本发明涉及一种具有抗溃疡性结肠炎功效的组合物及其制备方法,组合物由药材醇提物40‑49份、药材水提物50‑59份及益生菌1‑5份组成;药材包括苦瓜20‑40份、桑叶20‑40份、葛根10‑30份、山药8‑12份、山楂8‑12份。本发明制备方法简单,步骤易于操作,原料来源广泛,能够进行大规模量产;采用醇提和水提的方式将苦瓜、桑叶等原料中的有效成分充分提取,辅以益生菌,组合物表现出良好的抗溃疡性结肠炎功效,具有较好的推广应用前景。

技术领域
本发明涉及一种抗溃疡性结肠炎的组合物及其制备方法,属于天然产物提取与应用技术领域。
背景技术
溃疡性结肠炎是一种临床常见且难治的疾病,其发病机制复杂,临床症状表现为带血的腹泻、腹痛、黏脓性粪便、体重骤降等,整个病程漫漫且反复,目前尚无治疗溃疡性结肠炎的理想方案。内服中药能够有效减轻肠粘膜屏障损伤程度,苦瓜、桑叶、葛根、山药、山楂是临床常用的清热养阴中药组方,前期研究发现,该配方具有明显的降血糖和润肠通便的效果,本发明通过对该配方的进一步研究,对其抗溃疡性结肠炎进行研究分析,为使其应用价值进一步推广,提高产品的竞争力提供研究基础。
发明内容
本发明是为了解决上述问题,提供了一种抗溃疡性结肠炎的组合物及其制备方法。
本发明采用如下技术方案:一种具有抗溃疡性结肠炎功效的组合物,组合物由药材醇提物40-49份、药材水提物50-59份及益生菌1-5份组成;
所述药材包括苦瓜20-40份、桑叶20-40份、葛根10-30份、山药8-12份、山楂8-12份。
进一步的,所述益生菌为婴儿双岐杆菌、嗜酸乳杆菌或植物乳杆菌中的一种或几种。
一种具有抗溃疡性结肠炎功效的组合物的制备方法,包括如下步骤:
(1) 药材醇提物的制备:将药材原料按重量份数:苦瓜20-40份、桑叶20-40份、葛根10-30份、山药8-12份、山楂8-12份,混合后粗粉碎,按料液比1:10~15kg/L加入70-75%乙醇溶液充分搅拌,浸提1-2次,结束分离药液和药渣,药液于50-60℃下减压浓缩,喷雾干燥制成粉末;
(2)药材水提物的制备:将步骤(1)中醇提后所剩余药渣,按照料液比1:10~15kg/L加入去离子水充分搅拌,离心分离药液,将药液60-90℃减压浓缩,喷雾干燥制成粉末得到药材水提物;
(3)混合:按重量份数计将药材醇提物40-49份、药材水提物50-59份及益生菌1-5份混合搅拌均匀。
进一步的,所述步骤(1)中喷雾温度为150-180℃,喷雾压力为0.4-0.5Mpa。
进一步的,将所述步骤(1)中剩余药渣经过50-80℃烘干处理后用于步骤(2)中药材水提物的制备。
苦瓜,其苦味独特,同时也是一味药食同源的中药,含有生物碱、皂苷、多肽、维生素和矿物质等多种生物活性化合物,可为人体提供基本营养素、预防及治疗各种疾病,全株均可入药。传统的本草书籍中曾记载,苦瓜味苦,性微寒;归肺、心、脾三经,清热泻火、除烦止渴、清晷益气、清心明目;主治热病烦渴引饮,中暑,赤眼疼痛,痢疾等;外用治疗疔疮疖肿等。现代研究发现,苦瓜除了在传统中药理论中有良好的清热解毒效果外,也能治疗糖尿病、腹痛、肾脏结石、发烧和疥疮等,也可作为药膳辅助疾病的治疗,而且不良反应少,用药更为安全。
桑叶是国家***指定的“药食同源”材料。在最早的药学专著《神农本草经》中记载: 桑叶“除寒热”《本草纲目》中称桑叶“治劳热咳嗽,明目”。现代药理学研究发现,桑叶提取物中含有黄酮类、生物碱类、植物甾醇、γ-氨基丁酸及桑叶多糖等多种活性化学成分,桑叶、葛根、山楂等亦在抗氧化、抑制β细胞凋亡、减轻胰岛素抵抗、调节肠道菌群及延缓糖尿病发生发展中具有重要功能。葛根、山药、山楂含有大量黄酮类化合物,具有良好的抗炎作用。
本发明制备方法简单,步骤易于操作,原料来源广泛,能够进行大规模量产;采用醇提和水提的方式将苦瓜、桑叶等原料中的有效成分充分提取,辅以益生菌,组合物表现出良好的抗溃疡性结肠炎的功效,具有较好的推广应用前景。

实施例1:
一种具有抗溃疡性结肠炎功效的组合物:组合物由药材醇提物40份、药材水提物59份及婴儿双岐杆菌1份组成
制备方法,包括如下步骤:
(1) 药材醇提物的制备:将药材原料按重量份数:苦瓜20份、桑叶20份、葛根30份、山药12份、山楂12份,混合后粗粉碎,按料液比1:12kg/L加入75%乙醇溶液充分搅拌,浸提1次,结束分离药液和药渣,药液于60℃下减压浓缩,喷雾干燥制成粉末,喷雾温度为150℃,喷雾压力为0.4Mpa;
(2)药材水提物的制备:将步骤(1)中醇提后所剩余药渣,经过50℃烘干处理后,按照料液比1:12kg/L加入去离子水充分搅拌,离心分离药液,将药液80℃减压浓缩,喷雾干燥制成粉末得到药材水提物,喷雾温度为150℃,喷雾压力为0.4Mpa;
(3)混合:按重量份数计将药材醇提物40份、药材水提物59份及益生菌1份混合搅拌均匀。
实施例2:
一种具有抗溃疡性结肠炎功效的组合物,组合物由药材醇提物45份、药材水提物52份及婴儿双岐杆菌3份组成;
制备方法,包括如下步骤:
(1) 药材醇提物的制备:将药材原料按重量份数:苦瓜30份、桑叶30份、葛根20份、山药10份、山楂10份,混合后粗粉碎,按料液比1:10kg/L加入75%乙醇溶液充分搅拌,浸提2次,结束分离药液和药渣,药液于50℃下减压浓缩,喷雾干燥制成粉末,喷雾温度为180℃,喷雾压力为0.5Mpa;
(2)药材水提物的制备:将步骤(1)中醇提后所剩余药渣,经过60℃烘干处理后,按照料液比1: 15kg/L加入去离子水充分搅拌,离心分离药液,将药液90℃减压浓缩,喷雾干燥制成粉末得到药材水提物,喷雾温度为180℃,喷雾压力为0.5Mpa;
(3)混合:按重量份数计将药材醇提物45份、药材水提物52份及婴儿双岐杆菌3份混合搅拌均匀。
实施例3:一种具有抗溃疡性结肠炎功效的组合物:组合物由药材醇提物49份、药材水提物50份及婴儿双岐杆菌1份组成。
制备方法包括如下步骤:
(1) 药材醇提物的制备:将药材原料按重量份数:苦瓜40份、桑叶40份、葛根10份、山药8份、山楂8-12份,混合后粗粉碎,按料液比1:10kg/L加入75%乙醇溶液充分搅拌,浸提1-2次,结束分离药液和药渣,药液于55℃下减压浓缩,喷雾干燥制成粉末,喷雾温度为180℃,喷雾压力为0.5Mpa;
(2)药材水提物的制备:将步骤(1)中醇提后所剩余药渣,经过80℃烘干处理后,按照料液比1:10kg/L加入去离子水充分搅拌,离心分离药液,将药液90℃减压浓缩,喷雾干燥制成粉末得到药材水提物,喷雾温度为180℃,喷雾压力为0.5Mpa;
(3)混合:按重量份数计将药材醇提物49份、药材水提物50份及婴儿双岐杆菌1份混合搅拌均匀。
实施例4:一种抗溃疡性结肠炎的组合物及其制备方法:
药材醇提物、药材水提物的制备方法,同实施例1。取药材醇提物40份、药材水提物55份及婴儿双岐杆菌5份得到组合物。
抗溃疡性结肠炎的效果研究
以实施例1制备得到的组合物,研究组合物对DSS大鼠的保护作用。
研究方法:
研究选取雄性SD大鼠,分为空白组、模型组、组合物高剂量组、组合物低剂量组,每组各6只。空白组正常饮水,模型组及两组给药组以3%的葡聚糖硫酸钠(DSS)溶液代替日常饮水以诱导溃疡性结肠炎模型;高、低剂量给药组每日按每百克体重1ml的药量分别灌胃0.05g/ml、0.025g/ml浓度的组合物,空白组及模型组每日灌胃等量生理盐水,连续7天。记录患病动物的体重下降、大便黏稠度和大便出血评分,计算疾病活动指数(DAI): DAI=(体重指数+大便形状+出血情况)/3。记录脾脏指数:脾脏指数=脾脏重量/体重。HE染色观察病理组织变化,AB-PAS染色检测黏蛋白。
研究结果:
结果见表1。

由表1的DAI指数结果可知,相比于空白组,模型组大鼠的疾病活动指数有明显的升高,在两种剂量的组合物给药治疗后,DAI指数的升高被明显遏制。本发明制备得到的组合物给药能明显改善溃疡性结肠炎大鼠的患病症状。
(2) HE染色结果见图1;
由图1可知,相比正常组大鼠,模型组在造模之后出现部分隐窝结构缺失,并有大量炎症浸润,给药组给药治疗之后,相比于模型组,隐窝结构完整,炎症浸润程度降低,杯状细胞数量较多。组合物给药可以明显改善溃疡性结肠炎所致的炎症浸润和隐窝结构缺失。
(3)AB-PAS染色结果见图2
由图2可知,与空白组相比,模型组的结肠黏液面积较小且颜色较浅,组合物黏蛋白染色结果有所改善并趋近于空白组。
(4)脾脏指数图结果见图3
由图3可知,与空白组相比,模型组的脾脏指数有明显的上升,组合物低剂量组大鼠的脾脏指数有下降的趋势,组合物高剂量组大鼠脾脏指数显著下降,组合物可以使溃疡性结肠炎大鼠的免疫情况更趋近于正常值。

(5) 由图4可知,与空白组相比,模型组的厚壁菌门/拟杆菌门(F/B) 值明显升高,组合物高剂量处理后,大鼠F/B值显著降低,低剂量组合物处理后F/B值出现降低趋势。表明发明组合物可以改善溃疡性结肠炎发生后肠道内菌群微生物失衡的情况。

1.一种具有抗溃疡性结肠炎功效的组合物,其特征在于:组合物由药材醇提物40-49份、药材水提物50-59份及益生菌1-5份组成;
所述药材由苦瓜20-40份、桑叶20-40份、葛根10-30份、山药8-12份、山楂8-12份组成,所述益生菌为婴儿双岐杆菌。
2.一种具有抗溃疡性结肠炎功效的组合物的制备方法,其特征在于:包括如下步骤:
(1) 药材醇提物的制备:将药材原料按重量份数:苦瓜20-40份、桑叶20-40份、葛根10-30份、山药8-12份、山楂8-12份,混合后粗粉碎,按料液比1:10~15kg/L加入70-75%乙醇溶液充分搅拌,浸提1-2次,结束分离药液和药渣,药液于50-60℃下减压浓缩,喷雾干燥制成粉末;
(2)药材水提物的制备:将步骤(1)中醇提后所剩余药渣,按照料液比1:10~15kg/L加入去离子水充分搅拌,离心分离药液,将药液60-90℃减压浓缩,喷雾干燥制成粉末得到药材水提物;
(3)混合:按重量份数计将药材醇提物40-49份、药材水提物50-59份及婴儿双岐杆菌1-5份混合搅拌均匀。
