Description

Alcohol-free mouth wash and preparation method thereof
Technical Field
The invention belongs to the technical field of mouthwash, and particularly relates to alcohol-free mouthwash and a preparation method thereof.
Background
The oral health is one of the signs of human health and also one of the important signs of civilization in the modern society. Food scraps and the like remained in the oral cavity provide proper conditions for breeding of oral bacteria, so that a series of oral diseases such as dental pulp diseases, periodontal diseases and the like can be caused, in addition, oral odor also brings inconvenience to people, and brings more embarrassing experiences in work and life, and particularly, people who smoke for a long time have higher expectation and demand on breath freshening products. The mouthwash has better mobility and permeability compared with traditional oral care products, can clean any position in the oral cavity all-round in the oral cavity to more effective clean nursing to the oral cavity, in addition, contains antibacterial component in the mouthwash also can prevent the emergence of periodontal disease. At present, traditional oral care products represented by toothpaste and dental floss still occupy the market leading position, but with the improvement of living standard, the mouth wash which is deeply loved by consumers abroad is gradually popularized in China.
Most of the existing mouthwash products in the market have active ingredients mainly comprising chemical ingredients, and the mouthwashes taking natural active substances as main raw materials are rare, but most of the existing mouthwash products contain alcohol, and the effect of cleaning the oral cavity is achieved by adding other auxiliary materials and surfactants, so that the alcohol-containing mouthwashes are obviously not suitable for alcohol allergic people, drivers, overhead workers and other special groups; in addition, the existing mouthwash products mainly play a role in cleaning oral cavities, the quantity of the mouthwash with the antibacterial effect is limited, the market space is large, and the unique smell of the natural active substances also has unique advantages for refreshing breath.
Blumea balsamifera (L.). DC) is a herbaceous plant of Blumea genus of Compositae family, and is mainly distributed in provinces such as Guizhou, Yunnan, Guangxi, Guangdong, Fujian and Taiwan areas in China, wherein Blumea balsamifera produced in the Guizhou Ludian obtains a national geographical sign product. The blumea balsamifera mainly contains chemical components such as borneol, camphor, aromatic camphor, longifolene, caryophyllene oxide, myrcene and the like, and is pungent and bitter in taste. The blumea balsamifera root can be used as a medicine, and has remarkable pharmacological activities of analgesia, anti-inflammation, antibiosis, hemostasis and the like. The drug blumea balsamifera tablets collected in the 2020 edition of Chinese pharmacopoeia use blumea balsamifera as the only source. Blumea balsamifera is widely applied in the field of medicines and daily chemical products, is mainly used for treating scalds and infantile eczema, and products of daily chemical products taking blumea balsamifera as a raw material, such as toothpaste, soap and the like, are also reported in documents.

Solid medium: weighing 1.0g of yeast extract powder, 2.0g of tryptone, 2.0g of sodium chloride and 2.5g of agar, placing the yeast extract powder, adding 200ml of distilled water into a conical flask, heating, stirring for dissolving, adjusting the pH value to about 7.3 by using NaOH with the concentration of 1mol/L, covering by using a silicon rubber plug, and wrapping the bottleneck of the conical flask by using newspaper.
Sterilizing, namely wrapping the culture dish prepared in advance with newspaper, and sterilizing the wrapped culture dish and the liquid culture medium in a high-pressure steam sterilizing pot at the temperature of 121 ℃ for 20 min.
And (4) pouring the plate, namely putting the sterilized instrument and the culture solution in an oven to dry the moisture. And (5) opening an ultraviolet lamp of the ultra-clean bench to sterilize the ultra-clean bench for about 15 min. After the sterilization of the super clean bench is finished, the instruments and the culture medium in the oven are moved into the super clean bench and are subjected to ultraviolet sterilization again for about 15 min. Wearing gloves, wiping the gloves and the super clean bench surface by alcohol cotton soaked by 75% alcohol, igniting an alcohol lamp after a culture medium is not scalded, starting to pour the plate, shaking the liquid culture medium uniformly when each plate is poured, and burning the mouth of the conical flask by using outer flame of the alcohol lamp to perform local sterilization.
And (3) inoculation, namely after the culture medium is completely solidified, dipping the preserved staphylococcus aureus by using an inoculating ring which is burned and cooled, carrying out streak inoculation on the surface of the solid culture medium, and after the inoculation is finished, putting the culture medium in a constant-temperature incubator at 37 ℃ for 24 hours.
The preparation of the bacterial suspension comprises weighing yeast extract powder 0.5g, tryptone 1.0g and sodium chloride 1.0g, placing in a conical flask, adding 100ml distilled water, heating, stirring for dissolving, adjusting pH to about 7.3 with 1mol/L NAOH, covering with a silicon rubber plug, and wrapping the bottle mouth with newspaper. And (3) sterilizing, after the liquid culture medium is cooled, lightly scraping staphylococcus aureus growing on the solid culture medium into the liquid culture medium by using an inoculating loop after burning and cooling in a super clean bench, and placing the liquid culture medium in a constant-temperature culture shaker at the temperature of 37 ℃ and the rotating speed of 180rpm for culturing for 24 hours.
Antibacterial experiment, preparing the solid culture medium, wrapping with blumea balsamifera extract, pipette tip, coater, culture dish, 2 layers of filter paper sheets with diameter of 6mm and beakers with newspaper paper, and sterilizing in a high-pressure steam sterilization kettle at 121 deg.C for 20 min. After the sterilization is finished, putting the sterilized instruments and the culture medium into an oven, and drying the water. Pouring the plate in a clean bench, standing for the culture medium to solidify, soaking 6 sterilized filter paper sheets with diameter of 6mm in 6 parts of blumea balsamifera extract with different concentrations of 125.00mg/ml, 62.50mg/ml, 31.25mg/ml, 15.63mg/ml, 7.81mg/ml and 3.91mg/m l, numbering No. 1, No. 2, No. 3, No. 4, No. 5 and No. 6 in sequence, taking one part of the filter paper sheets soaked with 75% ethanol as a blank control group, and soaking in the blumea balsamifera extract for 10 min. And transferring 60 mu l of bacterial suspension on a solid culture medium by using a liquid transfer gun, uniformly smearing the bacterial suspension by using a cooled spreader after firing, clamping and sticking the soaked filter paper piece on the solid culture medium coated with the bacterial suspension by using forceps, slightly pressing, and then putting in a constant-temperature incubator at 37 ℃ for culturing for 24 hours.
