Abstract
The invention provides a plant beverage for improving sleep and a preparation method thereof, and belongs to the technical field of functional beverages. The invention is based on medicinal and edible raw materials (blighted wheat, spina date seed, Chinese date, tuckahoe and liquorice), and the blighted wheat, spina date seed and Chinese date are strictly prepared according to monarch, minister, assistant and guide compatibility, thereby having the effects of nourishing the heart, soothing the nerves and regulating the middle warmer and relieving urgency; the tuckahoe has the effects of promoting water and dampness, strengthening the spleen and calming the heart, and the liquorice has the effect of harmonizing the medicines; meanwhile, the casein hydrolysate peptide and new resource food (gamma-aminobutyric acid and tea theanine) are added to synergistically improve the sleep, so that the sleep-aiding effect is obviously improved, the taste is good, the functions are rich, and the sleep quality of a drinker can be improved.

Technical Field
The invention relates to the technical field of functional drinks, in particular to a plant beverage for improving sleep and a preparation method thereof.
Background
Insomnia means a state of unsatisfactory continuous sleep quality, even affects social functions in the daytime, and is clinically manifested by difficulty in falling asleep, low sleep, easy wakening, early waking, dreaminess, tiredness after waking, or lack of changes in wakefulness, alertness, energy, cognitive function, behavior, emotion, and the like. Insomnia is common and seriously affects normal work and life of people.
Sleep disorders are manifestations of alternating disorders of the normal rhythmicity of sleep and wakefulness. The data show that the incidence rate of the sleep disorder of children is 10% -45%, the incidence rate of the sleep disorder of the elderly is about 20% -60%, and the sleep disorder tends to rise continuously with the age.
At present, most of sleep-aiding products sold in the market at home mainly use sleep-aiding medicines or health-care food taking melatonin as a concept, the biggest problem of taking the sleep-aiding medicines and the melatonin for a long time is that dependence is easy to form, and the dosage of the sleep-aiding products is larger and larger after long-term application, and depression can be aggravated. At present, melatonin, hypnotics and other liquid beverages with sleep improving effect exist in the market, most of the beverages contain one or more non-medicinal and edible raw materials, the functions are single, and the effect of helping sleep cannot be achieved.

The semen Ziziphi Spinosae is dry mature seed of Ziziphus jujuba Mill.var. spinosa (Bunge) Hu ex H.F.Chou belonging to Rhamnaceae. Sweet in nature, sour and neutral; it enters liver, gallbladder and heart meridians; has effects in nourishing heart, nourishing liver, calming heart, tranquilizing mind, arresting sweating, and promoting fluid production, and can be used for treating vexation, insomnia, palpitation, dreaminess, asthenia, hyperhidrosis, body fluid deficiency, and thirst.
The preparation raw materials of the plant beverage for improving sleep provided by the invention comprise 5-15 g of Chinese date, preferably 6-12 g, and more preferably 8g based on 100mL of the plant beverage. The fructus Jujubae is dried mature fruit of Ziziphus jujuba Mill of Rhamnaceae. Sweet in nature and warm; spleen, stomach and heart meridians entered; has the effects of invigorating spleen and replenishing qi, nourishing blood and tranquillizing. Can be used for treating spleen deficiency, anorexia, asthenia, loose stool, and hysteria of women. The red dates are rich in protein, fat, sugar, carotene, B vitamins, vitamin C, vitamin P, calcium, phosphorus, iron, cyclic adenosine monophosphate and other nutritional ingredients.
The preparation raw materials of the plant beverage for improving sleep provided by the invention comprise 1-5 g, preferably 2-3 g, and more preferably 3g of poria cocos based on 100mL of the plant beverage. Poria is dried sclerotium of Wolf of Poria cocos (Schw.) belonging to Polyporaceae. Sweet, bland and mild in nature; it enters heart, lung, spleen and kidney meridians. Has the effects of promoting diuresis, invigorating spleen, and calming heart; can be used for treating edema, oliguria, phlegm and fluid retention, dizziness, palpitation, spleen deficiency, anorexia, loose stool, diarrhea, uneasiness, palpitation, and insomnia.
The preparation raw material of the plant beverage for improving sleep provided by the invention comprises 0.5-3 g, preferably 1-2 g, and more preferably 1g of liquorice based on 100mL of the plant beverage. The Glycyrrhrizae radix is perennial herb of Leguminosae, has mild and sweet nature, and has effects of invigorating spleen and replenishing qi, moistening lung for arresting cough, clearing heat and detoxicating, relieving spasm and pain, and harmonizing the drugs.
The invention combines the blighted wheat, the liquorice and the Chinese date into a whole, has the characteristics of nourishing the heart, regulating the liver and tonifying the heart, further plays the role of nourishing the heart and soothing the nerves, and has mild prescription, liver softening and emergency relief.
The preparation raw material of the plant beverage for improving sleep provided by the invention comprises 0.1-0.4 g of gamma-aminobutyric acid, preferably 0.2-0.4 g, and more preferably 0.3-0.4 g based on 100mL of the plant beverage. Gamma-aminobutyric acid is the most important inhibitory neurotransmitter in the central nervous system of mammals, participates in various neurophysiological activities in organisms, and has certain curative effects on convulsion and insomnia.

The casein hydrolysate peptide can regulate sleep and improve sleep quality, and is mainly characterized by shortening sleep onset period, prolonging sleep time, prolonging slow wave sleep, improving memory and increasing intelligence.
The preparation raw materials of the plant beverage for improving sleep provided by the invention comprise 0.1-0.3 g, preferably 0.15-0.3 g, and more preferably 0.3g of theanine of tea leaves based on 100mL of the plant beverage. The theanine is water soluble non-protein amino acid, and can improve excitability of cerebral cortex and accelerate signal transmission speed between neurons. In addition, theanine has the effects of improving cognition, enhancing immunity, relieving psychological stress, regulating glucose, lipid and protein metabolism, improving sleep, etc.
The preparation raw materials of the plant beverage for improving sleep provided by the invention comprise 3-15 g, preferably 4-12 g, and more preferably 4g of wild jujube concentrated juice based on 100mL of the plant beverage. The present invention is not particularly limited to specific components and contents of the wild jujube juice concentrate, and any commercially available product known in the art may be used.
The preparation raw materials of the plant beverage for improving sleep provided by the invention comprise 1-3 g of sweetener based on 100mL of the plant beverage; the sweetener preferably comprises xylitol, maltitol or white granulated sugar, when the sweetener is xylitol, the preparation raw material preferably also comprises sucralose, and the mass ratio of the sucralose to the xylitol is preferably (0.003-0.006): 1-3), more preferably (0.004-0.006): 1.5-2.5). The sucralose is used as a sweetener and can reach 400-800 times of the sweetness of the sucrose; the stability is high, and the stability is very stable to light, heat and pH; has the characteristics of no energy, high sweetness, pure sweetness, high safety and the like.
The preparation raw material of the plant beverage for improving sleep provided by the invention comprises water based on 100mL of the plant beverage; the amount of the water is such that the amount of the raw materials required to be prepared per 100mL of the plant beverage reaches the above range.
The invention provides a preparation method of the plant beverage in the technical scheme, which comprises the following steps:
mixing fructus Tritici Levis, semen Ziziphi Spinosae, fructus Jujubae, Poria and Glycyrrhrizae radix with water, and decocting to obtain extractive solution;
concentrating the extracting solution, mixing the obtained concentrated solution with the wild jujube concentrated juice, and sequentially standing and separating to obtain a supernatant;

The invention mixes light wheat, spine date seed, Chinese date, tuckahoe and liquorice with water, and carries out decoction extraction to obtain an extracting solution. In the present invention, before the mixing, it is preferable to soak the blighted wheat, the spina date seed, the Chinese date, the poria cocos and the licorice in water (the amount of water exceeds 5cm of the surface of all the raw materials) for 30min, and then wash the soaked materials with water.
The invention has no special limit on the mixing process of the blighted wheat, the spina date seed, the Chinese date, the tuckahoe and the liquorice with water, and the materials are uniformly mixed according to the process known in the field.
In the invention, the number of times of decoction and extraction is preferably 2, the ratio of the mass of water used for each decoction and extraction to the total mass of the blighted wheat, the spina date seed, the Chinese date, the poria cocos and the liquorice is preferably (8-10): 1, more preferably 9:1, and the time for each decoction and extraction is preferably 1-2 hours independently; after each decoction and extraction, the obtained mixed material is preferably screened by a screen of 80-200 meshes, and the extracting solution is collected. After the first decoction extraction is finished, the obtained material residue is preferably mixed with water for the second decoction extraction, and the two extracting solutions are combined for the subsequent steps.
After the extracting solution is obtained, the invention concentrates the extracting solution, mixes the obtained concentrated solution with the wild jujube concentrated juice, and sequentially stands and separates to obtain the supernatant. In the invention, the relative density of the concentrated solution is 1.02 +/-0.0025 g/mL; the concentration process is not particularly limited in the present invention, and the concentration to the above-mentioned relative density may be carried out according to a process known in the art.
The process of mixing the concentrated solution and the wild jujube concentrated juice is not specially limited, and the materials are uniformly mixed according to the process known in the field.
In the invention, the standing temperature is preferably 0-4 ℃, more preferably 2-3 ℃, and the time is preferably 6-12 hours, more preferably 8-10 hours; the separation mode is preferably centrifugal separation, the rotating speed of a centrifugal machine used for centrifugal separation is preferably 10000-17000 r/min, and the liquid inlet speed is preferably 150-200L/h.
After the supernatant is obtained, the plant beverage is obtained by mixing the supernatant, casein hydrolysate peptide, water, gamma-aminobutyric acid, tea theanine and a sweetening agent and sterilizing. In the present invention, the water is preferably purified water; preferably, the process of mixing the supernatant, the casein hydrolysate peptide, the water, the gamma-aminobutyric acid, the theanine in tea leaves, the xylitol and the sucralose is to add the casein hydrolysate peptide into the purified water, stir for 10min while adding the casein hydrolysate peptide, add the supernatant, add the gamma-aminobutyric acid, the theanine in tea leaves and the sweetener into the obtained mixed solution, stir for 20min while adding the casein hydrolysate peptide, the theanine in tea leaves and the sweetener, pump the mixed solution into a 202 stirring tank, add the purified water to a constant volume, and stir for 30min to obtain the preparation solution. In the invention, the dosage of the purified water and the supernatant preferably meets the proportion of preparation raw materials in the plant beverage; the specific volume of the preparation liquid is not specially limited, and the preparation liquid is adjusted according to actual production requirements to meet the mixture ratio of different preparation raw materials in the plant beverage. The stirring process is not particularly limited in the present invention, and the stirring is performed uniformly according to a process known in the art.
In the invention, the sterilization temperature is preferably 105-120 ℃, and the time is preferably 10-30 min; the sterilization process is not particularly limited in the present invention, and may be performed according to a process well known in the art.
The plant beverage is preferably placed in an oral liquid bottle, and the oral liquid bottle is preferably sterilized and dried in sequence and then contains the plant beverage; the temperature of the sterilization is preferably 150 ℃, and the time is preferably 30 min.
In the invention, the plant beverage is preferably filled, inspected by a lamp, packaged, labeled and inspected in sequence according to 30 mL/piece to obtain a finished product. The process of filling, lighting inspection, packaging, labeling and inspection is not particularly limited in the present invention and may be performed according to processes well known in the art.

A plant beverage for improving sleep comprises the following raw materials in parts by mass in a 100mL plant beverage: 9g of blighted wheat, 8g of spina date seed, 8g of Chinese date, 3g of tuckahoe, 1g of liquorice, 0.4g of gamma-aminobutyric acid, 0.125g of casein hydrolysate peptide, 0.3g of theanine, 4g of wild jujube concentrated juice, 2g of xylitol, 0.006g of sucralose and water;
the preparation method comprises the following steps:
weighing fructus Tritici Levis, semen Ziziphi Spinosae, fructus Jujubae, Poria and Glycyrrhrizae radix, adding water (5 cm above the surface of the raw materials), soaking for 30min, and washing with water;
mixing five cleaned raw materials of blighted wheat, spina date seed, Chinese date, tuckahoe and liquorice with water, extracting, adding water for the first extraction, decocting for 2 hours, wherein the mass of the added water is 10 times of the total mass of the five raw materials, sieving the obtained material with a 80-mesh sieve, collecting an extracting solution, and extracting filter residues for the second time; decocting the second extraction with water for 2h, sieving with 80 mesh sieve, collecting extractive solution, and mixing the two extractive solutions;
concentrating the extracting solution to the relative density of 1.02g/mL, adding the wild jujube concentrated juice, uniformly mixing, standing at 4 ℃ for 12h, centrifuging the liquid after standing by a centrifugal machine at the rotating speed of 17000r/min and the liquid inlet speed of 200L/h, and keeping the supernatant;
adding 200L of purified water into a 102 tank, adding casein hydrolysate peptide, and stirring for 10min while adding; adding 400L of the supernatant, respectively adding gamma-aminobutyric acid, theanine, xylitol and sucralose, stirring for 20min while adding, pumping into a 202 stirring tank, adding water to a constant volume, and stirring for 30min to obtain a preparation solution;
cleaning the oral liquid bottles, sterilizing at 150 ℃ for 10min, drying, filling the prepared liquid in each bottle by 30mL, sterilizing the obtained bottled liquid at 105 ℃ for 30min under high pressure, performing light inspection, and sequentially packaging, labeling and inspecting the obtained semi-finished products to obtain the finished plant beverage.

A plant beverage for improving sleep comprises the following raw materials in parts by mass in a 100mL plant beverage: 15g of blighted wheat, 12g of spina date seed, 10g of Chinese date, 4g of tuckahoe, 3g of liquorice, 0.4g of gamma-aminobutyric acid, 0.125g of casein hydrolysate peptide, 0.3g of theanine, 3g of wild jujube concentrated juice, 1g of maltitol and water; the procedure is as in example 1 except that xylitol and sucralose are replaced with maltitol.

A plant beverage for improving sleep, 100mL plant beverage comprises the following raw materials, by mass, 15g of blighted wheat, 12g of spina date seed, 10g of Chinese date, 4g of tuckahoe, 1g of liquorice, 0.4g of gamma-aminobutyric acid, 0.125g of casein hydrolysate peptide, 0.15g of theanine, 3g of wild jujube concentrated juice, 1.5g of white granulated sugar and water; the preparation method is the same as that of example 1, and xylitol and sucralose are replaced by white granulated sugar.
Example 4
A plant beverage for improving sleep, 100mL plant beverage comprises the following raw materials, by mass, 12g of blighted wheat, 8g of spina date seed, 10g of Chinese date, 2g of tuckahoe, 2g of liquorice, 0.4g of gamma-aminobutyric acid, 0.15g of casein hydrolysate peptide, 0.3g of tea theanine, 4g of wild jujube concentrated juice, 1.5g of white granulated sugar and water; the preparation method is the same as that of example 1, and xylitol and sucralose are replaced by white granulated sugar.
Example 5
A plant beverage for improving sleep comprises the following raw materials in parts by mass in a 100mL plant beverage: 9g of blighted wheat, 8g of spina date seed, 8g of Chinese date, 2g of tuckahoe, 1g of liquorice, 0.4g of gamma-aminobutyric acid, 0.125g of casein hydrolysate peptide, 0.3g of theanine, 4g of wild jujube concentrated juice, 1.5g of xylitol and water; the preparation method is the same as example 1, and the process of adding sucralose is abandoned.

Comparative example 2
A plant beverage for improving sleep comprises the following raw materials in parts by mass in a 100mL plant beverage: 9g of blighted wheat, 8g of spina date seed, 8g of Chinese date, 3g of tuckahoe, 1g of liquorice, 0.125g of casein hydrolysate peptide, 4g of wild jujube concentrated juice, 2g of xylitol, 0.006g of sucralose and water; the preparation method is the same as example 1, and the process of adding the gamma-aminobutyric acid and the theanine in the tea leaves is abandoned.
Sensory evaluation
1. Experimental materials: the plant beverages prepared in examples 1 to 5 and comparative examples 1 to 2.
2. The test method comprises the following steps: 70 volunteers were randomly divided into 7 groups, 10 persons in each group were drunk by the volunteers respectively, and the taste was scored as 1-10 points, and the higher the score, the better the taste.
3. The experimental results are as follows: the test results are shown in table 1 below.

From the results in table 1, the consumption of 5 medicinal and edible raw materials and the type of sweetener are important factors influencing the mouthfeel of the plant beverage. Compared with comparative examples 1-2, except for example 2, the number of other examples is more than 8, and the example 2 has slightly bitter taste after adding maltitol, but the taste is better than that of the comparative example 1 and slightly worse than that of the comparative example 2, because the comparative example 2 moderately reduces the consumption of homologous plant food and medicine, the bitter taste of the product is reduced.
Compared with the examples 2-5, the taste score of the example 1 is the highest and is 9.4, which shows that the effect of the taste is better than that of the addition of maltitol or white granulated sugar by adding xylitol; in addition, the taste of the product can be improved by reducing the consumption of medicinal and edible raw materials, so that the consumption of the blighted wheat, the tuckahoe and the liquorice can be reduced without influencing the effect of the product; meanwhile, the taste of the beverage can be further improved by blending the wild jujube concentrated juice.
Survey of crowd's effects
60 insomnia patients are selected as study objects, the study objects are aged 20-60 years and are randomly divided into A, B groups, the two groups of subjects respectively eat the plant beverage (group A) prepared in example 1 and the plant beverage (group B) prepared in comparative example 2 before sleeping every day, the eating amount is the same when 2 beverages (30 mL/beverage) are drunk every night, the period of the experiment is 2 months, the sleep condition performance of the two groups of subjects is observed and recorded, and the specific results are shown in table 2. All subjects keep the original dietary habits during taking, and do not eat other traditional Chinese medicines or western medicines with the sleep-aiding effect.

Note: the sleep condition is considered to be improved obviously, and the sleep condition is considered to be improved slightly but not obvious; and C, the sleep condition is not considered to be improved.
As can be seen from table 2, through evaluation of the subjects, the plant beverage for improving sleep provided in example 1 of the present invention has an obvious sleep-aiding effect compared to comparative example 2, which indicates that the traditional Chinese medicine has a good effect in improving sleep, and the sleep-aiding effect is further improved and better after the gamma-aminobutyric acid and the theanine are added.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
Claims (10)
1. The plant beverage for improving sleep is characterized by comprising the following preparation raw materials based on 100mL of the plant beverage: 5-20 g of blighted wheat, 5-15 g of spina date seed, 5-15 g of Chinese date, 1-5 g of poria cocos, 0.5-3 g of liquorice, 0.1-0.4 g of gamma-aminobutyric acid, 0.05-0.13 g of casein hydrolysate peptide, 0.1-0.3 g of theanine, 3-15 g of wild jujube concentrated juice, 1-3 g of sweetener and water.
2. The plant beverage according to claim 1, wherein the sweetener comprises xylitol, maltitol or white granulated sugar.
3. The plant beverage according to claim 2, wherein when the sweetener is xylitol, the preparation raw material further comprises sucralose, and the mass ratio of the sucralose to the xylitol is (0.003-0.006): 1-3).

The plant beverage according to any one of claims 1 to 3, comprising the following preparation raw materials: 5-20 g of blighted wheat, 5-15 g of spina date seed, 5-15 g of Chinese date, 1-5 g of poria cocos, 0.5-3 g of liquorice, 0.1-0.4 g of gamma-aminobutyric acid, 0.05-0.13 g of casein hydrolysate peptide, 0.1-0.3 g of theanine, 3-15 g of wild jujube concentrated juice, 1-3 g of xylitol, 0.003-0.006 g of sucralose and water.

The plant beverage according to claim 4, which comprises the following preparation raw materials based on 100mL of the plant beverage: 9g of blighted wheat, 8g of spina date seed, 8g of Chinese date, 3g of tuckahoe, 1g of liquorice, 0.4g of gamma-aminobutyric acid, 0.125g of casein hydrolysate peptide, 0.3g of theanine, 4g of wild jujube concentrated juice, 2g of xylitol, 0.006g of sucralose and water.
6. A method for preparing a vegetable beverage according to any one of claims 1 to 5, comprising the steps of:
mixing fructus Tritici Levis, semen Ziziphi Spinosae, fructus Jujubae, Poria and Glycyrrhrizae radix with water, and decocting to obtain extractive solution;
concentrating the extracting solution, mixing the obtained concentrated solution with the wild jujube concentrated juice, and sequentially standing and separating to obtain a supernatant;
and mixing the supernatant, casein hydrolysate peptide, water, gamma-aminobutyric acid, tea theanine and a sweetening agent, and sterilizing to obtain the plant beverage.
7. The preparation method of claim 6, wherein the number of times of decoction and extraction is 2, the ratio of the mass of water used for each decoction and extraction to the total mass of the blighted wheat, the spina date seed, the Chinese date, the poria cocos and the liquorice is (8-10): 1, and the time for each decoction and extraction is independently 1-2 h.
8. The method of claim 6, wherein the concentrate has a relative density of 1.02 ± 0.0025 g/mL; the standing temperature is 0-4 ℃, and the standing time is 6-12 hours.
9. The preparation method according to claim 6, wherein the separation mode is centrifugal separation, the rotation speed of the centrifugal separation is 10000-17000 r/min, and the liquid inlet speed is 150-200L/h.
10. The method according to claim 6, wherein the temperature of the sterilization is 105 to 120 ℃ and the time is 10 to 30 min.
