Abstract
本发明公开了一种含抗氧化物质的隐形眼镜护理液及其制备方法,属于眼睛保健领域。含有以下各组分的水溶液:VC 0.01‑0.2wt%、麦角硫因0.01‑0.2wt%、保湿剂0.1‑2wt%、抗菌剂0.01‑0.5wt%、缓冲剂0.05‑0.95wt%、表面活性剂0.01‑0.1wt%、渗透压调节剂0.35‑1.0wt%,注射用水93‑98wt%;所述隐形眼镜护理液的pH值为6‑8。本发明提供的隐形眼镜护理液,采用发酵工艺所得无毒副作用的VC和麦角硫因作为抗氧化剂,使用所述隐形眼镜护理液能够提升所佩戴隐形眼镜的抗氧化性能,保护眼睛抵御自由基损伤,并具有抗炎修复作用,提升佩戴舒适度。

技术领域
本发明涉及一种含抗氧化物质的隐形眼镜护理液及其制备方法,属于眼睛保健领域。
背景技术
世界卫生组织研究报告显示,中国近视患者人数达6亿,青少年总体近视率达50%以上,中国已成为世界第一近视大国。眼健康问题关系到青少年健康成长,也是关系国家未来的大问题。目前近视患者绝大多数采用光学矫正,光学矫正包括框架眼镜和隐形眼镜,隐形眼镜是根据人眼角膜的形态制成,直接附着在角膜的泪液层上,因其轻便、美观、视物逼真而广受近视患者的青睐和使用。但隐形眼镜会使角膜无法接触空气,长时间佩戴使眼睛无法正常代谢,会因缺氧导致眼部自由基增加从而产生眼干涩疲劳、抵抗力下降,如果隐形眼镜保养不到位,还会引起细菌等在镜片上滋生,长时间佩戴就会引发眼球过敏、视神经麻痹、角膜损伤等一系列的眼健康问题。因此需要一款具有清除自由基,同时具有杀菌消炎成分的隐形眼镜护理液。
发明内容
现有技术中隐形眼镜护理存在液不能有效清除长期佩戴隐形眼镜导致眼睛缺氧所致氧化损伤、且抗菌消炎效果差的技术问题。
鉴于此,本发明提供一种含抗氧化物质的隐形眼镜护理液,使用所述隐形眼镜护理液能够及时清除佩戴隐形眼镜角膜缺氧而产生自由基,且能够改善长期佩戴隐形眼镜引起的红肿、炎症等现象。
具体的,本发明提供了一种隐形眼镜护理液,以所述隐形眼镜护理液的总质量为基准,所述隐形眼镜护理液为含有以下各组分的水溶液:
维生素C:0.01-0.5%;
麦角硫因:0.01-0.2%;
保湿剂:0.1-2%;
杀菌剂:0.01-0.5%
缓冲剂:0.05-0.95%;
表面活性剂:0.01-0.1%;
渗透压调节剂:0.35-1.0%
所述隐形眼镜护理液的pH值为6-8。
在本发明的实施方式中,所述保湿剂包括依克多因、玻璃酸钠、甲基纤维素或羟丙基甲基纤维素(HPMC)中的一种或多种,优选依克多因和玻璃酸钠。
在本发明的实施方式中,所述缓冲剂包括硼酸、硼砂、硼酸钠、磷酸氢二钠、乙二胺四乙酸二钠中的一种或多种,优选硼酸、硼酸钠。
在本发明的实施方式中,所述表面活性剂可采用现有技术中常用的各种表面活性剂,如可选自泊洛沙姆、癸基葡萄糖苷、甘油酯、硫代甜菜碱等,优选为泊洛沙姆407。
在本发明的实施方式中,所述的抗菌剂选自甘草酸二钾、聚亚己基缩二胍、山梨酸中的一种或多种,优选为甘草酸二钾。
在本发明的实施方式中,所述渗透压调节剂可采用现有技术中常用的各种等渗调节剂,如可选自氯化钠、氯化钾或氯化钙中的一种或多种。优选情况下,所述等渗调节剂为氯化钠。
在本发明的一种实施方式中,所述隐形眼镜护理液的pH优选为6.5-7.5。
在本发明的一种实施方式中,维生素C的含量优选为0.02-0.1%。
在本发明的一种实施方式中,麦角硫因的含量优选为0.02-0.1%。
在本发明的一种实施方式中,所述隐形眼镜护理液的渗透压为260-320osmol/Kg。
本发明还提供了所述隐形眼镜护理液的制备方法,所述方法包括:将维生素C、麦角硫因、保湿剂、抗菌剂、表面活性剂按比例混合后溶解注射于水中,缓冲剂调节pH值为6-8,渗透压调节剂调节渗透压至260-320osmol/kg得到所述隐形眼镜护理液。
本发明于现有技术对比,具有以下优点:
本发明中,与现有的隐形眼镜护理液相比,本发明添加了维生素C和麦角硫因用作隐形眼镜护理液的抗氧化剂,VC不仅具有清除氧自由基的作用,还是晶状体的必须微量元素,对维持晶状体的透明有重要作用。
本发明中的抗氧化物质为维生素C和麦角硫因的组合物,麦角硫因作为天然抗氧化剂在眼部组织中含量较高,具有保护眼部神经的功效,麦角硫因的联合使用不仅扩宽了VC的抗氧化谱,而且能够阻断VC的氧化来维持VC的功效,二者协同作用能够同时清除多种自由基保护眼睛免受氧化损伤。
另外,本发明添加的抗氧化剂VC及麦角硫因,为眼部组织的固有成分,对人体无毒副作用,同时还具有消炎、营养眼部神经的作用,且本发明提供的隐形眼镜护理液的制备方法,工艺简单,易于实施。

下面结合实施例对本发明作进一步的描述,但本发明的实施方式不限于此。
本发明提供了一种隐形眼镜护理液,以所述隐形眼镜护理液的总质量为基准,所述隐形眼镜护理液为含有以下各组分的水溶液:
维生素C:0.01-0.5%;
麦角硫因:0.01-0.2%;
保湿剂:0.1-2%;
杀菌剂:0.01-0.5%;
缓冲剂:0.05-0.95%;
表面活性剂:0.01-0.1%;
渗透压调节剂:0.35-1.0%;
所述隐形眼镜护理液的pH值为6-8。
本发明提供的隐形眼镜护理液,通过采用维生素C和麦角硫因联合使用作为抗氧化剂,能同时清除氧自由基和多种自由基,减少眼睛因佩戴隐形眼镜缺氧导致的氧化损伤;另外,通过采用依克多因、玻璃酸钠等保湿润滑剂提升佩戴眼镜的舒适度。VC和麦角硫作为眼部组织的固有和必需成分,无毒副作用减少对眼睛的刺激,同时还具有消炎、营养眼部神经的作用。
VC作为我们人体必需的微量元素,参与体内氧化还原反应,能够清除氧自由基的作用,素有抗氧化之王之称;维生素C还是晶状体的重要营养成分,减弱紫外线与氧气对眼睛晶状体的损害,同时VC安全性高,对眼睛无刺激性,补充VC可有效预防眼部疾病,VC在保护眼睛健康中发挥重要作用。
优选情况下,以所述隐形眼镜护理液的总质量为基准,VC的含量为0.02-0.1wt%。
麦角硫因,为水溶性天然氨基酸,具有强大的抗氧化性能。麦角硫因受体在人体许多组织中广泛分布,在眼部组织中含量较高,是眼部组织的主要抗氧化剂。研究表明麦角硫因对视神经细胞具有保护作用。VC作为抗氧化剂的主要缺点之一是易被氧化变色。本发明中,麦角硫因与VC联合使用,在扩宽抗氧化谱的同时,麦角硫因进一步能够阻断VC的氧化来维持VC的功效。
优选情况下,以所述隐形眼镜护理液的总质量为基准,麦角硫因的含量为0.02-0.1wt%。
本发明中,所述保湿剂用于隐形眼镜护理液的保湿润滑作用,缓解佩戴隐形眼镜时的干涩不适症状,选自依克多因、玻璃酸钠、羟丙基甲基纤维素(HPMC)中的一种或多种,其中依克多因除具有亲水锁水特质外还具有强大的修复功效,优选依克多因,以所述隐形眼镜护理液的总质量为基准,保湿剂的含量优选为0.2-1wt%。
在本发明中,所述的抗菌剂选自甘草酸二钾、聚亚己基缩二胍、山梨酸中的一种或多种,具有抑制多种细菌及真菌生长的作用,优选为甘草酸二钾,以所述隐形眼镜护理液的总质量为基准,杀菌剂的含量为0.01-0.5wt%。
本发明中,所述缓冲剂可采用现有技术中常用的各种缓冲剂,例如可以选自硼酸、硼酸钠、磷酸氢二钠、乙二胺四乙酸二钠中的一种或多种,但不局限于此。本发明中,所述缓冲剂优选硼酸、硼酸钠用于保证体系的pH值为6-8,优选情况下,所述隐形眼镜护理液的pH值为6.5-7.5,优选情况下,以所述隐形眼镜护理液的总质量为基准,缓冲剂的含量0.05-0.95wt%。
本发明中,通过采用等渗调节剂,从而调节所述隐形眼镜护理液的渗透压为260-320osmol/Kg。所述等渗调节剂可采用现有技术中常用的各种等渗调节剂,如可选自氯化钠、氯化钾或氯化钙中的一种或多种。优选情况下,所述等渗调节剂为氯化钠,等渗调节剂的含量为0.35-1.0wt%。
本发明中,通过添加表面活性剂降低隐形眼镜护理液的表面张力,所述表面活性剂可采用现有技术中常用的各种表面活性剂,如可选自泊洛沙姆、癸基葡萄糖苷、甘油酯、硫代甜菜碱等,优选为泊洛沙姆407。优选情况下,以所述隐形眼镜护理液的总质量为基准,表面活性剂的含量为0.01-0.1wt%。
本发明提供的隐形眼镜护理液的制备方法,工艺简单,易于实施。只需将上述各组分溶解分散于水中,缓冲剂调节体系pH值为6-8,渗透压调节剂调节渗透压为260-320osmol/Kg,检验合格后即可进行分装,密封保存。
为了更清楚的阐述本发明的整体构思,下面将结合实施例,对本发明进行进一步详细说明。实施例及对比例中所采用原料均通过商购得到。其中本发明中的抗氧化剂麦角硫因和保湿剂依克多因均为深圳中科欣扬生物科技有限公司产品。
实施例
按表1所示的比例将各组分溶解于注射水形成混合溶液,过滤后得到实施例S1-S5的隐形眼镜护理液S1-S5。表1中均为质量百分比含量,余量为水。
对比例C1-C3采用与实施例S1-S5相同的步骤制备隐形眼镜护理液样品,不同之处在于各组分配比,具体如表1所示。

细胞毒性测试
将浓度为1×105细胞/mL的角膜基质细胞悬浮液以100μl/孔加入96孔板中,在含有5%CO2、37℃培养箱中培养24小时将培养基吸出。而后将配方S1-S5及C1-C3与完全培养基按1:10比例加入96孔板中细胞继续培养24h,培养箱继续培养24小时。阳性(10%DMSO)、阴性(完全细胞培养液)对照组与隐形眼镜护理液稀释液皆设置6个重复。加20μl/孔MTT继续孵化3h。而后加入100μl DMSO振荡10min于酶标仪上于570nm波长测定吸光值,细胞活性计算:细胞活性=(受试物孔OD值-空白对照孔OD值)/阴性对照孔OD值。
细胞毒性测试结果显示:实施例S1-S5及对比例C1-C3组合物培养角膜基质细胞24h,细胞存活率在95%以上,说明所选的实施例配方及对比例配方组合物对角膜细胞是安全无毒的。
抗氧化效果测试
佩戴隐形眼镜缺氧导致的ROS自由基产生过多特别是超氧阴离子(O2-)的积累,VC可以清除超氧阴离子及羟自由基,保护眼睛免受ROS的损伤。
麦角硫因是具有强大的抗氧化性能的稀有天然氨基酸,补充麦角硫因可显著提高机体对氧化应激的预防,麦角硫因有助于保护眼睛免受氧化损伤具有保护眼部神经的功效。
超氧阴离子通过邻苯三酚自氧化法来测定,羟自由基清除实验采用Fenton比色法,DPPH自由基清除法是使用最广泛的间接检测所测物质抗氧化能力的方法。以上反应通过检测吸光度的变化判断所测物质的抗氧化性能。结果如图1-3所示。
抗氧化结果显示:VC具有较强的超氧阴离子和羟自由基清除性能,麦角硫因具有很强的DPPH自由基清除性能,二者复配不仅可以扩展抗氧化谱,还可以提升对DPPH自由基和羟自由基的清除率。
抑菌效果测试
以大肠埃希菌、金黄色葡萄球菌、铜绿假单胞菌、白色念珠菌为试验菌,测试S1-S5的隐形眼镜护理液对4种菌株的抗菌效果。
4种菌株分别培养2天使菌液浓度达106cfu/ml。取0.1ml菌液加到实施例S1-S5及对比例C1-C3的眼科用组合物中,充分混合使菌完全分散。细菌组(大肠杆菌、金黄色葡萄球菌、铜绿假单胞菌)25℃培养,真菌组(白色念珠菌)置于35℃培养。第3-7天,取1ml样品液,依次稀释103、102、101倍计数菌量,以评估抗菌效果。
从表2可知,本发明实施例及对比例制备得到的隐形眼镜护理液对于4种菌株均具有良好的抗菌效果。

使用效果评价
随机抽取42名因长期佩戴隐形眼镜引起眼睛干涩、疲劳、炎症等不适症状的志愿者,男女不限,分为6组。试戴经过S1-S5和C1-C3浸泡的隐形眼镜,连续测试3天,对测试结果进行评分,评价标准为:1分-差,2分-较差,3分-一般,4分-较好,5分-很好。测试结果如表3所示。

使用效果显示:含有抗氧化剂VC和麦角硫因的隐形眼镜护理液可以显著缓解佩戴长期隐形眼镜导致的角膜缺氧所致的眼睛干涩疲劳和炎症等不适症状,提升佩戴舒适度。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。

1.一种隐形眼镜护理液,其特征在于,包括维生素C、麦角硫因、保湿剂、缓冲剂、表面活性剂、渗透压调节剂和注射水。
2.根据权利要求1所述的隐形眼镜护理液,其特征在于,以所述隐形眼镜护理液的总质量为基准,所述隐形眼镜护理液为含有以下各组分的水溶液:
维生素C:0.01-0.5%;
麦角硫因:0.01-0.2%;

3.根据权利要求2所述的隐形眼镜护理液,其特征在于,所述的维生素C为L-抗坏血酸,维生素C的含量为0.02-0.1%。
4.根据权利要求2所述的隐形眼镜护理液,其特征在于,以所述隐形眼镜护理液的总质量为基准,麦角硫因的含量为0.02-0.1%。
5.根据权利要求1~4任一项所述的隐形眼镜护理液,其特征在于,以所述保湿剂包括依克多因、玻璃酸钠、甲基纤维素或羟丙基甲基纤维素(HPMC)中的一种或多种,优选依克多因和玻璃酸钠。
6.根据权利要求1~4任一项所述的隐形眼镜护理液,其特征在于,所述缓冲剂包括硼酸、硼砂、硼酸钠、磷酸氢二钠、乙二胺四乙酸二钠中的一种或多种,优选硼酸、硼酸钠。
7.根据权利要求1~4任一项所述的隐形眼镜护理液,其特征在于,所述表面活性剂可选自泊洛沙姆、癸基葡萄糖苷、甘油酯、硫代甜菜碱,优选为泊洛沙姆407。
8.根据权利要求1~4任一项所述的隐形眼镜护理液,其特征在于,所述的抗菌剂选自甘草酸二钾、聚亚己基缩二胍、山梨酸中的一种或多种所述渗透压调节剂为氯化钠、氯化钾、氯化钙中的一种或多种。
9.权利要求1~8任一项所述的隐形眼镜护理液的制备方法,其特征在于,将维生素C、麦角硫因、保湿剂、抗菌剂、表面活性剂按比例混合后溶解注射于水中,缓冲剂调节pH值为6-8,渗透压调节剂调节渗透压至260-320osmol/kg得到所述隐形眼镜护理液。
10.权利要求1~8任一项所述的隐形眼镜护理液在隐形眼镜领域的应用。
