According to an embodiment of the invention, the lyophilized formulation comprises: the composition comprises 3-10 parts by weight of active ingredients (for example, 5 parts, 7 parts or 9 parts), 10-60 parts by weight of hydrophilic agents (for example, 15 parts, 20 parts, 25 parts, 30 parts, 35 parts, 40 parts, 45 parts, 50 parts or 55 parts), and 1-30 parts by weight of binding agents (for example, 3 parts, 5 parts, 8 parts, 10 parts, 15 parts, 20 parts or 25 parts), wherein the active ingredients comprise at least one selected from a first active ingredient, a second active ingredient and a third active ingredient, the first active ingredient is an active substance for preventing and/or treating dentin damage, the second active ingredient is an active substance for preventing and/or treating dental plaque generation, and the third active ingredient is an active substance for preventing and/or treating inflammation of periodontal tissues and/or damage of dental tissues.
The inventor finds that the freeze drying causes less damage to substances, especially the loss of heat-sensitive nutritional factors in the drying process is less than that of the heat treatment process for the drying purpose, the biological activity of related natural source components can be ensured to the maximum extent, and the loss of the component activity caused in the production process can be reduced; furthermore, due to the lack of moisture in the lyophilized formulation, the possibility of contamination of the product by microorganisms and related pathogens is minimized, which may increase the shelf life of the product and reduce the quality risk of the product to some extent. Moreover, the freeze-dried preparation can form microscopic pores due to sublimation of ice crystals in the freeze-dried process, so that the freeze-dried preparation can be rehydrated (recombined) more quickly and easily, the advantage of rehydration and dissolution in water is obvious, the dissolution effect is good, the active ingredients can be better applied to periodontal sites, and the loss of the active ingredients due to overlong retention time of the active ingredients in the oral cavity can be avoided. In addition, under the current daily care scene, the related oral care modes provided for different periodontal disease onset stages are mostly the same, the general care mode can cause further deterioration of the disease condition of a patient, and the composition of the active ingredients can be flexibly selected according to actual conditions by adopting the freeze-dried preparation of the invention.
In conclusion, according to the embodiment of the application, by controlling the ratio of the components of the freeze-dried preparation, the composition of the active ingredients can be selected according to actual needs such as the onset stage of the periodontal disease, so that the freeze-dried preparation has a targeted positive effect on prevention and treatment of different periodontal diseases, and the loss of the active ingredients can be reduced by adopting the freeze-dried preparation, and the selection range of the freeze-dried preparation on the selectable active ingredients can be widened. Simultaneously, this product of making by freeze-drying process can also reach better instant effect and nursing effect, can dissolve in solution such as water fast, and portable, the solution is taken conveniently, can make active ingredient fully flow everywhere to the oral cavity with its good flow effect during the use, form abundant nursing effect, at portable oral care agent, oral care product fields such as tooth flushing device have extensive application prospect, for example can arrange tooth flushing device and use, utilize tooth flushing device can further send the accurate pump of active ingredient to the oral cavity everywhere, and this freeze-drying preparation can disintegrate fast, can not block up tooth flushing device's pump liquid pipeline or at the inside scale deposit of tooth flushing device water tank because of dissolving inadequately.
The lyophilized formulation for oral care according to the above embodiment of the present invention will be described in detail below.
According to the embodiment of the invention, the inventor finds that the rapid disintegration effect of the freeze-dried preparation can be realized by increasing the proportion of the water-absorbing components, wherein a large amount of hydrophilic factors are rapidly lost in the freeze-drying process, so that the formation of a hydrated micro-molecular microporous structure is facilitated, and the better instant dissolution effect of the freeze-dried preparation can be ensured by controlling the dosage range of the hydrophilic agent in the freeze-dried preparation. Preferably, the amount of the hydrophilic agent used in preparing the lyophilized preparation may be 15 to 40 parts by weight or 15 to 20 parts by weight, etc., based on 100 parts by weight of the lyophilized liquid, whereby the dissolution rate and dissolution effect of the lyophilized preparation in water may be further improved. In addition, it should be noted that the kind of the hydrophilic agent in the present invention is not particularly limited, and may be selected by those skilled in the art according to actual needs, for example, the hydrophilic agent may include, but is not limited to, at least one selected from mannitol, sucrose, maltodextrin, polyethylene glycol, and polypropylene glycol.
According to the embodiments of the present invention, the inventors have found that a binder may be added to a lyophilized preparation in order to provide the lyophilized preparation with certain mechanical properties and physical hardness while maintaining its rapid disintegration properties, wherein the binder may be added in an amount of 1 to 30 parts by weight, such as 2 to 10 parts by weight, based on 100 parts by weight of a lyophilized solution, preferably, the lyophilized preparation may be prepared by controlling the addition amount of the binder in the lyophilized preparation according to the present invention so as to provide the lyophilized preparation with good mechanical properties and physical properties without affecting the instant dissolution effect of the lyophilized preparation. In addition, it should be noted that the kind of the binder in the present invention is not particularly limited, and may be selected by those skilled in the art according to actual needs, and for example, the binder may include, but is not limited to, at least one selected from the group consisting of hydroxyethyl starch, sodium alginate, methyl cellulose, gelatin, gum arabic, xanthan gum, gellan gum, sclerotium rolfsii gum, pullulan and carob gum.
According to the embodiment of the invention, different active substance choices can be provided according to the actual conditions of different periodontal disease users, and the instant dissolution rate of the freeze-dried preparation is greatly improved. Wherein, the different periodontitis development stages refer to one or more of dentin damage, dental plaque formation, inflammation of periodontal tissues and damage of periodontal tissues. The inventors have found that the first active ingredient for use in the dentin damage stage may comprise one or a combination of several selected from hydroxyapatite, sodium fluoride, calcium lactate, milk extract, sugar alcohol, thereby achieving a certain preventive or therapeutic effect against problems such as dental calculus, enamel damage and caries.
