(Cy)处理鼠的NK细胞的活性。枸杞子可激活巨噬细胞产生IL-1、激活T淋巴细胞产生IL-2、活化NK细胞、使T淋巴细胞增殖及CD3+、CD4+、CD8+表达增高,枸杞多糖与植物凝集素(PHA)协同作用后可使淋巴细胞增殖,PHA是T细胞非特异性有丝***原,能与T细胞表面CD/TCR复合体结合,导致包括CD3+、CD4+、CD8+以及LAF-1等细胞表面分子磷酸化,通过激活PKC途径引起细胞活化,促进淋巴细胞进入增殖周期而发生增殖。枸杞可使ConA激发的T淋巴细胞增殖反应明显增强,同时对B细胞免疫功能具有明显增强作用,促进脾细胞产生IL-2水平。枸杞多糖(LBPs)能够促进体外培养的树突状细胞(DC)的分化,成熟提高其表面表达MHC-II、CD83、CD86(B7-2)以及CDIla(LFA-I)的水平,促进DC分泌IL-12P40,增强T细胞的增殖能力,并使T淋巴细胞转化为杀伤性T细胞,提高T细胞的杀伤活性。
党参:性甘,平,归脾、肺经。健脾益肺,养血生津。可替代人参与茯苓、白术、炙甘草同用以健脾气,有胃气则生,无胃气则死。可替代人参与黄芪、五味子、紫菀、桑白皮等同用有补肺益气、止咳平喘之效,其尤可贵者:则健脾而不燥;滋胃阴而不湿;润肺而不犯寒凉;养血而不偏滋腻;鼓舞清阳,振动中气而无刚燥之弊。胆碱乙酰转移酶(CHAT)重组蛋白,生成ACH的量增加。增强SOD活性,使巨噬细胞数量明显增加,细胞体积增大,伪足增多,吞噬能力增强,细胞内的DNA、RNA、糖类、ACP酶、ATP酶、酸性酯酶及琥珀酸脱氢酶活性均显增强,对用环磷酰胺造成的免疫抑制状态的小鼠,能明显地增强淋巴细胞转化,抗体形成的功能,提高抗体滴度的水平。对CY所致免疫受抑小鼠AMAE+LC率及DTH有恢复作用。对环磷酰胺(CY)免疫抑制大鼠脾淋巴细胞产生白细胞介素-2有升提作用,能使IL-2生长指数升高。党参多糖对体液免疫和细胞免疫有较强的促进作用,对刀豆蛋白A刺激的淋巴细胞同样有明显的促进作用。
大枣:味甘,温,归脾、胃、心经。安中养神,助十二经,补阴阳、气血、津液、脉络、筋俞、骨髓一切虚损,调畅中脘虚滞之痰。善补阴阳、气血、津液、脉络、筋俞、一切虚损。凡内伤肝脾之病,土虚木燥,风动血耗者,非此不可。大枣多糖能明显增加小鼠红细胞C3b受体(RBC-C3bR)花环率和红细胞免疫复合物(RBC-IC)花环率,并能使Cy引起的下降的RBC-C3b花环率和RBC-C花环率显著回升。
黑芝麻油和花生芽:天然维生素E,抗细胞氧化,补肝肾、益精血,用于新冠肺炎便秘。
萝卜:抗病毒,缓解大便,治新冠肺炎引起的体弱恶寒发热,头痛鼻塞、咳嗽痰多。
木糖醇:用于糖尿病患者的糖代用品,不经胰岛素就可直接吸收利用,防蛀牙。
食盐:主要成分是氯化钠,为细胞外液中的主要电解质,是维持渗透压的重要成分,调节肌体的酸碱平衡。
本发明中,所述药物组合物以鸡血藤、紫苏、柴胡、大黄、涡虫、肉桂、干姜、细辛、麻黄、蜀椒和附子共为君药,乌梅、五味子、月桂子、夏枯草、天花粉、甘草、半夏、葛根和枳实为臣药,乌贼墨粉、白芷、车前子、杏仁、石膏、桑白皮、黄芩、黄连、葶苈子、土茯苓和白术为佐药,花生芽、萝卜、党参、山药、枸杞、大枣、泥鳅肽、牡蛎肽、黑芝麻油、木糖醇和食盐为使药,以上诸药通过合理的配伍关系,共同产生高温熔浸的药理作用,将盘踞在呼吸道上皮细胞即毛柱状上皮组织的多种复合细胞组成的细胞膜半表半里的、深部气道和肺泡半表半里、肺泡膈半表半里,遍布皮里肉外、空腔管道、经筋、经脉、经络、淋巴组织及五脏六腑腔系相连及脂类油膜、腠理毫毛和凝痰停血之中的新冠病毒熔出,使新冠囊膜病毒内压升高、体壳热涨膨大、裂解、破碎、凋亡,清除所有的新冠病毒,从而防止新冠病毒的变异和复阳。
本发明提供了上述药物组合物的制备方法,所述方法包括如下步骤:
将大枣、鸡血藤、紫苏、柴胡、大黄、肉桂、干姜、细辛、麻黄、附子、夏枯草、天花粉、甘草、半夏、白芷、车前子、石膏、桑白皮、黄芩、黄连、葶苈子、枳实、土茯苓、白术混合水提得到第一混合物;
将蜀椒、五味子、月桂子超微粉碎得到第二混合物;
将乌梅、杏仁、涡虫、花生芽、萝卜、党参、山药、枸杞蒸制、研磨得到第三混合物;
将乌贼墨粉、泥鳅肽、牡蛎肽、葛根、木糖醇、食盐混合得到第四混合物;
将第一混合物与加热后的黑芝麻油混合,搅拌均匀后与第三混合物混合,持续升温后加入第四混合物,经搅拌后加入第二混合物,混匀后即得所述药物组合物。
本发明中,所述大枣在浸泡前优选的经过炒制,所述炒制的温度优选为50-70℃,更优选为58-64℃。本发明对所述大枣炒制方法和时间并没有特殊限定,根据本领域的常规炒制方法炒至焦黄即可。本发明中,所述水提的料液比优选为1g:(7-9)mL,更优选为1g:8mL;所述水提的次数优选为1-3次,更优选为2次;所述每次水提的时间优选为1-1.5h,更优选为1.2-1.3h。本发明中,所述每次水提后优选的包括过滤,所述过滤后所得的提取液混合即为所述的第一混合物。
本发明中,所述蜀椒在超微粉碎前优选的经过炒制,本发明对所述炒制的方法并没有特殊限定,采用本领域对蜀椒的常规炒制方法即可。本发明中,所述超微粉碎后第二混合物的粒径优选的≤74μm,更优选的≤70μm。本发明对所述蒸制和研磨的方法并没有特殊限定,采用本领域常规的蒸制和研磨方法即可。本发明中,所述加热后的黑芝麻油的温度优选为40-60℃,更优选为48-52℃;所述升温后的温度优选为70-85℃,更优选为80-82℃。
本发明中,加入第二混合物后优选的需要继续炒制并搅拌,所述炒制并搅拌至成品软硬适宜即可得到所述的药物组合物。本发明对所述热油炒制的设备并没有特殊限定,在本发明的具体实施例中,所述热油炒制的设备优选为全自动行星搅拌炒锅。
本发明还提供了上述的药物组合物在制备防治新冠病毒肺炎变异或复阳药物中的应用。本发明对所述药物的剂型或辅料并没有特殊限定,根据剂型选择合适的辅料和制备工艺即可。
本发明中,所用原料、试剂与设备均为已知产品,采用常规市售产品即可。
为了进一步说明本发明,下面结合实施例对本发明提供的技术方案进行详细地描述,但不能将它们理解为对本发明保护范围的限定。
实施例1
1)将大枣20份、鸡血藤15份、紫苏15份、柴胡50份、大黄50份、肉桂15份、干姜15份、细辛10份、麻黄15份、附子15份、夏枯草15份、天花粉15份、甘草15份、半夏15份、白芷15份、车前子70份、石膏50份、桑白皮15份、黄芩15份、黄连10份、葶苈子15份、土茯苓15份、枳实15份、白术15份混合,加入8倍量的水浸泡,并置于提取罐中提取2次,每次提取1小时,过滤除去药渣,合并两次提取液得到第一混合物;
2)将蜀椒15份炒香,然后与清洗干净干燥后的五味子15份、月桂子30份共同进行超微粉碎得到粒径70μm的第二混合物;
3)将醋泡乌梅30份去核,杏仁30份去皮浸泡,然后与涡虫30份、花生芽30份、萝卜25份、党参50份、山药50份、枸杞30份共同入锅蒸熟,然后研磨成浓浆得到第三混合物;
4)将乌贼墨粉10份、泥鳅肽30份、牡蛎肽30份、葛根提取物25份、木糖醇30份、食盐5份混合得到第四混合物;
5)将第一混合物与50份的50℃黑芝麻油热油混合,搅拌均匀后与第三混合物混合,持续升温至80℃时添加第四混合物,持续搅拌炒制,然后再加入第二混合物继续搅拌,搅拌均匀后即得本发明所述药物组合物。
实施例2
1)将大枣15份、鸡血藤10份、紫苏10份、柴胡20份、大黄60份、肉桂10份、干姜10份、细辛5份、麻黄10份、附子5份、夏枯草10份、天花粉10份、甘草10份、半夏10份、白芷8份、车前子50份、石膏40份、桑白皮10份、黄芩10份、黄连5份、葶苈子10份、枳实5份、土茯苓10份、白术10份混合,加入7倍量的水浸泡,并置于提取罐中提取1次,提取1.5小时,过滤除去药渣得到第一混合物;
2)将蜀椒10份炒香,然后与清洗干净干燥后的五味子10份、月桂子10份共同进行超微粉碎得到粒径74μm的第二混合物;
3)将醋泡乌梅20份去核,杏仁20份去皮浸泡,然后与涡虫20份、花生芽10份、萝卜15份、党参20份、山药25份、枸杞15份共同入锅蒸熟,然后研磨成浓浆得到第三混合物;
4)将乌贼墨粉1份、泥鳅肽10份、牡蛎肽10份、葛根提取物15份、木糖醇15份、食盐1份混合得到第四混合物;
5)将第一混合物与25份的40℃黑芝麻油热油混合,搅拌均匀后与第三混合物混合,持续升温至70℃时添加第四混合物,持续搅拌炒制,然后再加入第二混合物继续搅拌,搅拌均匀后即得本发明所述药物组合物。
实施例3
1)将大枣30份、鸡血藤25份、紫苏25份、柴胡60份、大黄30份、肉桂30份、干姜30份、细辛20份、麻黄30份、附子26份、夏枯草25份、天花粉25份、甘草25份、半夏25份、白芷25份、车前子83份、石膏65份、桑白皮30份、黄芩30份、黄连25份、葶苈子30份、枳实35份、土茯苓35份、白术35份混合,加入9倍量的水浸泡,并置于提取罐中提取3次,每次提取1小时,过滤除去药渣,合并两次提取液得到第一混合物;
2)将蜀椒30份炒香,然后与清洗干净干燥后的五味子26份、月桂子40份共同进行超微粉碎得到粒径74μm的第二混合物;
3)将醋泡乌梅42份去核,杏仁40份去皮浸泡,然后与涡虫43份、花生芽45份、萝卜35份、党参68份、山药66份、枸杞40份共同入锅蒸熟,然后研磨成浓浆得到第三混合物;
4)将乌贼墨粉25份、泥鳅肽40份、牡蛎肽40份、葛根提取物40份、木糖醇40份、食盐15份混合得到第四混合物;
5)将第一混合物与65份的60℃黑芝麻油热油混合,搅拌均匀后与第三混合物混合,持续升温至85℃时添加第四混合物,持续搅拌炒制,然后再加入第二混合物继续搅拌,搅拌均匀后即得本发明所述药物组合物。
实施例4
1.实验对象
在某医院收集新冠肺炎患者轻型3例,临床表现发热、肺弥散、乏力、咳嗽、胸闷气短、体差、核酸检测阳性,肺通气及运动均不正常,肺CT有磨玻璃样(S刺突蛋白),如图1。
2.治疗方法
上述所有病例服用本发明实施例1所述的药物组合物进行治疗,其中,每日口服本发明实施例1组合物3次,早中晚各1次,每次服用18g,连续服用7日。
3.实验结果
3例患者的肺部CT结果如图2。结果显示:所有患者肺部磨玻璃样影变浅、条索状影变淡,纤维化吸收,肺活量功能改善。同时,所有患者均呼吸通畅,咳嗽咳痰症状消失,核酸结果显示阴性。
实施例5
1.实验对象
在某医院收集新冠肺炎核酸检测阴性,但存在康复期后遗症者30例,其中所有存在后遗症病例在实验前进行肺部CT,结果显示:肺弥散,肺CT依然有磨玻璃样,同时伴有气短、咳嗽、乏力,运动后呼吸不畅等症状。
2.治疗方法
上述实验对象均采用本发明实施例1所述的药物组合物进行治疗,其中,每日服用本发明实施例1组合物2次,早晚各1次,每次服用18克,嚼服即可,连续服用15日。
3.实验结果
30例病例在服用本发明实施例1所述的药物组合物后观察其气短、咳嗽、乏力,运动后呼吸不畅等症状的缓解情况,同时进行肺部CT,其中2例结果如图3所示。
可以看出,服用本发明实施例1的组合物后,所有病例肺部磨玻璃样影变浅,纤维化较前吸收,肺呼吸吸通畅,咳嗽症状消失,痰消失,能够有效清除所有的新冠病毒,防止新冠病毒的变异和复阳。
综合分析,本发明利用鸡血藤、柴苏、柴胡、大黄、乌梅、涡虫、五味子、半夏、葛根、枳实、乌贼黑粉、白芷、车前子、杏仁、石膏、桑白皮、黄芩、黄连、葶苈子、土茯苓、白术、泥鳅肽、牡蛎肽、花生芽、萝卜、党参、山药、枸杞、大枣、黑芝麻油、木糖醇这些辛味药和营养能量药组分给细胞贮备营养,源源不断的提供能量,助温热六虎药“肉桂、干姜、细辛、麻黄、蜀椒、附子”发挥强大的温里作用,使细胞产生巨大热能,如图4。
可以看出,本发明依据各味药的熔点和沸点,通过上述诸药的合理配伍使其产生高温熔浸、温阳散结的效果,从而将盘踞在遍及皮里肉外、空腔管道、经筋、经脉、络、淋巴组织、五脏六腑的脂类油膜及腠理毫毛和深入凝痰停血之中及深部气道和肺泡半表半里、肺泡膈半表半里的所有新冠病毒熔出,无论新冠病毒怎样变异都会使这些病毒蛋白壳膜发生热涨内压升高,膨大裂解、破碎凋亡,如图5,从而将入侵三焦膜原的病毒全部驱除,实现新冠肺炎的治疗,同时防止新冠病毒的变异和复阳。

1.一种治疗新型冠状病毒肺炎的药物组合物,其特征在于,由如下重量份的原料组成:
鸡血藤15份、紫苏15份、柴胡50份、大黄50份、涡虫30份、肉桂15份、干姜15份、细辛10份、麻黄15份、蜀椒15份、附子15份、乌梅30份、五味子15份、月桂子30份、夏枯草15份、天花粉15份、甘草15份、半夏15份、葛根25份、枳实15份、乌贼墨粉10份、白芷15份、车前子70份、杏仁30份、石膏50份、桑白皮15份、黄芩15份、黄连10份、葶苈子15份、土茯苓15份、白术15份、花生芽30份、萝卜25份、党参50份、山药50份、枸杞30份、大枣20份、泥鳅肽30份、牡蛎肽30份、黑芝麻油50份、木糖醇30份、食盐5份。
2.根据权利要求1所述的药物组合物,其特征在于,所述葛根为葛根提取物,所述乌梅为醋泡乌梅,所述附子为炮附子,所述甘草为炙甘草。
3.权利要求1或2所述的药物组合物的制备方法,其特征在于,所述方法包括如下步骤:
将大枣、鸡血藤、紫苏、柴胡、大黄、肉桂、干姜、细辛、麻黄、附子、夏枯草、天花粉、甘草、半夏、白芷、车前子、石膏、桑白皮、黄芩、黄连、葶苈子、枳实、土茯苓、白术混合水提得到第一混合物;
将蜀椒、五味子、月桂子超微粉碎得到第二混合物;
将乌梅、杏仁、涡虫、花生芽、萝卜、党参、山药、枸杞蒸制、研磨得到第三混合物;
将乌贼墨粉、泥鳅肽、牡蛎肽、葛根、木糖醇、食盐混合得到第四混合物;
将第一混合物与加热后的黑芝麻油混合,搅拌均匀后与第三混合物混合,持续升温后加入第四混合物,经搅拌后加入第二混合物,混匀后即得所述组合物。
4.根据权利要求3所述的制备方法,其特征在于,所述大枣在浸泡前经过炒制,所述炒制的温度为50-70℃。
5.根据权利要求3所述的制备方法,其特征在于,所述水提的料液比为1g:(7-9)mL;所述水提的次数为1-3次,每次水提的时间为1-1.5h。
6.根据权利要求3所述的制备方法,其特征在于,所述蜀椒在超微粉碎前经过炒制,所述超微粉碎后第二混合物的粒径≤74μm。
